        Wnt/β-Catenin Promotes the Osteoblastic Potential of BMP9 Through Down-Regulating Cyp26b1 in Mesenchymal Stem Cells

        BACKGROUND: All-trans retinoic acid (ATRA) promotes the osteogenic differentiation induced by bone morphogenetic protein 9 (BMP9), but the intrinsic relationship between BMP9 and ATRA keeps unknown. Herein, we investigated the effect of Cyp26b1, a critical enzyme of ATRA degradation, on the BMP9-induced osteogenic differentiation in mesenchymal stem cells (MSCs), and unveiled possible mechanism through which BMP9 regulates the expression of Cyp26b1. METHODS: ATRA content was detected with ELISA and HPLC–MS/MS. PCR, Western blot, and histochemical staining were used to assay the osteogenic markers. Fetal limbs culture, cranial defect repair model, and micro–computed tomographic were used to evaluate the quality of bone formation. IP and ChIP assay were used to explore possible mechanism. RESULTS: We found that the protein level of Cyp26b1 was increased with age, whereas the ATRA content decreased. The osteogenic markers induced by BMP9 were increased by inhibiting or silencing Cyp26b1 but reduced by exogenous Cyp26b1. The BMP9-induced bone formation was enhanced by inhibiting Cyp26b1. The cranial defect repair was promoted by BMP9, which was strengthened by silencing Cyp26b1 and reduced by exogenous Cyp26b1. Mechanically, Cyp26b1 was reduced by BMP9, which was enhanced by activating Wnt/b-catenin, and reduced by inhibiting this pathway. b-catenin interacts with Smad1/5/9, and both were recruited at the promoter of Cyp26b1. CONCLUSIONS: Our findings suggested the BMP9-induced osteoblastic differentiation was mediated by activating retinoic acid signalling, viadown-regulating Cyp26b1. Meanwhile, Cyp26b1 may be a novel potential therapeutic target for the treatment of bone-related diseases or accelerating bone-tissue engineering.

        소셜 커머스 샤오홍슈 콘텐츠 특성, 사이트 신뢰성, 리뷰 제작자의 전문성이 소비자 만족도와 구매의도에 미치는 영향

        본 연구는 소셜 커머스 샤오홍슈 콘텐츠 특성(정보성, 오락성, 신뢰성), 샤오홍슈 사이트 신뢰성, 리뷰 제작자의 전문성이 소비자 만족도와 구매의도에 미치는 영향을 알아보기 위해, 샤오홍슈 사용 경험자 총 300명의 설문을 분석하였다. 분석은 SPSS 24를 통해 요인분석과 변수들 간의 차이를 알아보기 위해 회귀분석을 실시하였다. 분석결과 첫째, 샤오홍슈 콘텐츠 특성(정보성, 오락성, 신뢰성)이 소비자 만족도와 구매의도에 미치는 영향을 살펴본 결과 콘텐츠 특성 중 오락성, 정보성은 소비자 만족도와 구매의도에 통계적으로 유의한 영향을 미치는 것으로 확인되었으나 신뢰성은 영향을 주지 않았다. 둘째, 샤오홍슈 사이트의 신뢰성은 사용자 만족도와 구매의도에 긍정적인 영향을 미치는 것으로 나타났다. 셋째, 샤오홍슈 콘텐츠 제작자의 전문성은 소비자 만족도와 구매 의도에 긍정적인 영향을 미치는 것으로 나타났다. 본 연구는 샤오홍슈와 같은 소셜 커머스가 소비자 태도에 미치는 영향 관계를 분석하였으며, 향후 소셜 커머스 사용자의 태도를 이해하는데 기초 자료가 될 것이다. In order to investigate the influence that social commerce Xiaohongshu contents property (information, amusement, reliability), reliability of Xiaohongshu site, review producer’s expertise have on consumer satisfaction & purchase intention, this study analyzed the questions of 300 persons experiencing in using Xiaohongshu. The result of analysis is as follows. First, it was found that amusement, information out of contents property (information, amusement, reliability) have statistically significant influence on consumer satisfaction & purchase intention, but reliability has no influence. Second, it was found that the reliability of Xiaohongshu site has positive influence on consumer satisfaction & purchase intention. Third, it was found that the expertise of Xiaohongshu contents producer has positive influence on consumer satisfaction & purchase intention. As this study analyzed the influential relation that social commerce like Xiaohongshu has on consumer attitude, it will be basic materials in understanding attitude of social commerce users later.

