        제대 및 신생아 혈중 cytokine(IL₁β, IL6, TNF) 수치의 변화

        분만방법에 따른 제대혈과 양수 그리고 감염 및 stress가 있는 신생아혈중과 세포성분을 배양한 상청액의 cytokine치를 ELISA법으로 측정하여 다음과 같은 성적을 얻었다. 제대혈중 IL₁β치는 질식 유도분만때가 질식 자연 분만, 응급 제왕절개 및 계획 제왕절개때보다 유의하게 높았다(p<0.001). 신생아 혈중 IL₁β치는 감염이 동반된 경우와 stress가 동반된 경우에 높았으며, 감염이 동반되었을 때가 stress보다 더 높게 나타났고, 저체중 출생아에서 높았다. 계획 제왕절개시의 제대혈 IL₁β치는 양수보다 낮았다. 질식자연 분만시 제대혈 세포상청액의 IL₁β 혈장치보다 높았고, 양자간에 유의한 상관관계를 보였다(r=0.92, p<0.05). 감염이 동반된 신생아에서 IL₁β치는 혈장치가 높은 경우 세포 상청액치는 낮고, 혈장치가 낮은 경우 세포 상청액치는 높았다. IL6는 감염이 있는 신생아 혈장과 세포 상청액, 양수 등에서 검출되었다. TNF는 감염이 있는 신생아 세포 상청액 8례중 3례에서 측정되었다 The cytokines, interleukin-1β(IL₁β), interleukin-6(IL6) and tumor necrosis factor(TNF) are important mediators of host response to stress and infection. Impaired immune reaction and febrile responses to the infection in newborn period may result from abnormal cytokine regulation and there were difference in the responses between newborn and adult due to the different cytokine regulation system. To investigate the cytokine regulation in different circumstances of the delivery and newborn conditions, the measurement of the levels of IL₁β,IL6 and TNF in cord blood, amniotic fluid and the blood of the newborn were performed, using enzyme linked immunosorbent assay(ELISA,R&D Co.). Cytokines of the supernatant of incubated cellular pellet were also measured. Cord blood IL₁β level was significantly higher in induced vaginal delivery than that of the normal vaginal, emergency, and elective cesarian section delivery(p<0.001). Plasma IL₁β level of the newborn with infection or stress was elevated and the level was higher in infection than that of the newborn with stress during delivery. Premature baby had higher level than normal newborn. Cord blood IL₁βlevel was lower than that of amniotic fluid in elective cesarian section delivery. In normal vaginal delivery, IL₁βlevel of the cellular supernatant was higher than that of the plasma and showed good correlation(r=0.92, p<0.05). In newborn with infection, plasma IL₁βlevel was high in the cases with low supernatant level and the plasma level was low in the cases with high supernatant IL₁β. IL6 was detcted from the plasma and cellular supernatant of the newborn with infection and the amniotic fluid. TNF was detected in cellular supernatant of 3 among 8 newborn with infection.

        만성두통 환자에서 한방 치료 후 인영혈 부위의 뇌혈류에 대한 연구

        Objectives: This study was performed to change of blood flow in In-Young(ST9) after treatment on chronic daily headache patients. Methods: Monitoring of TCD was examined in 10`s chronic daily headache patients before and after treatment. Mean velocity flow, systolic velocity and pulsatility index were analyzed from TCD at In-Young(ST9). The patients was laid for 20 minutes before treatment and also for treatment. Results: The results showed a significant(p<0.05: Paired T-test) decrease in mean velocity flow and systolic velocity. but there were no significant differences in pulsatility index. Conclusions: These results suggest that there were significant change on blood flow in In-young of chronic daily headache patients.

