        In this study 8 male athletes and 8 female athletes for matches requiring endurance were examined for the purpose of comparison and analysis on the effect of absorption of sugars and effect of absorption of sugars+protein+fats restoration of muscular glycogen after elapse of 4 hours subsequent to closing 90 minutes exercise. (Amount of energy absorbed was same). Examinees were tested with 3 kinds of performance such as CPF Performance whereby they were made to take ① sugars (0.75 g/㎏) and protein (0.1g/㎏) and fats (0.02 g/㎏) after elapse of 1 hour subsequent to long hours of exercise and CHO Performance whereby examines were made to take ② sugars (1g/㎏) alone and Performance whereby they were made to take ③ placebo (which does not contain any sugars, protein or fats). Also the examinees took meals which had same amount of energy and nitrogen during the Performance on the date prior to experiment or on the date of experiment. Women during the exercise showed significantly higher rate of oxidation of fats, basic element of energy than that of men. In CHO Performance and CPF Performance value of blood sugar and density of insulin were significantly higher in comparison with performance with placebo. At such cases there was no difference between different genders. Same kind of phenomena were confirmed with restoration of muscular glycogen. From such fact it could be observed that absorption of fats alone and absorption of sugars+protein+fats in comparison with absorption of placebo after exercise promoted relatively higher level of restoration of muscular glycogen for both men and women.

        류마티스 관절염에 대한 한약의 면역학적 연구동향

        Objective : Rheumatoid arthritis is an autoimmune disease that pathogenesis is not fully understood and one of the most intractable musculoskeletal diseases. The concern in the immunopathogenesis of rheumatoid arthritis has been increased since 1980's and many immunotherapeutic agents including disease-modifying antirheumatic drugs (DMARDs) were developed and became the mainstay of treatment of rheumatoid arthritis. However, the cure of the disease has hardly been achieved. In oriental medicine, rheumatoid arthritis is related to Bi-Zheng(痺證), that presents pain, swelling, andlor loss of joint function as major clinical manifestations, and also known to be deeply involved in suppression of immune function related to weakness of Jung-Ki(正氣). The herbal medicine, empirically used, could be a potential resource of development of new immunotherapeutic agents for rheumatoid arthritis. Methods : We developed a search strategy using terms to include "rheumatoid arthritis and herbal medicine" combined with "Chinese medicine" and/or "Oriental medicine". The search was focused on experimental studies of herbal medicine (January 1999 to May 2004), which is known to have effects on immune function of patients with rheumatoid arthritis. Computerized search used Internet databases including KISS and RISS4U (Korea), CNKI (China), MOMJ (Main Oriental Medicine Journal, Japan), and PubMed. The articles were selected from journals of universities or major research institutes. Results : The literature search for experimental studies on effects of herbal medicine on immunity of rheumatoid arthritis retrieved a total of 21 articles (Korea; 8, China ; 12, Japan ; 1). Of 21 articles, 10 were related to single-drug formula, 2 to drug interaction, and 9 to multi-drug formula. Single-drug formula was mainly used for aqua-acupuncture and researches on active components. Studies of drug interaction emphasized harmony of Ki-Hyul(氣血) and balance of Han-Yeul(寒熱). Multi-drug regimen was mainly found among formulas for Bo-Ki-Hyul(補氣血) and Bo-Sin(補腎). Conclusion : Studies on rheumatoid arthritis were performed both in vitro and in vivo in vitro study, LPS-stimulated splenocytes and synoviocytes were treated with herbal medicine, resulting in proliferation and activation of immune cells and suppression of cytokine activities in vivo study CIA animal model demonstrated that herbal medicine decreased antibody production and improved function of immune cells. In cellular and molecular study herbal medicine showed profound effects on the level of mRNA expression of certain cytokines related to immune function. This study revealed that herbal medicine has significant immune modulatory action and could be used for recovery of immune dysfunction of rheumatoid arthritis patients.

