        Whole-genome resequencing analysis of 20 Micro-pigs

        Background Miniature pigs have been increasingly used as mammalian model animals for biomedical research because of their similarity to human beings in terms of their metabolic features and proportional organ sizes. However, despite their importance, there is a severe lack of genome-wide studies on miniature pigs. Objective In this study, we performed whole-genome sequencing analysis of 20 Micro-pigs obtained from Medi Kinetics to elucidate their genomic characteristics. Results Approximately 595 gigabase pairs (Gb) of sequence reads were generated to be mapped to the swine reference genome assembly (Sus scrofa 10.2); on average, the sequence reads covered 99.15% of the reference genome at an average of 9.6-fold coverage. We detected a total of 19,518,548 SNPs, of which 8.7% were found to be novel. With further annotation of all of the SNPs, we retrieved 144,507 nonsynonymous SNPs (nsSNPs); of these, 5968 were found in all 20 individuals used in this study. SIFT prediction for these SNPs identified that 812 nsSNPs in 402 genes were deleterious. Among these 402 genes, we identified some genes that could potentially affect traits of interest in Micro-pigs, such as RHEB and FRAS1. Furthermore, we performed runs of homozygosity analysis to locate potential selection signatures in the genome, detecting several loci that might be involved in phenotypic characteristics in Micro-pigs, such as MSTN, GDF5, and GDF11. Conclusion In this study, we identified numerous nsSNPs that could be used as candidate genetic markers with involvement in traits of interest. Furthermore, we detected putative selection footprints that might be associated with recent selection applied to miniature pigs.

        Purpose: The purpose of this descriptive study is to examine the correlation between nursing students’ perception of nurses and the level of their satisfaction with the major. Method: The survey was done with the total number of 192 nursing students from 10 different universities in S city selected by a convenience samplingmethod. In order to measure the perception of nurses, the measurement which was invented by Lee(1992) and revised by Jang(1998) was used. Also, the measurement designed by Shim, and modified by Kim(2007) was used for measuring of the students’ satisfaction in their major. The collected data was analyzed by SPSS 17.0. Results: 1.The most decisive factor of the conceptualization of the image of nurses for the nursing college students is their experience of meeting nurses as a patient. 2. Students reconsider the role of nurses by experiencing immediate relationships with nurses in field training. 3. Other factors of the survey such as the students’academic year, religion, the field training situation (individual experience, type of the hospital where they experienced the training, and a relationship with the nurse in charge of the training) or the existence of nurses among their family members do not affect the students’ satisfaction. 4. There is a positive correlation between the image of nurses of nursing college students and their satisfaction with the major(0.66). Conclusion: The positive image of nurses is directly related to the students’ satisfaction in the nursing major, which hugely affects on their academic achievements and decisions of career path. Therefore, there should be more efforts to increase the positive perception of nurses among nursing students.

