        Isolation and Charaterization of Bioactive Peptides from Hwangtae (yellowish dried Alaska pollack) Protein Hydrolysate

        Hwangtae, dried Alaska pollack, is a major storage product in the fish processing industry. Hwangtae is prepared by removing the internal organs and drying outdoors during the cold witner months by allowing it to thaw during the daytime and re-freeze at night under sub-zero ($-10^{\circ}C$) conditions and gradually dry from December until the next April for around 5 months from Myungtae. In this study, ground Hwangtae was hydrolyzed using two proteolytic enzymes (pepsin and alcalase) which produced five soluble active peptides from Hwangtae (yellowish dried Pollack, Theragra chalcogramma) protein. Two different peptides with strong antioxidative activity were isolated from the hydrolysate using consecutive chromatographic methods of Sephadex G-25 gel, ion-exchange chromatography on a Sepharose-Sephadex C-25 gel, and high-performance liquid chromatography. The isolated peptides, APO1 and APO2, were composed of 16 and 13 amino acid residues, respectively. Both peptides contained a Gly residue at the C-terminus and the repeating motif Gly-Pro-Hyp. The peptide with a molecular weight less than 1,000 Daltons (APACE) obtained from enzymatic hydrolysates of Hwangtae exhibited the highest ACE inhibitory activity. The APACE peptides was composed of 4 amino acid residues (Gly-Leu-Leu-Pro). These results suggest that Hwangtae hydrolysates could be a good source of peptides with ACE inhibitory activity. Biochemical analysis indicated that two 70 kDa peptides (APG1 and APG2) isolated from the hydrolysate had gelatinoytic activity, which was shown to be a calcium dependent protease type as showed by gelatin SDS PAGE.

        Fish protein hydrolysates (FPHs) with different degrees of hydrolysis by treatment with alcalase, pronase, flavourzyme and trypsin and isolated peptide were prepared from Hwangtae (yellow dried pollack, Theragra chalcogramma). Hwangtae protein hydrolysate was fractionated according to the molecular weight into six major types of APO1 (1.3 kDa), APO2 (1 kDa), APO3 (<1 kDa), APACE (<1 kDa), APG1 (70 kDa) and APG2 (70 kDa) isolated from the hydrolysate using consecutive chromatographic methods. Soluble peptide were produced from Hwangtae and evaluated for their nutritional and functional properties. Some functional properties of FPHs were assessed and compared with those of egg albumin or the soybean protein. APO2 had the highest nitrogen solubility value (94.2%), emulsion capacity and emulsion stability of the Alaska Pollack peptide ranged from 12.4 to 39.5 (mL of oil per 200 mg of protein) and 44.0% to 77.5%, respectively. Highest and lowest fat adsorption values were observed for APG1 (9.9 mL of oil per gram of protein) and APO3 (3.8 mL of oil per gram of protein), respectively.

        This report studied 31 male patients who were in one hospital in Kwangju or came to a prosthesis clinic and are 15 normal control group to analyze the gait characteristics according to the stump length in the prosthesis users with the unilateral below-knee amputations. The patient group was divided into a short stump subgroup of 13 patients and a long stump subgroup of 18 patients according to the stump length, the control group was randomly sampled from 15 males who have similar age, height and weight to the experimental group. The results of analysis are summarized as follows. 1. There were no significant statistical differences of the general characteristics between patient and control groups and there was no difference of characteristics by patient groups except the mean gait distance a day. 2. Of 33 patients with amputation, 18 patients (57.8%) had phantom pain and 25 cases (80.6%) had neuroma. 3. Gait velocity, stride length of the affected side, step length of the sound side and base support among gait characteristics show a significant difference according to the stump length in the state excluding the effects of other factors (p<05) and the group with long stump showed closer gait characteristics to normal persons than those with short stumps. In conclusion, among the patients of unilateral below-knee amputations, the stump length affects the gait characteristics such as gait velocity, stride length, step and base of support and it is found that as the stump length gets longer, it shows the similar characteristics to gait characteristics of normal persons and long unilateral below-knee amputation can secure better gait characteristics.

