        정신분열병과 Neurotensin 수용체 유전자 다형성의 연합 연구

        연구목적: Neurotensin(NT)은 NT수용체와 결합하여 그 효과를 나타내는 neuromodulator 혹은 neurotransmitter로서 대뇌에서 도파민의 분비를 조절하는데 중요한 역할을 한다. 근래의 연구에 의하면 NT와 그 수용체는 대뇌에서 항정신병 약물의 효과를 매개하는 것으로 생각되고 있으며 약물치료를 받지 않은 정신분열병 환자의 뇌척수액에서 NT의 양이 적으로 보고되고 있어 이들은 정신분열병과 깊은 관련을 가지고 있을 것으로 추정된다. 최근 NT수용체의 유전자의 3`인접영역에서 정보가치가 높은 4 염기반복 다형성이 발견되어 이를 유전 표지자로 하여 정신분열병과의 연합을 알아보았다. 방 법: 서로 혈연관계에 있지 않은 정신분열병 환자 120명(남자 91명, 여자 29명)과 정상 대조군 106명(남자 84명, 여자 22명)을 대상으로 하였다. PANSS를 사용하여 양성 및 음성을 알아보았다. 말초혈액에서 DNA를 분리한 후에 중합효소연쇄반응을 사용하여 3`인접영역에 있는 4 염기 반복 다형성을 증폭하였고 silver staining한 후에 유전자형을 관찰하였다. chi-square 검증과 Bonferroni`s correction을 사용하여 환자군과 정상 대조군간의 대립유전자 빈도의 차이를 알아보았다. 또한 양성 및 음성 환자군으로 나누어 차이를 알아보았다. 결 과: 총 23개의 대립유전자가 관찰되었으며, 399bp의 대립유전자(A10)의 빈도가 환자군보다 정상대조군에서 통계적으로 유의하게 높았다(χ²=16.49, df=1, p<0.001). 음성 정신분열병 환자군과 정상대조군 사이의 비교에서는 정상대조군의 A10의 빈도가 환자군보다 유의하게 높았다(χ²=21.33, df=1, p<0.001). 성별 비교에서 남자 정신분열병 환자군은 대조군에 비하여 A10의 분포가 유의하게 적었다. (χ²=13.71, df=1, p<0.001) 결 론: NT 수용체 유전자와 정신분열병사이에 음성연합이 관찰되었다. NT 수용체 유전자가 일부 정신분열병의 발병과정에서 확실하지는 않지만 어떤 종류의 보호기능을 할 수도 있다는 것을 암시한다. Objectives: Neurotensin(NT), of which functions are evoked by its interaction with neurotensin receptors(NTR), coexists with mesolimbic dopamine and regulates endogenous dopamine release. Recent studies have shown that NT with NTR exerts neuroleptic-like activity within the central nervous system and may play an important role in the pathogenesis and in the treatment of schizophrenia. We have examined the gentic association between schizophrenia and tetranucleotide repeat polymorphism in the 3-flanking region of the NTR gene to investigate the possible contribution of the NTR gene to the schizophrenia susceptibility. Methods: Among 23 alleles identified, the subjects were 120 patients(male 91, female 29)with schizophrenia and 106 normal healthy controls(male 84, female 22). They were unrelated native Korean. PANSS was used to determine positive or negative subgroup in the schizophrenic patients. Using polymerase chain reaction and polyacrylamide gel electrophoresis, tetranucleotide repeat polymorphism(CCTT and CTT) in the 3`-flanking region of NTR gene was observed. For a comparison of NTR gene`s allelic frequencies between patients with schizophrenia and normal healthy controls, chi-square test and Bonferroni`s correction was performed. Results: The frequency of A10 allele(base pair size=399)was significantly higher in normal healthy controls than schizophrenia(χ²=16.4902, df=1, p<.000). In the comparison between schizophrenic patients with negative symptoms and normal controls, the frequency of A10 allele was significantly higher in normal healthy control subjects than patients with schizophrenia(χ²=21.33, df=1, p<0.001). In the case of male, the frequency of A10 allele of schizophrenia was significantly higher than normal controls(χ²=13.71, df=1, p<0.001). Conclusions: NTR gene was negatively associated with schizophrenia. NTR gene`s tetranucleotide repeat polymorphism may provide some protective function against schizophrenia.

