        MicroRNA-27a Inhibits Cell Migration and Invasion of Fibroblast-Like Synoviocytes by Targeting Follistatin-Like Protein 1 in Rheumatoid Arthritis

        Fibroblast-like synoviocytes (FLS) with aberrant expression of microRNA (miRNA) are critical pathogenic regulators in rheumatoid arthritis (RA). Previous studies have found that overexpression or silencing of miRNA can contribute to the development of miRNA-based therapeutics in arthritis models. In this study, we explored the effects of miR-27a on cell migration and invasion in cultured FLS from RA patients. We found that miR-27a was markedly downregulated in the serum, synovial tissue, and FLS of RA patients. Meanwhile, the expression of follistatin-like protein 1 (FSTL1) was upregulated, which suggests that FSTL1 plays a key role in RA development. The results of a Transwell assay showed that miR-27a inhibited FLS migration and invasion. However, miR-27a inhibition promoted the migration and invasion of FLS. In addition, the down-regulated expression of matrix metalloproteinases (MMP2, MMP9, and MMP13) and Rho family proteins (Rac1, Cdc42, and RhoA) was detected after treatment with miR-27a in RA-FLS by quantitative reverse transcription-PCR and western blot analysis. Then, a luciferase reporter assay validated that miR-27a targeted the 3-untranslated region (3'-UTR) of FSTL1. Moreover, miR-27a caused a significant decrease of FSTL1. In addition, the expression of TLR4 and $NF{\kappa}B$ was inhibited by miR-27a but increased by FSTL1 overexpression. In conclusion, we found that miR-27a inhibited cell migration and invasion of RA-FLS by targeting FSTL1 and restraining the $TLR4/NF{\kappa}B$ pathway.

        Analysis of 12 Chinese Patients with Proline-to-Leucine Mutation at Codon 102-Associated Gerstmann-Sträussler-Scheinker Disease

        Background and Purpose Gerstmann-Sträussler-Scheinker disease (GSS) with a prolineto- leucine mutation at codon 102 (P102L) in the PRNP gene is the most frequently reported GSS subtype worldwide. This study aimed to determine the epidemiological, clinical, genetic, and laboratory characteristics of 12 Chinese patients with P102L-associated GSS (henceforth P102L GSS). Methods The enrolled P102L GSS cases were analyzed according to the diagnostic criteria for Creutzfeldt-Jakob disease (CJD) issued by the China National Health Commission. Results The median onset age was 50 years (range 34 to 67 years) and sex ratio was 1:2 (males:females). Most patients displayed more than one foremost symptom. Movement symptoms were frequently reported (9 of the 12 cases), followed by rapidly progressing dementia (7 cases), mental problems (5 cases), and slowly progressing dementia (2 cases). Almost all cases displayed more sporadic CJD (sCJD)-associated neurological symptoms and signs as time progressed. Five (45.5%) of 11 cases were cerebrospinal fluid 14-3-3 positive, and 2 (25%) of 8 cases exhibited periodic sharp wave complexes in electroencephalograms. MRI abnormalities were detected in all 11 of the scanned patients. Methionine homozygous genotype at codon 129 (M129M) and glutamic acid homozygous at codon 219 (E219E) homozygosity was present in 11 cases, while 1 case was M129M homozygous and glutamic acid/lysine heterozygous at codon 219 (E219K) heterozygous. Ten of the 12 cases recalled a disease-related family history during the clinical interviews. The median survival from symptom onset of the seven dead cases was 16 months (range 10 to 44 months). Patients showing the sCJD phenotype (rapidly progressing dementia) appeared to be associated with a shorter survival time. Conclusions The indistinguishable clinical features of P102L GSS patients with sCJD, especially in the early stage, support the importance of PRNP testing for diagnosing GSS.

        Influence of track irregularities in high-speed Maglev transportation systems

        Track irregularities of high-speed Maglev lines have significant influence on ride comfort. Their adjustment is of key importance in the daily maintenance of these lines. In this study, an adjustment method is proposed and track irregularities analysis is performed. This study considers two modules: an inspection module and a vehicle-guideway coupling vibration analysis module. In the inspection module, an inertial reference method is employed for field-measurements of the Shanghai high-speed Maglev demonstration line. Then, a partial filtering, integration method, resampling method, and designed elliptic filter are employed to analyze the detection data, which reveals the required track irregularities. In the analysis module, a vehicle-guideway interaction model and an electromagnetic interaction model were developed. The influence of the measured line irregularities is considered for the calculations of the electromagnetic force. Numerical integration method was employed for the calculations. Based on the actual field detection results and analysis using the numerical model, a threshold analysis method is developed. Several irregularities modalities with different girder end's deviations were considered in the simulations. The inspection results indicated that long-wavelength irregularities with larger girder end's deviations were the dominant irregularities. In addition, the threshold analysis of the girder end's deviation shows that irregularities that have a deviation amplitude larger than 6 mm and certain modalities (e.g., M- and N-shape) are unfavorable. These types of irregularities should be adjusted during the daily maintenance.