        RON and MET Co-overexpression Are Significant Pathological Characteristics of Poor Survival and Therapeutic Targets of Tyrosine Kinase Inhibitors in Triple-Negative Breast Cancer

        Purpose Triple-negative breast cancer (TNBC) is highly malignant and has poor prognosis and a high mortality rate. The lack of effective therapy has spurred our investigation of new targets for treating this malignant cancer. Here, we identified RON (macrophage-stimulating 1 receptor) and MET (MET proto-oncogene, receptor tyrosine kinase) as a prognostic biomarker and therapeutic targets for potential TNBC treatment. Materials and Methods We analyzed RON and MET expression in 187 primary TNBC clinical samples with immunohistochemistry. We validated the targeted therapeutic effects of RON and MET in TNBC using three tyrosine kinase inhibitors (TKIs): BMS-777607, INCB28060, and tivantinib. The preclinical therapeutic efficacy of the TKIs was mainly estimated using a TNBC xenograft model. Results Patients with TNBC had widespread, abnormal expression of RON and MET. There was RON overexpression, MET overexpression, and RON and MET co-overexpression in 63 (33.7%), 63 (33.7%), and 43 cases (23.0%), respectively, which had poor prognosis and short survival. In vivo, the TKI targeting RON ant MET inhibited the activation of the downstream signaling molecules, inhibited TNBC cell migration and proliferation, and increased TNBC cell apoptosis; in the xenograft model, they significantly inhibited tumor growth and shrank tumor volumes. The TKI targeting RON and Met, such as BMS-777607 and tivantinib, yielded stronger anti-tumor effects than INCB28060. Conclusion RON and MET co-overexpression can be significant pathological characteristics in TNBC for poor prognosis. TKIs targeting RON and MET have stronger drug development potential for treating TNBC.

      • Performance Analysis of Two-Way AF Cooperative Networks with the Nth Relay Selection

        In this letter, we propose a novel two-way amplify- and-forward (AF) relay selection scheme over independent flat Nakagami-m fading channels. In the proposed schemes, two users first sequentially broadcast their respective information to all the relays. Then, the Nth best-relay perform AF protocol on the received signals sent from the two users and forward them to both users. The Nth best-relay is selected to optimize the end-to- end performance of the system transmission. The performance of the proposed system is quantified by deriving the lower bound of the overall outage probability and average symbol error rate (SER). The effectiveness of our proposed selection scheme and analytical results is verified by simulations.

      • Secure Communication in Two-Way Relay Networks with Channel Estimation Errors

        In this paper, we investigate the secrecy outage probability (SOP) for the cooperative amplify-and-forward (AF) two-way relay networks (TWRN) with channel estimation (CE) errors under Rayleigh fading channels. In the proposed system, two sources simultaneously broadcast their respective information to all the relays firstly. Then, the optimal relay performs AF protocol on the received signals sent from the two sources and forward them to both sources. In both phases, An eavesdropper receives the signals from sources and relay nodes illegally. The performance of the proposed system is quantified by deriving the upper bound of the overall outage probability. Impact of CEs on the system is illustrated by simulations.

        Induction of Forkhead Class box O3a and apoptosis by a standardized ginsenoside formulation, KG-135, is potentiated by autophagy blockade in A549 human lung cancer cells

