        Disruption of rsmA Gene of Pectobacterium carotovorum subsp. carotovorum LY34 and Effect on Pathogenicity

        The rsmA gene was cloned from soft-rot bacterium Pectobacterium carotovorum subsp. carotovorum LY34 (Pcc LY34), and its role in pathogenicity was investigated by marker exchange mutagenesis. From a cosmid library of Pcc LY34 genomic DNA, a positive clone carrying the rsmA gene was selected, and the gene was cloned by polymerase chain reaction (PCR) amplification. The gene is 186 bp in size and encodes a protein of 62 amino acids with a predicted molecular mass of 6,839 Da. The calculated pI of the RsmA is 8.16. The phylogenetic tree showed that the RsmA of Pcc LY34 appeared genetically identical to the CsrA of Pectobacterium atrosepticum SCRI1043 (100% identity) and similar to the CsrA of Yersinia pestis KIM10+(98.3%). The gene was disrupted by the $Km^r$ gene, and the cells became mutated (i.e., $RsmA^-$ mutant). The pathogenicity test revealed that the disease rating of the $RsmA^-$ mutant only differed slightly from that of the wild type on a slice of potato tuber and a Chinese cabbage stalk. These results suggest that RsmA is not an essential factor for the pathogenicity of Pcc LY34 and that the rsmA gene of Pcc LY34 is not completely derepressed in the $RsmA^-$ mutant for virulence-related genes, contrary to the results of Erwinia carotovora subsp. carotovora $RsmA^-$ mutant, which proved hypervirulent for celery petioles. These results showed that the microenvironmental conditions of the host and/or strain of pathogen are important for the coordination of virulence gene expression.

        Effect of Developmental Lead Exposure on the Expression of Hippocampal NMDA Receptor Subunit mRNA

        목적: in vivo 및 vitro에서 해마 신경세포의 발생단계별 NMDA 수용체 아단위 mRNA 발현에 대한 연 폭로 영향을 알아보고자 하였다. 방법: 흰쥐 해마 신경세포의 발생단계별 NMDA 수용체 NR2A, NR2B 아단위 mRNA 발현에 대한 연의 영향은 정상군과 연 폭로군의 출생 후 7일, 14일, 22일 흰쥐의 해마에서 in situ hybridization으로 mRNA 발현 정도를 densitometer로 측정하여 비교하였고, 연과 NMDA 의 세포독성은 해마 신경세포 일차배양 후 도립현미경을 이용한 형태학적인 관찰과 LDH 활성도를 이용하여 측정하였다. 결과: 연 과 NMDA 에 의한 세포독성에 대한 in vitro 실험에서 형태학적 소견과 LDH 활성도에서 해마 미성숙 신경세포와 성숙 신경세포사이의 차이가 있었으므로, 연과 NMDA 독성효과는 해마 신경세포의 발달 단계에 따라 차이가 있다. 정상군의 해마에서의 NR2A mRNA 발현은 출생 후 연령이 증가함에 따라 점진적으로 증가하였으나, NR2B mRNA 발현은 출생 후 연령이 증가함에 따라 점진적으로 증가하였으나 NR2B mRNA 발현은 연령의 증가에 따른 변화가 없었다. 연 폭로에 희한 NR2A mRNA 발현은 유의하게 감소하였으나(p<0.05), NR2B mRNA 발현은 변화가 나타나지 않았다. 만성적 연 폭로는 NR2A를 포함하는 NMDA 수용체를 감소시킬 수 있음을 알 수 있다. 결론: 연은 해마신경세포의 발생단계에서 NMDA 수용체 아단위 특히 NR2A mRNA 발현의 변화를 야기하여 시냅스 신호 전달에 영향을 나타냄을 알 수 있었다.       • Impact of Interferon-Based Treatment on Quality of Life and Work Related Productivity from the Korean Cohort in the MOSAIC Study

