        A DC Charging Pile for New Energy Electric Vehicles

        New energy electric vehicles will become a rational choice to achieve clean energy alternatives in the transportation field, and the advantages of new energy electric vehicles rely on high energy storage density batteries and efficient and fast charging technology. This paper introduces a DC charging pile for new energy electric vehicles. The DC charging pile can expand the charging power through multiple modular charging units in parallel to improve the charging speed. Each charging unit includes Vienna rectifier, DC transformer, and DC converter. The feasibility of the DC charging pile and the effectiveness of the control strategies of each component of the charging unit are verified by simulation and experimental results. This DC charging pile and its control technology provide some technical guarantee for the application of new energy electric vehicles.

        Application of cascaded H-bridge multilevel inverter in the speed regulation of medium- and high-voltage asynchronous motor

        Medium- and high-voltage motors are characterized by high power and large inertia, and are widely used in industrial frequency conversion. The cascaded H-bridge multilevel (CHB-ML) inverter adopts a modular design concept to realize high-voltage and high-power functions by cascading multiple identical low-voltage conversion units. Moreover, the harmonic content of the output current of the inverter is small. For this purpose, a multilevel inverter consisting of eight H-bridge inverter units cascaded in each phase is introduced to drive medium- and high-voltage asynchronous motors for starting and frequency regulation. The carrier phase-shifted sinusoidal pulse-width modulation technique, DC voltage equalization control of power unit, and speed sensorless vector control strategy based on a voltage–current hybrid chain observer are mainly introduced. The simulation and experimental results of the real-time digital simulation system (RTDS) of the asynchronous motor dragged by the inverter verify that the CHB-ML inverter with the proposed control strategy has good starting and frequency regulation performance. These findings provide a technical guarantee for the field of medium- and high-voltage high-power drives.

        Vitellogenin’s putative role in Tegillarca granosa’s cadmium detoxification

        The bloody clam, Tegillarca granosa, is a commercial benthic bivalve, having a strong accumulation ability and torrelence to cadmium. To investigate whether vitellogenin (Vg) is involved in cadmium (Cd) detoxification, the full-length cDNA of T. granosa Vg was cloned, and its expression pattern in response to cadmium exposure was studied compared with the reference metallothionein (MT) gene. The full T. granosa Vg sequence consisted of 8988 bp, including a 6930-bp open reading frame that encoded a 2309 amino acid polypeptide. The deduced Vg protein contained a Vg N-terminal domain, domain of unknown function (DUF1943), SbcC domain, and von Willebrand factor typeD domain (VWD). Multiple metal-binding sites were predicted in the deduced T. granosaVg protein, suggesting its potential in functioning as a metal-binding protein. In addition, Vg expression increased in the T. granosa digestive gland and hemolymph in time-dependent manner after exposure to 1, 3, 6 and 9 lg/L Cd for 28 days.MTexpression was measured in parallel with Vg expression, and the latter was more sensitive to Cd induction than the former. Together, results of the present research suggested that Vg may play an important role in T. granosa metal detoxification.

        Performance and Mechanism Analysis of Succinate Production under Different Transporters in Escherichia coli

        Succinic acid is a platform chemical with potential for bio-based synthesis. However, the production of bio-based succinate is limited because of insufficient succinate efflux capacity in the late stage of fermentation. In the present study, three different transporters, which have been reported to be responsible for C4-dicarboxylates transport, were employed for investigation of the transport capacity of succinate in Escherichia coli. After engineered strains were constructed, the fermentative production of succinic acid was studied in serum bottles and 3 L of fermentor. The results demonstrated that engineered strain showed better efflux capacity than control strain under high concentration of succinate. The highest production of succinate was 68.66 g/L, while the NCgl2130 transporter may be the best candidate for succinate export in E. coli. Further research showed that the expression levels and relative enzyme activities involved in the metabolic pathway all increased markedly, and the maximum activities of PPC, PCK, PYK, and MDH increased by 1.50, 1.38, 1.28, and 1.27-fold in recombinant E. coli AFP111/pTrc99a- NCgl2130, respectively. Moreover, the maximum level of intracellular ATP increased by 23.79% in E. coli AFP111/ pTrc99a-NCgl2130. Taken together, these findings indicated that engineered transporters can improve succinate production by increasing key enzyme activities and intracellular ATP levels. To the best of thew authors’ knowledge, this is the first report on a mechanism to improve succinate production by engineered transporters. This strategy set up a foundation for improving the biosynthesis of other C4-dicarboxylates, such as fumaric acid and malic acid.
