Three compounds, vanillic acid, p-hydroxylcinnamic acid, p-hydroxybenzaldehyde have been isolated from the ethylacetate extract of Sparganium stoloniferum Buch.-Ham roots using silica gel open column chromatography, preparative thin-layer chromatography (pTLC) and reverse phase high performance liquid chromatography. The structures of the compounds were established on the basis of IR, extensive 1D NMR, and MS analyses. The ethylacetate (EtOAc) extract, vanillic acid, and p-hydroxybenzaldehyde showed α-glucosidase inhibition activity of 72.71%, 20.13%, and 30.42%, at the concentration of 10 ㎍/mL, respectively. The EtOAc extract exhibited strong antioxidant activity with an IC50 value of 24.37 ㎍/mL against DPPH radical scavenging activity, the vanillic acid, p-hydroxylcinnamic acid, and p-hydroxybenzaldehyde with an IC50 value of 2.10 μM, 1.59 μM, and 2.72 μM against DPPH, respectively.
Could Tumor Size Be A Predictor for Papillary Thyroid Microcarcinoma: a Retrospective Cohort Study

Background: Central lymph node metastasis(CLNM) is common in papillary thyroid microcarcinoma (PTMC). The aim of this study was to define the pathohistologic risk grading based on surgical outcomes. Materials and Methods: Statistical analysis was performed to figure out the optimal cut-off values of size in preoperative ultrasound images for defining the risk of CLNM in papillary thyroid microcarcinoma. Receiver operating characteristic curves (ROC) studies were carried out to determine the cutoff value(s) for the predictor(s). All the patients were divided into two groups according to the above size and the clinic-pathological and immunohistochemical parameters were compared to determine the significance of findings. Results: The optimal cut-off value of tumor size to predict the risk of CLNM in papillary thyroid microcarcinoma was 0.575 cm (area under the curve 0.721) according to the ROC curves. Significant differences were observed on the multifocality, extrathyroidal extension and central lymph node metastasis between two groups which were divided according to the tumor size by the cutoff values. Patients in two groups showed different positive rate and intensity of Ki67. Conclusions: The size of PTMC in ultrasound images are helpful to predict the aggressiveness of the tumors, it could be an easy predictor for PTMC prognosis and assist us to choose treatment.

Association Between the Ku70-1310C/G Promoter Polymorphism and Cancer Risk: a Meta-analysis

Ku70 plays an important role in DNA double-strand break repair. Studies revealing conflicting results on the role of the Ku70-1310C/G promoter polymorphism on cancer risk led us to perform a meta-analysis to investigate this relationship. Ten case-control studies with 2566 cases and 3058 controls were identified. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to assess the strength of associations. The overall results suggested no association between the Ku70-1310C/G promoter polymorphism and total cancer risk. However, on stratified analysis, significantly increased risks were observed among the Asian population (GG vs. CC: OR=1.50, 95%CI=1.10-2.06; GG vs. CC/CG: OR=1.47, 95%CI=1.07-2.01) and population-based case-control studies (GG vs. CC: OR=1.57, 95%CI=1.12-2.22; CG vs. CC: OR=1.35, 95%CI=1.11-1.64; CG/GG vs. CC: OR=1.37, 95%CI=1.14-1.65). Additionally, variant genotypes were associated with a significantly increased breast cancer risk (GG vs. CC: OR=1.80, 95%CI=1.26-2.56; GG vs. CC/CG: OR=1.40, 95%CI=1.01-1.95).
OsCML4 improves drought tolerance through scavenging of reactive oxygen species in rice

Calmodulin-like (CML) genes function in regulating plant responses to different abiotic stresses, such as high salt and drought. Using japonica type rice seedlings (‘Nipponbare’), we induced the expression of OsCML4 (Accession No. NM_001057768), which encodes a rice CML protein. Under drought conditions, transgenic plants over-expressing that gene exhibited obviously improved growth performance and higher survival rates than the wild type (WT). Activity by reactive oxygen species (ROS)- scavenging enzymes superoxide dismutase (SOD) and catalase (CAT), as well as proline concentrations, were significantly increased in transgenic plants. Expression by ROS scavenging-related genes APXI and Cat-B and the stress-related gene OsP5CS1 was also enhanced in the transgenics under drought conditions. However, the sensitivity to abscisic acid (ABA) was similar between over-expressing and WT plants. These results suggest that OsCML4 confers drought tolerance partially through ROS-scavenging and also by inducing other stress-related genes in an ABAindependent manner.
Study on the Internal Pipeline Inspection Devices with Energy Acquisition Methods

The pipeline conveying natural gas must be regularly inspected by the detection device, and the cracks and corrosion condition inside the pipeline should be detected. Generally, the detection device consists of three parts: drive part, detection part and energy part. Since pipeline transportation is long-distance transportation and cannot realize wired detection, the energy part needs to provide enough electric energy for the detection part to meet the energy consumption of the detection part. The method of obtaining energy for the energy part is very important. In this paper, the energy part is studied, and an efficient energy part is designed. By analyzing the change law of the positive pressure on the inner wall of the energy acquisition wheel on the pipeline, the corresponding torque and power changes are obtained when the detector moves at different speeds. Through the test platform system, the output power of the energy part at different speeds is collected, and the accuracy of the theoretical calculation is verified.

