      • Clinical Evaluation of Human Papillomavirus Detection by careHPV<sup>TM</sup> Test on Physician-Samples and Self-Samples using The Indicating FTA Elute® Card

        Objective: To make the clinical evaluation of a solid-state human papillomavirus (HPV) sampling medium in combination with an economical HPV testing method ($careHPV^{TM}$) for cervical cancer screening. Methods: 396 women aged 25-65 years were enrolled for cervical cancer screening, and four samples were collected. Two samples were collected by woman themselves, among which one was stored in DCM preservative solution (called "liquid sample") and the other was applied on the Whatman Indicating FTA $Elute^{(R)}$ card (FTA card). Another two samples were collected by physician and stored in DCM preservative solution and FTA card, respectively. All the samples were detected by $careHPV^{TM}$ test. All the women were administered a colposcopy examination, and biopsies were taken for pathological confirmation if necessary. Results: FTA card demonstrated a comparable sensitivity of detecting high grade Cervical Intraepithelial Neoplasia (CIN) with the liquid sample carrier for self and physician-sampling, but showed a higher specificity than that of liquid sample carrier for self-sampling (FTA vs Liquid: 79.0% vs 71.6%, p=0.02). Generally, the FTA card had a comparable accuracy with that of Liquid-based medium by different sampling operators, with an area under the curve of 0.807 for physician &FTA, 0.781 for physician &Liquid, 0.728 for self & FTA, and 0.733 for self &Liquid (p>0.05). Conclusions: FTA card is a promising sample carrier for cervical cancer screening. With appropriate education programmes and further optimization of the experimental workflow, FTA card based self-collection in combination with centralized $careHPV^{TM}$ testing can help expand the coverage of cervical cancer screening in low-resource areas.

      • Geometric Optimization of a Mathematical Model of Radiofrequency Ablation in Hepatic Carcinoma

        Radio frequency ablation (RFA) is an effective means of achieving local control of liver cancer. It is a particularly suitable mode of therapy for small and favorably located tumors. However, local progression rates are substantially higher for large tumors (>3.0 cm). In the current study, we report on a mathematical model based on geometric optimization to treat large liver tumors. A database of mathematical models relevant to the configuration of liver cancer was also established. The specific placement of electrodes and the frequency of ablation were also optimized. In addition, three types of liver cancer lesion were simulated by computer guidance incorporating mathematical models. This approach can be expected to provide a more effective and rationale mechanism for employing RFA in the therapy of hepatic carcinoma.

      • Dysregulated Fatty Acid Metabolism in Hepatocellular Carcinoma

        Aims: Studies are urgently needed on it molecular pathogenesis and biological characteristics of hepatocellular carcinoma (HCC). Dysregulation of fatty acid (FA) metabolism, in which aberrant activation of oncogenic signaling pathways alters the expression and activity of lipid-metabolizing enzymes, is an emerging hallmark of cancer cells, and it may be involved in HCC development and progression. Methods: We summarize the characteristics of FA metabolism in HCC, focusing on the pathways of FA synthesis, oxidation, uptake and transport. We also provide a brief review of the relationship between NAFLD and HCC development. Results: The current review summarizes the dysregulated FA metabolism in HCC and pathways through which this dysregulation may regulate HCC survival and growth. Aberrant activation of oncogenic signaling pathways regulates the expression and activity of lipid-metabolizing enzymes, thus reprogramming FA metabolism to promote HCC development and progression. Intracellular FAs are required for biosynthesis of most biological membrane lipids and signaling molecules, and are also used to provide energy to support HCCs survival and proliferation, when necessary, through β-oxidation process. HCC cells can employ appropriate metabolic pathways as different situation demands. Intrahepatic cholangiocarcinoma (ICC) and HCC exhibits differential requirement for de novo lipogenesis and distinct response to therapeutic approaches focusing on inhibition of exogenous FA uptake. Non-alcoholic fatty liver disease related obesity and diabetes have increasingly emerged as two major factors responsible for the rise in prevalent of HCC. Conclusions: Our understanding of dysregulated FA metabolism and associated signaling pathways may contribute to the development of novel and efficient anti-tumor approaches for patients with HCC.

        Influence of Preparation Conditions on the Formation of Copper (II) Architectures with Pyrazine-2,3,5-tricarboxylic Acid

        Three new metal-organic copper(II) complexes, [Cu(H_2PZTC)_2]_n·2nH_2O (1), [Cu(HPZTC)·2H_2O]_n·2nH_2O (2),and Cu_2[(PZHD)(OH)(H_2O)_2]_n (3) (H_3PZTC = pyrazine-2,3,5-tricarboxylic acid, PZHD^3− = 2-hydroxypyrazine-3,5-dicarboxylate), have been synthesized from Cu(II)/H3PZTC system under different synthetic conditions, and characterized by single-crystal X-ray diffraction, elemental analysis, IR spectroscopy and thermogravimetric analysis. In complexes 1 and 2, H_3PZTC ligands loose one and two protons, which were transformed into H_2PZTC− anion and HPZTC^2− dianion under different preparation condition, respectively. Furthermore, two ligands coordinate with Cu(II) cations in different modes, leading to the formation of the different chain structures. In complex 3, H_3PZTC ligand was converted into a new ligand-PZHD by in situ decarboxylation and hydroxylation under a higher pH value than that for complexes 1 and 2. PZHD ligands link the Cu(II) cations to form a 2D layer structure. These results demonstrate that the preparation conditions,including pH value and reaction temperature etc, play an important role in the construction of complexes based on H_3PZTC ligand.