      • 모델동물의 병태.생리 유발 유전자 기능 연구 : 당뇨모델동물의 췌장 β-세포를 이용한 비만발생에 관한 연구 Effect of development of diabetes by fatty acid in animal model

        체장 β세포에 지방이 축적되는 lopotoricity 현상은 인슬린 비의존성 당뇨 (Non InsuBin Dependent Diabetes Mellitus :NIDDM)의 발생과 상당히 밀접한 관계가 있으며, 췌장 β세포의 lipoapoptosis는 세포막형성에 관여하는 sphingomyelin의 전군체안 ceramide의 과생산에 의해 유도된다는 보고가 있다. 그러나, 지방산 종류에 따른 췌장 β세포에서의 세포사멸 (apoptosis)유발 기작에 대해서는 야직까지 보고된 바가 없파. 또한, 지방세포에서 생성되며, 지질대사, 음식섭취 및 체중조절에 관여하는 렙틴 (leptin)과 렙틴 수용체와의 관계 역시 체장 세포에서 명착하게 밝혀져 있지 않다. 따라서 본 연구에서는 6주령 53와 Zucker rat의 췌장으로부터 췌도를 분리하는 기술을 확립하고, 배양된 췌장 췌도 세포와 MS-1세포주에 불포차즌방산 (n-3 linolenic acid; n-6 :linoleic acid), 포화지방산 (stearic acid), conjugated linoleic acid (CLA)를 포함하는 4 가지의 지방산을 처리한 후 이들이 췌장 』세포의 사멸에 미치는 영향을 다양한 apoptosis 관련 유전자의 발현을 분석함으로써 확인하고자 하였다. 그 결과 SD rat으로부터 마리 당 평균 약 500개의 췌장 췌도를 분리하는데 성공하였으며, 4 종류의 지방산 중 포화지방산인 stearic acid를 처리하여 ONA fragmentation 증가를 관찰하였다. 또한 RT-PCR 방법을 이용하여 분리된 췌도 세포에사의 Bcl-2, Bax, iNOS가 지방산의 종류에 따라 서료 다른 발현양상을 나타낸다는 」라실을 관귿하였다. 이러한 떤구 결과는 지방산의 종류에 따라 췌장 췌도 세포에서의 세포사멸 유발 효과를 겋증하였으며, 이러한 연구결과는 복잡한 당뇨발병 원인 규명에 중요한 정보를 제공할 것으로 사료된다. Fat contents within the pancreatic islets are closely related to lipotoxicity which induces apoptosis, resulting in non insulin dependent mellituse(NIDDM). High level of fat may contribute to both insulin resistance and obesity in rodent and humans but the evidence is limited. To determine whether dietary fatty acids affect pancreatic cell function, the INS-1 cells and the pancreatic islets isolated from SD and Zucker obese(fa/fa) rats(6 wk, prediabetic stage) were cultured with stearic acid and CLA. As a result, DNA fragmentation laddering was substantially decreased in the INS-1 cells and the isolated pancreatic islets cultured with 2mM CLA compared with those with stearic aced. To investigate the mechanism by which CLA alleviates cell apoptosis as indicated by DNA fragmentation assay, we examined mRNA expressions of apoptosis-related proteins including Bax and Bcl-2 associated with cell death agonist and antagonist, respectively in the both INS-1 cells and islets cultured with 2 mM fatty acids. Bax mRNA expression was not markedly altered by either stearic acid or CLA, whereas Bcl-2 mRNA expression was noticeably enhanced by CLA compared with stearic acid. However, there were no changes in cell apoptosis and cell apoptotic-regulating gene products in the both INS-1 cells and isolated islets treated without or with 2 mM CLA. It is concluded that CLA may have a beneficial effect for maintenance of cell viability via increased Bcl-2 protein expression compared with steraic aced, which lead to delay or prevent, at least somewhat, the onset of NIDDM in the physiological status.

