        Effect of night light regimen on growth performance, antioxidant status and health of broiler chickens from 1 to 21 days of age

        Objective: The study was conducted to evaluate the effects of night light regimen on growth performance, antioxidant status and health of Lingnan Yellow broiler chickens from 1 to 21 days of age. Methods: A completely randomized factorial design involved 2 photoperiods (constant lighting [CL], 24 L:0 D and intermittent lighting [INL], 17 L:3 D:1 L:3 D)×2 light intensities (10 lx and 30 lx). A total of one thousand six hundred and eighty 1-d-old Lingnan Yellow broiler chicks were randomly divided into 4 treatments with 6 replicates (70 birds per replicate). The experiment lasted for 21 d. Results: Photoperiods and light intensities had no effect on average daily gain, feed conversion ratio, and mortality of the broiler chickens (p>0.05). The INL had a significant effect on average daily feed intake (p<0.05) of broiler chickens compared with CL. Photoperiod and light intensity had an interactive effect on melatonin (MT) concentration (p<0.05). At CL, reducing light intensity increased MT concentration; INL birds had higher MT but MT concentration was not affected by light intensity. There was an interactive effect on glutathione peroxidase (GPx) and catalase (CAT) in serum and total antioxidant capability (T-AOC) in liver between photoperiod and light intensity. With the decrease of light intensity, the activities of GPx and CAT in serum and T-AOC in liver increased in CL group (p<0.05). Broiler chickens reared under INL had better antioxidant status and 10 lx treatments had higher activities of CAT in serum than 30 lx (p<0.05). Different photoperiods and light intensities had no effect on malondialdehyde. There was an interaction between photoperiod and light intensity on serum creatine kinase (CK) concentration (p<0.05). At CL, the elevated light intensity resulted in an increase in CK content; INL birds had lower CK concentration especially in low light intensity group. Besides, INL and low light intensity significantly reduced the concentration of serum corticosterone and heat shock protein 70 (p<0.05). Serum immunoglobulin M contents were increased in broiler chickens reared under the INL compared with CL group (p<0.05). Conclusion: Results above suggest that the night light regimen of INL and 10 lx could be beneficial to the broiler chickens from 1 to 21 days of age due to the better health status and electricity savings.

        Anti-adherence of Antibacterial Peptides and Oligosaccharides and Promotion of Growth and Disease Resistance in Tilapia

        Four hundred and fifty tilapias ($6.77{\pm}0.23$ g) were assigned randomly to six groups to evaluate the feasibility of the tested antibacterial peptides (ABPs) and oligosaccharides as substitutes for antibiotics. The control group was fed with a commercial tilapia diet; other five groups were fed with the same commercial diet supplemented with konjac glucomannan (KGLM), cluster bean galactomannan (CBGAM), and three animal intestinal ABPs derived from chicken, pig and rabbit at 100 mg/kg respectively. After 21 days of feeding, growth, disease resistance, and in vivo anti-adherence were determined. Furthermore, the inhibitory effect of tested agents on adhesion of Aeromonas veronii biovar sobria (A.vbs) strain BJCP-5 to tilapia enteric epithelia in vitro was assessed by cell-ELISA system. As a result, the tested agents supplemented at 100 mg/kg show significant benefit to tilapia growth and disease resistance (p<0.05), and the benefit may be correlated with their interfering in the contact of bacteria with host mucosal surface. Although none of the tested agents did inhibit the growth of BJCP-5 in tryptic soy broth at $100{\mu}g/ml$, all of them did inhibit the adhesion of A.vbs to tilapia enteric epithelia in vivo and in vitro. In vitro mimic assays show that three ABPs at low concentrations of $25{\mu}g/ml$ and $2.5{\mu}g/ml$ have the reciprocal dose-dependent anti-adherence effect. The inhibition of ABPs may be correlated with a cation bridging and/or receptor-ligand binding, but not with hydrophobicity. The KGLM and CBGAM inhibited the adherence of BJCP-5 to tilapia enteric epithelia with dose-dependent manner in vitro, and this may be through altering bacterial hydrophobicity and interfering with receptor-ligand binding. Our results indicate that the anti-adherence of the tested ABPs and oligosaccharides may be one of the mechanisms in promoting tilapia growth and resistance to A.vbs.

        Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

        This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

        Hydrogen Behaviors with different introduction methods in SiC-C Films

        SiC-C films were deposited with r. f. magnetron sputtering on substrates followed by argon ion bombardment. These films were then permeated by hydrogen gas under the pressure of $3.23\times10^{7}$ Pa for 3 hours at temperature of 500K or bombarded with hydrogen ion beam at 5 keV and a dose of $1\times10^{18}$ ions/$\textrm{cm}^2$. SIMS, AES and XPS were used to analyze hydrogen related species, chemical bonding states of C, Si as well as contamination oxygen due to hydrogen participation in the SiC-C films in order to study the different behaviors of hydrogen in carbon-carbide films due to different hydrogen introduction. Related mechanism about the effects of hydrogen on the element of the SiC-C films was discussed in this paper.

        Expression Profiles of Cellular Retinol-binding Protein, Type II (CRBP II) in Erlang Mountainous Chickens

        Cellular retinol-binding protein II (CRBP II) belongs to the family of cellular retinol-binding proteins and plays a major role in absorption, transport, and metabolism of vitamin A. In addition, because vitamin A is correlated with reproductive performance, we measured CRBP II mRNA abundance in erlang mountainous chickens by real-time PCR using the relative quantification method. The expression of CRBP II showed a tissue-specific pattern and egg production rate-dependent changes. The expression was very high (p<0.05) in jejunum and liver, intermediate in kidney, ovary, and oviduct, and lowest (p<0.05) in heart, hypothalamus, and pituitary. In the hypothalamus, oviduct, ovary, and pituitary, CRBP II mRNA abundance were correlated to egg production rate, which increased from 12 wk to 32 wk, peaked at 32 wk relative to the other time points, and then decreased from 32 wk to 45 wk. In contrast, the expression of CRBP II mRNA in heart, jejunum, kidney, and liver was not different at any of the ages evaluated in this study. These data may help to understand the genetic basis of vitamin A metabolism, and suggest that CRBP II may be a candidate gene to affect egg production traits in chickens.

        <i>K</i>-mixing in the doubly mid-shell nuclide <sup>170</sup>Dy and the role of vibrational degeneracy

        <P><B>Abstract</B></P> <P>A detailed study of the structure of the doubly mid-shell nucleus Dy 104 1 66 170 has been carried out, following isomeric and <I>β</I> decay. We have measured the yrast band up to the spin-parity <SUP> J π </SUP> = <SUP> 6 + </SUP> state, the K = 2 <I>γ</I>-vibration band up to the <SUP> 5 + </SUP> state, a low-lying negative-parity band based on a <SUP> 2 − </SUP> state that could be a candidate for the lowest energy octupole vibration state within this nucleus, and a candidate for the <SUP> K π </SUP> = <SUP> 6 + </SUP> two quasi-particle isomer. This state was determined to have an excitation energy of 1643.91(23) keV and a half life of 0.99(4) μs, with a reduced hindrance for its decay to the ground-state band an order of magnitude lower than predicted by <SUB> N p </SUB> <SUB> N n </SUB> systematics. This is interpreted as being due to <I>γ</I>-vibrational mixing from a near degeneracy of the isomer and the <SUP> 6 + </SUP> state of the <I>γ</I> band. Furthermore, the parent nucleus <SUP>170</SUP>Tb has been determined to have a half-life of 0.91 ( − 13 + 18 ) s with a possible spin-parity of <SUP> 2 − </SUP> .</P>

      • Effect of lysophospholipids in diets differing in fat contents on growth performance, nutrient digestibility, milk composition and litter performance of lactating sows

