        Hypoglycemic and Hypolipidemic Activity of Polygonatum sibiricum Fermented with Lactobacillus brevis YM 1301 in Diabetic C57BL/6 Mice

        Polygonatum sibiricum (PS) has been used as herbal medicine to treat type 2 diabetes mellitus (T2DM). However, how lactic acid fermentation of PS influences glucose and lipid metabolism remains unclear. The current study was undertaken to evaluate the hypoglycemic and hypolipidemic effects of PS fermented with Lactobacillus brevis YM 1301 (YM 1301) in streptozotocin and high-fat diet-induced T2DM mice. Biochemical analysis revealed that supplementation with metformin, PS, or fermented Polygonatum sibiricum (FPS) lowered the fasting blood glucose, insulin, total cholesterol, triglyceride, and low-density lipoprotein cholesterol of diabetic mice. FPS showed relatively more potency to reduce the homeostasis model assessment-insulin resistance and glycated hemoglobin than PS. Moreover, a high dosage of FPS protected against glucose intolerance and insulin resistance by increasing the ratio of phosphor-AKT/AKT. Histological examination and quantitative polymerase chain reaction results showed that dietary FPS ameliorated the lipid accumulation in liver and white adipose tissue (WAT) by inhibiting lipogenesis, enhancing lipolysis, and fatty acid oxidation. FPS exhibited greater efficacy than PS on improving the transcriptional expression of adipose triacylglyceride lipase, carnitine palmitoyltransferase 1, and uncoupling protein 1. In addition, FPS exerted a striking anti-inflammatory effect by suppressing the expression of interleukin 6, interleukin 1β, tumor necrosis factor-α, and transforming growth factor-β in WAT of diabetic C57BL/6 mice. Finally, FPS supplementation enhanced the activation of AMPK. In conclusion, these results suggest that the FPS may be more promising than PS as a potential therapeutic agent for diabetes and obesity.

        Effect of lactoferrin on ram sperm motility after cryopreservation

        Objective: The objective of this study was to analyse the differentially abundant proteins caused by freeze–thawing of ram sperm and explore candidate proteins of interest for their ability to improve ram sperm cryopreservation outcomes in vitro. Methods: Sperm were from three mature Dorper. Fresh and frozen sperm proteins were extracted, and the differentially abundant proteins were analysed by mass spectrometry. Among these proteins, lactoferrin (LTF) was selected to be added before cryopreservation. Next, sperm samples were diluted in Tris extender, with the addition of 0, 10, 100, 500, and 1,000 μg/mL of LTF. After thawing, sperm quality was evaluated by motility, plasma membrane integrity, mitochondrial activity and reactive oxygen species (ROS). Results: Cryopreservation significantly altered the abundance of 40 proteins; the abundance of 16 proteins was increased, while that of 24 proteins was decreased. Next, LTF was added to Tris extender applied to ram sperm. The results showed that sperm motility and plasma membrane integrity were significantly improved (p<0.05) by supplementation with 10 μg/mL LTF compared to those in the control group. There was no significant difference in mitochondrial activity between the 0 μg/mL group and other groups (p>0.05). Supplementation of the cryoprotective extender with 10 μg/mL LTF led to decreased ROS levels compared with those in the control and other groups (p<0.05). Conclusion: The LTF is an important protein during cryopreservation, and the addition of 10 μg/mL LTF to a cryoprotective extender can significantly improve the function of frozen ram sperm.         Effects of Dietary Copper Source and Level on Performance, Carcass Characteristics and Lipid Metabolism in Lambs

        An experiment was conducted to determine the effect of dietary copper (Cu) on performance, carcass characteristics and lipid metabolism in lambs. Fifty DorperMongolia wether lambs (approximately 3 month of age; 23.80.6 kg of body weight) were housed in individual pens and were assigned randomly to one of five treatments. Treatments consisted of 1) control (no supplemental Cu), 2) 10 mg Cu/kg DM from Cu-lysine, 3) 20 mg Cu/kg DM from Cu-lysine, 4) 10 mg Cu/kg DM from tribasic copper chloride (Cu2(OH)3Cl; TBCC), 5) 20 mg Cu/kg DM from tribasic copper chloride. The Cu concentration was 6.74 mg/kg DM in the basal diet. Body weight was measured on two consecutive days at the start and the end of the 60-day experimental period. Blood samples were collected and then the lambs were slaughtered on d 60. Performance was not affected (p>0.05) by dietary Cu treatment. Cu-supplemented and control lambs had similar hot carcass weight, dressing percentage and longissimus muscle area, but Cu supplementation, regardless of source and level, reduced (p<0.01) 12th rib backfat and kidney fat in lambs. Plasma tumor necrosis factor-alpha (TNF-) and serum triglyceride concentrations were increased (p<0.05), total cholesterol concentrations were decreased (p<0.05) and nonesterified fatty acids (NEFA) concentrations tended to be increased (p<0.07) by Cu supplementation. However, Serum concentrations of HDL-cholesterol and LDL-cholesterol were not affected (p>0.05) by dietary treatment. Fatty acid profile of longissimus muscle was similar across treatments. These results indicate that Cu-lysine and TBCC are of similar availability in lambs. Cu supplementation given to DorperMongolia wether lambs altered lipid metabolism. The reduction in backfat depth may be due to copper altering TNF- metabolism in lambs. Supplementation of 10 or 20 mg Cu/kg DM showed similar effects on lipid metabolism in lambs.