        온도와 광주기 조건이 코드링나방 수컷의 활동력에 미치는 영향

        광주기 16:8(명기:암기) 온도 이상 이하의 실험실 조건에서 코드링나방(Laspeyresia pomonella) 수컷의 활동리듬을 활동력 측정장치로 조사한 바, 소등시에 한번(제1활동기)과 점등시에 한번(제2활동기)의 두 번의 활동최성기를 나타내었다. 제1활동기는 소등 시간 전에 시작되었으며 소등신호에 의해 활동시작시간이 결정되는 것으로 나타났다. 또한 계속조명조건에서는 활동주기가 24시간보다 약간 길어지는 경향이었으며 이는 곤충의 일주활동성의 특성에 부합되는 것으로 나타났다. 광주기 16:8 하에서 부터 까지 8단계의 온도조건을 두어 숫나방의 활동리듬을 조사한 결과 에서는 전혀 활동하지 못하였다. 또한 이하의 온도에서는 제2활동기가 나타나지 않았으며 이하에서는 제1활동기 중 암기에서의 활동이 완전히 억제되었다. 온도범위 하에서는 온도가 상승함에 따라 제2활동기와 제1활동기 중 암기에서의 활동성이 증대되었다. 이러한 온도상승에 따른 제1활동기의 암기에서의 활동력 증대로 인하여 활동 평균시간이 암기쪽으로 이동하였다. 코드링나방의 온도에 따른 평균 활동시간의 변동은 상기한 저온에서의 암기에 활동력 감퇴에 의해 나타나는 것으로 추정된다. The male activity in Laspeyresia pomonella (L.) measured by an activity recording device in New York had two distinct peaks, the first peak at lights-off and the second one at ligts-on signal, under the defined conditions of temperature above and light:dark (LD) 16:8 regime. The activity initiation of the first activity was observed four to six hours prior to the onset of scotophase and seened to be entraind by lights-off cue. Under the continuous photophase (LL) the activity period freeran with a period slightly greater than 24 hours, indicating that the rhythmicity is circadian The activity pattern was measured in eight different temperature conditions ranging from under LD 16:8 regime. No activity was observed at which seems to be temperature threshold for activity. The second peak of activity at lights-on signal disappeared at the temperature below and the activity in scotophase was also suppressed at the temperature lower than . At the temperature range from , as temperature increased the second peak in the morning became larger and the activty in the scotophase was also increased. Because of the activity increase in the scotophase with rising temperature, the mean time of activity shifted towards the scotophase. The shift of the moth male activity period with the change of ambient temperature appears to be due to the suppression of activity under cool temperature (below ) in scotophase.

        Phosphorylation of Hepatitis C Virus RNA Polymerases Ser29 and Ser42 by Protein Kinase C-Related Kinase 2 Regulates Viral RNA Replication

        <P>Hepatitis C virus (HCV) nonstructural protein 5B (NS5B), an RNA-dependent RNA polymerase (RdRp), is the key enzyme for HCV RNA replication. We previously showed that HCV RdRp is phosphorylated by protein kinase C-related kinase 2 (PRK2). In the present study, we used biochemical and reverse-genetics approaches to demonstrate that HCV NS5B phosphorylation is crucial for viral RNA replication in cell culture. Two-dimensional phosphoamino acid analysis revealed that PRK2 phosphorylates NS5B exclusively at its serine residues <I>in vitro</I> and <I>in vivo</I>. Using <I>in vitro</I> kinase assays and mass spectrometry, we identified two phosphorylation sites, Ser29 and Ser42, in the Δ1 finger loop region that interacts with the thumb subdomain of NS5B. Colony-forming assays using drug-selectable HCV subgenomic RNA replicons revealed that preventing phosphorylation by Ala substitution at either Ser29 or Ser42 impairs HCV RNA replication. Furthermore, reverse-genetics studies using HCV infectious clones encoding phosphorylation-defective NS5B confirmed the crucial role of these PRK2 phosphorylation sites in viral RNA replication. Molecular-modeling studies predicted that the phosphorylation of NS5B stabilizes the interactions between its Δ1 loop and thumb subdomain, which are required for the formation of the closed conformation of NS5B known to be important for <I>de novo</I> RNA synthesis. Collectively, our results provide evidence that HCV NS5B phosphorylation has a positive regulatory role in HCV RNA replication.</P><P><B>IMPORTANCE</B> While the role of RNA-dependent RNA polymerases (RdRps) in viral RNA replication is clear, little is known about their functional regulation by phosphorylation. In this study, we addressed several important questions about the function and structure of phosphorylated hepatitis C virus (HCV) nonstructural protein 5B (NS5B). Reverse-genetics studies with HCV replicons encoding phosphorylation-defective NS5B mutants and analysis of their RdRp activities revealed previously unidentified NS5B protein features related to HCV replication and NS5B phosphorylation. These attributes most likely reflect potential structural changes induced by phosphorylation in the Δ1 finger loop region of NS5B with two identified phosphate acceptor sites, Ser29 and Ser42, which may transiently affect the closed conformation of NS5B. Elucidating the effects of dynamic changes in NS5B phosphorylation status during viral replication and their impacts on RNA synthesis will improve our understanding of the molecular mechanisms of NS5B phosphorylation-mediated regulation of HCV replication.</P>

