Identification of molecular signatures involved in radiation-induced lung fibrosis

<P><B>Abstract</B></P><P>In radiotherapy, radiation (IR)-induced lung fibrosis has severe and dose-limiting side effects. To elucidate the molecular effects of IR fibrosis, we examined the fibrosis process in irradiated mouse lung tissues. High focal IR (90 Gy) was exposed to a 3-mm volume of the left lung in C57BL6 mice. In the diffused irradiation, 20 Gy dose delivered with a 7-mm collimator almost covered the entire left lung. Histological examination for lung tissues of both irradiated and neighboring regions was done for 4 weeks after irradiation. Long-term effects (12 months) of 20Gy IR were compared on a diffuse region of the left lung and non-irradiated right lung. Fibrosis was initiated as early as 2 weeks after IR in the irradiated lung region and neighboring region. Upregulation of <I>gtse1</I> in both 90Gy-irradiated and neighboring regions was observed. Upregulation of <I>fgl1</I> in both 20Gy diffused irradiated and non-irradiated lungs was identified. When <I>gtse1</I> or <I>flg1</I> was knock-downed, TGFβ or IR-induced epithelial-mesenchymal transition was inhibited, accompanied with the inhibition of cellular migration, suggesting fibrosis responsible genes. Immunofluorescence analysis using mouse fibrotic lung tissues suggested that fibrotic regions showed increased expressions of Gtse1 and Fgl1, indicating novel molecular signatures of <I>gtse1</I>and <I>fgl1</I> for IR-induced lung fibrosis. Even though their molecular mechanisms and IR doses or irradiated volumes for lung fibrosis may be different, these genes may be novel targets for understanding IR-induced lung fibrosis and in treatment strategies.</P><P><B>Key messages</B></P><P><P>Upregulation of <I>gtse1</I> by 90Gy focal irradiation and upregulation of <I>fgl1</I> by 20Gy diffused irradiation are identified in mouse lung fibrosis model.</P><P>Gtse1 and Fgl1 are involved in radiation or TGFβ-induced epithelial-mesenchymal transition.</P><P>Radiation-induced fibrotic regions of mouse lungs showed increased expressions of Gtse1 and Fgl1.</P><P>Gtse1 and Fgl1 are suggested to be novel targets for radiation-induced lung fibrosis.</P></P><P><B>Electronic supplementary material</B></P><P>The online version of this article (10.1007/s00109-018-1715-9) contains supplementary material, which is available to authorized users.</P>

Our understanding of the differential effects between specific omega-3 fatty acids is incomplete. Here, we aimed to evaluate the effects of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) on T-helper type 1 (Th1) cell responses and identify the pathways associated with these responses. Naïve CD4<sup>+</sup> T cells were co-cultured with bone marrow-derived dendritic cells (DCs) in the presence or absence of palmitate (PA), DHA, or EPA. DHA or EPA treatment lowered the number of differentiated IFN-γ-positive cells and inhibited the secretion of IFN-γ, whereas only DHA increased IL-2 and reduced TNF-α secretion. There was reduced expression of MHC II on DCs after DHA or EPA treatment. In the DC-independent model, DHA and EPA reduced Th1 cell differentiation and lowered the cell number. DHA and EPA markedly inhibited IFN-γ secretion, while only EPA reduced TNF-α secretion. Microarray analysis identified pathways involved in inflammation, immunity, metabolism, and cell proliferation. Moreover, DHA and EPA inhibited Th1 cells through the regulation of diverse pathways and genes, including Igf1 and Cpt1a. Our results showed that DHA and EPA had largely comparable inhibitory effects on Th1 cell differentiation. However, each of the fatty acids also had distinct effects on specific cytokine secretion, particularly according to the presence of DCs. [BMB Reports 2021; 54(5): 278-283]

Proprotein Convertase Subtilisin/Kexin type 9 (PCSK9) is a pivotal regulator of low lipoprotein-cholesterol (LDL-C) metabolism, and the interest in PCSK9 has increased in cardiovascular diseases. PURPOSE: Exercise reduces blood LDL-C via PCSK9-LDLR pathway in liver. However, the mechanism of exercise-induced inhibition of PCSK is not unclear. The aim of this review is to describe the role of exercise on PCSK9-LDLR axis in cardiovascular diseases. METHODS: This study reviews 34 previous studies focusing on the effect on exercise on PCSK9 in the human and animal. RESULTS: The effects of exercise and lifestyle intervention on hepatic and circulating PCSK9 are controversial. However, exercise consistently increases hepatic LDLR, and inhibits atherosclerosis via suppression of PCSK9 and LOX-1 in atherosclerotic region. CONCLUSIONS: Even though experimental data are still very limited, exercise training can improves blood LDL-C via inhibition of PCSK9 and enhancement of LDLR in liver and vasculature. The study of exercise on PCSK9 are urgently needed.

정보통신기술의 급속한 발달과 더불어 인터넷이 널리 사용되게 되고, 인터넷을 통한 음성과 데이터를 통합한 서비스 사업이 크게 확산되어 가고 있다. 이에 따라 IP망에서 전송되는 음성의 품질은 현재 상용화되고 있는 VoIP의 대중화를 위해서 가장 시급히 해결되어야 할 문제이다. 본 연구에서는 IP망에서 전송되는 음성의 품질에 영향을 미치는 요소들과 음성품질 측정 방법을 분석하고, 차세대 네트워크에서 사용될 VoIP의 음성품질을 측정할 수 있는 측정방식을 제안 및 구현하였다. 제안된 방식을 이용하여 VoIP에서 음성품질을 측정한 결과, 음성품질은 코덱과 인터넷서 버스방식에 따라 달라지는 것으로 분석된다. Due to the rapid growth of the Internet, voice and data integrated service over Internet Protocol(IP) network is growing very fast. In order to popularize the commercialized IP telephony service, voice transmission quality through IP network must be enhanced. In this paper, we analyze various elements that interfere the voice transmission quality over the IP network and propose the measurement scheme that can measure voice quality over the IP network effectively. Using proposed scheme, we implement the scheme and measure the voice quality of VoIP. The result show voice quality is influenced by codec and internet service method.

Integratable Micro-Doherty Transmitter

We propose Doherty power amplifier structure which can be integrated in Silicon RF ICs. Doherty power amplifiers are widely used in RF transmitters, because of their high Power Added Efficiency (PAE) and good linearity. In this paper, it is proposed that a method to replace the quarter wavelength coupler with IQ up-conversion mixers to achieve 90 degree phase shift, which allows on-chip Doherty amplifier. This idea is implemented and manufactured in CMOS 5 ㎓ band directconversion RF transmitter. We measured a 3㏈ improvement output RF power and linearity.
