        An integrated dendrite-free zinc metal electrode for corrosion inhibition in aqueous system

        Zinc ion batteries have gotten increasing attention as a potential candidate for lithium-ion batteries, due totheir high specific capacity (820 mAh·g1), energy density, and safety. Inevitably, dendrite and corrosion create sometrouble for this system. Herein, an integrated Zn electrode coated by Zn-Al metal oxides prepared by a simple spincoatingmethod was utilized to increase the rechargeability for aqueous zinc ion batteries. By coating the Zn anodewith an artificial electrolyte interface, the wettability of Zn anodes was improved and impedance was reduced. Thecoating suppressed not only the appearance of dendrite but also the formation of corrosion products. The symmetricalcells with coating have a low overpotential (43mV) and an excellent life span. Meanwhile, the applied full batteriesexhibit an improved capacity retention rate (86.67% after 120 cycles), great rate performance and low apparent activationenergy (24.6 KJ·mol1). The simple production methods and superior corrosion suppression effects provide newideas for the anode protection of aqueous system batteries.

        Albumin-to-Fibrinogen Ratio as an Independent Prognostic Parameter in Untreated Chronic Lymphocytic Leukemia: A Retrospective Study of 191 Cases

        Purpose Chronic lymphocytic leukemia (CLL) is one of the most frequent type of B-cell chronic lymphoproliferative disorders and chronic inflammation takes part in the development of CLL. However, there has been no valid immune biomarker to predict the prognosis of untreated CLL patients. Materials and Methods In this retrospective study, we analyzed the clinical correlations and prognostic value of albumin-to-fibrinogen ratio (AFR) detected at diagnosis in 191 CLL patients. Results The cut-off value of AFR was 9.7 calculated by X-tile. Patients who were more than 65 years old were often accompanied by low level of AFR (p < 0.001). Survival analysis showed that patients with low level of AFR had shorter overall survival (OS) than patients with high level of AFR (p < 0.001). Multivariate analysis illustrated that AFR had a negative impact on OS (p=0.003) and was independent of parameters involved in CLL international prognostic index and other prognostic markers such as CD38 and ZAP-70. Conclusion These data provide a comprehensive view of AFR and shows that AFR at diagnosis is an adverse prognostic factor in untreated CLL patients.

        Effects of the silencing of CmMET1 by RNA interference in chrysanthemum (Chrysanthemum morifolium)

        DNA methylation is an epigenetic modification involving many biological processes. It is known that epigenetic mechanisms such as cytosine methylation play a pivotal role in regulating plant development. In this current study, a conservative domainspliced hairpin RNA (ihpRNA) plant expression vector aimed at gene of DNA METHYLTRANSFERASE1 (CmMET1) has been constructed. Transgenic chrysanthemum materials (Zijingling) were obtained by Agrobacterium-mediated transformation with expression vectors. Transgenic plants were used as rootstock, grafted onto non-transgenic the scions [Guoqinghong (GQH) and Huanshuijinqiu (HSJQ)], which silencing CmMET1 gene, and exhibited the early flower phenotypes. A highperformance liquid chromatography analysis indicated that the decrease of CmMET1 expression decreased the methylation level of genomic DNA. Similarly, a quantitative real-time polymerase chain reaction analysis revealed that CmMET1 expression levels decreased in transgenic chrysanthemum plants and in the scions grafted onto transgenic plants. This decrease of CmMET1 expression upregulated the expression of the methyltransferase gene, METHYLTRANSFERASE2 (CmDRM2), but downregulated the expression of the demethylating enzyme gene, DEMETER (CmDME), while the CHROMOMETHYLASEA (CmCMT3) expression level remained low and could be almost undetectable. Among the CmFT-likes genes that affect flowering time, CmFTL1 expression was downregulated, as well as CmFTL2 and CmFTL3 expression levels which were upregulated. Our data indicated that silencing CmMET1 could decrease plant height, change the phenotype of chrysanthemum, and promote earlier flowering. The transgenic plants bloomed 8 days earlier. GQH and HSJQ scions grafted onto transgenic plants were 12 and 9 days earlier than the scions grafted onto non-transgenic plants, respectively. Overall, the results have some meanings for promoting the flowering of chrysanthemum scion varieties using genetic modified rootstock.