      • 외래도입 병솔꽃나무(Callistemon citrinus)의 결과와 개화습성에 관한 연구

        This study was performed to obtain basic information on fruit setting, propagation and flowering habitats of exotic ornamental tree, bottle brush tree (Callistemon citrinus). The results are as follows; It has been prolonged four years since the fruit had set. Fruit capsules have enlarged for three years but there was no difference in seed number in the fruit capsule between 2-year-old seeds and 3-year-old ones. To separate the seeds from capsules, capsules were dried on three different temperature 30, 50 and 70℃. Seeds, which dried in 50 and 70℃, lost 25% of water in seeds within 24 hours. Two-year-old seeds dried on 30℃ germinated well than others. Removing the seed capsule from the branch, 14 flowers per tree were induced in average. Whereas a flower was bloom when the branch had seed capsule on it. In cutting for propagation, semi-hard wood branches dipped into 100 mg/ℓ IBA for 2 hours rooted 86.4%, and had 12.6 roots and long roots.

        We are currently in the actual circumstances to be lost as 30 percents our country leakage. A surrounding environment of the water pipe was infected, and increased quality of water pollution phenomenen in conduit pipe by water leakage. And, by water leakage, caused huge economical damage. Is ceilinged in leakage of water detection to using leakage of water detection method in our country now. As well as can not search correct position even if water leakage happens, because leakage of water detection is impossible by real time. In this paper we solved difficult of existent product and difficulty of existent sensing method and maintenance through research of leakage sensing system DB and S/W design in leakage sensign system. And, at the same time, we simulated leakage sensign system and implemented of DB and S/W design.

      • A Novel Analysis of <tex> ${L}_{\text{gd}}$</tex> Dependent-1/ <tex> ${f}$</tex> Noise in In<sub>0.08</sub>Al<sub>0.92</sub>N/GaN

        <P>In<SUB>0.08</SUB>Al<SUB>0.92</SUB>N/GaN fin-type high-electron mobility transistors (fin-HEMTs) with different gate-to-drain lengths ( <TEX>${L} _{\text {gd}}$</TEX>) are fabricated and characterized by dc and low-frequency noise (LFN) measurements. The fabricated device with the largest <TEX>${L} _{\text {gd}}$</TEX> exhibits the degradation of the maximum drain current and transconductance with a positive shift of the threshold voltage. LFN measurements of the In<SUB>0.08</SUB>Al<SUB>0.92</SUB>N/GaN fin-HEMTs reveal clear 1/ <TEX>${f}$</TEX> behavior of the noise spectra, and the minimum value is observed in the device at <TEX>${L} _{\text {gd}} = {20}\,\,\mu \text{m}$</TEX>. The devices with smaller <TEX>${L} _{\text {gd}}$</TEX> follow a carrier number fluctuation noise model owing to electron trapping/detrapping into the In<SUB>0.08</SUB>Al<SUB>0.92</SUB>N barrier layer from the 2-D electron gas (2DEG) channel. In contrast, the device with the largest <TEX>${L} _{\text {gd}}$</TEX> shows correlated mobility fluctuations due to the large 2DEG mobility fluctuations in the large access area.</P>

