        In order to investigate the fungal flora of Mt. Chilgab, Chungnam Provincial Park, the wild mushroom were collected in the area from July to September in 1984 and 1985. 21 genera and 37 species of Basidiomycetes were added to the first report which 7 orders, 14 families, 22 genera, 40 species of Basidiomycetes and Ascomycetes were recorded in 1983. Russula, Lactarius and Amanita were dominants ant they were widely distributed over the whole area.

        한국산 무당버섯과의 분류학적 연구 (제1보) : Lactarius 속의 분류

        Mushrooms were collected at Mt. Gyeryong, Mt. Chilgab, Mt. Ducyu, Mt. Chiri, Mt Songri and the regions of Chungnam Province for 15 months from July, 1983 to October, 1984. They were classified into 19 species, four varieties and one form of Lactarius. These species were grouped into six sections according to Hesler and Smith. Of these Lactarii, nine species and four varieties appeared to be newly found ones.

        Detection of Fever with Subcutaneously Implanted Thermo-Loggers in Cattle Administered with Lipopolysaccharide

        The aim of this study is to determine whether subcutaneous temperature (ST) was correlated with rectaltemperature (RT) in cattle with inducing artificial fever. In order to determine the correlation between their temperatures,the experiment was performed as follow: Among nine Holstein steers, lipopolysaccharide (LPS) was intravenouslyadministered at a dose of 0.5 μg/kg of body weight to six Holstein steer, then, 6 ml of saline was administrated tothree steers as a control group. After LPS injection, ST was recorded using subcutaneously implanted thermo-loggersensors at 10-min intervals, and RT was measured using a digital thermometer at 0, 1, 2, 3, 4, 8 and 12 h. In steerswith LPS injection, RT was highest at 3 to 4 h and recovered to a pre-challenge temperature at 8-22 h. A similarfluctuation was shown in ST except for an unexpected decrease at 1 h, and a positive correlation between RT andST was observed in LPS-challenged steers (r = 0.497, P = 0.04). This result suggests that ST could be utilized as anindex for early detection of infectious diseases or physiological events.

      • The Fungal Flira of Mt. Gyeryong National Park(Ⅱ)

        To investigate the funfal flora of Mt. Gyeryong National Park, the wild mushrooms were collected in the area from July to October in 1982 and 1983. They were classified into 20 families, 43 genera, 63 species of Basidiomycotion and 3 families, 4 genera, 4 species of Ascomycotiona. Species of Marasmius, Amanita, Agaricus, Boletus, Suillus, Russula, Lastarius, Ramaria and Clavaria were dominantly observed in this area. Lactarius lignyotus var. marginatus, Russla pulchella, Suillus placidus, and Pulveroboletus auriflammeus were reported for the first time in Korea.

        한국산 무당버섯속의 분류학적 연구

        Mushrooms were collected from those grown at Mt. Gyeryong, Mt. Chilgab, Mt. Ducyu, Mt. Chiri, Mt. Songri and at the regions of Chungnam Province during the period from July, 1983 to October, 1984. They were classified into 30 species, two varieties and one form of Russula. These species were grouped into eleven sections. Of the classified species, Russula subnigricans Hongo, R. albonigra (Kromb.) Fries, R.. cyanoxantha (Kromb.) Fries, R. cyanoxantha var. peltereaui Singer and R. compacta Prost et Pk were newly recorded in Korea.

        Pichia pastoris(Methylotrophic Yeast)를 이용한 항균성 양이온 펩타이드의 발현

        Antimicrobial cationic peptides have been received increasing attention as natural antibiotics for their broad spectrum of antimicrobial activities and remarkably low cytotoxicity against normal mammalian cells. Pichia pastoris, a methylotrophic yeast, is an outstanding host for high degree of heterologous gene expression. Four candidates of antimicrobial cation peptides (CPs; Lactofemicin, Magainin, Protegrin-1 and Indolicidin) were expressed using pPIC9K-MPM vector containing AOXI promoter and MPM (modified Promagainin) as an acidic fusion partner. The MPM-CPs were expressed and induced by methanol induction method. The cell extracts, solubilized, were subjected to cyanogen bromide cleavage. SDS-PAGE (16.5% tricine) analysis showed the size of Protegrin-1 at 2.1kDa and of Indolicidin at 1.4kDa. The antimicrobial activity of Protegrin-1 or Indolicidin gene integrated transformant was observed by measuring clearing zones on 1% bacto-peptone agar plate against Gram-negative bacteria Escherichia coli XL-1 blue (10^5 CFU/㎖) or Gram-positive bacteria Staphylococcus aureus (10^5 CFU/㎖).

