Prevention of UV-Induced Skin Damages by 11,14,17-Eicosatrienoic Acid in Hairless Mice In Vivo

Polyunsaturated fatty acids (PUFAs) are known to play important roles in various physiological and pathological processes. Recent studies have shown that some omega-3 (w-3) PUFAs, such as eicosapentaenoic acid (EPA) and dodecahexaenoic acid (DHA), have protective effects on acute and chronic UV-induced changes. However,the effects of other w-3 PUFAs including 11,14,17-eicosatrienoic acid (20:3)(ETA) on UV-induced skin damages are poorly understood. In this study, we investigated the cutaneous photoprotective effects of ETA in hairless mice in vivo. Female HR-1 hairless mice were topically treated with vehicle (ethanol:polyethylene glycol=30:70) only, 0.1% ETA, or 1% ETA once a day for 3 successive days after one time UV irradiation (200 mJ/cm2) on dorsal skins. Skin biopsy was carried out on the fourth day (72 hr after UV irradiation). We found that topical treatment with ETA attenuated UV-induced epidermal and dermal thickness and infiltration of inflammatory cells,and impairment of skin barrier function. In addition, ETA suppressed the expression of IL-1b, COX-2, and MMP-13 induced by UV irradiation. Our results show that the topical application of ETA protects against UV-induced skin damage in hairless mice and suggest that ETA can be a potential agent for preventing and/or treating UVinduced inflammation and photoaging.

<P>The hepatoprotective effects of a diterpenoid acanthoic acid isolated from <I>Acanthopanax koreanum</I> N<SMALL>AKAI </SMALL>were evaluated in a <SMALL>D</SMALL>-galactosamine/lipopolysaccharide-induced fulminant hepatic failure mouse model. Mice were pretreated orally with acanthoic acid 12 and 1 h before intraperitoneal injection of <SMALL>D</SMALL>-galactosamine and lipopolysaccharide. Pretreatment with the compound markedly reduced lethal liver injury in experimental animals. The effects were likely associated with a significant decrease in serum tumor necrosis factor (TNF)-α levels, which are correlated not only with those of alanine aminotransferase and aspartate aminotransferase but also with the reduced number of apoptotic hepatocytes in the liver as confirmed using the terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling method and DNA fragmentation assay. These results suggest that acanthoic acid protects against <SMALL>D</SMALL>-galactosamine/lipopolysaccharide-induced fulminant liver failure at least in part by a mechanism associated with the down-regulation of TNF-α secretion.</P>
Mutations in AP22.65 Accelerate Flowering in Arabidopsis thaliana

Identification of the gene(s) responsible for floweringtime in Arabidopsis has significant implications. We used theT-DNA insertion library of Arabidopsis thaliana to screen anearly-flowering mutant that exhibits accelerated floweringunder short-day conditions. AP22.65, a novel flowering-timegene in that species, was isolated and identified via genomewalkingand bioinformatics analysis. The flowering time ofAP22.65-complementing plants was similar to that of theCol-0 wild type (WT). Conversely, its overexpression delayedflowering. Consistent with this phenotype, expression ofAP22.65 was decreased in the ap22.65-1 mutant, recoveredin AP22.65-complementing plants, and increased in AP22.65-overexpressing plants. Compared with the WT, expressionlevels of critical genes in different flowering pathways, i.e.,SPY, FLC, GI, CO, FT, and LFY, were down-regulated inloss-of-function mutants. Expression of AP22.65 was distributedin flowers, siliques, rosette leaves, and whole seedlings. Therefore, this gene may be a negative regulator of Arabidopsisflowering.
Deep Learning-based Target Masking Scheme for Understanding Meaning of Newly Coined Words