      • 당뇨병 백서의 간세포에서 Glucokinase 활성도 및 유전자 발현에 대한 인슐린의 영향

        목적 당대사의 조절 상태에 따른 생체 변화를 분자 수준에서 이해하고자 식이 조건을 달리한 정상 백서와 화학적으로 유도된 당뇨병 백서의 간조직에서 혈당수준과 인슐린치료 정도에 따라 나타나는 글루코키나제 활성도 및 유전자 발현을 분석하였다. 방법 스트렙토조토신 정맥투여 후 당뇨병의 유발을 확인하고, 인슐린을 1일 3회 3일간 복강내로 투여하여 상태를 안정시킨 후, 인슐린 투여군은 인슐린 투여 6시간이내에 그리고 인슐린 비투여군은 24시간 후 단두하여 채혈하고 복강을 열어 간조직을 채취하였다. 채취한 간조직에서 글루코키나제 활성도는 인산화된 포도당에서 NADH의 형성을 형광분광계로 측정하였으며, 글루코키나제 유전자 mRNA발현은 Northern 분석법을 이용하였다. 성적 정상 백서에서 공복상태와 식이를 섭취한 경우에 간조직의 글루코키나제 효소의 활성은 차이가 없었으나, 글루코키나제 유전자 mRNA 발현은 증가되었다. 당뇨병이 유발된 백서의 간조직에서 글루코키나제 효소의 활성 및 글루코키나제 유전자의 mRNA 발현은 정상 백서에 비하여 낮았다. 인슐린 투쳐 후 글루코키나제 효소의 활성 및 글루코키나제 유전자의 mRNA 발현이 증가되었고, 특히 혈당이 정상화된 경우에서 글루코키나제 유전자의 mRNA 발현이 증가도었다. 결론 인슐린에 의한 간조직에서 글루코키나제 효소의 활성 및 글루코키나제 유전자의 mRNA을 증가를 볼수 있었다. 당뇨병 백서에서 인슐린 투여 후에 혈당조절이 안된 경우 간조직의 글루코키나제 유전자의 mRNA 발현이 증가가 없는 것으로 보아 글루코키나제 mRNA의 발현에는 인슐린 이외의 다른 요소가 관여할 것으로 생각된다. The liver-specific hexokinase isoenzyme, referred to as glucokinase, is thought to play a key reglulatory role in hepatic glucose metabolism. The glucokinase gene is, therefore, of interest both because of its tissue-specific expression and because of the several regulatory processes that can be analyzed. The level of hepatic glucokinase activity appears to be determined essentially by regulation of the rate of enzyme synthesis, with insulin playing a leading role as an inducer. We investigated the role of insulin for the induction of glucokinase in the liver of diabetic rats. Experimental diabetes was induced by injection of streptozotocin 7 days before the experiment. Regular insulin was given by three days intraperitoneal injection at 8-h interval. The glucokinase mRNA in the liver was estimated by Nothern blot assay, as well as by fluorometric enzyme activity assay. Glucokinase activity was not reduced in the liver of normal fasting rats as compared to normal fed rats. And glucokinase activity was reduced in the liver of diabetic rats as compared to normal rats. In diabetic rats treated with insulin, glucokinase enzyme activity were increased. But glucokinase mRNA expression was only increased in normoglycemic diabetic rat with treated with insulin as compared to hyperglycemic rat. These data indicate that insulin stimulates hepatic glucokinase enzyme activity and mRNA expression. But other hormonal or metabolic factors may be contribute to regulation of glucokinase mRNA expression.

      • 누수감지 시스템의 시뮬레이션

        Population's city centralism phenomenon happened by continuous increase in population and ancient city industry socialization development. According to this, increased demand of capital is state been accelerating radually. Specially, source of a stream security by water lack phenomenon and quality of water change for the worse is difficult. There is the limit in a leakage of water detection method which is using in my country. Therefore, water leakage detection monitoring system that use TDR can search for water leakage point of small quantity as well as correct water leakage point and enforce repair. In this paper we solved difficult of existent product and difficulty of existent sensing method and maintenance through research of special leakage sensing pipe and leakage sensing system. And, we did system of correct most suitable simulation in our country real condition using TDR.

        We have developed PCB pattern inspection system using CCD camera for detecting PCB pattern defects. There are two main techniques in detecting PCB defects: image differencing and design-rule verification. In this paper, we use image differencing method. We separate object and background using color segmentation method. We use sobel operator to find edges, and using the edges register reference image and test image. The proposed method provides reduced cost, fast inspection speed, efficient performance, flexibities.

      • 박하세포의 현탁배양에 대한 Fungal Elicitor의 영향

        The effect of fungal effector on suspension culture of M. piperita cells was investigated using shake flasks. About two fold increase in oil production was observed in response to the treatment of fungal elicitor prepared from Rhodotorura rubra. Also, maximum accumulation of oil in M. piperita cells occurred at 48 hrs of incubation and 14 day-old peppermint cells were best in peppermint oil production.
