        Comparative profiling of plasma proteome from breast cancer patients reveals thrombospondin-1 and BRWD3 as serological biomarkers

        Breast cancer is the most common cancer in women worldwide. It is necessary to identify biomarkers for early detection, to make accurate prognoses, and to monitor for any recurrence of the cancer. In order to identify potential breast cancer biomarkers, we analyzed the plasma samples of women diagnosed with breast cancer and age-matched normal healthy women by mTRAQ-based stable isotope-labeling mass spectrometry. We identified and quantified 204 proteins including thrombospondin-1 (THBS1) and bromodomain and WD repeat-containing protein 3(BRWD3) which were increased by more than 5-fold in breast cancer plasma. The plasma levels of the two proteins were evaluated by Western blot assay to confirm for their diagnostic value as serum markers. A 1.8-fold increase in BRWD3 was observed while comparing the plasma levels of breast cancer patients (n = 54) with age-matched normal healthy controls (n = 30), and the area under the receiver operating characteristic curve (AUC) was 0.917. THBS1 was detected in pooled breast cancer plasma at the ratio similar to mTRAQ ratio (>5-fold). The AUC value for THBS1 was 0.875. The increase of THBS1 was more prominent in estrogen receptor negative and progesterone receptor negative patients than receptor-positive patients. Our results are evidence of the diagnostic value of THBS1 in detecting breast cancer. Based on our findings, we suggest a proteomic method for protein identification and quantification lead to effective biomarker discovery.

      • Doping of donor-acceptor polymers with long side chains via solution mixing for advancing thermoelectric properties

        <P><B>Abstract</B></P> <P>One-step doping of conjugated polymers by solution mixing is typically performed instead of sequential doping because of its simplicity. However, doped polymer solutions often exhibit poor solubility, and the presence of dopants in the produced films can disturb the molecular ordering of polymer structures. In this work, effective pairs of two donor-acceptor (D-A) type polymers and a molecular dopant characterized by high solution stability and good thermoelectric properties of the prepared thin films have been reported. The presence of long side chains in the polymer structures preserves their original solubilities and crystallinity in the solution and thin-film states, respectively, even at large amounts of added dopant (up to 38 mol%). Furthermore, the relatively shallow levels of the highest occupied molecular orbitals of the selected D-A polymers enable efficient charge transfer from the dopant species. Owing to their good charge transport properties, the doped D-A polymers exhibit outstanding thermoelectric properties with a maximum power factor of 31.5 μW m<SUP>−1</SUP> K<SUP>−2</SUP>, which is more than an order of magnitude higher than those of the control samples prepared from donor-only poly(3-hexylthiophene).</P> <P><B>Highlights</B></P> <P> <UL> <LI> Doping of two donor-acceptor polymers (PCDTFBT and PCDTPT) is performed by simple one-step solution mixing. </LI> <LI> The long side chains of the polymers allow high solution stabilities of the polymer-dopant mixtures. </LI> <LI> The polymer films retain structural coherence even after doping with high dopant ratios up to 38 mol%. </LI> <LI> Owing to their good charge transport properties, the doped polymers exhibit high thermoelectric power factors up to 31.5 μW m<SUP>-1</SUP> K<SUP>−2</SUP>. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>Outstanding thermoelectric properties of two D-A polymers are demonstrated by molecular doping with F<SUB>4</SUB>TCNQ via a simple one-step solution mixing process.       • Performance Comparison of Tracking Filters in Terms of Predicted Hitting Point

        This article describes the performance comparison of several tracking filters from the viewpoint of gun fire control. It is common that filter performance can be represented by the RMSE of the current state. However, for the design of a gun fire control system, the performance of the tracking filter in terms of prediction is more important since the current state proceeds with a designed dynamic model to a certain future which is used for the final gun order. It is shown that the predicted results amplify the current state RMSE and can be a good measure.

