S-Adensoyl-L-methionine : Protein-carboxyl O-methyltransferase (E.C. 2.1. 1.24, protein methylase Ⅱ) was purified from bovine heart approximately 13,100-fold with a 17% yield by combination of ammonium sulfate precipitate, Sephadex G-75 chromatogrphy, S-adenosyl-L-homoeystein-sepharose 4B chromatography and hydroxyapatite chromatography, and then its properties and physiological roles were investigated. 1. The purified enzyme appeared single protein band by SDS/polyacrylamide gel electrophoresis. 2. The enzyme showed a pH optimum around 5.8-6.4. It was heat labile, inactivated by heat-treatment for 10 minutes at 60℃, and when stored at-20℃ in the presence 10% glycerol, approximately 40% of its activity has been lost in a year. 3. Copper ion (Cu^2+) was a potent inhibitor, the activity being inhibited upto 85% at 2mM and the inhibition was recovered 25% of its activity by addition of 4mM EDTA, and ferrous ion (Fe^2+) also inhibited up to 40% at 2 mM. The other ions did not almost inhibit. 4. The apparent Km value for S-adenosyl-L-methionine was 5×10 ∧(-6) M and kinetic analysis of the enzyme in the presence of 0.125 mM copper ion showed that the nature of the inhibition to the enzyme was uncompetitive, considering that Km and Vmax were decreased. 5. Histone IIA, immunoglobulin G and histone H3 were relatively good substrates for the enzyme. 6. The enzyme activity was completely inhibited by 0.5 mM of p-hydroxymercuribenzoic acid, but all of the activity was recovered in adding 10 mM mercaptoethanol. 7. The molecular weight of the enzyme was 24,000. 8. The methylate calmodulin inhibited about 20% of Ca^2+-calmodulin-induced activation of phosphodiesterase, compared with that of nomethylated calmodulin 9. Methylation rate of calmodulin was constant, independent of Ca^2+ -presence, and Ca^2+ -calmodulin-induced activation of phosphodiesterase by methylated calmodulin was completely inhibited in the presence of 1 mM EGTA. 10. The binding of calmodulin to TNS was inhibited by the methylation of calmodulin. 11. Ventricular plasma membrane was a good substrate for this enzyme and ^3H-CH_3 group was incorporated to 6 protein bands (M. W. : 100,000, 41,000, 22,000, below 14,000 etc) on SDS/polyacrylamide gel electrophoresis. 12. Phosphorylation of plasma membrane by endogenous protein kinase occurred in the protein of molecular weight 100,000 41,000, 22,000, below 14,400 and amomg these molecular weight 22,000 was strongly phosphorylated. 13. Endogenous cAMP-dependent phosphorylation was inhibited by carboxylmethylation of plasma membrane. These results suggest that protein methylase Ⅱ may play some role to regulate Ca^2+ -calmodulin-induced activation of phosphodiesterase and endogenous cAMP-dependent phosphorylation of plasma membrane.

Abstract - In this thesis, we propose a QoS supporting method on wireless ad hoc-based active network for improving reliability. On the loss of route path, the existing DSR routing protocol has several problems with wireless network communication because of decease of data reception rate and increase of network load. The proposed method improves reliability of data transfer by using functions of active network. That is, by using functions of active nodes and active packets, the proposed method supports QoS with reservation transfer method that saves data packets on transfer at intermediate nodes with lost path and then re-transmit the packets after route reconfiguration.

In this paper, we propose a bidirectional mobile Ad Hoc routing protocol based on AODV(ad-hoc on-demand distance vector routing) with QoS(quality of service) support. At presence, wireless mobile communication focuses on how to efficiently support mobility of users more than QoS guarantee. However, in order to satisfy requirement of various applications which have been or will be served, QoS guarantee between end points is a very important issue. The existing AODV routing protocol is implemented to use only BE (Best Effort) service. However, actual wireless mobile environment requires to reduce a waste of communication resources and meet with the real-time change of data paths by setting up routes with QoS support, considering available communication resources at each node in advance. Therefore, in this thesis, we propose an adaptive QoS support method using hello message in order to rapidly meet with the change of available communication resources and to smoothly perform route maintenance. For performance evaluation, we analyzed the average data reception rate. At that time, we used evaluation parameters such as node's mobility-stop time, hello message period, and packet priority, considering the node's mobility and the number of best effort/QoS data flows. As results, we found that with our proposed method the average data reception rate was increased 16.293%, 15.914%, 17.238% for 300 mobility-stop second case and 4.809%, 4.264%, 2.956% for 300 mobility-stop second case.