      • Optical Design of Porous ZnO/TiO<sub>2</sub> Films for Highly Transparent Glasses with Broadband Ultraviolet Protection

        <P>We present a design of a bilayer porous film structure on a glass substrate for the highly efficient ultraviolet (UV) protection with high visible-light transparency. To effectively block UVB (280–315 nm) and UVA (315–400 nm), titanium dioxide (TiO2) and zinc oxide (ZnO) are used as absorbing layers having the appropriate coverages in different UV ranges with extinction coefficients, respectively. We show the process of refractive index (RI) matching by controlling porosity (<SUB>Pr</SUB>). Effective RIs of porous media with TiO2 and ZnO were calculated based on volume averaging theory. Transmittances of the designed films with different effective RIs were calculated using rigorous coupled-wave analysis method. Using admittance loci method, the film thickness was optimized in center wavelengths from 450 to 550 nm. The results show that the optimal design provides high UV shielding performance at both UVA and UVB with high transparency in the visible range. We also analyze electrical field distributions in each layer and angle dependency with 3D HSV color map.</P>

        미국 북서부지역에 발생하는 서부양벚과실파리의 발생 월동 후 발생 동태에 관한 연구

        서부양벚과실파리(Rhagolettis indifferens Curran)은 미국 북서부지역 및 California 북부에서 재배되는 단체리(Prunus avium)에 가장 큰 피해를 주는 해충이다. 체리의 수출을 위한 식물검역에서 Zero Tolerance의 규제를 받고 있는 이 해충의 방제를 위해 농가에서는 월동 후 우화 시점부터 지속적으로 약제를 살포하고 있으며, 살충제 처리의 적기를 예측할 수 있는 모형이 절실히 필요한 실정이다. 본 연구는 서부양벚과실파리를 대상으로 월동 후 성충의 우화 및 발생시기, 유충의 밀도변화, 번데기의 용화시기 및 밀도변화 등을 정량적으로 추적하여 개체군 밀도의 경시적 변동과 월동 후 우화시기를 예측하는 모형 검정의 기초자료로 사용코자 수행하였다. 이를 위하여 황색끈끈이트랩, 우화케이지, 용화트랩 등을 이용하여 실제 과원에서 각 태별 발생경과를 경시적으로 조사하였으며, 체리 과실이 달리는 시기부터 일정 간격으로 과실을 수거하여 시기별 과실 내부의 유충 수를 조사하였고, 실내에서 용화케이지를 이용하여 용화시기를 조사하였다. 그 결과 월동 성충의 우화는 5월 중순에 시작하여 6월 초순에 정점에 도달하였고, 6월 중순부터 7월 상순까지 과실 당 1마리 이상의 유충이 존재하였다. 7월 중순에 번데기의 수가 정점에 도달하였으며, 월동 중에 토양습도 등의 조건에 따른 번데기의 발육속도 및 생존율을 측정하기 위해 대량의 번데기를 확보하였다. 이 연구에서 얻어진 자료에서 나타난 과실파리의 월동 후 개체군 밀도변동과 용화시기를 Song et al.(2003)의 모형에서 예측한 결과와 비교한 결과 모형에 의해 예측된 발생일과 실측 발생일과는 1~2일 차이로 매우 정확하게 예측이 되었다. 이로 미루어 볼 때 본 연구에서 획득한 포장 실측자료는 누적 우화일, 유충의 발육단계, 산란일 등 다른 중요한 생물학적 사건을 예측하는 모형의 정확도 검정에도 잘 활용될 수 있을 것으로 생각된다. The western cherry fruit fly, Rhagoletis indifferens Curran (Diptera: Tephritidae), is the most important pest of cultivated cherries in the Pacific Northwest area of the United States, being widely distributed throughout Oregon, Washington, Montana, Utah, Idaho, Colorado and parts of Nevada. The control of R. indifferens has been based on calendar sprays after its first emergence because of their zero tolerance for quarantine. Therefore, a good prediction model is needed for the spray timing. This study was conducted to obtain the empirical population dynamic information of R. indifferens after overwintering in the major cherry growing area of the Pacific Northwest of the United States, where the information is critically needed to develop and validate the prediction model of the fruit fly. Adult fly populations were monitored by using yellow sticky and emergence traps. Larvae growth and density in fruits were observed by fruit sampling and the pupal growth and density were monitored by pupal collection traps. The first adult was emerged around mid May and a large number of adults were caught in early June. A fruit had more than one larva from mid June to early July. A large number of pupae were caught in early July. The pupae were collected in various period of time to determine the effect of pupation timing and the soil moisture content during the winter. A series of population density data collected in each of the developmental stage were analyzed and organized to provide more reliable validation information for the population dynamic models.