      • 고립성 폐결절의 조직학적 진단에 있어서 경기관지폐생검의 진단적 가치

        Transbronchial lung biopsy(TBLB) is a relatively simple and convenient diagnostic tool in solitary pulmonary nodule(SPN) on chest radiograph, which is cannot be diagnosed through routine tests. To investigate the diagnostic value and diagnostic rate of TBLB according to the variable factor, we analyzed 69 patients with radiologically SPN lesion on chest radiograph which was confirmed histologically. A histologic diagnosis of TBLB was made in 47 of 69 patients(68.1%), in which 26 of 37(70.3%) with malignancy and 21 of 32(65.6%) with benign lesion. There were no significant differences in diagnostic yield according to age, sex and location of the lesion. The diagnostic rate tended to increase with the more than 5 pieces to acquired by TBLB. When the size of SPN was less than 3 ㎝, it cannot be diagnosed. In conclusion, TBLB is a diagnostic procedure with good yield and safety, but percutaneous needle aspiration, transbronchial needle aspiration, and bronchial washing fluid cytology may be a complementary procedure to overcome the limitation of TBLB and get higher diagnostic yield for SPN.

        Creep-Fatigue Crack Growth Behavior of a Structure with Crack Like Defects at the Welds

        A study on a creep-fatigue crack growth behavior has been carried out for a cylindrical structure with weldments by using a structural test and an evaluation according to the assessment procedures. The creep-fatigue crack growth behavior following the creep-fatigue crack initiation has been assessed by using the French A16 procedure and the conservatism for the present structural test has been examined. The structural specimen is a welded cylindrical shell made of 316 L stainless steel (SS) for one half of the cylinder and 304 SS for the other half. In the creep-fatigue test, the hold time under a tensile load which produces the primary nominal stress of 45 MPa was one hour at $600^{\circ}C$ and creep-fatigue loads of 600 cycles were applied. The evaluation results for the creep-fatigue crack propagation were compared with those of the observed images from the structural test. The assessment results for the creep-fatigue crack behavior according to the French Al6 procedure showed that the Al6 is overly conservative for the creep-fatigue crack propagation in the present case with a short hold time of one hour.

        Comprehensive Residual Stress Distributions in a Range of Plate and Pipe Components

        A comprehensive review of through thickness transverse residual stress distributions in a range of as-welded and mechanically bent components made up of a range of steels has been carried out, and simplified generic transverse residual stress profiles for a plate and pipe components have been proposed. The geometries consisted of welded pipe butt joints, T-plate joints, tubular T-joints, tubular Y-joints and a pipe on plate joints as well as cold bent tubes and pipes. The collected data covered a range of engineering steels including ferritic, austenitic, C-Mn and Cr-Mo steels. Measured residual stress data, normalised with respect to the parent material yield stress, has shown a good linear correlation versus the normalised depth of the region containing the residual stress resulting from the welding or cold-bending process. The proposed simplified generic residual stress profiles based on the mean statistical linear fit of all the data provides a reasonably conservative prediction of the stress intensity factors. Whereas the profiles for the assessment procedures are fixed and case specific, the simple bilinear profiles for the residual stresses obtained by shifting the mean and bending stress from the mean regression line have been proposed and validated.

