      • Overlapping and unique toxic effects of three alternative antifouling biocides (Diuron, Irgarol 1051<sup>®</sup>, Sea-Nine 211<sup>®</sup>) on non-target marine fish

        <P><B>Abstract</B></P> <P>The use of alternative biocides has increased due to their economic and ecological relevance. Although data regarding the toxicity of commercial alternative biocides in marine organisms are accumulating, little is known about their toxic pathways or mechanisms. To compare the toxic effects of commercial alternative biocides on non-target pelagic fish (flounder) embryos, we investigated the adverse effects of developmental malformation and transcriptional changes. Three biocides including Diuron, Irgarol 1051<SUP>®</SUP> and Sea-Nine 211<SUP>®</SUP> produced a largely overlapping suite of developmental malformations, including tail-fin fold defects and dorsal body axis curvature. In our test, the potencies of these biocides were ranked in the following order with respect to malformation and mortalities: Sea-Nine 211<SUP>®</SUP> > Irgarol 1051<SUP>®</SUP> > Diuron. Consistent with the toxicity rankings, the expression of genes related to heart formation was greater in embryonic flounder exposed to Sea-Nine 211<SUP>®</SUP> than in those exposed to Irgarol 1051<SUP>®</SUP> or Diuron, while expression of genes related to fin malformation was greater in the Irgarol 1051<SUP>®</SUP> exposure group. In analyses of differential gene expression (DEG) profiles (fold change of genes with a cutoff <I>P < 0.05</I>) by high-throughput sequencing (RNA-seq), genes associated with nervous system development, transmembrane transport activity, and muscle cell development were significantly changed commonly. Embryos exposed to Diuron showed changes related to cellular protein localization, whereas genes associated with immune system processes were up-regulated significantly in embryos exposed to Irgarol 1051<SUP>®</SUP>. Genes related to actin filament organization and embryonic morphogenesis were up-regulated in embryos exposed to Sea-Nine 211<SUP>®</SUP>.         Antilock Brake Sysytem(ABS) si the device for getting the braking force and the cornering force of the car. The ABS sensor consists of a permanent magnet and pickup coil. When the rotor and the sensor changes, The air gap between the rotor and the sensor changes. This disturbs the magnetic flux emanating from the permanent magnet and generates ac voltage in the pickup coil. The frequency fo this ac voltage changes with the speed of rotation, and in this way the system is able to measure the rotation speed of the wheel. The purpose of this report is to describe a development method of a new type sensor after analyzing two kinds of typical sensors.

        Sphingosine 1-Phosphate and Sphingosine Kinase Activity during Chicken Embryonic Development

        The chicken embryo has been weil used in studies of the developmental process, and during development sphingosine and sphingosine 1-phosphate (So1P) are considered critical mediators of cell death and survival. In this study, we compared the sphingolipid contents of chicken embryos during the early embryonic development period from day 3 to day 6. HPLC analyses of sphingosine and So1P in chicken embryos revealed that sphingosine levels were greatly reduced on day 4 whereas So1P levels were not significantly changed. Sphingosine kinase(Sphk) activities, which require sphingosine as substrate to produce So1P, were also greatly reduced on day 4. Collectively, we found sphingosine levels and Sphk activities, but not So1P levels are changed in early stage of chicken embryos development.

        In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria gonorrhoeae(PPNG), we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by menas of the chromogenic cephalosporin method. In 1998, 93 strians of N. genorrhoeae were isolated, among which 60(64.5%) were PPNG. The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 64.5% in 1998.

        Study on the Fire Hazard and Risk Analysis Derived from the Plant Configuration Change During the Shutdown Period at Nuclear Power Plants

        Fire hazard and risk analysis at Nuclear Power Plants is implemented on the basis of the normal operational configuration. This steady configuration, however, can be changed due to the temporary displacement of equipment, electric cable and irregular movement of workers through the fire compartments when the on-line maintenance is processed during the power operation mode or the scheduled outage mode for the refueling. With the consequence of this configuration change, the fire analysis condition and the evaluation result will be different from those that were analyzed based on the steady configuration. In this context, at this paper, the general items for the reassessment are categorized when the configuration has changed. The contemporary zone models for the detail fire analysis are also illustrated for their application for each classified condition.