        직업훈련 프로그램이 장애인 직업준비도에 미치는 효과

        직업재활을 위해서는 무엇보다 장애인의 직업적 능력을 최대한으로 개발하고 적응시키기 위한 직업훈련이 필수적이며, 이러한 훈련을 통해 직업인으로서의 바탕이 되는 자질과 기능을 충분히 갖도록 해야 한다. 직업훈령에 있어서 단순한 직업적 기능의 훈련만으로는 효과를 거둘 수 없고 직업인으로서의 전인적 훈련이 필요하다. 따라서 장애인에게 적용되고 있는 직업훈련프로그램이 직업적응, 직업유지를 위해 가장 바탕이 되는 직업준비도에 어느 정도 영향을 미치는 효과가 있는지를 파악하여 직업훈련서비스 제공에 있어서 개선을 위한 기초자료로 활용하고자 하였다. 구체적인 연구문제로 직업훈련프로그램이 장애인의 직업태도, 대인관계 및 사회기술, 직업생활 유지에 필요한 직업에 대한 인식과 직업적응에 미치는 효과와 장애인 직업준비도 요인간의 영향에 대해서 알아보았다. 본 연구에서는 WPP, VALPAR 17, 진로태도성숙도, 자기효능감 측정 도구를 사용하여 대구직업전문학교 입학생 16명을 대상으로 4-5개월 동안 직업훈련프로그램 적용 전과 적용 이후 직업인성, 대인관계 및 사회기술, 직업인식, 자기효능감을 측정하여 직업준비도의 변화정도를 파악하였다. 직업준비도에 미치는 효과를 알아보기 위해 실시한 연구를 통해 나타난 결과들은 다음과 같다. 첫째, 직업준비도 요인 변화에 있어서 직업에서 요구하는 일에 적응하는데 중요한 요인이 되는 직업인성은 직업훈련프로그램 적용 이후 향상되었고, 직업준비를 위한직업인성 형성에 효과가 있는 것으로 나타났다. 둘째, 직업훈련프로그램 적용이 장애유형에 따른 직업준비도에 미치는 영향은 직업인성과 직업인식, 자기효능감에서 장애유형에 따라 효과의 차이가 발생하였다. 셋째, 직업준비도 요인간의 상관관계는 직업인성이 높으면 자기효능감도 높아지며, 또한 자기효능감이 높을수록 직업인식이 높아지는 것으로 나타났다. 넷째, 직업준비도 요인에 대한 독립변수들의 상대적 기여도를 알아보기 위해 실시한 회귀분석 결과에서 대상자들의 나이와 장애유형, 대인관계와 사회기술이 직업인성에 큰 영향을 미치는 것으로 나타났으며, 대인관계 및 사회기술은 아니와 직업인성이 큰 영향을 미치는 것으로 나타났다. 결론적으로 직업생활 유지에 있어서 직업적응을 위한 작업준비도를 높이기 위해서는 직업인성을 향상시킬 수 있는 직업훈련프로그램이 필요하며, 이러한 직업인성을 향상시킬 수 있는 방법은 대인관계 및 사회기술 영역에 비중을 둘 수 있는 직업훈련프로그램이 필요하다. 그리고 직업훈련프로그램의 직업유지나 직업적응에 대한 효과성 조사와 훈련교사들에 대한 교육도 지속적으로 이루어져야 하며, 이러한 것들이 프로그램구성이나 교과과정 작성 시 반영하는 연구설계가 필요하다. It is essential for rehabilitating persons with disabilities, to develop employment skills. To maximize vocational potential and abilities of persons with disabilities, it is required for them to obtain work attitudes and skills. However, the work skills training alone may not be enough for persons with severe disabilities, requiring them t o have work preparation services. Therefore, this study was aimed to understand whether the vocational training program provided by Dae-Gu Metropolitan Training Institute for Persons with Disabilities had impact on the improvement of both work skills and job readiness skills for them. Data gathered included the following areas. The effect of the disabled's vocational behaviour. the personal relations, the social skills, the vocational consciousness and the job readiness areas. The WPP, VALPAR 17, Course Attitude Maturity, Self-Efficacy Feeling Measurement Tool were utilized for the data gathering purposes. For vocational Rehabilitation, first of all, it is essential for disabled trainees to develop and adapt through individualized service provisors. They are required to have enough appropriate attitudes and skills which basic to average workers. For the vocational training, a simple vocational skills training alone may not be enough. Therefore, this study was aims to figure out whether the vocational training program which have applied to the trainees in Dae-Gu Vocational Training Institute (DGVTI) was effective or not. This training program was composed of the work adaptation training and job readiness training. Initial study questions posed are as follows. Whether this training program had effect on the research participants' vocational behaviours, personal relationship skills, social skills, the vocational consciousness and the job readiness skills. The current researcher utilized the WPP, VALPAR 17, Course Attitude Maturity. Self-Efficacy Feeling Measurement Tool. They measure types of vocational personality. personal relations and social skills, vocational and self-efficacy feeling in comparison with before and after the application of training program. The 16 research participants had exposed to 4-5 months in Dae-Gu Vocational Training Institute(DGVTI), operated by Korea Employment Promotion Agency for the Disabled (KEPAD). Statistical analyses focused on the means, standard deviations, and frequencies. Futher analyses included t-tests, correlation Analyses, regression analyses. The results of this study can be summarized as follows. First, in the job readiness skills area, after training, vocational personality (which is the important factor adapting to assignment demanding occupation) is improved, and reveals that the formation of vocational personality for job readiness and, in the vocational recognition and self-efficacy feeling are positively changed. Second. job readiness skills had effect to improve research participants' vocational adaptation skills, vocational recognition and self-efficacy feeling. Third. "correlation between job readiness factors" appeared that if scores of the vocational personality is high, then the self-efficacy feeling also rises. When the self-efficacy scores are high, the vocational recognition rises. Fourth, the results of regression analyses indicated that the age and the types of disabilities of the research participants had influenced on the occupational personality. Also, research participants who showed good personal relation skills and social skills had good job readiness. It is conclusive that the method of improving occupational personality would influence on personal relations and social skills. In conclusion, it is necessary for the rehabilitation practitioners to provide skills and interpersonal skills with their clients and trainees. The implication of this study is that there is a strong need to improve quality of vocational training programs.

