<P><B>Abstract</B></P><P>We investigated the mechanism for the induction of a chemokine, IL-8, by bacterial flagellins in the human alveolar type II epithelial cell line, A549. Bacterial flagellin induced expression of IL-8 mRNA and protein in dose- and time-dependent manners. IL-8 expression was inhibited by nystatin (a lipid rafts inhibitor) but not by chlorpromazine (a clathrin-coated pits inhibitor). Interestingly, Toll-like receptor 5 (TLR5) recognizing flagellins was found in the intracellular compartment of A549 but rarely on the cell surface. Flagellin-induced IL-8 expression appears to be mediated through TLR5 as determined by <I>in vitro</I> transient transfection experiment in HEK-293 cells expressing TLR5 using a reporter gene construct containing IL-8 promoter. IL-8 expression was attenuated by inhibitors for protein kinase C (PKC) and mitogen-activated protein (MAP) kinases. Furthermore, NF-κB and NF-IL6 transcription factors played an important role in the flagellin-induced IL-8 gene expression in A549 cells. Collectively, these results suggest that flagellin-induced IL-8 expression requires formation of lipid rafts, intracellular TLR activation, and subsequent activation of PKC and MAP kinases leading to the activation of the transcription factors NF-κB and NF-IL6 in human alveolar type II epithelial cells.</P>

<P><B>Abstract</B></P> <P>The insulator-metal transition (IMT) in vanadium dioxide (VO<SUB>2</SUB>) which occurs above room temperature (67 °C) is highly sensitive to atomic defects caused by oxygen stoichiometry. The strained growth and the degree of oxygen deficiency in VO<SUB>2</SUB> epitaxial films result in lowering of transition temperature below room temperature as well as the broadening of transition parameters such as transition width and hysteresis width, which limit its application potential. Here we demonstrate the growth of highly oriented strain-relaxed VO<SUB>2</SUB> thin films on (001)-oriented TiO<SUB>2</SUB> substrates at various oxygen partial pressures, exhibiting the narrow transition and hysteresis width. The cross-sectional transmission electron microscopy and x-ray diffraction analyses of the films reveal the highly oriented growth of insulating monoclinic VO<SUB>2</SUB>. The IMT parameters associated with temperature-dependent phase transition vary with the oxygen partial pressure used during the deposition. The presence of multiple and mixed valence states of vanadium in the films was confirmed by Raman and XPS analyses. We have achieved a narrow transition width (2.3 °C) and hysteresis width (1.2 °C) through controlling the oxygen stoichiometry during the growth of VO<SUB>2</SUB>/TiO<SUB>2</SUB> films.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Highly oriented epitaxial VO<SUB>2</SUB> films were grown on (001)-oriented TiO<SUB>2</SUB> substrate. </LI> <LI> Insulator to metal transition (IMT) parameters of the VO<SUB>2</SUB> film varies with the oxygen stoichiometry. </LI> <LI> IMT in VO<SUB>2</SUB> varied depending on the degree of crystallinity and microstructure of the films. </LI> <LI> The VO<SUB>2</SUB> film grown at high deposition oxygen pressure showed a sharp transition with a narrow hysteresis width. </LI> </UL> </P>
The Numerical Study on the Flow Characteristics in Two-Dimensional Moonpool in Waves

The objective of this study is to examine the nonlinear fluid characteristics near and inside a moonpool in various sea conditions. We estimate the flow of the free surface in a moonpool taking into account the viscosity effect and the hydrodynamic forces that affects a moonpool and hull through CFD calculations. The comparison of horizontal forces per wave length shows that the hydrodynamic force is greater for the long wave length than short wave length, and the greatest hydrodynamic force acts on the moonpool when the wave length is equal to the ship’s length. The horizontal force decreases as the wave amplitude decreases, and the hydrodynamic force acting on the moonpool in λ=LBP is 10 times that in λ=LBP/3 The free surface demonstrates the piston mode, in which it oscillates up and down while remaining essentially flat, and the rise of the free surface level increases as the wave length increases. We can assume that the hydrodynamic force acting on the moonpool increases owing to the effect of a strong vortex for λ=LBP and owing to the rise of the free surface level for λ=LBP×2.
Risk Factors for Postoperative Recurrence in Korean Patients with Crohn’s Disease

Background/Aims: A considerable number of patients with Crohn’s disease still need intestinal resection surgery. Postoperative recurrence is an important issue in Crohn’s disease management, including the selection of high-risk patients. Eastern Asian patients showed several differences from Caucasian patients. Therefore, we investigated the postoperative surgical recurrence outcome and identified risk factors in Korean patients. Methods: Clinical data of 372 patients with Crohn’s disease who underwent first intestinal resection between January 2004 and August 2014 at 14 hospitals in Korea were retrospectively reviewed. Results: Over the follow-up period, 50 patients (17.1%) showed surgical recurrence. The cumulative surgical recurrence rate was 6.5% at 1 year and 15.4% at 7 years. Age under 16 (p=0.011; hazard ratio [HR], 5.136; 95% confidence interval [CI], 1.576 to 16.731), colonic involvement (p=0.023; HR , 2.011; 95% CI, 1.102 to 3.670), and the presence of perianal disease at surgery (p=0.008; HR, 2.239; 95% CI, 1.236 to 4.059) were independent risk factors associated with surgical recurrence. Postoperative thiopurine treatment (p=0.002; HR, 0.393; 95% CI, 0.218 to 0.710) was a protective factor for surgical recurrence. Conclusions: Among the disease characteristics at surgery, younger age, colonic location, and perianal lesions were independent risk factors for surgical recurrence. Postoperative thiopurine treatment significantly reduced the incidence of surgical recurrence.

Cellular Uptake Properties of the Complex Derived from Quantum Dots and G8 Molecular Transporter

The biotin-attached G8 molecular transporter (5) was synthesized and used together with quantum dots in preparing the complexes (QD-MT). The QD-MT complexes were studied in terms of the cellular uptake and the internalization mechanism in live HeLa cells with the aid of various known endocytosis inhibitors. It has been concluded that the QD-MT complex is internalized largely by macropinocytosis. The mouse tissue distribution of the QD-MT complex by i.p. and i.v. routes showed some organ selectivity and a good ability to cross the BBB.