To overcome the hyperacute immune rejection during pig-to-non-human primates xenotranasplantation, we have produced and bred α-1,3-galactosyltransferase knock-out (GalT —/—) pigs. In this study, the somatic cells and tissues from the GalT —/— pigs were characterized by an analysis of the expression of Galα-1,3-Gal (α-Gal) epitope. Briefly, ear fibroblast cell lines of 19 homozygous GalT —/— pigs were established and cryopreserved. The expression of α-Gal epitope in the cells was measured by fluorescence activated cell sorter (FACS) analysis using BS-I-B4 lectin. Also, the homozygous (GalT —/—) cells and tissues samples were immunostained with BS-I-B4 lectin for analysis of α-Gal epitope expression. The results showed that the expression of α-Gal epitope in GalT —/— cells (0.2 %) were significantly (p< 0.05) down-regulated to the range of cynomolgus monkey fibroblast (0.2 %) cells compared to heterozygous (GalT —/+) (9.3 %) and wild type (GalT +/+) (93.7 %) fibroblast cells. In the immunostaining results, while the expression of α-Gal epitope was detected a partly in GalT —/+ cells and mostly in GalT +/+ cells, it was almost not detected in the GalT —/— cells. Also, immunostaining results from various tissues of the GalT —/— pig showed that the expression of α-Gal epitope was not detectable, whereas various tissues from GalT +/+ pig showed a strong expression of α-Gal epitope. Our results demonstrated that α-Gal epitope expressions from GalT —/— pigs were successfully knocked out to prevent hyperacute immune rejection for further study of xenotransplantation.

Plant-derived multiple monoclonal antibody (mAb) <TEX>$CO17-1A{\times}BR55$</TEX> (<TEX>$mAb^P$</TEX> <TEX>$CO17-1A{\times}BR55$</TEX>) produced in transgenic tobacco plants were cross-pollinated with mAb CO17-1A and mAb BR55. Human anti-colorectal cancer multiple mAb <TEX>$CO17-1A{\times}BR55$</TEX> was cloned using pBI121 vector. Mice were given a single intraperitoneal injection of azoxymethane (AOM) with 10 mg/kg body weight. Starting 1 week after the injection, mice received 2% dextran sulfate sodium (DSS) in the drinking water for 1 week. In addition, the mice were injected intraperitoneal with mAbs dissolved in phosphate buffered saline (<TEX>$100{\mu}g/mouse$</TEX>) twice per week for 4 weeks. Apoptotic cell death, expression of pro-apoptotic proteins, activity of inflammatory cytokines and ERK pathway phosphorylation were assayed by Western blot and TUNEL kit. <TEX>$mAb^P$</TEX> <TEX>$CO17-1A{\times}BR55$</TEX> meaningfully and efficiently suppressed the development of AOM/DSS-induced colorectal inflammation and colorectal tumors, as determined by a reduced activation of inflammatory cytokines, number of colorectal tumor-induced mouse, number of tumor per mouse colon than other mAbs. Cell death by apoptosis was much increased in the <TEX>$mAb^P$</TEX> <TEX>$CO17-1A{\times}BR55$</TEX>-treated tumor compared with negative control. Apoptotic cell death and expression of pro-apoptotic proteins including Bax and cleaved caspase-3 were highest in treatment with <TEX>$mAb^P$</TEX> <TEX>$CO17-1A{\times}BR55$</TEX>. In addition, mAbP <TEX>$CO17-1A{\times}BR55$</TEX> was meaningfully decreased the expression of inflammatory cytokines, including COX-2, iNOS, p50 and p65, but the expression of <TEX>$PPAR{\gamma}$</TEX> was significantly increased compared with AOM/DSS-induced carcinogenesis negative control. Moreover, <TEX>$mAb^P$</TEX> <TEX>$CO17-1A{\times}BR55$</TEX> meaningfully repressed the ERK1/2 phosphorylation in AOM/DSS-induced colorectal tumors. Therefore, our results suggest that multiple <TEX>$mAb^P$</TEX> <TEX>$CO17-1A{\times}BR55$</TEX> have meaningful effects of anti-inflammation related with the anti-carcinogenesis in AOM/DSS-induced colorectal tumor by inhibition of ERK1/2 phosphorylation.

<P>Organic bistable devices fabircated utilizing SnO2 nanoparticles embedded in a poly(methyl methacrylate) (PMMA) polymer layer were formed by using a spin coating method. Transmission electon microscopy images and photoluminescence spectra showed that synethized SnO2 nanoparticles were randomly distributed in the dibutyl ehter solution. Current-voltage (I-V) measurements on the Al/SnO2 nanoparticles embedded in PMMA layer/ITO devices at 300 K showed current bistability due to the existence of SnO2 nanoparticles. Current-time (I-t) results showed the memory retention characteristic of the device. Carrier transport mechanisms of the device are described on the basis of the I-V experimental results and electronic structures.</P>
