      • Prognostic Role of Hepatoma-derived Growth Factor in Solid Tumors of Eastern Asia: a Systematic Review and Meta-Analysis

        Hepatoma-derived growth factor (HDGF) is a novel jack-of-all-trades in cancer. Here we quantify the prognostic impact of this biomarker and assess how consistent is its expression in solid tumors. A comprehensive search strategy was used to search relevant literature updated on October 3, 2014 in PubMed, EMBASE and WEB of Science. Correlations between HDGF expression and clinicopathological features or cancer prognosis was analyzed. All pooled HRs or ORs were derived from random-effects models. Twenty-six studies, primarily in Eastern Asia, covering 2,803 patients were included in the analysis, all of them published during the past decade. We found that HDGF overexpression was significantly associated with overall survival (OS) ($HR_{OS}=2.35$, 95%CI=2.04-2.71, p<0.001) and disease free survival (DFS) ($HR_{DFS}=2.25$, 95%CI =1.81-2.79, p<0.001) in solid tumors, especially in non-small cell lung cancer, hepatocellular carcinoma and cholangiocarcinoma (CCA). Moreover, multivariate survival analysis showed that HDGF overexpression was an independent predictor of poor prognosis ($HR_{OS}=2.41$, 95%CI: 2.02-2.81, p<0.001; $HR_{DFS}=2.39$, 95%CI: 1.77-3.24, p<0.001). In addition, HDGF overexpression was significantly associated with tumor category (T3-4 versus T1-2, OR=2.12, 95%CI: 1.17-3.83, p=0.013) and lymph node status (N+ versus N-, OR=2.37, 95%CI: 1.31-4.29, p=0.03) in CCA. This study provides a comprehensive examination of the literature available on the association of HDGF overexpression with OS, DFS and some clinicopathological features in solid tumors. Meta-analysis results provide evidence that HDGF may be a new indicator of poor cancer prognosis. Considering the limitations of the eligible studies, other large-scale prospective trials must be conducted to clarify the prognostic value of HDGF in predicting cancer survival.

      • MiR-21 Upregulation Induced by Promoter Zone Histone Acetylation is Associated with Chemoresistance to Gemcitabine and Enhanced Malignancy of Pancreatic Cancer Cells

        Background and Aims: MicroRNA-21 (miR-21) is reported to be overexpressed and to contribute to proliferation, apoptosis and gemcitabine resistance in pancreatic ductal adenocarcinomas (PDACs). The aims of this study were to explore regulation of miR-21 expression by epigenetic change and its impact on chemoresistance and malignant properties of of pancreatic cancer. Materials and methods: We retrospectively collected 41 cases of advanced pancreatic cancer patients who were sensitive or resistant to gemcitabine and assessed levels of serum circulating miR-21 for correlation with cytotoxic activity. Histone acetylation in the miR-21 promoter was also studied in gemcitabine-sensitive and gemcitabine-resistant PDAC cells. Gemcitabine-resistant HPAC and PANC-1 cells were transfected with pre-miR-21 precursors (pre-miR-21) and antisense oligonucleotides (anti-miR-21), and were treated with TSA. Finally, invasion and metastasis assays were performed and alteration in mir-21, PTEN, AKT and pAKT level was evaluated in these cells. Results: Serum miR-21 levels were increased in gemcitabine-resistant PDAC patients compared with gemcitabine-sensitive subjects. The miR-21 levels were increased in 6 PDAC cells treated with gemcitabine significantly, associated with 50% inhibitory concentrations ($IC_{50}s$). Histone acetylation levels at miR-21 promoter were increased in PDAC cells after treatment with gemcitabine. Enhanced invasion and metastasis, increased miR-21 expression, decreased PTEN, elevated pAKT level were demonstrated in gemcitabine-resistant HPAC and PANC-1 cells. Pre-miR-21 transfection or TSA treatment further increased invasion and metastasis ability, decreased PTEN, and elevated pAKT levels in these two lines. In contrast, anti-miR-21 transfection could reverse invasion and metastasis, and PTEN and pAKT expressions induced by gemcitabine. Conclusions: MiR-21 upregulation induced by histone acetylation in the promoter zone is associated with chemoresistance to gemcitabine and enhanced malignant potential in pancreatic cancer cells.