        The causes of pyogenic liver abscess has been known as biliary tract disease or intrabadominal infection but the large proportions of the patients has no apparent underlying disorders. Recently colonic mucosal lesions were reported in patients with cryptogenic liver abscess and it has been suggested that colonic mucosal break may play a role in developing liver abscess in otherwise healthy patients. We experienced a patient of severe recurrent liver abscess complicated with endophthalmitis only 3 months after successful treatment of initial cryptogenic liver abscess and a polypoid colon cancer was discovered by chance. It seems prudent to proceed colonoscopic examination in patients with cryptogenic liver abscess especially when it is recurrent.

        Reports : Structure-activity relationships of the intramolecular disulfide bonds in coprisin, a defensin from the dung beetle

        Defensins, which are small cationic molecules produced by organisms as part of their innate immune response, share a common structural scaffold that is stabilized by three disulfide bridges. Coprisin is a 43-amino acid defensin-like peptide from Copris tripartitus. Here, we report the intramolecular disulfide connectivity of cysteine-rich coprisin, and show that it is the same as in other insect defensins. The disulfide bond pairings of coprisin were determined by combining the enzymatic cleavage and mass analysis. We found that the loss of any single disulfide bond in coprisin eliminated all antibacterial, but not antifungal, activity. Circular dichroism (CD) analysis showed that two disulfide bonds, Cys20-Cys39 and Cys24- Cys41, stabilize coprisin’s α-helical region. Moreover, a BLAST search against UniProtKB database revealed that coprisin’s α-helical region is highly homologous to those of other insect defensins. [BMB Reports 2014; 47(11): 625-630]

        Phase diagram of Ag–Pd bimetallic nanoclusters by molecular dynamics simulations: solid-to-liquid transition and size-dependent behavior

        <P>This report on the solid-to-liquid transition region of an Ag–Pd bimetallic nanocluster is based on a constant energy microcanonical ensemble molecular dynamics simulation combined with a collision method. By varying the size and composition of an Ag–Pd bimetallic cluster, we obtained a complete solid–solution type of binary phase diagram of the Ag–Pd system. Irrespective of the size and composition of the cluster, the melting temperature of Ag–Pd bimetallic clusters is lower than that of the bulk state and rises as the cluster size and the Pd composition increase. Additionally, the slope of the phase boundaries (even though not exactly linear) is lowered when the cluster size is reduced on account of the complex relations of the surface tension, the bulk melting temperature, and the heat of fusion. The melting of the cluster initially starts at the surface layer.       • Fabrication of PVDF film sensors for fatigue damage monitoring of single-lap adhesive joints

        <P><B>Abstract</B></P> <P>Polyvinylidene fluoride (PVDF) has been widely used in piezoelectric applications because of its outstanding properties such as low permittivity, high thermal stability, high chemical resistance, and flexibility. In this study, PVDF film sensors were prepared for fatigue damage monitoring of single-lap adhesive joints. PVDF films were fabricated using the solvent casting method, and post treatment, including mechanical stretching and heat setting, was conducted to increase the β-phase crystallinity of the films. Subsequently, the PVDF films were polarized to align the molecular dipoles. The crystal structures of the PVDF films were investigated using X-ray diffraction (XRD) and the thermal behaviors were analyzed using differential scanning calorimetry (DSC). The piezoelectric constants of the prepared PVDF films were measured using a d<SUB>33</SUB> meter. The sensing performance of the films for fatigue damage monitoring of single-lap adhesive joints was assessed by fatigue tests under various stress levels. In addition, a fatigue failure manual based on the PVDF sensor signal, which can predict the failure of the adhesive joints, was constructed.</P>

      • Characterizing the potential estrogenic and androgenic activities of two disinfection byproducts, mono-haloacetic acids and haloacetamides, using <i>in vitro</i> bioassays

