        Temperature field and demagnetization analysis of in‑wheel motors based on magneto‑thermal two‑way coupling

        Due to the narrow working space of an in-wheel motor, the heat generated by the motor loss is difficult to dissipate. This makes it easier for the in-wheel motor to demagnetize the permanent magnet due to the mega-temperature, which affects the output efficiency. To solve this problem, an external rotor hub motor is studied. First, in accordance with the theory of magnetic field modulation, the in-wheel motor to be studied is designed. By analyzing the electromagnetic characteristics of the motor, the correctness of the motor design is verified, and the losses of the motor under different working conditions are calculated. To acquire a more rigorous temperature increase record, the magnetic-thermal bidirectional coupling method is utilized to analyze the temperature field under different load conditions. The mechanism of the demagnetization of permanent magnets is analyzed, and demagnetization at different temperatures is obtained by magneto-thermal two-way coupling. Research shows that when the motor is overloaded for a long time, the temperature can reach a maximum of 220 °C. At this temperature, the permanent magnet undergoes irreversible demagnetization, which results in a 93.44% decrease in torque. Finally, temperature increase tests of a permanent magnet motor are carried out to verify the validity of the magneto-thermal two-way coupling analysis.

        Map-based cloning and functional analysis of the chromogen gene C in rice (Oryza sativa L.)

        The chromogen gene C is critical for anthocyanin regulation in rice, and apiculus color is an important agronomic trait in selective breeding and variety purification. Mapbased cloning and in-depth functional analysis of the C gene will be useful for understanding the molecular mechanism of anthocyanin biosynthesis and for rice breeding. Japonica landrace Lijiangxintuanheigu (LTH) has red apiculi and purple stigmas. Genetic analysis showed that red apiculus and purple stigma in LTH co-segregated indicating control by a single dominant gene, or by two completely linked genes. Using 1,851 recessive individuals from two F2 populations, the target gene OsC was delimited to a 70.8 kb interval on chromosome 6 that contains the rice homologue of the maize anthocyanin regulatory gene C1. When the entire OsC gene and its full-length cDNA cloned from LTH were transformed into japonica cultivar Kitaake with colorless apiculi and stigmas all positive transformants had red apiculi but non-colored stigmas, validating that OsC alone was responsible for the apiculus color and represented the functional C gene. OsC was constitutively expressed in all tissues examined, with strongest expression in leaf blades. These results set a foundation to clarify the regulatory mechanisms of OsC in the anthocyanin biosynthetic pathway.

        FLOURY ENDOSPERM12 Encoding Alanine Aminotransferase 1 Regulates Carbon and Nitrogen Metabolism in Rice

        Starch is a major storage substance in cerealgrains, and starch biosynthesis is a complex process. In orderto elucidate regulation of the starch biosynthesis pathway, wescreened a series of rice (Oryza sativa L.) endospermmutants. In this study, we identified a floury white-coreendosperm mutant named floury endosperm12 (flo12). Theflo12 mutant exhibited loosely packed starch granules and alower thousand kernel weight compared to wild type. Semithinsections revealed that compound starch grains (SG) inflo12 interior endosperm cells were developed abnormally. Furthermore, amylose content was decreased, while totalprotein content was significantly increased in flo12 grains. Map-based cloning showed that FLO12 encodes rice alanineaminotransferase 1 (OsAlaAT1). OsAlaAT1 is highly expressedin developing endosperm. Subcellular localization showedthat OsAlaAT1 is localized in the cytosol. Moreover, theexpression of most starch synthesis-related genes wasdecreased, while most of the storage protein coding geneshad elevated expression levels in the flo12 mutant. Inaddition, overexpression of the OsAlaAT1 gene increasedgrain weight. In brief, we demonstrated that OsAlaAT1regulates carbon and nitrogen metabolism, which provides anew insight for the improvement of rice quality and yield.

      • Upright Orientation of 3D shapes via Tensor Rank Minimization

        In general, the upright orientation of a model is beneficial for human to recognize this model and is widely used in geometry processing and computer graphics. However, the orientation of the model obtained by existing technologies, such as 3D scanning systems or modeling, may be far away from the right orientation. In order to solve this problem, a robust and efficient upright method is needed. We observe that when the model is aligned with the three axes, the rank of the three-order tensor constructed by this model is the lowest usually. Inspired by this observation, we formulate the alignment of the 3D model with axes as a low-rank tensor optimization problem which is a global and unsupervised method. After the 3D model has been aligned with the three axes, some geometric properties can be used to pick out the best upright orientation from the six candidate supporting bases easily. The three-order tensor is constructed by voxelizing the bounding box of the 3D model, and then filling the voxel element with zero or one based on whether it contains the points of the model or not. The experimental results demonstrate that our method is robust, efficient and effective for all kinds of the models (manifold or non-manifold, man-made or non-artificial, or point cloud).