        <P>It is well known that energy plays an important role in sow growth and development. Increasing the utilization of lipids will be beneficial to sows. Emulsifiers are substances which stabilize mixtures and prevent oil and water from separating, thereby enhancing the digestion of lipids. This study was conducted to evaluate the effect of dietary emulsifier (lysophospholipids (LPL)) supplementation in diets differing in fat contents on growth performance, nutrient digestibility and milk composition in lactating sows, as well as performance and fecal score in piglets. A total of 32 multiparous sows (Landrace×Yorkshire) were used in a 21-day experiment. On day 110 of gestation, sows were weighed and moved into the farrowing facility, randomly assigned in a 2×2 factorial arrangement according to their BW with two levels of LPL (0 and 30 mg/kg) and two levels of fat (4.75% and 2.38% fat; 13.66 and 13.24 MJ/kg). BW loss and backfat thickness loss were decreased (<I>P</I><0.05) by LPL supplementation. Backfat thickness at weaning was higher (<I>P</I><0.05) in sows fed LPL supplementation diets. The apparent total tract digestibility of dry matter, nitrogen, gross energy and crude fat in sows fed LPL diets was increased (<I>P</I><0.05) compared with those fed non-LPL diets. Sows fed the high-fat diets had higher (<I>P</I><0.05) milk fat on day 10 and milk lactose on day 20 than those fed the low-fat diets. Milk fat and lactose concentrations in LPL supplementation treatments was increased (<I>P</I><0.05) compared with non-LPL treatments on day 10 and day 20, respectively. Positive interaction effects (<I>P</I><0.05) between fat and LPL were observed for milk fat concentration on day 10. In conclusion, LPL addition decreased BW loss and backfat thickness loss, improved nutrient digestibility and milk fat as well as milk lactose concentrations. In addition, there was a complementary positive effect of dietary fat and LPL supplementation on milk fat concentration in lactating sows.</P>

      • Prolonged Insulin Independence After Islet Allotransplants in Recipients with Type 1 Diabetes

        <P>We sought to determine the long-term outcomes in type 1 diabetic recipients of intraportal alloislet transplants on a modified immunosuppressive protocol. Six recipients with hypoglycemia unawareness received one to two islet infusions. Induction therapy was with antithymocyte globulin (ATG) plus etanercept for tumor necrosis factor-alpha blockade. Recipients received cyclosporine and everolimus for maintenance immunosuppression for the first year posttransplant, with mycophenolic acid or mycophenolate mofetil subsequently substituted for everolimus. Recipients have been followed for 1173 +/- 270 days since their last infusion for islet graft function (insulin independence, hemoglobin A(1c) levels and C-peptide production) and for adverse events associated with the study protocol. Of the six recipients, five were insulin-independent at 1 year, and four continue to be insulin-independent at a mean of 3.4 +/- 0.4 years posttransplant. None of the six recipients experienced recurrence of severe hypoglycemia. Measured glomerular filtration rate decreased from 110.5 +/- 21.2 mL/min/1.73 m(2) pretransplant to 82.6 +/-19.1 mL/min/1.73 m(2) at 1 year posttransplant. In conclusion, islet transplants restored insulin independence for a mean of >3 years in four of six recipients treated with ATG and etanercept induction therapy and with cyclosporine and, initially, everolimus for maintenance. Our results suggest this immunosuppressive protocol may allow long-term graft survival.</P>

      • A Semiconducting Organic Radical Cationic Host–Guest Complex

        <P>The self-assembly and solid-state semiconducting properties of single crystals of a trisradical tricationic complex composed of the diradical dicationic cyclobis(paraquat-<I>p</I>-phenylene) (CBPQT<SUP>2(•+)</SUP>) ring and methyl viologen radical cation (MV<SUP>•+</SUP>) are reported. An organic field effect transistor incorporating single crystals of the CBPQT<SUP>2(•+)</SUP>⊂MV<SUP>•+</SUP> complex was constructed using lithographic techniques on a silicon substrate and shown to exhibit <I>p</I>-type semiconductivity with a mobility of 0.05 cm<SUP>2</SUP> V<SUP>–1</SUP> s<SUP>–1</SUP>. The morphology of the crystals on the silicon substrate was characterized using scanning electron microscopy which revealed that the complexes self-assemble into “molecular wires” observable by the naked-eye as millimeter long crystalline needles. The nature of the recognition processes driving this self-assembly, radical–radical interactions between bipyridinium radical cations (BIPY<SUP>•+</SUP>), was further investigated by resonance Raman spectroscopy in conjunction with theoretical investigations of the vibrational modes, and was supported by X-ray structural analyses of the complex and its free components in both their radical cationic and dicationic redox states. These spectroscopic investigations demonstrate that the bond order of the BIPY<SUP>•+</SUP> radical cationic units of host and guest components is not changed upon complexation, an observation which relates to its conductivity in the solid-state.