The antioxidant and antiproliferation effects of fractions from methanol extract of Sparganiun stoloniferum were investigated in this study. The ethyl acetate fraction of the methanol extract showed significant antioxidant activity against α,?α-diphenyl-picrylhydrazyl (DPPH) and hydroxyl ( · OH) radical scavenging, and it showed greater reducing power than the n-hexane, dichloromethane, n-butanol and water fractions. Qualitative HPLC analysis was performed on the extractions. The high content of phenolics in ethyl acetate fractions were believed to be responsible for its antioxidant activity, which was the strongest in all five fractions. Moreover, the ethyl acetate fraction showed a dose-dependent inhibition of cell proliferation with an EC?? of 139.21 ± 0.54 μg · mL?¹ against the human colon cell, HT-29. The anti-proliferative effects of the ethyl acetate fraction were associated with apoptosis. Semi-quantitative RT-PCR analysis indicates that, after treatment with the ethyl acetate fraction, the expression of caspase-3, p53, and Bax in HT-29 cells was significantly up-regulated and c-myc expression was induced. These findings demonstrate that the ethyl acetate fraction of S. stoloniferum has potent antioxidant and antiproliferation effects and induces apoptosis. Thus, the extract of S. stoloniferum may be considered as a potential functional food resource and complementary medicinal supplement.

BACKGROUND/OBJECTIVES: Sonchus asper is used extensively as an herbal anti-inflammatory for treatment of bronchitis, asthma, wounds, burns, and cough; however, further investigation is needed in order to understand the underlying mechanism. To determine its mechanism of action, we examined the effects of an ethyl acetate fraction (EAF) of S. asper on nitric oxide (NO) production and prostaglandin-E2 levels in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. MATERIALS/METHODS: An in vitro culture of RAW264.7 macrophages was treated with LPS to induce inflammation. RESULTS: Treatment with EAF resulted in significant suppression of oxidative stress in RAW264.7 macrophages as demonstrated by increased endogenous superoxide dismutase (SOD) activity and intracellular glutathione levels, decreased generation of reactive oxygen species and lipid peroxidation, and restoration of the mitochondrial membrane potential. To confirm its anti-inflammatory effects, analysis of expression of inducible NO synthase, cyclooxygenase-2, tumor necrosis factor-α, and the anti-inflammatory cytokines IL-1β and IL-6 was performed using semi-quantitative RT-PCR. EAF treatment resulted in significantly reduced dose-dependent expression of all of these factors, and enhanced expression of the antioxidants MnSOD and heme oxygenase-1. In addition, HPLC fingerprint results suggest that rutin, caffeic acid, and quercetin may be the active ingredients in EAF. CONCLUSIONS: Taken together, findings of this study imply that the anti-inflammatory effect of EAF on LPS-stimulated RAW264.7 cells is mediated by suppression of oxidative stress.

Radiation proctitis is a common complication after radiotherapy for pelvic malignant tumors. This study was conducted to assess the efficacy of novel almagate enemas in hemorrhagic chronic radiation proctitis (CRP) and evaluate risk factors related to rectal deep ulcer or fistula secondary to CRP. All patients underwent a colonoscopy to confirm the diagnosis of CRP and symptoms were graded. Typical endoscopic and pathological images, risk factors, and quality of life were also recorded. A total of 59 patients were enrolled. Gynecological cancers composed 93.1% of the primary malignancies. Complete or obvious reduction of bleeding was observed in 90% (53/59) patients after almagate enema. The mean score of bleeding improved from 2.17 to 0.83 (P<0.001) after the enemas. The mean response time was 12 days. No adverse effects were found. Moreover, long-term successful rate in controlling bleeding was 69% and the quality of life was dramatically improved (P=0.001). The efficacy was equivalent to rectal sucralfate, but the almagate with its antacid properties acted more rapidly than sucralfate. Furthermore, we firstly found that moderate to severe anemia was the risk factor of CRP patients who developed rectal deep ulcer or fistulas (P= 0.015). We also found abnormal hyaline-like thick wall vessels, which revealed endarteritis obliterans and the fibrosis underlying this disease. These findings indicate that almagate enema is a novel effective, rapid and well-tolerated method for hemorrhagic CRP. Moderate to severe anemia is a risk factor for deep ulceration or fistula.