        Effects of aflatoxin B1 combined with ochratoxin A and/or zearalenone on metabolism, immune function, and antioxidant status in lactating dairy goats

        Objective: This experiment investigated the effects of aflatoxin B1 (AFB1) alone or mixed with ochratoxin A (OTA) and/or zearalenone (ZEA) on the metabolism, immune function, and antioxidant status of dairy goats. Methods: Fifty lactating Laoshan dairy goats were randomly assigned to one of five treatment groups (n = 10) for 14 days. Goats were fed no additive (control) or administered with $50{\mu}g\;AFB1/kg$ dry matter (DM) (AFB1), $50{\mu}g\;AFB1/kg$ $DM+100{\mu}g\;OTA/kg$ DM (AFB1+OTA), $50{\mu}g\;AFB1/kg$ $DM+500{\mu}g\;ZEA/kg$ DM (AFB1+ZEA), or $50{\mu}g\;AFB1/kg$ $DM+100{\mu}g\;OTA/kg$ $DM+500{\mu}g\;ZEA/kg$ DM (AFB1+OTA+ZEA). Results: Dry matter intake and milk production were lower in goats fed AFB1+OTA+ZEA than in controls. Supplementation with AFB1, OTA, and ZEA significantly decreased red blood cell count, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin, and mean platelet volume, and significantly increased white blood cell count, when compared with the control group. Compared with control, the combination of AFB1, OTA, and ZEA significantly increased alanine aminotransferase (ALT) and alkaline phosphatase (ALP) activities, total bilirubin (TBIL), interleukin-6, and malondialdehyde (MDA), but significantly reduced immunoglobulin A concentration, the activities of superoxide dismutase (SOD) and glutathione peroxides (GSH-Px), and total antioxidant capacity (T-AOC) in serum. Administration of AFB1 combined with OTA led to higher ALP, ALT, TBIL, and MDA, as well as lower milk production, SOD and GSH-Px activities, and T-AOC, than administration of AFB1 combined with ZEA. Conclusion: The mixture of AFB1, OTA, and ZEA exerted the greatest adverse effects on dairy goats, meanwhile the deleterious damage of the other mycotoxin combinations were in varying degrees. The findings of this study could provide guidance for the prevention and treatment of the consequences of contamination of animal feeds with combinations of mycotoxin.

      • High Resolution Image Reconstruction with Compressed Sensing based on Iterations

        This paper proposes a new method of efficient image reconstruction based on the Modified Frame Reconstruction Iterative Thresholding Algorithm (MFR ITA) developed under the compressed sensing (CS) domain by using total variation algorithm. The new framework is consisted of three phases. Firstly, the input images are processed by the multilook processing with their sparse coefficients using the Discrete Wavelet Transform (DWT) method. Secondly, the measurements are obtained from sparse coefficient by using the proposed fusion method to achieve the balance resolution of the pixels. Finally, the fast CS method based on the MFR ITA is proposed to reconstruct the high resolution image. The proposed method achieved superior results on real images, and demonstrate qualitative improvements in terms of PSNR and SSIM values. Furthermore, achieved good reconstruction SNR in the presence of noise.

        Genome-wide scan for runs of homozygosity identifies candidate genes in Wannan Black pigs

        Objective: Runs of homozygosity (ROH) are contiguous lengths of homozygous genotypes that can reveal inbreeding levels, selection pressure, and mating schemes. In this study, ROHs were evaluated in Wannan Black pigs to assess the inbreeding levels and the genome regions with high ROH frequency.Methods: In a previous study, we obtained 501.52 GB of raw data from resequencing (10×) of the genome and identified 21,316,754 single-nucleotide variants in 20 Wannan Black pig samples. We investigated the number, length, and frequency of ROH using resequencing data to characterize the homozygosity in Wannan Black pigs and identified genomic regions with high ROH frequencies.Results: In this work, 1,813 ROHs (837 ROHs in 100 to 500 kb, 449 ROHs in 500 to 1,000 kb, 527 ROHs in >1,000 kb) were identified in all samples, and the average genomic inbreeding coefficient (FROH) in Wannan Black pigs was 0.5234. Sixty-one regions on chromosomes 2, 3, 7, 8, 13, 15, and 16 harbored ROH islands. In total, 105 genes were identified in 42 ROH islands, among which some genes were related to production traits.Conclusion: This is the first study to identify ROH across the genome of Wannan Black pigs, the Chinese native breed of the Anhui province. Overall, Wannan Black pigs have high levels of inbreeding due to the influence of ancient and recent inbreeding due to the genome. These findings are a reliable resource for future studies and contribute to save and use the germplasm resources of Wannan Black pigs.