      • 효모 세포벽으로부터 MANNOPROTEIN 과 β-GLUCAN 의 분리

        본 연구는 효모 세포벽으로부터 manoprotein과 β-glucan을 분리하는 방법을 확립하기 위하여 실시하였다. 효모로부터 분리된 manoprotein과 β-glucan은 동물의 면역기능을 강화시키는 기능성 가축용 면역증강제로 개발하기 위한 목적을 가지고 있다. 따라서 본 연구에서는 이러한 목적을 달성하기 위하여 알칼리 추출 원리에 근거한 Catley의 방법 (Yeast, a practical approach, 1988, edited by Campbell and Duffus, IRL Press)을 변형하여 사용하였으며, 효모의 처리는 파쇄한 것과 파쇄하지 않은 것을 이용하여 manoprotein, β-glucan 및 protein 함량과 순도를 조사하였다. 세포건물로부터 추출된 세포벽 함량은 파쇄한 세포와 파쇄하지 않은 세포에서 각각 15.4%와 28.8%로 나타났으며, mannan의 함량은 파쇄하지 않을 경우 단백질을 제외한 mannan의 함량이 28.5%, 파쇄할 경우 52%로 나타나 mannoprotein은 파쇄된 효모에서 높은 추출율을 보였다. 효모세포벽으로 부터 추출된 mannan 순도에 있어서 파쇄된 효모와 파쇄되지 않은 효모에서 각각 97.6%와 77.6%로 나타나 파쇄할 경우 mannan의 추출 순도를 높이는 효과를 보였다. 그러나 glucan 추출율에 있어서는 파쇄할 경우와 파쇄하지 않은 경우 7.2%와 18.2%로 나타나 파쇄하지 않을 경우 추출율이 높게 나타난 경향을 보였으며, glucan의 순도는 70.8%와 71.7%로 비슷한 결과를 나타내었다. 따라서 본 연구에서 확립한 방법은 알칼리 용해성 glucan을 제외하였으므로 실제로 분리할 수 있는 glucan의 함량은 더 높을 것으로 생각된다. 결론적으로 본 연구에서 확립한 방법은 효모 세포벽으로부터 mannoprotein을 분리할 수 있는 효과적인 방법이라고 생각된다. In this study the extraction method of both mannoprotein and β-glucan was established from yeast cell wall fractions. From this method, both mannoprotein and β -glucan was used as an immune potentiator for domestic animals by activating the overall immune activity. Both mannoprotein and β-glucan were extracted from yeast cell wall fractions or total cell using basically Catley's method (Yeast, a practical approach, 1988, edited by Campbell and Duffus, IRL Press) with minor modification and simplification to be fit into an industrial application. With our modified extraction method. the extractable cell wall contents from mechanically broken cells and unbroken cells were 15.4% and 28.8%, respectively. Mannan contents of cell wall fractions from unbroken and broken cells were 28.5% and 52%, respectively, showing much higher extraction efficiency when the yeast cells were mechanically broken. Mannan contents extracted from cell wall fractions from unbroken and broken cells were 77.4% and 97.6%, respectively. For a higher purity of mannan, thus, it is necessary to break up the yeast cells. On the other hand, the extraction of glucan moiety was obtained 18.2% and 7.2% extraction rates from the cell wall preparation of unbroken and broken yeast cells, respectively, with similar sample purity in both samples (71.7%, 70.8%). In this study, alkali-extractable glucan was not included in the extraction process for the sake of simplicity necessary for the industrial application. However if this step is included in our modified protocol, the glucan yield will be significantly increased. In conclusion, the mannoprotein extraction method from yeast cell wall established in this study may be very effective and applicable in the industrial scale.