      • Comparison analysis of aromatic compounds in the aromatic rice germplasm by Gas Chromatography and Mass Spectrometry

        The volatile and semi-volatile compounds of 5 accessions of domestic and 25 accessions of foreign aroma rice were extracted by solid phase microextraction (SPME) and analyzed by gas chromatography-mass spectrum (GC-MS). A total of 158 volatile and semi-volatile compounds were identified from 30 accessions of aroma rice germplasm, including 32 alcohols, 25 acids, 25 ketones, 21 hydrocarbon, 18 esters, 16 aldehydes, 4 ethers, 5 amines, 2 phenols, 2 bases, and 8 miscellaneous compounds. By UPGMA/Neighbor-join tree analysis, the thirty aroma rice germplasms could be classified into five groups according to the major odor or aroma compounds. Group Ⅰ included Indica type of 'Basmati' varieties. GroupⅡ included Japonica type foreign aroma rice except WAR 16 (domestic aroma rice). GroupⅢ and GroupⅤ included Indica type of Basmati and non-Basmati varities. GroupⅣ included domestic aroma rice except WAR34 and WAR35(Basmati varieties).

        In vivo quantitative measurement of arthritis activity based on hydrophobically modified glycol chitosan in inflammatory arthritis: more active than passive accumulation.

        <P>We demonstrated that arthritis could be visualized noninvasively using hydrophobically modified glycol chitosan nanoparticles labeled with Cy5.5 (HGC-Cy5.5) and an optical imaging system. Activated macrophages expressing Mac-1 molecules effectively phagocytosed HGC-Cy5.5, which formed spherical nanoparticles under physiologic conditions. We estimated the applicability of HGC-Cy5.5 to quantitative analysis of arthritis development and progression. Near-infrared fluorescence images, captured after HGC-Cy5.5 injection in mice with collagen-induced arthritis, showed stronger fluorescence intensity in the active arthritis group than in the nonarthritis group. According to the progression of arthritis in both collagen-induced arthritis and collagen antibody-induced arthritis models, total photon counts (TPCs) increased in parallel with the clinical arthritis index. Quantitative analysis of fluorescence after treatment with methotrexate showed a significant decrease in TPC in a dose-dependent manner. Histologic evaluation confirmed that the mechanism underlying selective accumulation of HGC-Cy5.5 within synovitis tissues included enhanced phagocytosis of the probe by Mac-1-expressing macrophages as well as enhanced permeability through leaky vessels. These results suggest that optical imaging of arthritis using HGC-Cy5.5 can provide an objective measurement of disease activity and, at the same time, therapeutic responses in rheumatoid arthritis.</P>

        국내 분리 닭 전염성 기관지염 바이러스 성상에 관한 연구

        In order to investigate the biological properties, pathogenicity and immune responses in artficially infected SPF chickens with Avian infectious bronchitis virus that was isolated from chickens showing IB like signs in southern region of Chung buk. Results obtained throuth the experiments are summarized as follows. 1. From 15 IB suspected cases, two strains of IB virus were isolated, one each from the tracheas and lungs. 2. Infectious bronchitis specific embryo lesions were observed after four serial passages of the isolates in chicken embryos. 3. The field isolates and M-41 strain of IB virus interfered with the replication of Newcastle disease virus in chicken embryos. 4. When specific pathogen free chickens, two week old, were inoculated with the TB virus isolates, clinical respiratory signs as dyspnea, coughing were observed. Airsacculitis was observed by necropsy. 5. AGP antibody positive rates of inoculated SPF chickens were highest on day 14 and lowest on day 36, while HI antibody responses were detected on day 14 in all Groups, the reinoculated Group was shown highest titers. 6. By Indirect immunofluorescence antibody assay of artificially infected SPF chickens, the viral antigens were detected in tissues of larynx, trachea and lung on the 4th to 7th days post inoculation.

        Sex pheromone composition of the variegated cutworm, Peridroma saucia (Lepidoptera: Noctuidae), in Korea

        This study was conducted to investigate the sex pheromone composition of the variegated cutworm (Peridroma saucia <TEX>$H{\ddot{u}}bner$</TEX>) in Korea, The sex pheromone components of P. saucia were identified as (Z)-9-tetradecenyl acetate (Z9-14:Ac) and (Z)-11-hexadecenyl acetate (Z11-16:Ac) through GC-EAD and GC-MS analysis. EAG tests of the male antennae revealed that the Z9-14:AC exerted significantly larger responses than other compounds. The female moths primarily called and copulated between 6 hand 7 h after the lights off, and the ratio of two pheromone components, Z9-14:Ac and Zl1-16:Ac, in the sex pheromone gland during this period was 1:2.1 to 1:2.4. In the field trapping studies, a large number of male moths were caught in the traps baited with the mixtures of Z9-14:Ac and Z11-16:Ac at the ratios ranging from 2.3:1 to 1:4, with the highest trap catches at 1:1 to 1:2.3 ratios of the two components. The seasonal flight activities of P. saucia monitored by using pheromone lures revealed complicated patterns in Korea. Specifically, the first flight period was spread over a long period and irregular, while the second flight period differed among the localities examined.