        The effects of <i>Chamaecyparis obtusa</i> essential oil on pain-related behavior and expression of pro-inflammatory cytokines in carrageenan-induced arthritis in rats

        <P>Chamaecyparis obtusa essential oil (COE) has been widely used to treat allergic diseases and was suggested to exert anti-inflammatory, antioxidant, and antimicrobial effects. This study evaluated the effects of COE on pain-related behavior and pro-inflammatory cytokines in rats with carrageenan (CGN)-induced arthritis. Reduced dynamic weight load on inflamed joint in voluntarily walking rats was used as the behavior test for arthritic pain; 10% COE-treated group was significantly attenuated pain (6-8h post-CGN injection) compared to VEH (mineral oil)-treated group. In addition, the protein levels of interleukin (IL)-1, tumor necrosis factor-, IL-6 (6-8h), and cyclooxygenase (COX)-2 (8h) within the synovial membrane, as well as IL-1, COX-2 (6-8h), and IL-6 (5-7h) within the meniscus, of 10% COE-treated group were significantly reduced. The current results implicate that COE has anti-inflammatory and anti-nociceptive effects on arthritis in rats.</P>

      • 강원도 국민학생의 베타용혈성 연쇄구균 보균율에 관한 연구

        Group A β-hemolytic streptococcal pharyngitis has been studied very widely in area pediatrics, because of its severe sequelae, that is, acute rheumatic fever and acute glomerulonephritis. Throat culture is gold standard to confirm group A streptococcal (GAS) pharyngitis. Antistreptolysin-O (ASO) test is also performed to investigate the immune state of GAS pharyngitis. In this study, the carrier rate of β-hemolytic streptococci (BHS) was evaluated on 798 children of three elementary schools located in Kangweondo on March and April, 1992. The carriers of BHS were 108 (13.5%) of which male were 63 (14.1%), female 45 (12.8%) respectively. The carrier rate of BHS was high in 12 (20.3%) and 11 (19.4%) year old children, compared to 2.4% in 13 year old. The carrier rate was ranged from 13.0% to 15.8% in three schools. Of 108 strains, 79 (73.2%) were identified as group A, 14 (13.0%) as group G, 6 (5.6%) as group B, 3 (2.9%) as group C, and 6 as non-group A,B,C,G respectively. The demography of positive rate of-GAS showed similar pattern to the one of BHS. The carriers of GAS were 79 (9.9%) of which male were 49 (11.1%), female 30 (8.5%) respectively. ASO of 85 children haboring BHS were screened by Rapidia-ASO(Fujirebio, Japan). The positive rate of ASO was 79.4% in GAS cases, 65% in non-GAS cases respectively. This result means that ASO is not sufficient to differentiate true infection from carrier and can be raised in non-GAS cases. This study will help clinician know present bacteriologic and epidemiologic knowledge of BHS in Korean children.

      • Unasyn에 대한 임상검체에서 분리한 세균의 감수성

        Recent clinical isolates of bacteria are frequently resistant to various antimicrobial agents including β-lactam antibiotics. Sulbactam is an β-lactamase inhibitor which can restore the activity of the β-lactam antibiotics when combined. Clinical isolates of bacteria from three university hospitals in Seoul and one in Taegu were compared for their susceptibility to sulbactam plus ampicillin (Unasyn), by the disk diffusion method. Among the penicillin G-resistant Staphylococcus aureus isolated at Kyungpuk University Hospital in Taegu and Kyung Hee University Hospital in Seoul 80.0% and 92.9% respectively were susceptible to Unasyn. Among the strains from Seoul National University Hospital, 90.7% of the penicillin G-resistant and methicillin-susceptible S. aureus and 34.3% of methicillin-resistant S. aureus were susceptible to Unasys. Among the ampicillin-resistant gram-negative bacilli isolated at Kyung Hee University Hospital 75.0% were susceptible to Unasys, while 53.6% of gentamicin-susceptible ampicillin-resistant gram-negative bacilli, and 42.5% of gentamicin-and-ampicillin resistant gram-negative bacilli were susceptible to Unasyn. Unasyn was more active against Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis and Acinetobacter anitratus than to other speices.

      • Forecast Driven Simulation Model for Service Quality Improvement of the Emergency Department in the Moses H. Cone Memorial Hospital

        Patient satisfaction with the Emergency Department(ED) in a hospital is related to the length of stay, and especially to the amount of waiting time for medical treatments. ED over-crowding decreases quality and efficiency, therefore affecting hospitals` profitability. This paper presents a forecasting and simulation model for resource management of the ED at Moses H. Cone Memorial Hospital. A linear regression forecasting model is proposed to predict the number of ED patient arrivals, and then a simulation model is provided to estimate the length of stay of ED patients, system throughput, and the utilization of resources such as triage nurses, patient beds, registered nurses, and medical doctors. The near future load level of each resource is presented using the proposed models.