        Aims: Chronic Hepatitis C Virus (HCV) infection increases the risk for progressive liver disease, hepatocellular carcinoma and negatively impacts the patient’s quality of life. HCV treatment is evolving with direct acting antivirals but IFN based therapy has been the standard of care for many years and remains available in some countries. The MOSAIC study aims to characterize patients with chronic HCV infection and assess the impact of IFN-containing treatment on health-related quality of life, work related productivity and health care utilization. Methods: MOSAIC is an international prospective multicenter observational study that has been conducted in 20 countries. Consecutive patients with chronic HCV infection were enrolled and those who initiated an IFN based regimen were prospectively followed for 48 weeks. We report results from the Korean cohort Results: 100 patients were enrolled: 86 were treatment naïve and 14 were treatment experienced. 33 patients initiated an IFN based regimen: 6 patients started IFN + RBV, 26 patients started Peg-IFN + RBV, none started Peg-IFN + RBV + DAA and 1 patient received other treatment. Among the treated cohort, demographic and disease characteristics were the following: the mean age was 54.5 years; 14 patients were male. 14 had minimal or no fibrosis, 2 portal fibrosis, 3 bridging fibrosis and 6 patients suffered from cirrhosis. HCV Genotype distribution was as follows: genotype 1: 11; genotype 2: 19 and genotype 3: 3. Table 1 describes the results at baseline and changes over 4, 12 and 48 weeks and end-of-treatment (EOT) for the quality of life and work productivity outcome measures (EQ-5D-5L, HCV-PRO and WPAI). Conclusions: Results from the Korean cohort of the MOSAIC study show a moderate trend for deterioration of health-related quality of life and work productivity associated with IFN based treatment for patients with chronic HCV infection during treatment period.         Quantitative comparison of mRNA expression of glucosyltransferase(GTF) between xylitol-resistant(X^(R)) and xylitol-sensitive(X^(s)) mutans streptococci

        Since the long-term exposure of mutans streptococci to xylitol is known to select for xylitol-resistant(X^(R)) natural mutants, the occurrence and survival of such X^(R) strains were performed in batch culture methods. The aim of the study was to compare the differentiation and quantification of mRNA expression of the gtf genes of X^(R) and X^(S) mutans streptococci. Using a real-time reverse-transcription polymerase chain reaction, the expression of each gtf was determined. In X^(R) strains, the relative levels of transcription of gtfB and gtfC were decreased while that of gtfD was increased, suggesting the presence of independent promoters. It also suggested that mutation related to production of glucosyltransferase occurred under the exposure of xylitol could explain the caries-preventive mechanisms of xylitol. 자일리톨에 장기간 노출된 mutans streptococci는 자일리톨에 내성이 발현되어 자일리톨 내성균주가 생성된다고 알려져 있다. 본 연구의 목적은 mutans streptococci에서 자일리톨 내성균주와 감성균주의 gtf 유전자 발현량을 각각의 유전자별로 정량적으로 분석하고 비교하는 것이다. 실시간 역전사 중합효소연쇄반응법을 이용하여 각각의 gtf 발현을 조사한 결과 gtfD는 증가한 반면. gtfB와 gtfC는 감소하였는데 이는 각 유전자의 독립된 조절기전이 존재함을 보여주는 것이다. 또한 자일리톨에 노출된 mutans streptococci에서의 glucosyltransferase와 연관된 유전자변형이 자일리톨의 치아우식증 예방효과의 작용기전 중 하나임을 알 수 있었다.

        여성노인의 저항운동 시 세트구성 방법의 차이가 Myokine에 미치는 영향

        연구목적: 본 연구는 여성노인을 대상으로 저항운동 시 사용되는 set구성 방법에 중 ascending set, descending set, drop set를 각각 적용시켜 Myokine의 변화를 알아보고자 하였다. 연구방법: 연구대상은 노인복지시설을 이용하고 있는 만 65-72세 사이의 여성노인 32명을 대상으로 ascending set, 8명, descending set 8명, drop set 8명, control group 8명으로 구분하였다. 자료처리는 반복측정에 의한 이원변량분석(two-way ANOVA with repeated measure)을 사용하였으며, 유의도는 .05로 하였다. 결론: 운동프로그램은 12주간 주 3회 실시하였으며, 신체구성과 IL-15, BDNF, VEGF, Irisin의 변화를 관찰하였다. 연구결과 신체구성의 체중, 체지방률, 제지방량 모두 유의한 차이가 없었다. IL-15와 BDNF는 운동 전·후의 비교결과 Descending set와 Drop set에서 운동 후 유의하게 증가하였고, 집단 간의 차이에서 Descending set와 Drop set 집단이 대조집단에 비하여 높게 나타났다. VEGF는 운동 전·후의 비교결과 Descending set와 Drop set 집단이 유의하게 증가하였으며, 집단 간의 차이에서 Descending set와 Drop set가 Ascending set와 control group 보다 높게 나타났다. Irisin은 집단 간 차이는 없었지만 Ascending, Descending, Drop set집단에서 운동 전에 비하여 운동 후 유의한 차이로 증가하였다.. Purpose: The purpose of this study is to identify the change of Myokine by applying ascending set, descending set, and drop set which are set composition used during elderly women’s resistance training. Method: 32 participants aged 65 to 72 who were users of welfare facilities for the aged were divided into 4 groups. They were ascending set group of 8, descending set group of 8, drop set group of 8, and control group of 8. The training program was conducted 3 times a week for 12 weeks and the change of body composition, IL-15(interleukin-15), BDNF(brain derived neurotrophic factor), VEGF(Vascular endothelial growth factor) and Irisin was observed. Two-way ANOVA with repeated measure was used for data processing. Result: The result of this study is as follows. Weight, %fat, and LBM(lean body mass) without fat showed no significant change. IL-15 and BDNF increased significantly after training in Descending set group(p<.05; p<.01)and Drop set group(p<.01; p<.05). VEGF significantly increased in descending set group(p<.01) and drop set group(p<.05). Among groups, VEGF was higher in descending set and drop set groups than ascending set and control groups. Irisin showed no difference between groups, but it showed significant increase after training in ascending(p<.05), descending(p<.05), and drop set groups(p<.05).