      • Ultrasound Targeted Microbubble Destruction for Novel Dual Targeting of HSP72 and HSC70 in Prostate Cancer

        The aim was to determine whether ultrasound targeted microbubble destruction (UTMD) promotes dual targeting of HSP72 and HSC70 for therapy of castration-resistant prostate cancer (CRPC), to improve the specific and efficient delivery of siRNA, to induce tumor cell specific apoptosis, and to find new therapeutic targets specific of CRPC.VCaP cells were transfected with siRNA oligonucleotides. HSP70, HSP90 and cleaved caspase-3 expression were determined by real-time quantitative polymerase chain reaction and Western blotting. Apoptosis and transfection efficiency were assessed by flow cytometry. Cell viability assays were used to evaluate safety. We found HSP72, HSC70 and HSP90 expression to be absent or weak in normal prostate epithelial cells (RWPE-1), but uniformly strong in prostate cancerous cells (VCaP). UTMD combined with dual targeting of HSP72 and HSC70 siRNA improve the efficiency of transfection, cell uptake of siRNA, downregulation of HSP70 and HSP90 expression in VCaP cells at the mRNA and protein level, and induction of extensive tumor-specific apoptosis. Cell counting kit-8 assays showed decreased cellular viability in the HSP72/HSC70-siRNA silenced group. These results suggest that the combination of UTMD with dual targeting HSP70 therapy for PCa may be most efficacious, providng a novel, reliable, non-invasive, safe targeted approach to improve the specific and efficient delivery of siRNA, and achieve maximal effects.

      • Therapeutic Regimens and Prognostic Factors of Brain Metastatic Cancers

        Objective: This work aims to investigate the therapeutic regimen of brain metastatic cancers and the relationship between clinical features and prognosis. Methods: Clinical data of 184 patients with brain metastatic cancers were collected and analysed for the relationship between survival time and age, gender, primary diseases, quantity of brain metastatic foci, their position, extra cranial lesions, and therapeutic regimens. Results: The average age of onset was 59.1 years old. The median survival time (MST) was 15.0 months, and the patients with breast cancer as the primary disease had the longest survival time. Females had a longer survival time than males. Patients with meningeal metastasis had extremely short survival time. Those with less than 3 brain metastatic foci survived longer than patients with more than 3. The MST of patients receiving radiotherapy only and the patients receiving chemotherapy only were all 10.0 months while the MST of patients receiving combination therapy was 16.0 months. Multiple COX regression analysis demonstrated that gender, primary diseases, and quantity of brain metastatic foci were independent prognostic factors for brain metastatic cancers. Conclusions: Chemotherapy is as important as radiotherapy in the treatment of brain metastatic cancer. Combination therapy is the best treatment mode. Male gender, brain metastatic cancers originating in the gastrointestinal tract, more than 3 metastatic foci, and involvement of meninges indicate a worse prognosis.

      • Isolation of adipose-derived stem cells by using a subfractionation culturing method

        <P><B><I>Objective:</I></B> Adipose-derived stem cells (ASCs) isolated from subcutaneous adipose tissue have been tested in clinical trials. However, ASCs isolated by enzyme digestion and centrifugation are heterogeneous and exhibit wide variation in regenerative potential and clinical outcomes. Therefore, we developed a new method for isolating clonal ASCs (cASCs) that does not use enzyme digestion or centrifugation steps.</P><P><B><I>Research design and methods:</I></B> In addition to cell surface markers and differentiation potential, we compared the mitogenic, paracrine and hair growth-promoting effects of ASCs isolated by the gradient centrifugation method (GCM) or by the new subfractionation culturing method (SCM).</P><P><B><I>Results:</I></B> We selected three cASCs isolated by SCM that showed high rates of proliferation. The cell surface markers expressed by ASCs isolated by GCM or SCM were very similar, and SCM-isolated ASCs could potentially differentiate into different cell lineages. However, cASC lines exhibited better mitogenic and paracrine effects than ASCs isolated by GCM. The expression of Diras3, Myb, Cdca7, Mki67, Rrm2, Cdk1 and Ccna2, which may play a key role in cASC proliferation, was upregulated in cASCs. In addition, cASCs exhibited enhanced hair growth-promoting effects in dermal papilla cells and animal experiments.</P><P><B><I>Conclusions:</I></B> SCM generates a highly homogeneous population of ASCs via a simple and effective procedure that can be used in therapeutic settings.</P>

      • Establishment and Characterization of MTDH Knockdown by Artificial Micro RNA Interference - Functions as a Potential Tumor Suppressor in Breast Cancer

        Background: Considerable evidence suggests that metadherin (MTDH) is a potentially crucial mediator of tumor malignancy and an important therapeutic target for simultaneously enhancing chemotherapy efficacy and reducing metastasis risk. Inhibition of MTDH expression by RNA interference has been shown in several previous research, but silencing MTDH expression by microRNA (miRNA) interference in breast cancer has not been established. In the present study, we investigated the role of MTDH-miRNA in down-regulation of proliferation, motility and migration of breast carcinoma cells. Methods: Expression vectors of recombinant plasmids expressing artificial MTDH miRNA were constructed and transfected to knockdown MTDH expression in MDA-MB-231 breast cancer cells. Expression of MTDH mRNA and protein was detected by RT-PCR and Western blot, respectively. MTT assays were conducted to determine proliferation, and wound healing assays and transwell migration experiments for cell motility and migration. Results: Transfection of recombinant a plasmid of pcDNA-MTDH-miR-4 significantly suppressed the MTDH mRNA and protein levels more than 69% in MDA-MB-231 breast cancer cells. This knockdown significantly inhibited proliferation, motility and migration as compared with controls. Conclusions: MTDH-miRNA may play an important role in down-regulating proliferation, motility and migration in breast cancer cells, and should be considered as a potential small molecule inhibitor therapeutic targeting strategy for the future.

