Background: Caveolin-1, the scaffolding protein of cholesterol-rich invaginations, plays an important rolein store-operated Ca2þ influx and its phosphorylation at Tyr14 (p-caveolin-1) is vital to mobilize protectionagainst myocardial ischemia (MI) injury. SOCE, comprising STIM1, ORAI1 and TRPC1, contributesto intracellular Ca2þ ([Ca2þ]i) accumulation in cardiomyocytes. The purified extract of steamed Panaxginseng (EPG) attenuated [Ca2þ]i overload against MI injury. Thus, the aim of this study was to investigatethe possibility of EPG affecting p-caveolin-1 to further mediate SOCE/[Ca2þ]i against MI injury in neonatalrat cardiomyocytes and a rat model. Methods: PP2, an inhibitor of p-caveolin-1, was used. Cell viability, [Ca2þ]i concentration were analyzedin cardiomyocytes. In rats, myocardial infarct size, pathological damages, apoptosis and cardiac fibrosiswere evaluated, p-caveolin-1 and STIM1 were detected by immunofluorescence, and the levels ofcaveolin-1, STIM1, ORAI1 and TRPC1 were determined by RT-PCR and Western blot. And, release of LDH,cTnI and BNP was measured. Results: EPG, ginsenosides accounting for 57.96%, suppressed release of LDH, cTnI and BNP, and protectedcardiomyocytes by inhibiting Ca2þ influx. And, EPG significantly relieved myocardial infarct size, cardiacapoptosis, fibrosis, and ultrastructure abnormality. Moreover, EPG negatively regulated SOCE viaincreasing p-caveolin-1 protein, decreasing ORAI1 mRNA and protein levels of ORAI1, TRPC1 and STIM1. More importantly, inhibition of the p-caveolin-1 significantly suppressed all of the above cardioprotectionof EPG. Conclusions: Caveolin-1 phosphorylation is involved in the protective effects of EPG against MI injury viaincreasing p-caveolin-1 to negatively regulate SOCE/[Ca2þ]i.
Causal Relations between Exposome and Stroke: A Mendelian Randomization Study

Background and Purpose To explore the causal relationships of elements of the exposome with ischemic stroke and its subtypes at the omics level and to provide evidence for stroke prevention. Methods We conducted a Mendelian randomization study between exposure and any ischemic stroke (AIS) and its subtypes (large-artery atherosclerotic disease [LAD], cardioembolic stroke [CE], and small vessel disease [SVD]). The exposure dataset was the UK Biobank involving 361,194 subjects, and the outcome dataset was the MEGASTROKE consortium including 52,000 participants. Results We found that higher blood pressure (BP) (systolic BP: odds ratio [OR], 1.02; 95% confidence interval [CI], 1.01 to 1.04; diastolic BP: OR, 1.03; 95% CI, 1.01 to 1.05; pulse pressure: OR, 1.03; 95% CI, 1.00 to 1.06), atrial fibrillation (OR, 1.18; 95% CI, 1.13 to 1.25), and diabetes (OR, 1.13; 95% CI, 1.07 to 1.18) were significantly associated with ischemic stroke. Importantly, higher education (OR, 0.69; 95% CI, 0.60 to 0.79) decreased the risk of ischemic stroke. Higher systolic BP (OR, 1.06; 95% CI, 1.02 to 1.10), pulse pressure (OR, 1.08; 95% CI, 1.02 to 1.14), diabetes (OR, 1.28; 95% CI, 1.13 to 1.45), and coronary artery disease (OR, 1.58; 95% CI, 1.25 to 2.00) could cause LAD. Atrial fibrillation could cause CE (OR, 1.90; 95% CI, 1.71 to 2.11). For SVD, higher systolic BP (OR, 1.04; 95% CI, 1.00 to 1.07), diastolic BP (OR, 1.06; 95% CI, 1.01 to 1.12), and diabetes (OR, 1.22; 95% CI, 1.10 to 1.36) were causal factors. Conclusions The study revealed elements of the exposome causally linked to ischemic stroke and its subtypes, including conventional causal risk factors and novel protective factors such as higher education.

<P>A Gram-stain-negative, aerobic bacterium, designated strain YIM 77409T, was isolated from the Niujie hot spring in the Eryuan county of Dali, Yunnan province, south-west China. Cells of the strain were rod-shaped and colonies were yellow and circular. The strain grew at pH?6.0-8.0 (optimum, pH?7.0) and 50-70C (optimum, 60-65C). The predominant menaquinone was MK-8 and the DNA G+C content was 66.4?mol%. Major fatty acids (>10?%) were iso-C15?:?0 and iso-C17?:?0.The polar lipids consisted of one aminophospholipid, one phospholipid and two glycolipids. 16S rRNA gene sequence analysis showed that strain YIM 77409T formed a cluster with Thermus scotoductus DSM 8553T, Thermus antranikianii DSM 12462T, Thermus caliditerrae YIM 77925T and Thermus tengchongensis YIM 77924T, with highest 16S rRNA gene sequence similarity to T. scotoductus DSM 8553T (97.57?%). However, DNA-DNA hybridization indicated that strain YIM 77409T should be viewed as a representative of a novel species, as there was only 30.6??1.6?% reassociation with T. scotoductus DSM 8553T. On the basis of the morphological and chemotaxonomic characteristics, as well as the genotypic data, it is proposed that strain YIM 77409T represents a novel species of the genus Thermus, with the name Thermus amyloliquefaciens sp. nov. The type strain is YIM 77409T (?=?DSM 25898T?=?KCTC 32024T).</P>

