Epigenetic Regulation of miR-129-2 Leads to Overexpression of PDGFRa and FoxP1 in Glioma Cells

miR-129-2 is frequently downregulated in multiple cancers. However, how it is silenced in cancers remains unclear. Here we investigated the expression profile and potential biological function of miR-129-2 in glioblastoma (GBM), the most common and lethal form of brain tumors in adults. We showed that miR-129-2 is lost in GBM patient specimens and cultured cell lines. miR-129-2 expression could be restored upon treatment with a histone deadetylase inhibitor (trichostatin A) but not a DNA methylation inhibitor (5-Aza-2'-deoxycytidine), and more profound effect was observed with the treatment of these two drugs in combination. Furthermore, forced expression of miR-129-2 repressed the expression of major oncogenic genes such as PDGFRa and Foxp1 in GBMs. Consistently, expression of miR-129-2 significantly inhibits GBM cell proliferation in vitro. These results reveal that miR-129-2 is epigenetically regulated and functions as a tumor suppressor gene in GBMs, suggesting it may serve as a potential therapeutic target for GBM treatment.
Cross-sectional analysis of arbitrary sections allowing for residual stresses

The method of cross-section analysis for different sections in a structural frame has been widely investigated since the 1960s for determination of sectional capacities of beam-columns. Many handcalculated equations and design graphs were proposed for the specific shape and type of sections in precomputer age decades ago. In design of many practical sections, these equations may be uneconomical and inapplicable for sections with irregular shapes, leading to the high construction cost or inadequate safety. This paper not only proposes a versatile numerical procedure for sectional analysis of beam-columns, but also suggests a method to account for residual stress and geometric imperfections separately and the approach is applied to design of high strength steels requiring axial force-moment interaction for advanced analysis or direct analysis. A cross-section analysis technique that provides interaction curves of arbitrary welded sections with consideration of the effects of residual stress by meshing the entire section into small triangular fibers is formulated. In this study, two doubly symmetric sections (box-section and H-section) fabricated by high-strength steel is utilized to validate the accuracy and efficiency of the proposed method against a hand-calculation procedure. The effects of residual stress are mostly not considered explicitly in previous works and they are considered in an explicit manner in this paper which further discusses the basis of the yield surface theory for design of structures made of high strength steels.
Determination of Ammonia-N in Environmental Water by Air-segmented FIA

A method for determination of ammonia-N in environmental water by air-segmented flow injection analysis using the colour reaction of phenol and sodium hypochlorite with ammonia was described in this paper. When the reaction temperature is $70^{\circ}C$ and the reaction residence time is 5 minutes, a sample frequency of $60-90h^{-1}$ can be achieved. The detection limit and relative deviation are $0.05mg.ml^{-1}$ and 4%, respectively. The recovery of this method is 96 - 110%, and the determination results of the method are greatly agreement with standard colorimetric method.
On the hydraulic axial thrust of Francis hydro-turbine

We present a new method for the computation of hydraulic axial thrust of Francis hydro-turbine. This approach combines the computation of axial thrust on the crown, band and blade by using the numerical simulation and calculation of the pressure distribution outside the crown and band based on the theory of fluid mechanics. The axial thrusts are calculated for different flow conditions with various suction head and flow rate. The calculated results are consistent with that of experimental test on the prototype hydro-turbine, and this combination is proven to be an effective way to assess the magnitude of hydraulic axial thrust for hydro-turbine. Also, the suction head of the turbine is shown to have significant influence on the hydraulic axial thrust.
IL-24 Contributes to Neutrophilic Asthma in an IL-17A-Dependent Manner and Is Suppressed by IL-37

Purpose: Neutrophilic asthma is associated with asthma exacerbation, steroid insensitivity, and severe asthma. Interleukin (IL)-24 is overexpressed in asthma and is involved in the pathogenesis of several allergic inflammatory diseases. However, the role and specific mechanism of IL-24 in neutrophilic asthma are unclear. We aimed to elucidate the roles of IL-24 and IL-37 in neutrophilic asthma, the relationships with IL-17A and the mechanisms regulating neutrophilic asthma progression. Methods: Purified human neutrophils were isolated from healthy volunteers, and a cell coculture system was used to evaluate the function of IL-24 in epithelium-derived IL-17A-dependent neutrophil migration. IL-37 or a small interfering RNA (siRNA) targeting IL-24 was delivered intranasally to verify the effect in a murine model of house dust mite (HDM)/lipopolysaccharide (LPS)-induced neutrophilic asthma. Results: IL-24 enhanced IL-17A production in bronchial epithelial cells via the STAT3 and ERK1/2 signaling pathways; this effect was reversed by exogenous IL-37. Anti-IL-17A monoclonal antibodies reduced neutrophil chemotaxis induced by IL-24-treated epithelial cells in vitro. Increased IL-24 and IL-17A expression in the airway epithelium was observed in HDM/LPS-induced neutrophilic asthma. IL-37 administration or IL-24 silencing attenuated neutrophilic asthma, reducing IL-17A levels and decreasing neutrophil airway infiltration, airway hyperresponsiveness, and goblet cell metaplasia. Silencing IL-24 inhibited T-helper 17 (Th17) immune responses, but not Th1 or Th2 immune responses, in the lungs of a neutrophilic asthma model. Conclusions: IL-24 aggravated neutrophilic airway inflammation by increasing epithelium-derived IL-17A production, which could be suppressed by IL-37. Targeting the IL-24/IL-17A signaling axis is a potential strategy, and IL-37 is a potential candidate agent for alleviating neutrophilic airway inflammation in asthma.
