        Pyrophosphate-triggered nanoaggregates with aggregation-induced emission

        <P><B>Abstract</B></P> <P>A novel tetraphenylethene-based probe bearing bis-imidazolium anion donors is herein reported for pyrophosphate anion recognition.       • Sources, distribution, bioavailability, toxicity, and risk assessment of heavy metal(loid)s in complementary medicines

        <P><B>Abstract</B></P> <P>The last few decades have seen the rise of alternative medical approaches including the use of herbal supplements, natural products, and traditional medicines, which are collectively known as ‘Complementary medicines’. However, there are increasing concerns on the safety and health benefits of these medicines. One of the main hazards with the use of complementary medicines is the presence of heavy metal(loid)s such as arsenic (As), cadmium (Cd), lead (Pb), and mercury (Hg). This review deals with the characteristics of complementary medicines in terms of heavy metal(loid)s sources, distribution, bioavailability, toxicity, and human risk assessment. The heavy metal(loid)s in these medicines are derived from uptake by medicinal plants, cross-contamination during processing, and therapeutic input of metal(loid)s. This paper discusses the distribution of heavy metal(loid)s in these medicines, in terms of their nature, concentration, and speciation. The importance of determining bioavailability towards human health risk assessment was emphasized by the need to estimate daily intake of heavy metal(loid)s in complementary medicines. The review ends with selected case studies of heavy metal(loid) toxicity from complementary medicines with specific reference to As, Cd, Pb, and Hg.         A new dibenzocyclooctadiene lignan, intermedin A (1), and a new natural bisabolane sesquiterpenoid, intermedin B (2), were isolated from the aerial parts of Schisandra propinqua var. intermedia. Their structures were elucidated on the basis of extensive spectroscopical analysis.

        Comparative Analysis of Water Stress-Responsive Transcriptomes in Drought-Susceptible and -Tolerant Wheat (Triticum aestivum L.)

        To understand better the mechanisms that regulate the water stress response in wheat, we conducted a comparative analysis of transcript profiles in roots from two wheat genotypes -- drought-tolerant ‘Luohan No. 2’ (LH)and drought-susceptible ‘Chinese Spring’ (CS). In LH roots,3831 transcripts displayed changes in expression of at least two-fold over the well-watered control when drought treatment was applied. Of these, 1593 were induced while 2238 were repressed. Relatively fewer transcripts were drought-responsive in CS; i.e., 1404 transcripts were induced and 1493 were repressed. In common between LH and CS,569 transcripts were induced and 424 transcripts were repressed. In all, 689 transcripts (757 probe sets) identified from LH and 537 transcripts (575 probe sets) from CS were annotated and classified into 10 functional categories. Among those annotated transcripts from LH and CS that had fold-change ratios of at least 4, 92 induced transcripts were common to both, while 23 transcripts were specifically induced in LH. Gene ontology analysis of these induced genes showed highly significant enrichment for multiple terms related to abiotic stimuli, organic acid biosynthesis,and lipid metabolism. This suggests that these gene groups play important roles during the stress response in LH and CS, and might also be responsible for differences in drought tolerance between those genotypes.

        6,7-di-O-acetylsinococuline (FK-3000) induces G <sub>2</sub> /M phase arrest in breast carcinomas through p38 MAPK phosphorylation and CDC25B dephosphorylation

        <P>We evaluated the cytostatic effect of 6,7-di-O-acetylsinococuline (FK-3000) isolated from <I>Stephania delavayi</I> Diels. against breast carcinoma cell lines MDA-MB-231 and MCF-7. FK-3000 suppressed CDC25B phosphorylation directly and indirectly via p38 MAPK phosphorylation. CDC25B dephosphorylation decreased levels of cyclin B and phospho-CDC-2, and ultimately induced cell cycle arrest at the G<SUB>2</SUB>/M phase. The p38 MAPK inhibitor, SB 239063 blocked FK-3000-induced p38 MAPK phosphorylation and nuclear accumulation, but did not completely rescue cell death. Conclusively FK-3000 exerts its antiproliferative effect through two pathways: i) G<SUB>2</SUB>/M cell cycle arrest via downregulation of cyclin B and phospho-CDC2 by p38 MAPK phosphorylation and CDC25B dephosphorylation, and ii) p38 MAPK-independent induction of apoptosis.</P>