        <P><B>Abstract</B></P> <P>Exposure to disinfection byproducts (DBPs) is potentially related to cytotoxic, genotoxic, mutagenic, and tumorigenic effects in humans, in addition to their adverse effects on the environment. However, their impacts on endocrine disruption, especially reproductive toxicity, remain largely unknown. In this study, the estrogenic and androgenic activities of DBPs and corresponding antagonistic activities were investigated using a yeast-based reporter assay, focusing on haloacetic acids and haloacetamides. We also examined the cytotoxicity of DBPs and mechanisms of antagonistic activities. Of the DBPs assayed, iodoacetamide (IAM) and bromoacetamide (BAM) were the most cytotoxic, with LC<SUB>50</SUB> values of 0.0462 and 0.0537 mM, respectively, followed by chloroacetic acid (CAA; LC<SUB>50</SUB> = 4.87 mM) and chloroacetamide (CAM; LC<SUB>50</SUB> = 5.28 mM). Iodoacetic acid (IAA) and bromoacetic acid (BAA) were the least cytotoxic, with LC<SUB>50</SUB> values of 5.52 and 6.35 mM, respectively. IAA (EC<SUB>10</SUB> = 0.00573 mM; EC<SUB>50</SUB> = 0.0215 mM) exhibited most potent estrogenic activity, and CAA (EC<SUB>10</SUB> = 0.0434 mM) and BAM (EC<SUB>10</SUB> = 0.0150 mM) showed weak estrogenic and androgenic activities, respectively. By contrast, IAM exhibited anti-estrogenic effects.       • Photoresponsive Micelle-Incorporated Doxorubicin for Chemo-Photodynamic Therapy to Achieve Synergistic Antitumor Effects

        <P>A combination of chemo-photodynamic therapy has been manifested as a promising strategy for efficient cancer treatment due to the enhanced therapeutic efficacy. Here, we designed doxorubicin (DOX)-loaded photoresponsive micelles (DPRMs) based on a combination of chlorin e6 (Ce6) and lipoic acid (LA) conjugated methoxy-poly(ethylene) glycol (mPEG-Ce6, mPEG-LA) to achieve effective drug delivery using a single system. DPRMs were optimized with different molar ratios of mPEG-Ce6 and mPEG-LA which showed uniformly spherical morphology of size ∼130 nm and approximately 9% of DOX loading contents. Photoresponsive lipoyl ring of mPEG-LA was incorporated in DPRMs in order to induce photomediated reduction resulting in 2-3-fold accelerated DOX release according to higher molar ratio of mPEG-LA and enhancement of light dose. The photoresponsive DOX release and ROS generation by Ce6 mediated cytotoxic effect of DPRMs were demonstrated in vitro using CT-26 (mouse colon cancer) and HCT-116 (human colon cancer) cells. We observed both the photosensitizer and the anticancer drug are colocalized in the tumor cells to achieve effective enhancement. Additionally, the DPRMs with laser irradiation successfully inhibited tumor growth in CT-26 tumor bearing mouse model and immunohistochemical staining verified apoptosis-mediated tumor growth inhibition. These observations demonstrated that the DPRMs showed a higher therapeutic effect than the other systems and PDT maximized the antitumor effect. Thus, DPRMs confirmed the advantages as a chemo-photodynamic dual-therapy with a synergistic therapeutic effect and great potential for cancer treatment.</P> [FIG OMISSION]</BR>

      • Topography of the Submental Artery That Should Be Considered in Bleeding During Dentoalveolar Surgery

        ABSTRACT: The purpose of the present study was to provide precise data regarding the branching pattern of the submental artery, which should be considered in occasions of bleeding during various dentoalveolar surgical procedures of the mandible, such as implant surgeries, tori removal, and iatrogenic injuries.Twenty-six embalmed adult hemifaces from Korean cadavers were used in this study. The vertical distance, horizontal distance, and diameter of the submental artery were measured from the site of the first premolar to the third molar. In cases where there was penetration of the mylohyoid muscle by the main branches of the submental artery, the same items were measured at that point.The vertical distance between the submental artery and the inferior border of the mandible decreased toward the premolar, whereas the horizontal distance from the lingual plate of the mandible increased gradually as it traveled in the anterior direction. The diameter of the artery narrowed slightly toward the premolar. The main branches of the submental artery perforated the mylohyoid muscle in 14 (54%) of the 26 specimens.As a result of this study, the submental artery is located higher from the inferior border and closer to the lingual plate of the mandible in the region of the molar than that of the premolar. Therefore, clinicians should be more careful of bleeding when performing surgery in the molar region compared with the premolar region. Where the mylohyoid muscle is perforated by the main branches of the submental artery, its point of insertion can be observed in diverse locations.