        신약 헬라어 본문 분석 방식의 새로운 연구

        The New Testament Greek is often a compulsory pre-requisite course in theological seminary education. Although it becomes heavy burden to most students who take the course, spending much time to learn to read the NT in Greek, most of them fail to maintain their Greek after graduation. I think we need to advance some useful tool for Greek study, particularly from the pedagogical point of view, such as a syntactic-semantic arranged method contributed by Johannes Louw, Douglas Stuart, Walter Kaiser, Gordon Fee, William Mounce, and others. These sorts of methods differ from the traditional way of interpretation, the grammatical-historical method, in terms of how to approach the text. The texts arranged for the purpose of visualization, letting the reader to see the text with much clarity in terms of grammatical function of each phrase and their connections between them. The visualized arrangement itself is applied from the area of syntax and semantics in modern linguistics. There are various types of syntactic arrangements of the Greek text. Each pattern has its own principles and rules to be arranged in certain ways. I have compared, in this paper, those models of syntactic arrangements to my model, the Kim's diagram. For the purposes of this paper, I have posed several questions to my students in my classes, regarding their experiences on the syntactic-analytic arrangement of the Greek Bible (SAGB) and their practical uses of both the method and its materials, analyzing their questions in order to show up how much this type of study to be needed for the student

        Q-행렬에 근거하는 치유교습 순서

        이 연구는 인지진단 결과로 주어지는 미성취 인지요소의 치유교습 순서에 Q-행렬이 어느 정도로 어떻게 결정하는지 탐색하였다. Tatsuoka(2009)가 소개한 인접행렬, 도달행렬, 그리고 영향도표를 이용하여 Q-행렬에 내재하는 인지요소 간의 인과관계를 치유교습의 순서로 개념화하였다. 이 연구 결과로 제안한 Q-행렬의 치유교습 순서 기여도는 그 행렬에 포함된 K개의 인지요소 간 있을 수 있는 모든 인과관계의 수에 대한 실제 Q-행렬에 내재하는 인과관계 수의 비율로 정의하였다. 분석 대상 Q-행렬은 이 이론적 배경에 사용된 예시적 Q-행렬과 실제 Q-행렬 2개다. 후자는 성태제(2018) 연구와 박세진·이현숙(2018) 연구에서 인용하였다. 연구 결론으로서 Q-행렬은 치유교습 순서 결정을 위한 보편적 출발점이 되며; 지식의 위계가 분명한 검사일수록 Q-행렬의 치유교습 순서 기여도는 높을 것이며; 인지진단 결과로 주어지는 인지상태는 Q-행렬의 가정과 충돌할 수 있음 등이 논의되었다. This study was to investigate how and how much does a Q-matrix contribute in deciding the sequence of remedial instruction of the unattained attributes being identified from cognitive diagnostic assessment. Applying the adjacency matrix, reachability matrix, and influence diagram introduced in cognitive assessment context by Tatsuoka (2009), the sequence was defined as the causal relationship among attributes forming a Q-matrix. Three Q-matrices were gone under analysis. They were an example one, one from Sung’s(2018) study, and the other from Park & Lee’s(2018) study. The results were discussed into three conclusions. Firstly, the Q- matrix could provide a objective starting point for a whole sequence of remedial instruction; secondly, the more hierarchical a Q- matrix was the more it helped deciding sequence of remedial instruction; thirdly, knowledge states from cognitive diagnosis might contradict the causal relationship assumptions implied by the Q-matrix.
