Nicotiana tabaccum 'Xanti', Petunia hybrida 'Blue Star' 그리고 Chrysanthemum morifolium 'Baeckwang'을 공시하여 엽조직 유래의 mesophyII protoplast(MP)와 paraveinal mesophyII protoplast (PVMP) 간의 세포 활성을 조사하기 위하여 urea 투과성을 측정하였다. 아울러 엽육 조직에서 원형질체 나출을 위한 각종 효소제를 처리하고 경과 시간별로 관찰했으며, 나출 원형질체로부터 식물체 재생을 위한 NAA와 thidiazuron의 효과를 조사하였다. Vibratome을 이용한 엽육조직 절단방법은 엽조직의 상해를 최소화하고 조직절편도 최소 50 $\mu\textrm{m}$까지 절단할 수 있기 때문에 urea 투과성 검정과 원형질체 나출을 위한 필수적인 기술이다. 세포활성에 대한 urea투과성 측정은 공시한 3종 식물의 PVMP에서 모두 Ks=2.Ox$10^{-5}$cm/sec 정도가 MP보다 높았다. 효소혼합 용액(1.5% Cellulase R-10, l% Driselase, 0.5% Macerozyme R-10, 0.05% Pectolyase)을 4-8 시간 처리하는 것이 PVMP 나출에 유효하였다. 나출 원형체로부터의 캘러스 유기와 식물체 재생에는 2 mg/L NAA + 0.01 mg/L thidiazuron 처리구에서 가장 양호하였으며 조직별로는 PVMP에서 월등한 결과를 나타내었다. 따라서 엽조직을 대상으로한 원형질체 배양에 있어서 엽록소의 함량이 많은 MP보다는 재생력이나 세포 활성이 강한 paraveinal 엽육조직 유래의 세포를 집약적으로 나출시켜 실험재료로 이용한다면 더 높은 식물체 재생률을 얻을 수 있을 것으로 사료된다. This study was rimed out to examine the difference in the cell vitality between mesophyII protoplast (MP) and paraveinal mesophyII protoplast (PVMP) of Nicotiana tabaccum 'Xanti', Petunia hybrida 'Blue Star' and Chrysanthemum morifolium 'Baeckwang' by using urea permeability technique. The effects of various enzyme solutions and incubation time, NAA and thidiazron on plant regeneration from isolated protoplasts were also investigated. The vibratome technique was used for protoplast isolation and urea permeability test because the fresh living, thin tissue stripes (50 ${\mu}{\textrm}{m}$ of thickness) could be obtained with minimal damage with the vibratome. For the three plants examined, the urea permeability on the tested tissue stripes was relatively higher in PVMP than in MP by about Ks = 2.0 $\times$ 10$^{-5}$ cm/sec. The treatment of an enzyme mixture of 1.5% cellulase R-10, 1% Driselase, 0.5% Macerozyme R-10, and 0.5% Pectinase for 4 to 8 h was effective on the isolation of PVMP. The highest frequency of callus formation and plant regeneration from the isolated protoplasts was obtained with NAA 2 mg/L and thidiazuron 0.01 mg/L. Furthermore, the results demonstrated that cell devision and plantlet regeneration was more frequent in the PVMP than in the MP of the same leaf or plant We, therefore, conclude that UM is an excellent experimental material for the callus formation and regeneration from isolated protoplasts.
Study of Active Water Absorption of Polystyrene-Based Ionomers

The active water absorption behavior of poly(styrene-co-methacrylate) PSMANa and sulfonated polystyrene PSSNa ionomers was studied. On one hand, the acidic copolymers did not absorb water noticeably. On the other hand, the amount of water absorbed by the ionomer increased with increasing ion content. Especially when the ion contents of the PSMANa and PSSNa ionomers exceeded 6 and 10 mol%, respectively, the maximum amount of the water absorbed by the ionomers increased rapidly as the ion content increased. This indicated that the cluster-dominant ionomers exhibited stronger water absorption behavior, compared to the matrix-dominant ionomers. In addition, when the ion contents of the PSMANa and PSSNa ionomers were less than 6 and 10 mol%, respectively, the volume and shape of the bulk ionomers did not change significantly by the water absorption. Morphological studies on the soaked ionomers showed that the SAXS peak shifted to lower angles as the water uptake increased with increasing ion content of the ionomers, which was consistent with the results obtained by the swelling method.
Saintpaulia 조직배양 중 광도와 배지의 양에 따른 생육반응

The growth responses and morphogenic function on the in vitro culture of Saintpaulia were investigated in various medium volumes and under the controlled light intensities. The experimental results obtained are shown as follows: 1. The smaller the medium volumes in the cultural vessels, the faster the growth responses and deteriolation of nutrients 2. In general. the optimum volumes for mass propagation was truned out to be 20% of the cultural vessels. 3. Many of the adventitious buds were developed under the 15 Klux light intensity, whereas no bud but callus growth and adventitious roots were developed under the 0.5 Klux. It was suggested that the mechanism of light effect on the adventitious bud formation be further expoured.
