Reactive aqueous two phase system(ATPS) using hydrophilic polymer and dextran was applied to recover the Copolymer consists of polyethlene oxide(PEO) and maleic acid anhdride (MA) was used to modify the cellulase. BY controlling the ratio of ethylene oxide, hydrophilic property of copolymer was controlled, and the MA functional group of copolymer covalently coupled with the amino groups of cellulase. The two phase system was mixed with dextran and copolymers with molecular weight of 505,000. The modified moved to the down phase,dextran As a result, most enzymes, approximately over 70%, moved to copolymer Therefore, reactive aqueous two phase partition by modification of cellulase is very effective to separate enzyme from the fermentation broth.

We describe the purification of native and modified enzymes using a reactive aqueous two-phase systems. The two-phase systems consist of a hydrophilic AKM copolymer and dextran. Cellulase was modified with the AKM copolymer; the degree of modification is directly proportional to the number of maleic acid anhydride groups present in the AKM copolymer. Partitioning of the native and modified cellulases was also investigated in the aqueous two-phase AKM copolymer/dextran systems; the partition coefficient increases as the degree of modification in creases. The AKM-053 1 copolymer is enzyme selective; the modified enzyme always shows separation that is better than that of the native enzyme.

Lipase was modified with hydrophilic and hydrophobic copolymers and, was reacted to the several substrates. In general, modified lipase was better than native one as it resolved the substrate. As lipase was modified with strongly hydrophobic copolymers, degree of modification decreased and, relative activity increased.

이 연구는 물리개념의 특성과 학습자의 정의적 특성을 고려한 인지갈등 수업전략에 관한 모형과 이 모형에 근거한 인지갈등과제 개발 연구의 기초 단계로서, 연구참여 교사들의 인지갈등 수업전략에 관한 인식과 현장적용 실태 및 그 문제점을 알아보는 데 목적이 있다. 대학원에서 과학교육을 전공하는 40명의 현직 교사를 대상으로 인지갈등 수업전략의 인식, 활용 실태, 그 문제점 및 해결방안을 조사한 후 면담을 이용하여 자료를 수집하고 응답결과를 분석하였다. 결론적으로 참여 과학교사들은 인지갈등 수업전략의 필요성과 학습효과를 높게 평가하고 있었으나, 그 전략의 적용에는 다양한 문제점을 느끼고 있었다. 그 중 교사요인과 교수·학습자료 요인의 문제를 해결하기 위해서는 교사들이 그 학습전략을 체계화하고 구체적으로 적용할 수 있도록 돕는 실천 중심의 교과교육과정과 갈등자료의 개발 및 공유 시스템 구축이 필요하다. 또한 질적 접근을 통해서 학습자의 특성과 과학개념의 특성을 고려한 구체적인 인지갈등 전략 모형과 과제 개발이 연구되어야 한다. This research is a foundational study for developing a cognitive conflict learning strategy model considering characteristics of physics concepts and students' affective variables. The purpose of this study is to investigate the science teachers' understanding of the cognitive conflict learning strategy and the practice of the learning strategy in science classes. The total of 40 science teachers who had taken the lectures of 'science pedagogy' and 'science learning and instruction: theory into practice' in graduate school were selected for this study. The participants were asked to answer a questionnaire and participated interviews. Data have been collected by the questionnaire and the interviews. The conclusion can be summarized as follows. First, Most science teachers recognized that the cognitive conflict learning strategy is important and has a good effect on students' conceptual changes when learning science. However, there are many kinds of difficulties in their applying it to classroom context. Among them, to solve the teacher's factor and the lack of teaching/learning materials, we need to develop not only the science pedagogical curriculum composed of practice and application related to concrete conceptual change learning strategy, but the systems to spread the developed materials. Second, qualitative approach seemed to be necessary to get further developing the cognitive conflict learning strategy model based the characteristics of physics concepts and student's affective variables.
THE DEVELOPMENT OF COURSE ON THE PARCTICE OF COGNITIVE CONFLIT STRATEGY FOR PHYSICS TEACHERS

The purpose of this study was to develop a course on the practice of cognitive conflict strategy for physics teachers. We designed a graduate education course in which twenty-one physics teachers enrolled. During the course, each teacher developed two cognitive conflict materials for demonstration and the teaching plans to apply those materials to instruction. For the improvement of this course, the participants were asked to answer questionnaire and participated interviews and discussion about cognitive conflict strategy. Data have been collected by the questionnaire, the interviews, videotapes, and observation documents. Most physics teachers recognised that the cognitive conflict strategy is important and has a good effect on students' conceptual changes when learning science. However, there are some difficulties in their applying it to classroom context and developing the cognitive conflict materials. This article suggested the implications for the science pedagogical curriculum composed of development and application of the cognitive conflict materials