      • Clinical Outcomes of Downregulation of E-cadherin Gene Expression in Non-small Cell Lung Cancer

        Objective: To investigate the promoter methylation status of the E-cadherin gene in non-small cell lung cancer (NSCLC) and its association with clinical pathological parameters, and to explore the relationship between downregulation of E-cadherin gene expression and the methylation status of its promoter region. Methods: Nested methylation-specific PCR was performed to examine CpG methylation within the 5' CpG island of the E-cadherin gene in lung cancer and para-cancerous tissue from 37 patients with primary non-small cell lung cancer. Quantitative real-time PCR was performed to measure the level of E-cadherin mRNA. Results: Of thirty-seven cases, 12 (32.4%) samples showed aberrant CpG methylation in tumor tissues compared with the corresponding normal tissues. In addition, a reduction in E-cadherin mRNA levels was observed in 11 of the 12 (91.7%) tumor tissues carrying a methylated E-cadherin gene. However, only 10 (43.5%) cases displayed reduced mRNA levels in tumor tissues from the remaining 23 cases (excluding 2 samples from which mRNA was unavailable) without methylation events. Downregulation of E-cadherin gene expression significantly correlated with the promoter methylation status of this gene. Conclusion: These results provide strong evidence that the methylation status of E-cadherin gene contributes to a reduction in the expression of E-cadherin mRNA, and may play a role in the development and progression of NSCLC.

        New Hydrophobic Charge-induction Resin with 2-mercaptoimidazole as the Ligand and Its Separation Characteristics for Porcine IgG

        New hydrophobic charge-induction chromatography(HCIC) resin coupled with 2-mercaptoimidazole(MI) as the ligand was prepared and used to purify IgGfrom porcine plasma. The cellulose matrix was activatedby divinylsulfone (DVS), and was then coupled with MI asthe functional ligand. The reaction conditions wereoptimized as pH: 11, volume ratio of DVS to matrix: 1.0,reaction time: 4 h. The ligand density reached about 100μmol/mL gel. The adsorption isotherms of porcine IgGwas determined at different pH values, and high saturatedadsorption capacities of 78.02 mg IgG/mL gel were foundat pH 8. The adsorption of IgG showed a typical pHdependentproperty of HCIC, and the adsorption capacitydecreased significantly in acidic conditions. The preparedresin was used to separate IgG from porcine plasma. Afterprecipitating the fibrinogen by salting-out, the supernatantwas loaded onto the column at pH 7 and the elution pHwas optimized. The results indicated that acidic elution pHwas necessary to recover the IgG. The purity of IgG in theelution fractions was in the range of 81 ~ 90%, whichdemonstrated that HCIC with the new ligand showed theexcited separation performs and is a potential effectivetechnique to purify IgG from the complex feedstock.

      • Visual Saliency Detection via Unsupervised Sparse Learning

        In this paper, a visual saliency detection method with unsupervised sparse learning is proposed. To achieve this goal, the information theoretic perspective of term frequency - inverse document frequency (TF-IDF) measure is introduced to learn a compact dictionary for salient components detection. Experimental results on benchmark dataset suggest that the proposed method works effectively for visual Saliency detection.

      • Overexpression of Cyclooxygenase-1 Correlates with Poor Prognosis in Renal Cell Carcinoma

        The aim of this study was to evaluate expression of COX-1 in renal cell carcinoma (RCC) and its prognostic value. mRNA of COX-1 was detected in 42 paired RCC and adjacent normal tissues with quantitative realtime polymerase chain reaction (qRT-PCR). Expression of COX-1 was also evaluated in 196 RCC sections and 91 adjacent normal tissues with immunohistochemistry. Statistical analysis was performed to assess COX-1 expression in RCC and its prognostic significance. The results of qRT-PCR showed mRNA levels of COX-1 in RCC tissues to be significantly higher than that in adjacent normal tissues (p < 0.001). Immunohistochemical assays also revealed COX-1 to be overexpressed in RCC tissues (p < 0.001). Statistical analysis demonstrated high expression of COX-1 was correlated with tumour size (p = 0.002), pathological stage (p = 0.003), TNM stage (p = 0.003, 0.007, 0.027, respectively), and tumour recurrence (p < 0.001). Survival analysis indicated patients with high expression of COX-1 had shorter survival time (p < 0.001), and COX-1 was an independent predictor. This is the first study to reveal overexpression of COX-1 in RRC and point to use as a prognostic marker in affected patients.
