Development of a novel chromosome dissection chip for chromosomal analysis with nanometer size

The dissection of chromosomes followed by extraction of specic DNA fragments constitutes a topic of high interest, especiallyfor cytogenetic studies in the post genome era. The dissected chromosomes can be used in various conventional applications, includ-ing establishing probes for uorescence in situ hybridization (FISH), the generation of chromosome band-specic libraries andphysical mapping for cytogenetic analysis. For this purpose, several reports using micro- and nano-dissection methods by micro-manipulators have been published.In the present study, we developed a functional chromosome dissection chip that can dissect various chromosomes in one-chip byusing the pattern chip with nanometer-scale porous anodic alumina. The porous anodic alumina that is to make use of self-orga-nized nano-array materials as a template for the pattern chip has a highly oriented porous structure with very uniform and nearlyparallel pores that can be organized in an almost precise close-packed hexagonal structure. The most characteristic feature of thismaterial has been emphasized with the extremely high aspect ratio of their channels that is dicult to achieve with conventionallithographic techniques. We have proceeded as follows for this study; the pattern chip with nanometer-scale porous anodic aluminawas fabricated. Then, metaphase chromosomes were prepared by dropping the cell suspension onto pattern chip and air-drying untilwere dissected with nanometer size. This technique will form the basis for the development of new system such as ‘‘Nano-scale chro-mosome dissection chip’’ that arrays chromosomal fragments with nanometer scale on a chip for DNA and protein nano-analysis.
백혈병 세포에서 Multidrug Resistance Gene-1 (mdr1)의 과발현이 ^99m Tc-sestaMIBI 섭취에 미치는 영향

Purpose: To determine whether 99mTc-MIBI is recognized by the multidrug resistant P-glycoprotein (Pgp), we have measured quantitatively 99mTc-MIBI uptake in cancer cells. The effects of various Pgp reversing agents on cellular 99mTc-MIBI uptake were also investigated in the presence of multidrug resistance gene-1(mdr1 gene) overexpression. Materisls and Methods: We measured percentage uptake of 99mTc-MIBI at different incubation temperatures both in mdr1 positive and negative cells. The effects of verapamil, cyclosporin, and dipyridamole on cellular uptake of 99mTc-MIBI were also evaluated with or withouts overexpression of mdr1 gene in Cultured murine leukemia L1210 cells. Results: The mdr1 gene expressing cell lines were effectively induced in in vitro with continuous application of low-dose adriamycin or vincristine. Cellular uptake of 99mTc-MIBI was higher in mdr1 negative L 1210 cells than those of mdr1 positive cells, and higher when incubated in 37℃ than 4℃. In the presence of verapamil, cyclosporin or dipyridamole, 99mTc-MIBI uptake was increased upto 604% in mdr1 positive cells. Conclusion: Cellular uptake of 99mTc-MIBI is lower in leukemia cells over-expressing mdr1 gene, and MDR-reversing agents increase cellular uptake. These results suggest that 99mTc-MIBI can be used for characterizing Pgp expression and developing MDR-reversing agents in vitro. (Korean J Nucl Med 1999;33: 152-62)

집단미술치료가 농촌지역 아동들의 자아존중감향상에 미치는 효과

본 연구는 농촌지역 아동에게 집단미술치료를 실시하여 자아존중감 향상에 긍정적인 변화가 있는지를 알아보기 위해 실시되었다. 본 연구의 대상자는 충청남도 면단위에 소재한 초등학교 1학년 학생 7명으로 학교의 지원과 부모의 허락 하에 집단미술치료를 실시하였다. 대상자는 특정종교 다문화가정 자녀 3명, 조손 가정 2명, 신체적으로 정상발달에 지장을 받고 있는 학생 2명이었다. 집단미술치료의 결과는 다음과 같다. 집단미술치료가 농촌지역 저학년의 자아존중감 중 가정적 자아존중감에서 통계적으로 유의미한 향상이 나타났다. 또한 K-HTP의 내용과 형식적분석에서 부정적인 표현이 줄어들고 긍정적 표현이 나타났으며 행동관찰기록에서 회기가 진행될수록 긍정적인 변화가 나타났다. 이상과 같은 결과로 부터 본 연구의 집단미술치료프로그램이 농촌지역 아동의 자아존중감 향상에 효과가 있었음을 알수 있다. The piece examines whether a group art therapy program brings some positive effects on elementary school children in rural area or not. Such elementary school children for the study include three from multicultural families, two from families under the protection of their grandmother and grandfather without their parents, and two with physical developmental disability in rural area. The author concludes as follows: (a) The group art therapy program has a good influence on such study participants in the family self-esteem, not the social and school self-esteem. (b) The study participants` negative expressions are being transformed into some positive expressions in the K-HTP. (c) The study participants` behavior comes to be positive little by little in the process of the study. Therefore, the group art therapy program for the article has some positive effects on elementary school children in order to improve their self-esteem in rural area away from downtown of Chungnam Province.
HBV : O-037 ; A nationwide seroprevalence of hepatitis B virus in 2009, Korea

Background/Aims: Hepatitis B virus (HBV) infection is the major cause of chronic liver disease and hepatocellular carcinoma in Korea. After hepatitis B vaccine era, the seroprevalence of hepatitis B surface antigen (HBsAg) is decreasing, especially in children and adolescents. In this study we estimated the nationwide, age-, sex- and area- adjusted seroprevalence of HBV infection in 2009. Methods: We reviewed data of a total of 290,212 subjects who had received health check-up examination in 27 institutions from January to December 2009. The estimated seroprevalence was adjusted by age, sex and area of the standard population based on the 2009 Census of Korea. Results: The overall seroprevalences of HBsAg and anti-HBs adjusted by age, sex and area were 4.0% and 73.5%, respectively. The area- and age- adjusted HBsAg positivity was higher in men (4.5%) than in women (3.4%) (p<0.001). The sex- and area- adjusted HBsAg positivity was the most common in forties and showed decreasing trend in age. The HBsAg positivity in Southern province including Jeju was significantly higher than that in central province(p<0.001). The area- and age- adjusted anti-HBs positivity was lower in men (71.6%) than in women (75.4%) (p<0.001). The seropositivity of anti-HBs was the lowest in Jeonnam. The concurrent positivity of HBsAg and anti-HBs was found in 0.1% of the subjects. The comparison between patients showing sole positive for HBsAg and patients showing concurrent positive for HBsAg and anti-HBs will be presented. Conclusions: The seroprevalence of HBV infection showed differences according to age, sex and areas in Korea. As the seroepidemiology is dynamically changing, and the overall seropositivity of HBsAg has shown continuously decreasing trend.