        Background: KG-135, a standardized formulation enriched with Rk1, Rg3, and Rg5 ginsenosides, has been shown to inhibit various types of cancer cells; however, the underlying mechanisms are not fully understood. In this study, we explored its effects in A549 human lung cancer cells to investigate the induction of Forkhead Class box O3a (FOXO3a) and autophagy. Methods: Cell viability was determined by sulforhodamine B staining. Apoptosis and cell cycle distribution were analyzed using flow cytometry. The changes of protein levels were determined using Western blot analysis. Autophagy induction was monitored by the formation of acidic vesicular organelles stained with acridine orange. Results: KG-135 effectively arrested the cells in G1 phase with limited apoptosis. Accordingly, a decrease of cyclin-dependent kinase-4, cyclin-dependent kinase-6, cyclin D1, and phospho-retinoblastoma protein, and an increase of p27 and p18 proteins were observed. Intriguingly, KG-135 increased the tumor suppressor FOXO3a and induced the accumulation of autophagy hallmark LC3-II and acidic vesicular organelles without an increase of the upstream marker Beclin-1. Unconventionally, the autophagy adaptor protein p62 (sequestosome 1) was increased rather than decreased. Blockade of autophagy by hydroxychloroquine dramatically potentiated KG-135-induced FOXO3a and its downstream (FasL) ligand accompanied by the cleavage of caspase-8. Meanwhile, the decrease of Bcl-2 and survivin, as well as the cleavage of caspase-9, were also drastically enhanced, resulting in massive apoptosis. Conclusion: Besides arresting the cells in G1 phase, KG-135 increased FOXO3a and induced an unconventional autophagy in A549 cells. Both the KG-135-activated extrinsic FOXO3a/FasL/caspase-8 and intrinsic caspase-9 apoptotic pathways were potentiated by blockade of autophagy. Combination of KG-135 and autophagy inhibitor may be a novel strategy as an integrative treatment for cancers.

      • High Power Density Asymmetrical Full-Bridge Soft-Switching Inverter

        This paper presents a Zero-Voltage-Switching (ZVS) technique eliminating the switching loss on a proposed single phase bi-directional full-bridge Buck inverter in which asymmetrical filter topology and control scheme are for each arm. Here Metal-Oxide-Field-Effect- Transistor(MOSFET) is chosen as the switching device, an L₁-C₁ filter is connected across the full-bridge, an L₂ filter is connected in series to load or grid. While the arm connecting with the C₁ is controlled by the polarity of 50Hz sinusoid reference command and the arm connecting with the L₁ is controlled by a Pulse-Width-Modulation(PWM) signal modulated by sinusoid, the L₁-C₁ then operates as an output filter of Buck, and the succeeding L₂ filter further attenuates ripple current flowing to load or grid. The ZVS technique sets the L₁ inductance such low that its current flows bi-directionally within a switching period, then the bidirectional current with sufficiently high peaks in turn charge and discharge the output capacitance Coss of MOSFETs of the switching arm within floating time, defined as the duration when both MOSFETs of an arm are off, to let the MOSFETs D-S channels be turned on or off at the state that Coss are fully discharged to zero voltage, so ZVS of MOSFET is achieved. Under the ZVS operation without frequency proportional switching loss, the switching frequency is possible to be raised to high to compact the sizes of passive components and heat sink of the inverter for achieving high power density. Simulation and experiment have been carried out to verify the ZVS operation.

        Colorful Oxide Film Formation on Titanium by using EDM Process

        This paper describes the influence of the main factor on the colorful titanium oxide reacted film fabricated by EDM (Electrical Discharge machining). In our process, copper, together with pure titanium sheet, was used as anode and cathode (workpiece)respectively, and dielectric medium involved in the study was tap water. From the figures and tables shown, we can observe that colors of titanium oxide reacted films change with the increase of open voltages (0~240) from yellow (10 V), purple (20 V), indigo (30 V),golden yellow (70 V), to peach (100 V), blue (130 V), yellow green (180 V), pink (240 V). The other open voltages produce mixed colors between those created by neighboring voltages. Moreover, XRD as well as EDS analysis have made it evident that the structure of the reacted film mentioned above is Ti2O other than TiO2 and the rise of pulse duration and interval will cause non-uniform color distribution. Compared with anodizing process, the Electrical Discharge machining approach we developed for generating colorful titanium oxide reacted film in the paper is much simpler, faster, cheaper and have plenty of advantages still in environment protection and hazard avoidance.

        The purpose of this study is to present characteristics by comparing the operation of the ethnic dance curriculum among the curriculum of the department of dance in Korea and China. The ethnic dance curriculum at Korean universities is established as a step-by-step in-depth course, and creative dance courses using ethnic dance are operated. The ethnic dance curriculum at Chinese universities has a variety of ethnic dance courses and courses on teaching methods for each ethnic dance. The Korean ethnic dance curriculum is a curriculum for the transmission of future-oriented national dance. The ethnic dance curriculum in China is a past-oriented curriculum.