        일문전(Stephania delavayi Diels.) 메탄올, 부탄올, 에틸아세테이트 분획물의 항산화 및 세포증식억제 효과

        본 연구에서는 중국에서 급성 위장염에 대한 약제로 사용되어 오던 일문전의 메탄올 추출물을 이용하여 물, 에틸 아세테이트, 부탄올 층으로 분획한 뒤 항산화 활성과 항암활성을 측정하였다. DPPH 라디칼 소거능은 1,000 ㎍/mL 농도에서 부탄올(75.23%)>메탄올(68.11%)>에틸아세테이(63.58%)>물(50.13%) 순으로 나타났으며 환원력의 경우 용매 분획물의 농도가 증가함에 따라 환원력이 증가하는 경향을 나타내었다. DPPH 라디칼 소거능 및 환원력 모두 부탄올 분획층에서 좋은 활성을 나타내었다. 항암활성 측정결과 메탄올 추출물과 부탄올 분획물, 에틸 아세테이트 분획물에서 MDA-MB-231 cell과 MCF-7 cell에 대한 세포증식억제효과를 나타내었다. 이상의 결과에서 일문전의 용매 분획물은 항암 후보물질로의 가능성이 있을 것으로 사료된다. Stephania delavayi Diels. (S. delavayi Diels.) has been used as a drug for pain-relieving and acute gastroenteritis treatment in China. Because the major therapeutic mechanism of anti-inflammatory drug is to inhibit the cyclooxygenase (COX)-2 and because COX-2 proteins inhibit apoptosis, COX-2 inhibitor has been thought as the anticancer drug candidate. For this reason, we examined S. delavayi Diels. as an anticancer drug. S. delavayi Diels. was fractionated with methanol and then partitioned with ethyl acetate, n-butanol and water. The antioxidant activity was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and reducing power. DPPH radical scavenging activities of the crude fractions at the concentration of 1,000 ㎍/mL were 75.23% (n-butanol), 68.11% (methanol), 63.58% (ethyl acetate), and 50.13% (water). The reducing power increased according to the concentration in dose-dependent manner. Also, when the antiproliferation effects of each fraction against human breast cancer cell-lines MDA-MB-231 and MCF-7 were examined, methanol extract, n-butanol fraction and ethyl acetate fraction exhibited cell proliferative inhibition effects in both cell-lines whereas water fraction did not. Among the crude fractions, the n-butanol fraction exhibited the most potent anti-proliferation effect. In conclusion, fractions from S. delavayi Diels. are promising anticancer drug candidates.

        Isolation of Chemical Constituents from the Aerial Parts of Verbascum thapsus and Their Antiangiogenic and Antiproliferative Activities

        Phytochemical investigation of Verbascum thapsus led to the isolation and identification of one new iridoid compound named verbathasin A, along with ten known compounds. The structure and relative stereochemistry of verbathasin A were elucidated by analysis of spectroscopic data. All the isolates except 10-deoxyeucommiol and ajugol were tested for antiangiogenic and antiproliferative activities, and compounds luteolin and 3-O-fucopyranosylsaikogenin F showed promising antiproliferative activities, with an obvious effect of inducing apoptosis of A549 lung cancer cells.

        Phylogeny and molecular evolution of the DMC1 gene within the StH genome species in Triticeae (Poaceae)

        To estimate the phylogeny and molecular evolution of a single-copy nuclear disrupted meiotic cDNA (DMC1) gene within the StH genome species, two DMC1 homoeologous sequences were isolated from nearly all the sampled StH genome species and were analyzed with those from seven diploid taxa representing the St and H genomes in Triticeae. Sequence diversity patterns and genealogical analysis suggested that (1) there is a close relationship among North American StH genome species;(2) the DMC1 gene sequences of the StH genome species from North America and Eurasia are evolutionarily distinct;(3) the StH genome polyploids have higher levels of sequence diversity in the St genome homoeolog than the H genome homoeolog;(4) the DMC1 sequence may evolve faster in the polyploid species than in the diploids; (5) high dN and dN/dS values in the St genome within polyploid species could be caused by low selective constraints or AT-biased mutation pressure. Our result provides some insight on evolutionary dynamics of duplicate DMC1 gene, the polyploidization events and phylogeny of the StH genome species.

      • DEPTOR Expression Negatively Correlates with mTORC1 Activity and Tumor Progression in Colorectal Cancer

        The mammalian target of rapamycin (mTOR) signaling pathway is upregulated in the pathogenesis of many cancers, including colorectal cancer (CRC). DEPTOR is an mTOR inhibitor whose expression is negatively regulated by mTOR. However, the role of DEPTOR in the development of CRC is not known. The aim of this study was to investigate the expression of DEPTOR and mTORC1 activity (P-S6) in a subset of CRC patients and determine their relation to tumor differentiation, invasion, nodal metastasis and disease-free survival. Here, Immunohistochemical expression of P-S6 (S235/236) and DEPTOR were evaluated in 1.5 mm tumor cores from 90 CRC patients and in 90 samples of adjacent normal mucosa by tissue microarray. The expression of P-S6 (S235/236) was upregulated in CRC, with the positive rate of P-S6 (S235/236) in CRC (63.3%) significantly higher than that in control tissues (36.7%, 30%) (p<0.05). P-S6 (S235/236) also correlated with high tumor histologic grade (p=0.002), and positive nodal metastasis (p=0.002). In contrast, the expression level of DEPTOR was correlated with low tumor histological grade (p=0.006), and negative nodal metastasis (p=0.001). Interestingly, P-S6 (S235/236) expression showed a significant negative association with the expression of DEPTOR in CRC (p=0.011, R= -0.279). However, upregulation of P-S6 (S235/236) (p=0.693) and downregulation of DEPTOR (p=0.331) in CRC were not significantly associated with overall survival. Thus, we conclude that expression of DEPTOR negatively correlates with mTORC1 activity and tumor progression in CRC. DEPTOR is a potential marker for prognostic evaluation and a target for the treatment of CRC.