        개에서 동소성 간이식술시의 대사 및 혈역학적 변화

        Skilled and experienced anestheia is of great importance for patients undergoing orthotopic liver transplantation, because of multiple preexisting medical problems in such patients as well as the intraoperative problems of rapid hemodynamic, metabolic, and coagulation changes. In this study, the intraoperative hemodynamic and laboratory data were analyzed in ten dogs that underwent an orthotopic liver transplantation procedure by veno-venous bypass using Biopump. Liver transplantation can be divided into three distinct periods: stage I, or preanhepatic stage, which begins with the induction of anesthesia and continues until cross clamping of portal vein and IVC; stage II, or anhepatic stage, which begins at the anhepatic time and continues until the donor liver is reperfused by the recipients circulating blood; and stage III, or postanhepatic stage, which begins at the time of reperfusion and continues until the end of surgical procedure. The hemodynamic changes at the time of IVC and portal vein cross clamping were decreases in CVP, PCWP, and pulmonary artery pressure in spite of using Biopump. The significant metabolic alternations during anhepatic stage were decrease in blood glucose levels and increase in blood lactate levels. The more significant hemodynamic changes occurred at the time of reperfusion. Systolic pressure decreased suddenly to 58±6 mmHg and cardiac output decreased to 1.08±0.1l L/min. However heart rate, pulmonary artery pressure, CVP, and PCWP did not change significantly. During stage III, hyperglycemia occurred quite frequently. Significant abnormal coagulation chages could not be found, probably because the dogs were healthy. In conclusion, during anhepatic stage, we have to compensate for alternations of fluid balance. At the time of reperfusion, we should prevent severe hemodynamic changes and treat them immediately if they occur. However, it seems that glucose administration is not necessary to the liver recipient during stage II because there is no significant hemodynamic depression e to hypoglycemia at this time and hyperglycemia occurs later

        Genome-Wide Analysis of DNA Methylation before- and after Exercise in the Thoroughbred Horse with MeDIP-Seq

        Athletic performance is an important criteria used for the selection of superior horses. However, little is known about exercise-related epigenetic processes in the horse. DNA methylation is a key mechanism for regulating gene expression in response to environmental changes. We carried out comparative genomic analysis of genome-wide DNA methylation profiles in the blood samples of two different thoroughbred horses before and after exercise by methylated-DNA immunoprecipitation sequencing (MeDIP-Seq). Differentially methylated regions (DMRs) in the pre-and post-exercise blood samples of superior and inferior horses were identified. Exercise altered the methylation patterns. After 30 min of exercise, 596 genes were hypomethy-lated and 715 genes were hypermethylated in the superior horse, whereas in the inferior horse, 868 genes were hypomethylated and 794 genes were hypermethylated. These genes were analyzed based on gene ontology (GO) annotations and the exercise-related pathway patterns in the two horses were compared. After exercise, gene regions related to cell division and adhesion were hypermethylated in the superior horse, whereas regions related to cell signaling and transport were hypermethylated in the inferior horse. Analysis of the distribution of methylated CpG islands confirmed the hypomethylation in the gene-body methylation regions after exercise. The methylation patterns of transposable elements also changed after exercise. Long interspersed nuclear elements (LINEs) showed abundance of DMRs. Collectively, our results serve as a basis to study exercise-based reprogramming of epigenetic traits.