        Cloning and characterization of a novel gene with alternative splicing in murine mesenchymal stem cell Line C3H/10T1/2 by gene trap screening

        A novel gene, designated mgt-6, containing four splicing variants, was isolated from a gene trap clone library of C3H/10T1/ 2 cells transfected with retroviral promoterless gene-trap vector, ROSAFARY. The transcript variants were differentially expressed in murine tissues and cell lines and differentially responded to diverse stimuli including TGF-β1 and mitogen-activated protein kinase (MAPK) inhibitors. The mgt-6 gene encoded a protein of 37 or 11 amino acid residuals with cytoplasmic distribution. However, when C3H/10T1/2 cells were treated with 5-azacytidine, the protein translocated into cell nucleus as indicated by fused LacZ or C-terminally tagged EGFP. Our preliminary results suggest that further study on the role of mgt-6 gene in cell transformation and differentiation may be of significance. [BMB reports 2010; 43(12): 789-794]

      • ppGalNAc T1 as a Potential Novel Marker for Human Bladder Cancer

        Objectives: To investigate the effect of glycopeptide-preferring polypeptide GalNAc transferase 1 (ppGalNAc T1 ) targeted RNA interference (RNAi) on the growth and migration of human bladder carcinoma EJ cells in vitro and in vivo. Methods: DNA microarray assays were performed to determine ppGalNAc Ts(ppGalNAc T1-9) expression in human bladder cancer and normal bladder tissues. We transfected the EJ bladder cancer cell line with well-designed ppGalNAc T1 siRNA. Boyden chamber and Wound healing assays were used to investigate changes of shppGalNAc T1-EJ cell migration. Proliferation of shppGalNAc T1-EJ cells in vitro was assessed using [3H]-thymidine incorporation assay and soft agar colony formation assays. Subcutaneous bladder tumors in BALB/c nude mice were induced by inoculation of shppGalNAc T1-EJ cells and after inoculation diameters of tumors were measured every 5 days to determine gross tumor volumes. Results: ppGalNAc T1 mRNA in bladder cancer tissues was 11.2-fold higher than in normal bladder tissues. When ppGalNAc T1 expression in EJ cells was knocked down through transfection by pSUPER-shppGalNAc T1 vector, markedly reduced incorporation of [3H]-thymidine into DNA of EJ cells was observed at all time points compared with the empty vector transfected control cells. However, ppGalNAc T1 knockdown did not significantly inhibited cell migration (only 12.3%). Silenced ppGalNAc T1 expression significantly inhibited subcutaneous tumor growth compared with the control groups injected with empty vector transfected control cells. At the end of observation course (40 days), the inhibitory rate of cancerous growth for ppGalNAc T1 knockdown was 52.5%. Conclusion: ppGalNAc T1 might be a potential novel marker for human bladder cancer. Although ppGalNAc T1 knockdown caused no remarkable change in cell migration, silenced expression significantly inhibited proliferation and tumor growth of the bladder cancer EJ cell line.

        Molecular Cloning and Characterization of a New C-type Lysozyme Gene from Yak Mammary Tissue

        Milk lysozyme is the ubiquitous enzyme in milk of mammals. In this study, the cDNA sequence of a new chicken-type (c-type) milk lysozyme gene (YML), was cloned from yak mammary gland tissue. A 444 bp open reading frames, which encodes 148 amino acids (16.54 kDa) with a signal peptide of 18 amino acids, was sequenced. Further analysis indicated that the nucleic acid and amino acid sequences identities between yak and cow milk lysozyme were 89.04% and 80.41%, respectively. Recombinant yak milk lysozyme (rYML) was produced by Escherichia coli BL21 and Pichia pastoris X33. The highest lysozyme activity was detected for heterologous protein rYML5 (M = 1,864.24 U/mg, SD = 25.75) which was expressed in P. pastoris with expression vector $pPICZ{\alpha}A$ and it clearly inhibited growth of Staphylococcus aureus. Result of the YML gene expression using quantitative polymerase chain reaction showed that the YML gene was up-regulated to maximum at 30 day postpartum, that is, comparatively high YML can be found in initial milk production. The phylogenetic tree indicated that the amino acid sequence was similar to cow kidney lysozyme, which implied that the YML may have diverged from a different ancestor gene such as cow mammary glands. In our study, we suggest that YML be a new c-type lysozyme expressed in yak mammary glands that plays a role as host immunity.

        Characteristics of Refractive Index Profiles at Different Temperatures in LiNbO₃ and KTiOPO₄ Waveguide Formed by 350 keV Light Ions

        Both LiNbO₃ and KTiOPO₄ samples were implanted with 350 keV H^+ and He^+ ions at different doses ranging from 1×10^(16) to of 5×10^(16) ions/㎠. Single and multi-energy implantations were performed at room temperature. Mono-mode or a few modes in both LiNbO₃ and KTiOPO₄ waveguides were observed. The effect of temperature on the refractive index profiles of LiNbO₃ and KTiOPO₄ waveguids was studied. The temperature covered from room temperature, 200℃, 194.5 K (dry ice) and 77K (liquid nitrogen). Different mechanisms are needed to interpret the observed behavior. A n_e increased mono-mode LiNbO₃ waveguide was formed by multi-energy keV He^+ ions.

      • Depth Distributions of Bi+ Ions Implanted into Ni, Si and SiO₂, Films

        Ni, Si and SiO₂ films were implanted by 350 keV Bi^+ ions at room temperature with fluences of 1×10^(16) and 2×10^(16) ions/㎤. The depth distributions of implanted Bi^+ ions in Ni, Si and SiO₂ films were by investigated by Rutherford backscattering. The results show that the depth distributions of implanted Bi^+ ions into Ni, Si and SiO₂ films have obeyed nearly Gaussian distributions. The maximum difference between experimental and calculated values is less than 18 % for mean projected range. Experimental range straggling deviated significantly from calculated value. The possible reasons are discussed.
