Lack of Influence of an XRCC3 Gene Polymorphism on Oral Cancer Susceptibility: Meta-analysis

Background: To systematically summarize the association between the X-ray repair cross complementing 3 (XRCC3) gene polymorphism and oral cancer susceptibility by meta-analysis. Materials and Methods: Databases including PubMed, EMbase, CNKI, VIP and WanFang Data were searched to identify case-control studies concerning the association between an XRCC3 gene polymorphism and the risk of oral cancer from the inception to June 2014. Two reviewers independently screened the literature according to the criteria, extracted the data and assessed the quality. Then meta-analysis was performed using Stata 11.0 software. Results: Seven published case-control studies including 775 patients with oral cancer and 1922 controls were selected. Associations between the rs861539 polymorphism and overall oral cancer risk were not statistically significant in all kinds of comparison models (CT vs CC: OR=0.94, 95%CI=0.74-1.18; TT vs CC: OR=0.94, 95%CI=0.64-1.38; dominant model: OR=0.95, 95%CI=0.76-1.18; recessive model: OR=0.94, 95%CI=0.69-1.29; allele T vs C: OR=0.97, 95%CI=0.84-1.11). In the stratified analysis by ethnicity, no significant associations were found among Asians and Caucasians. On stratification by tumor type, no significant associations were found for cancer and oral premalignant lesions. Conclusions: The XRCC3 gene polymorphism was not found to be associated with the risk of oral cancer. Considering the limited quality of the included case-control studies, more high quality studies with large sample size are needed to verify the above conclusion.
Free vibration analysis of continuous bridge under the vehicles

Free vibration analysis for continuous bridge under any number of vehicles is conducted in this paper. Calculation strategy for natural frequency and mode shape is proposed based on Euler-Bernoulli beam theory and numerical assembly method. Firstly, a half-car planar model is adopted; equations of motion and displacement functions for bridge and vehicle are established, respectively. Secondly, the undermined coefficient matrices for wheels, vehicles, intermediate support, left-end support and right-end support are derived. Then, the numerical assembly technique for conventional finite element method is adopted to construct the overall matrix of coefficients for whole system. Finally, natural frequencies and corresponding mode shapes are determined based on iterative method and overall matrix solution. Numerical simulation is presented to verify the effectiveness of the proposed method. The results reveal that the solutions of present method are exact ones. Natural frequencies and associate modal shapes of continuous bridge under different conditions of vehicles are investigated. The influences of vehicle parameters on natural frequencies are also demonstrated.
Assessment of the Cytotoxic and Apoptotic Effects of Chaetominine in a Human Leukemia Cell Line

Chaetominine is a quinazoline alkaloid originating from the endophytic fungus Aspergillus fumigatus CY018. In this study, we showed evidence that chaetominine has cytotoxic and apoptotic effects on human leukemia K562 cells and investigated the pathway involved in chaetominine-induced apoptosis in detail. Chaetominine inhibited K562 cell growth, with an $IC_{50}$ value of 35 nM, but showed little inhibitory effect on the growth of human peripheral blood mononuclear cells. The high apoptosis rates, morphological apoptotic features, and DNA fragmentation caused by chaetominine indicated that the cytotoxicity was partially caused by its pro-apoptotic effect. Under chaetominine treatment, the Bax/Bcl-2 ratio was upregulated (from 0.3 to 8), which was followed by a decrease in mitochondrial membrane potential, release of cytochrome c from mitochondria into the cytosol, and stimulation of Apaf-1. Furthermore, activation of caspase-9 and caspase-3, which are the main executers of the apoptotic process, was observed. These results demonstrated that chaetominine induced cell apoptosis via the mitochondrial pathway. Chaetominine inhibited K562 cell growth and induced apoptotic cell death through the intrinsic pathway, which suggests that chaetominine might be a promising therapeutic for leukemia.

Geosmin and 2-methylisoborneol (2-MIB) are two of the most common taint compounds that adversely affect the quality of aquacultural animals. In the present study, 94% of geosmin and 97% of 2-MIB in suspended growth reactors producing bioflocs (SGRs) with aquaculture waste were removed after inoculation with Bacillus subtilis, significantly higher than that of control SGRs (70% of geosmin and 86.4% of 2-MIB). The lowest concentrations of geosmin and 2-MIB achieved in the effluent of the SGRs were 2.43 ± 0.42 ng/l and 2.23 ± 0.15 ng/l, respectively. The crude protein content of the bioflocs produced in the SGRs was 35 ± 4%. The NH<sub>4</sub>+-N and NO <sub>2</sub>-N concentrations in the effluent of the reactors were 1.13 ± 0.21 mg/l and 0.42 ± 0.04 mg/l, respectively. These results suggest that inoculated with Bacillus subtilis, SGRs have a better performance to reuse the nitrogen in fish waste and to remove geosmin and 2-MIB from the culture water efficiently.
Crack-tip constraint analysis of SENB specimen under creep condition

A higher-order asymptotic C(t)-A2(t)-M approach was used to investigate the crack-tip stress of single edge notched bend (SENB)specimen in a power-law creeping material under plane strain conditions. Comprehensive calculations were made of SENB specimenwith a/W = 0.59 by using higher-order asymptotic C(t)-A2(t)-M approach, higher-order asymptotic C(t)-A2(t) approach and finite elementanalysis; the latter two methods were used to check the constraint significance and the calculation accuracy of C(t)-A2(t)-M approach,respectively. The results show that the stress components qq s obtained from the higher-order asymptotic C(t)-A2(t)-M solution agreedwell with that of finite element analysis, however, which gradually deviated from the results of C(t)-A2(t) approach as the distance faraway from the crack tip for small-scale creep stage.