최근 대량의 텍스트 분석을 위해 딥 러닝(Deep Learning)을 활용하는 연구들이 활발히 수행되고 있으며, 특히 대량의 텍스트에 대한 학습 결과를 특정 도메인 텍스트의 분석에 적용하는 사전 학습 언어 모델(Pre-trained Language Model)이 주목받고 있다. 다양한 사전 학습 언어 모델 중 BERT(Bidirectional Encoder Representations from Transformers) 기반 모델이 가장 널리 활용되고 있으며, 최근에는 BERT의 MLM(Masked Language Model)을 활용한 추가 사전 학습(Further Pre-training)을 통해 분석 성능을 향상시키기 위한 방안이 모색되고 있다. 하지만 전통적인 MLM 방식은 신조어와 같이 새로운 단어가 포함된 문장의 의미를 충분히 명확하게 파악하기 어렵다는 한계를 갖는다. 이에 본 연구에서는 기존의 MLM을 보완하여 신조어에 대해서만 집중적으로 마스킹을 수행하는 신조어 표적 마스킹(NTM: Newly Coined Words Target Masking)을 새롭게 제안한다. 제안 방법론을 적용하여 포털 ‘N’사의 영화 리뷰 약 70만 건을 분석한 결과, 제안하는 신조어 표적 마스킹이 기존의 무작위 마스킹에 비해 감성 분석의 정확도 측면에서 우수한 성능을 보였다. Recently, studies using deep learning to analyze a large amount of text are being actively conducted. In particular, a pre-trained language model that applies the learning results of a large amount of text to the analysis of a specific domain text is attracting attention. Among various pre-trained language models, BERT(Bidirectional Encoder Representations from Transformers)-based model is the most widely used. Recently, research to improve the performance of analysis is being conducted through further pre-training using BERT"s MLM(Masked Language Model). However, the traditional MLM has difficulties in clearly understands the meaning of sentences containing new words such as newly coined words. Therefore, in this study, we newly propose NTM(Newly coined words Target Masking), which performs masking only on new words. As a result of analyzing about 700,000 movie reviews of portal "N" by applying the proposed methodology, it was confirmed that the proposed NTM showed superior performance in terms of accuracy of sensitivity analysis compared to the existing random masking.
Hydrophilicity-controlled ordered mesoporous carbon for lithium-sulfur batteries.

<P>Ordered mesoporous carbon (OMC) materials were synthesized from a mesoporous silica KIT-6 (3-D cubic la3d meso-structure) as the hard-template via a nano-replication method. Hydrophilic and hydrophobic OMC materials were prepared using different carbon precursors including sucrose (suc-OMC) and phenanthrene (phe-OMC) at different carbonization temperatures of 700 degrees C and 1100 degrees C, respectively. The OMC materials thus obtained exhibit high surface areas, uniform mesopore sizes and highly ordered meso-structure. To investigate the hydrophilicity effect of OMC materials on the performance of lithium-sulfur battery, we prepared the samples having different ratios of the suc-OMC to phe-OMC, which were 100:0, 75:25, 50:50, 25:75 and 0:100. As a result, the mixed OMC materials (with ratios of 75:25, 50:50 and 25:75) exhibited better cycle performances, compared to those of the suc-OMC and phe-OMC.</P>

We investigated the alterations of major fatty acid components in epidermis by natural aging and photoaging processes, and by acute ultraviolet (UV) irradiation in human skin. Interestingly, we found that 11,14,17-eicosatrienoic acid (ETA), which is one of the omega-3 polyunsaturated acids, was significantly increased in photoaged human epidermis in vivo and also in the acutely UV-irradiated human skin in vivo, while it was significantly decreased in intrinsically aged human epidermis. The increased ETA content in the epidermis of photoaged human skin and acute UV-irradiated human skin is associated with enhanced expression of human elongase 1 and calcium-independent phophodiesterase A2. We demonstrated that ETA inhibited matrix metalloproteinase (MMP)-1 expression after UV-irradiation, and that inhibition of ETA synthesis using EPTC and NA-TCA, which are elongase inhibitors,increased MMP-1 expression. Therefore, our results suggest that the UV increases the ETA levels, which may have a photoprotective effect in the human skin.