        EBV-miR-BHRF1-1 Targets p53 Gene: Potential Role in Epstein-Barr Virus Associated Chronic Lymphocytic Leukemia

        Purpose The purpose of this study was to investigate the prognostic impact of Epstein-Barr virus (EBV)–microRNA (miRNA, miR)-BHRF1-1 with chronic lymphocytic leukemia (CLL) as well as role of EBV-miR-BHRF1-1 in p53 gene. Materials and Methods Quantitative reverse transcription–polymerase chain reaction and western blotting were used to quantify EBV-miR-BHRF1-1 and p53 expression in cultured CLL. Results p53 aberration was associated with the higher expression level of EBV-miR-BHRF1-1 (p < 0.001) which was also an independent prognostic marker for overall survival (p=0.028; hazard ratio, 5.335; 95% confidence interval, 1.193 to 23.846) in 97 newly-diagnosed CLL patients after adjusted with International Prognostic Index for patients with CLL. We identified EBV-miR-BHRF1-1 as a viral miRNA regulator of p53. EBV-miR-BHRF1-1 repressed luciferase reporter activity by specific interaction with the seed region within the p53 3- untranslated region. Discordance of p53 messenger RNA and protein expression was associated with high EBV-miR-BHRF1-1 levels in CLL patients and cell lines. EBV-miR-BHRF1- 1 inhibition upregulated p53 protein expression, induced cell cycle arrest and apoptosis and decreased cell proliferation in cell lines. EBV-miR-BHRF1-1 mimics downregulated p53 protein expression, decreased cell cycle arrest and apoptosis, and induced cell proliferation in cell lines. Conclusion This study supported the role of EBV-miR-BHRF1-1 in p53 regulation in vitro. Our results support the potential of EBV-miR-BHRF1-1 as a therapeutic target in EBV-associated CLL with p53 gene aberration.

        Identification, fine mapping and characterization of Rht-dp, a recessive wheat dwarfing (reduced height) gene derived from Triticum polonicum

        Semi-dwarfism is an agronomically important trait in breeding for resistance to damage by wind and rain (lodging resistance)and for stable high yields. Dwarf Polish wheat (Triticum polonicum L., 2n = 4x = 28, AABB AS304) is a potential donor of dwarfing and other traits for common wheat improvement. A genetic analysis using an F2 population derived from a cross of AS304 and tall cultivar AS302 and derived F2:3 lines indicated that AS304 carries a recessive dwarfing gene, temporarily designated Rht-dp. Molecular markers and bulked segregant analysis were used to characterize and map the gene. Eight polymorphic SSR markers (Xwmc511, Xgwm495, Xgwm 113, Xgwm192, Xgpw7026, Xgpw3017, Xgpw1108 and Xgpw7521) on chromosome arm 4BS and two AFLP markers (M8/E5 and M4/E3) were mapped relative to the dwarfing locus. The closest linked markers, Xgpw3017 and M8/E5 at 0.5 and 3.5 cM, respectively, from Rht-dp will enable its marker assisted transfer to wheat breeding populations. Allelic tests indicated that Rht-dp was allelic to Rht-B1b; hence it may be an alternative allele at the Rht-B1 locus.