In this study, 69 lactobacilli isolated from Tibetan Qula, a raw yak milk cheese, were screened for their potential use as probiotics. The isolates were tested in terms of: Their ability to survive at pH 2.0, pH 3.0, and in the presence of 0.3% bile salts; tolerance of simulated gastric and intestinal juices; antimicrobial activity; sensitivity against 11 specific antibiotics; and their cell surface hydrophobicity. The results show that out of the 69 strains, 29 strains (42%) had survival rates above 90% after 2 h of incubation at pH values of 2.0 or 3.0. Of these 29 strains, 21 strains showed a tolerance for 0.3% bile salt. Incubation of these 21 isolates in simulated gastrointestinal fluid for 3 h revealed survival rates above 90%; the survival rate for 20 of these isolates remained above 90% after 4 h of incubation in simulated intestinal fluid. The viable counts of bacteria after incubation in simulated gastric fluid for 3 h and simulated intestinal fluid for 4 h were both significantly different compared with the counts at 0 h (p<0.001). Further screening performed on the above 20 isolates indicated that all 20 lactobacilli strains exhibited inhibitory activity against Micrococcus luteus ATCC 4698, Bacillus subtilis ATCC 6633, Listeria monocytogenes ATCC 19115, and Salmonella enterica ATCC 43971. Moreover, all of the strains were resistant to vancomycin and streptomycin. Of the 20 strains, three were resistant to all 11 elected antibiotics (ciprofloxacin, erythromycin, tetracycline, penicillin G, ampicillin, streptomycin, polymyxin B, vancomycin, chloramphenicol, rifampicin, and gentamicin) in this study, and five were sensitive to more than half of the antibiotics. Additionally, the cell surface hydrophobicity of seven of the 20 lactobacilli strains was above 70%, including strains Lactobacillus casei 1,133 (92%), Lactobacillus plantarum 1086-1 (82%), Lactobacillus casei 1089 (81%), Lactobacillus casei 1138 (79%), Lactobacillus buchneri 1059 (78%), Lactobacillus plantarum 1141 (75%), and Lactobacillus plantarum 1197 (71%). Together, these results suggest that these seven strains are good probiotic candidates, and that tolerance against bile acid, simulated gastric and intestinal juices, antimicrobial activity, antibiotic resistance, and cell surface hydrophobicity could be adopted for preliminary screening of potentially probiotic lactobacilli.

Fumigaclavine C (FC), which is produced by Aspergillus fumigatus, is a conidiation-associated ergot alkaloid with significant medical benefits. However, its application is restricted by low yields from submerged cultures. In this study, the technical feasibility of using molasses as a cost-effective ingredient for FC production in a two-stage culture of A. fumigatus was evaluated. The results indicated that molasses supplementation significantly enhanced FC accumulation by promoting conidiation and up-regulating hydroxymethylglutaryl-CoA reductase activity. Via the optimization of the two-stage process in the presence of molasses, FC production in shake flasks reached 226.9 mg/L, which was approximately three times that in the original medium (75.9 mg/L). The use of molasses as a cost-effective ingredient for FC fermentation was also successfully reproduced in a lab-scale bioreactor system in which the maximum FC production reached 215.0 mg/L. The FC production obtained in this study is the highest ever reported. This increased efficiency will enable large-scale production of FC and extend the application of molasses as a low-cost substrate for producing other conidiation-related secondary metabolites.

High-temperature friction performances of graphite blocks (GBs) and zinc phosphate impregnated graphite blocks (IGBs) were evaluated under various friction temperatures. The surface of IGB exhibited extremely lower average friction coefficient values, that was 0.007 at 400 °C and 0.008 at 450 °C, in comparison to that of GB (0.13 at 400 °C and 0.16 at 450 °C, respectively). The worn surface of IGB in the high-temperature friction test was smoother and more complete than that of GB. The wear under high temperature and load caused the transformation of zinc pyrophosphate to zinc metaphosphate and the formation of a continuous large-area boundary lubrication layer combined with graphite and metallic element on the wear surface. The superior tribology property of IGB could be attributed to the digestion of iron oxides by tribo-chemical reactions and passivation of the exposed dangling covalent bonds. Specifically, the layered structure generated on the IGB wear interface effectively decreased the adhesive forces and prevented the surface from serious damage.