      • 수입 외국산 모발세정용 화장품의 중금속 농도에 관한 연구

        Objective : For the purpose of preparing the fundamental data and preventing the health impairment due yo heavy metals in hair cleaner. Methods and Material : The author determined the concentration of heavy metals(Fb, fn, Cr, Ni and Cu) in hair cleaner with flameless atomic absorption spectrophotometer. Results : The results were as follows : 1. The mean concentration of heavy metals in total hair cleaner were 0.046㎍/g for Pt, 0.O67㎍/g for Mn, 0.069㎍/g for Cr, 0.046㎍/g for Ni, and 0.022㎍/g for Cu, respectively. 2. There was not statistically significant difference to mean concentrations of each heavy metal in hair cleaner by producing nation. 3. The highest mean concentration of each heavy metal in hair cleaner by type of use were 0.140㎍/g for Mn in sensitive hair cleaner(p<0.05), 0.053㎍/g for Cu in others products(p<0.01). 4. The mean concentration of heavy metals in hair cleaner by type of hair cleaner were high at 0.O54㎍/g for Pb in shampoo products(p<0.O5), 0.120㎍/g for Cr and 0.06㎍/g for Cu in combination products(p<0.01). 5. The mean concentration of heavy metals in hair cleaner by acidity were high at 0.056㎍/g for Pb, 0.124㎍/g for Cr, and 0.032㎍/g for Cu in alkali products (p<0.05). 6. Correlation coefficient of heavy metal concentration in hair cleaner were 0.175 for Pb and Cr, 0.165 for Pb and Mn, 0.824 for Cr, and Mn, 0.189 for Cr, and Ni, 0.406 for Cr, and Cu, 0.197 for Mn and Ni, and 0.375 for Mn and Cu(p<0.05). 7. Exposure amount of heavy metals per when we used lOg of hair cleaner (onetime using amount) were 0.46㎍ for Pb, 0.67㎍ for Mn, 0.69㎍ for Cr, 0.46㎍ for Ni, and 0.22㎍ for Cu, respectively. Conclusion : As above results, there was very low level concentration of heavy metal in hair cleaner but we are using the hair cleaner everyday, therefore we can exposed to heavy metals as cronic exposure so the author suggest to prepare the program of preventing the health impairment due to heavy metals.

        From May to July 1993, we observed an epidemic of aseptic meningitis. We performed clinical and virological studies on 174 children with aseptic meningitis who had been admitted to the Department of Pediatrics, Asan Medical Center, from May to July, 1993 The results were as follows; 1) This epidemic of aseptic meningitis was most prevalent in June 1993. The most of the patients were 3 to 6 years old and male to female ratio was 1.6 : 1. 2) The most patients suffered from fever, vomiting, headache, and abdominal pain was noticed in 36% of patients, characteristically. 3) In 7 cases among 10 randomly sampled cases, enterovirus was cultured. Among them, we performed serotyping in 3 cases and Echovirus 9 was isolated.

        Critical role of bile acid (BA) in the cellular entry and permissiveness of Hepatitis B virus in vitro

        Background Lack of universal replication system for hepatitis B virus with narrow host range and organ tropism has hampered to uncover the pathogenesis of HBV. Previously, we reported the essentiality of humoral milieu and its components toward HBV and hepatitis C virus survival/viability in vitro. Of these components, the precise role of enterohepatic humoral milieu such as bile acid (BA) on HBV cultivation in vitro and in vivo is unknown. Objective We explored whether BA, specifically taurochenodeoxycholic acid (tCDCA) would directly regulate the viral DNA and surface antigen expression of HBV in vitro, consequently rendering HBV to enter into human or murine immortalized hepatocytes, and non-hepatocytes. Result We found that higher concentration of taurochenodeoxycholic acid (tCDCA) is able to preserve the genomic stability of HBV in cell-free DMEM, showing higher the surface antigenicity than taurocholic acid (tCA). In line, we found that in vitro cell culture condition (100 μmol/L of tCDCA coupled with 1 × 108 g e/mL HBV) would be optimal for HBV entry into target cells. Using this, human (HepG2, Huh7), and rodent (Hepa1c1c7, H4-II-E) hepatoma cell lines were infected by HBV, as evidenced by the presence of HBV biomarkers (HBsAg, and HBV DNA in culture supernatant, as well as HBcAg in cell). Further, cellular entry test revealed that HBV is able to infect 12 different non-hepatic cell lines regardless of species, and organ/tissue, consequently reproducing progeny as confirmed by HBV biomarkers. Last, reinfection test showed that the progenies of HBV from immortalized HepG2, and Hepa1c1c7 cells are able to enter into each or vice versa naïve HepG2, and Hepa1c1c7 cells with or without BA. Conclusion This study demonstrates that enterohepatic humoral milieu such as BA, specifically tCDCA would directly regulate HBV DNA and its surface antigen expression in vitro, consequently rendering HBV to enter into human or murine immortalized hepatocytes, and non-hepatocytes. This is the first note to render HBV permissive to human or rodent hepatic and non-hepatic cells via sole manipulation of humoral milieu, thus establishing the platform for in vitro robust replication system of HBV.