        Optimization of Brazing Conditions for OFHC Cu and ASTM A501 Low Carbon Steel by Taguchi Method

        This paper concerns the optimization of brazing conditions for joining the two dissimilar materials Oxygen-free high conductivity (OFHC) copper and ASTM A501 low carbon steel usually used for an Oil-Separator of an air-conditioner using Ag-based(BAg) alloy as a brazing filler metal (BFM). A mixture of 70% $N_2$ and 30% $H_2$ gases was used to prevent oxidation of the joints during furnace brazing process. Brazing joint clearance, length, temperature, and time were selected as design parameters that have significant effect on the bonding strength of a brazed joint. Taguchi method was used as a statistical technique for design of experiment (DOE) in an attempt to optimize brazing conditions in terms of shear strength. $L_9\;(3^4)$ orthogonal array was designed for conducting the experiments. The relative influence of design parameters and their interaction on the response were also discussed. The experimental results verified that the brazing conditions predicted in this study by Taguchi method could produce the brazed joint between OFHC copper and ASTM A501 low carbon steel with maximum shear strength.

      • Wear Mechanisms of Al₂O<SUB>3f</SUB>/SiC<SUB>p</SUB> Reinforced Al Metal Matrix Composites under Dry and Lubricant Wear Conditions at Room Temperature

        This study investigates the room temperature sliding wear mechanisms of Al₂O₃<SUB>f</SUB> and SiC<SUB>p</SUB> reinforced Al metal matrix composites (MMCs). The effects of fiber orientation and fiber to particle hybrid ratio on the wear behaviors at both the dry and lubricant conditions are discussed in details. The MMC specimens were fabricated by squeeze casting method with different hybrid ratios and fiber orientations. The tests were carried out by a pin-on-disk friction and wear tester at a constant sliding speed of 0.36 ㎧ (570 rpm) against a steel counter disk. The results of dry sliding tests showed that the F20P0 (unhybrid) MMCs with normal (N) orientation of fibers had better wear behaviors than those with planer-random (PR) orientation of fibers. However, for hybrid MMCs, the wear behavior was better for PR-orientation of fibers. On the contrary, the lubricant wear behaviors of F20P0 MMCs with PR-orientation of fibers were superior to those with N-orientation of fibers and the reverse lubricant wear behaviors were observed for hybrid MMCs. Finally, the scanning electron microscope (SEM) images of the wear surface and debris were used for investigating the modes of wear.

        Effects of inoculating different mycorrhizal fungi on rhizosphere soil fungi and nutrient uptake of blueberry

        The aim of this study was to explore the eff ects of inoculating with diff erent fungi on the structural characteristics of soil fungal communities and the nutrient absorption of blueberry plants. The fungi DSE ( Phialocephala fortinii ), SS ( Penicillium pinophilum ), ZB ( Cladosporium cladosporioides ), QMK ( Chaetomium globosum ), and LZ ( Schizophyllum commune ) were selected for this study. The soil fungal diversity and community structure in the blueberry rhizosphere were compared after inoculation with these diff erent mycorrhizal fungi. The fungal diversity in the blueberry rhizosphere soil was signifi cantly higher after inoculation with DSE, QMK, and ZB than after inoculation with SS and LZ. The dominant class, order, family, and genus of soil fungi were Eurotiomycetes (25.49–36.91%), Eurotiales (24.78–35.00%), Aspergillaceae (24.47–34.74%), and Penicllium (24.34–34.63%), respectively. Among all the fungal treatments, DSE led to the highest abundance of dominant classes, orders, families, and genera. Inoculation with SS, LZ, and DSE increased the contents of nitrogen, phosphorus, and potassium in blueberry plants. The uptake of nitrogen, phosphorus, and potassium were highest in DSE-inoculated plants. The nitrogen content was signifi cantly higher in ZB-inoculated plants than in the control. A TOPSIS analysis showed that among the fi ve fungi, DSE was the best strain to improve the diversity of the blueberry rhizosphere fungal community and promote nutrient uptake by blueberry plants, followed by LZ and ZB. These fi ndings provide scientifi c data for the develop ment and utilization of soil microbial resources, and provide useful information for improving the cultivation and production of blueberries.