Acifluorfen tolerance charateristics determined the involvement of absorption, translocation, and metabolism in acifluorfen tolerance. Less than 6% of the applied $^{14}C-acifluorfen$ was absorbed. There were no differences in acifluorfen absorption between susceptible and tolerant somaclones. More $^{14}C-acifluorfen$ was translocated in the susceptible than the tolerant somaclones. The susceptible somaclone did not metabolize acifluorfen while some somaclones (i.e.,EBN-3A) metabolized $^{14}C-acifluorfen$. <P>Adiponectin (APN) is an adipocyte-secreted adipokine that exerts well-characterized antidiabetic properties. Patients with type 2 diabetes (T2D) are characterized by reduced APN levels in circulation and impaired stem cell and progenitor cell mobilization from the bone marrow for tissue repair and remodeling. In this study, we found that APN regulates the mobilization and recruitment of bone marrow-derived mesenchymal stem cells (BMSCs) to participate in tissue repair and regeneration. APN facilitated BMSCs migrating from the bone marrow into the circulation to regenerate bone by regulating stromal cell-derived factor (SDF)-1 in a mouse bone defect model. More importantly, we found that systemic APN infusion ameliorated diabetic mobilopathy of BMSCs, lowered glucose concentration, and promoted bone regeneration in diet-induced obesity mice. In vitro studies allowed us to identify Smad1/5/8 as a novel signaling mediator of APN receptor (AdipoR)-1 in BMSCs and osteoblasts. APN stimulation of MC3T3-E1 osteoblastic cells led to Smad1/5/8 phosphorylation and nuclear localization and increased SDF1 mRNA expression. Although APN-mediated phosphorylation of Smad1/5/8 occurred independently from adaptor protein, phosphotyrosine interaction, pleckstrin homology domain, and leucine zipper containing 1, it correlated with the disassembly of protein kinase casein kinase 2 and AdipoR1 in immunoprecipitation experiments. Taken together, this study identified APN as a regulator of BMSCs migration in response to bone injury. Therefore, our findings suggest APN signaling could be a potential therapeutic target to improve bone regeneration and homeostasis, especially in obese and T2D patients.</P>

An efficient somatic embryogenesis and plant regeneration protocol was developed for Schisandra chinensis Baill, using embryogenic cell suspensions and optimized media conditions. Friable embryogenic callus was induced from cotyledonary leaf and hypocotyl explants of 7 days old seedlings on MS agar medium supplemented with 1.0 to 4.0 mg l-1 of 2,4-dichlorophenoxyacetic acid (2,4-D). Fast growing and well dispersed embryogenic cell suspensions were developed within two months when embryogenic calli were transferred to MS liquid medium containing 1.0 mg l-1 2,4-D. One third strength of MS medium was the best for both overall growth and development of somatic embryos in liquid culture. Over 3400 viable somatic embryos were produced from each 150 ml flask with an initial cell density of 30 mg in 30 ml medium. Germinated somatic embryos developed in liquid medium converted into plantlets after transferred to half-strength MS semi-solid medium. Approximately 90% of the converted plantlets were successfully transplanted to soil and grew into fertile plants.

Using the recently developed ABS algorithm for solving linear Diophantine equations we introduce an algorithm for solving a system of m linear integer inequalities in n variables, m≤n, with full rank coefficient matrix. We apply this result to solve linear integer programming problems with m≤n inequalities.

Parthenogenetic embryonic stem (pES) cells could provide a valuable model for research into genomic imprinting and X-linked diseases. In this study, pES cell lines were established from oocytes of hybrid offspring of Kunming and 129/Sv mice, and pluripotency of pES cells was evaluated. The pES cells maintained in the undifferentiated state for more than 50 passages had normal karyotypes with XX sex chromosomes and exhibited high activities of alkaline phosphatase (AKP) and telomerase. Meanwhile, these cells expressed ES cell molecular markers SSEA-1, Oct-4, Nanog, and GDF3 but not SSEA-3 detected by immunohistochemistry and RT-PCR. The pES cells could be differentiated into various types of cells from three germ layers in vitro by analysis of embryoid bodies (EBs) with immunohistochemistry and RT-PCR, and in vivo by observation of pES cell-derived teratoma sections. Therefore, the established pES cell lines contained all features of mouse ES cells. This work provides a new strategy for isolating pES cells from Kunming mice, and the pES cell lines could be applied as the cell model in research into genomic imprinting and epigenetic regulation of Kunming mice.

The results on the useful functional compounds of kenaf (Hibiscus cannabinus L.) leaves cultivated in reclaimed lands and the biological activity effects of extracts were as follows. On 98 days after sowing (DAS) Tainung-2 showed the highest total chlorophyll content (1.68 mg/g), and on 141 DAS Dowling showed higher chlorophyll content (1.50 mg/g) than the other two did. In all cultivars the total chlorophyll content was higher on 141 DAS than on 98 DAS. Total polyphenol and total flavonoid contents were the highest in Tainung-2 (30.50 mg/g and 57.03 mg/g, respectively), and total polyphenol and total flavonoid contents (30.50 mg/g and 57.03 mg/g, respectively) were the highest in 30% ethanol extraction. Ascorbic acid contents were higher on 141 DAS than on 98 DAS in three cultivars. SOD activities of kenaf leaf extract were generally over 90%. DPPH radical scavenging activity of Tainung-2 was higher than others.