        CRISPR/Cas9‑mediated editing of CsWRKY22 reduces susceptibility to Xanthomonas citri subsp. citri in Wanjincheng orange (Citrus sinensis (L.) Osbeck)

        Key message CRISPR/Cas9-mediated editing of CsWRKY22 repressed canker development in Wanjincheng orange. Abstract Citrus canker, a destructive disease of citrus, is threatening the citrus industry worldwide. Breeding resistant cultivars is the most economical and effective approach to control citrus canker. Recently, CRISPR/Cas9-mediated genome editing was demonstrated to be a powerful tool for the improvement of citrus resistance to the disease. In our previous works, we confirmed that CsWRKY22 is involved in the plant immunity response to citrus canker in Wanjincheng orange (Citrus sinensis (L.) Osbeck). In this study, we targeted this gene to improve the resistance of Wanjincheng orange against citrus canker by CRISPR/Cas9-mediated editing. Sanger sequencing confirmed that CsWRKY22 in Wanjincheng orange contains CsWRKY22G and CsWRKY22C alleles, and the ratio of CsWRKY22G to CsWRKY22C is approximately 2:1. Four sgRNAs, which targeted the first exon of CsWRKY22, were selected for testing. In vitro cleavage activity analysis showed that two (W1 and W2) of the four sgRNAs displayed robust cleavage activities using PCR amplicons from the Wanjincheng orange genome as template. Subsequently, two constructs, pCas9/WRKY22sgRNA-W1 and pCas9/WRKY22sgRNA-W2, were used to modify the CsWRKY22. Three mutant plants were identified from seven independent transgenic plants. Based on Sanger sequencing, the W1-1, W2-2, and W2-3 mutant lines displayed 85.7%, 79.2%, and 68.2% mutation rates, respectively. Resistance evaluation indicated that the mutant plants showed decreased susceptibility to citrus canker. These results indicate that CRISPR/Cas9-targeted gene modification is an efficient approach for enhancing disease resistance in citrus.

        Characteristics and risk assessment of atmospheric PM<SUB>2.5</SUB> heavy metals pollution near coal gangue sites in Huaibei, China

        To study the level of atmospheric PM<SUB>2.5</SUB> and its heavy metal pollution near the coal gangue mountain, this study analyzed the content of seven heavy metals (Zn, Pb, Cu, Cd, Hg, Ni, Cr) and As through the PM2.5 samples from the vicinity of a large-scale coal gangue filed of Tongting coal mine in Huaibei, and evaluated the level of pollution, sources, and health effects. The results showed that during the sampling period, the average concentration of PM2.5 near the coal gangue field was 169.83 μg·m-3, which was 2.26 times that of the national air quality level Ⅱ daily standard. The coal gangue field may be an important source of air pollution, with the degree of heavy metal and arsenic pollution in the order of Cd, Pb, Zn, Hg (extremely heavy pollution) > As, Cu (medium pollution) > Ni, Cr (light pollution) and coal gangue dust and mining dust contributed more. This study provides data for atmospheric particulate matter and heavy metal and arsenic pollution levels near coal gangue fields and provides a theoretical basis for air pollution prevention and control near coal gangue hills.

      • Experimental Study of Lead-acid Battery Remaining Capacity Measuring in Photovoltaic Energy Storage System

        The combination of ampere-hour method and open-circuit voltage method in the photovoltaic measurement system is studied in this paper. In order to achieve the purpose of online measurement for remaining capacity, the influence of internal battery discharge and temperature on the battery capacity should be considered. The battery capacity decreases by the increasing charging times, and the increasing terminal voltage difference causes the inaccurate measurement of the capacity of battery. In this paper, two experimental measurement methods are employed, which are complementary to each other, to make the system measurement more accurate and extend the life of lead-acid batteries. If the charging process of photovoltaic system is unstable, an optimization of lead-acid battery applications in photovoltaic energy storage system is explored at the same time. Finally, an illustrative example is presented to demonstrate the effectiveness of the proposed measuring method.

      • The Research of the Recommendation Algorithm in Online Learning

        Recommendation algorithm is a kind of method in information filtering and has been widely applied on Internet. Collaborative filtering is widely used in the recommendation systems and has turned out to be successful. With the growth of the resources, it is difficult for users to find learning resources that suit for themselves. The recommendation algorithm is required to analyze the behavior of the users and then recommend object of learning. Online judge is a kind of the online learning. People can evaluate their programming ability through online judge. The performance of the different recommendation algorithms is analyzed in this paper and it is proposed that the item-based collaborative filtering recommendation algorithms should be applied into the online learning system. Based on the algorithm, we propose that by pre-processing the data and using user data that have solve large number of problems, we can get a better recommendation result.