        Progressive Inelastic Deformation Characteristics of Cylindrical Structure with Plate-to-Shell Junction Under Moving Temperature Front

        A study on the progressive inelastic deformation behavior of the 316 L stainless steel cylindrical structure with plate-to-shell junction under moving temperature front was carried out by structural test and analysis. The structural test intends to simulate the thermal ratcheting behavior occurring at the reactor baffle of the liquid metal reactor as free surface of hot sodium pool moves up and down under plant transients. The thermal ratchet load that heats the specimen up to 550$^{\circ}C$ was applied repeatedly and residual deformation was measured. The thermal ratcheting test was carried out with two types of cylindrical structures, one with plate to-shell junction and the other without the junction to investigate the effects of the geometric discontinuities on the global ratcheting deformation. The temperature distributions of the test specimens were measured and were used for the ratcheting analysis. The ratchet deformations were analyzed with the constitutive equation of the non-linear combined hardening model. The analysis results were in good agreement with those of the structural tests.

      • Design and analysis of high-temperature piping system in the STELLA-2 sodium test facility

        Elevated temperature design and integrity evaluation have been conducted for the high-temperature piping systems in the STELLA(Sodium integral effect test loop for safety simulation and assessment)-2 sodium test facility which is under development at KAERI(Korea Atomic Energy Research Institute). There are several piping systems in the STELLA-2 sodium test loop. Piping systems were designed and design evaluation were conducted for the piping systems in IHTS(Intermediate Heat Transport System) and PSLS(Pump Simulation Loop System). Piping analyses for these sodium test facility have been performed according to the piping design codes of ASME B31.1 and integrity of the piping systems were confirmed.

        Treatment of residual stress in failure assessment procedure

        <P><B>Abstract</B></P><P>The effect of residual stress on component failure has been investigated using the distributions from current failure assessment procedures, and a residual stress profile simple to apply with less conservatism has been proposed for the weld geometries of T-plate and tubular T-joint. The stress intensity factors (SIFs) in the two weld geometries under various types of loads have been calculated using the Green’s function method. The Green’s functions were determined not only for the T-plate but also for the tubular T-joint with the built-in ends. The use of a linear (bending) stress profile, derived from an analysis of measured residual stress distributions in T-plate and tubular T-joints, has been examined. The profile was validated with experimentally measured residual stress distributions in two materials, a high strength and medium strength ferritic steel and two geometries, a T-plate joint and a tubular T-joint for crack lengths up to half the plate or pipe thickness. Whereas the recommended residual stress distributions are geometry and material specific, it is shown that a simplified linear bending profile provides a possible guideline, applicable to a range of materials and geometries, where detailed information on weld procedures or residual stress profiles are unavailable.</P>

      • Effect of Thermal Aging on Material Strength and Fracture Behavior in Mod.9Cr-1Mo Steel

        The material properties of heat resistant materials are known to be affected by thermal aging as operating time is accumulated at power plants. In this study, the influence of thermal aging on yield strength, ductility and J-Resistance behavior for Mod.9Cr-1Mo (ASME Grade 91) steel which is a material widely adopted for Generation IV nuclear energy system has been investigated. Service exposed Gr.91 steel materials sampled from a piping system of an ultrasupercritical (USC) plant in Korea with accumulated operation time of 73,716 hours were used for material testing. The test results of the service exposed material were compared with those of the virgin Gr.91 steel. Those test data were compared with the material properties of ASME code and RCC-MRx code that are international elevated temperature design codes. Conservatism of the material properties in design code has been quantified based on the comparison of virgin and service exposed materials.

      • Construction of a High Temperature Grade 91 Sodium Component Test Loop

        <P>A large scale sodium test facility of ‘CPTL’(Component Performance Test Loop) for simulating thermal hydraulic behavior of the Korean demonstration fast reactor components such as IHX(Intermediate Heat Exchanger), DHX(Decay Heat Removal Heat Exchanger) and sodium pump under development by KAERI is to be constructed. The design temperature of this test loop is 600°C and design pressure is 1MPa. The three heat exchangers are made of Grade 91 steel. Another sodium test facility of the ‘STEF’(Sodium Thermal-Hydraulic Experimental Facility) will be constructed next to the CPTL facility to simulate the passive decay heat removal behavior in the sodium cooled fast reactor. In this paper, the overall facility features of the CPTL and STEF are introduced and preliminary conceptual design of the facilities are carried out.</P>