      • Decreased Expression of FADS1 Predicts a Poor Prognosis in Patients with Esophageal Squamous Cell Carcinoma

        FADS1 (fatty acid desaturase 1) plays a crucial role in fatty acid metabolism, and it was recently reported to be involved in tumorigenesis. However, the role of FADS1 expression in esophageal squamous cell carcinoma (ESCC) remains unknown. In the current study, we investigated the expression and clinical pathologic and prognostic significance of FADS1 in ESCC. Immunohistochemical analyses revealed that 58.2% (146/251) of the ESCC tissues had low levels of FADS1 expression, whereas 41.8% (105/251) exhibited high levels of FADS1 expression. In positive cases, FADS1 expression was detected in the cytoplasm of cells. Correlation analyses demonstrated that FADS1 expression was significantly correlated with tumor location (p=0.025) but not with age, gender, histological grade, tumor status, nodal status or TNM staging. Furthermore, patients with tumors expressing high levels of FADS1had a longer disease-free survival time (p<0.001) and overall survival time (p <0.001). Univariate and multivariate analyses revealed that, along with nodal status, FADS1 expression was an independent and significant predictive factor (p<0.001). In conclusion, our study suggested that FADS1 might be a valuable biomarker and potential therapeutic target for ESCC.

      • Effect of Microstructure on Mechanical Property of Carbon Steel/Strainless Steel Rolling Clad Plate

        The bonding interface microstructure and mechanical property of clad plate with different hot rolling schedules were researched. The results show that the carbide layer thickness of bonding interface increases firstly and then decreases with increasing reduction ratio. The carbide layer of bonding interface is the major factor relating to tensile strength, so the experimental values of clad plate are slightly higher than the calculated ones. Moreover, the bending strength of clad plate is mainly influenced by the thickness ratio and the carbide quantity of SS, while the carbide layer thickness of bonding interface is an important factor affects the bending strength.

      • Supervisory Optimization of the MGT-CCHP System Using Model Predictive Control

        This paper proposes a supervisory optimal control structure for the micro-gas-turbine based combined cooling heating and power (MGT-CCHP) system. In the upper layer of the structure, a dynamic optimal reference governor is developed to calculate the optimal operating points according to the given economic performance indexes, and in the lower layer of the structure, model predictive control is utilized to track the given operating points, so that a dynamic optimality can be achieved under the input-output constraints during the optimization. Moreover, a disturbance term is introduced in the model to lump the effect of unmeasured disturbances and plant behavior variations, thus, their influences on the supervisory optimization can be removed. The advantages and effectiveness of the proposed method are demonstrated through the simulations on an 80kW MGT-CCHP simulator.

        Sp1-Induced SETDB1 Overexpression Transcriptionally Inhibits HPGD in a β-Catenin-Dependent Manner and Promotes the Proliferation and Metastasis of Gastric Cancer

        Purpose: Gastric cancer (GC) has high morbidity and mortality, the cure rate of surgical treatment and drug chemotherapy is not ideal. Therefore, development of new treatment strategies is necessary. We aimed to identify the mechanism underlying Sp1 regulation of GC progression. Methods and Methods: The levels of Sp1, β-catenin, SET domain bifurcated 1 (SETDB1), and 15-hydroxyprostaglandin dehydrogenase (HPGD) were detected by quantitative reverse transcription polymerase chain reaction and western blot analysis. The targets of SETDB1 were predicted by AnimalTFDB, and dual-luciferase reporter assay was used for confirming the combination of Sp1, β-catenin, and SETDB1. HGC27 or AGS cells (1×10<sup>6</sup> cells/mouse) were injected into mice via the caudal vein for GC model establishment. The level of Ki67 was detected using immunohistochemistry, and hematoxylin and eosin staining was performed for evaluating tumor metastasis in mice with GC. Results: HPGD was inhibited, while the protein levels of Sp1, β-catenin, and SETDB1 were up-regulated in GC tissues and cell lines. HPGD overexpression or SETDB1 silencing inhibited the proliferation, invasion, and migration of GC cells, and Sp1 regulated the proliferation, invasion, and migration of GC cells in a β-catenin-dependent manner. Furthermore, HPGD served as a target of SETDB1, and it was negatively regulated by SETDB1; additionally, Sp1 and β-catenin bound to the SETDB1 promoter and negatively regulated HPGD expression. We proved that Sp1 regulated GC progression via the SETDB1/HPGD axis. Conclusions: Our findings revealed that Sp1 transcriptionally inhibited HPGD via SETDB1 in a β-catenin-dependent manner and promoted the proliferation and metastasis of GC cells.
