      • MGMT-B Gene Promoter Hypermethylation in Patients with Inflammatory Bowel Disease - A Novel Finding

        Inflammatory bowel disease (IBD) is a disease strongly associated with colorectal cancer (CRC) as a well-known precancerous condition. Alterations in DNA methylation and mutation in K-ras are believed to play an early etiopathogenic role in CRC and may also an initiating event through deregulation of molecular signaling. Epigenetic silencing of APC and SFRP2 in the WNT signaling pathway may also be involved in IBD-CRC. The role of aberrant DNA methylation in precancerous state of colorectal cancer (CRC) is under intensive investigation worldwide. The aim of this study was to investigate the status of promoter methylation of MGMT-B, APC1A and SFRP2 genes, in inflamed and normal colon tissues of patients with IBD compared with control normal tissues. A total of 52 IBD tissues as well as corresponding normal tissues and 30 samples from healthy participants were obtained. We determined promoter methylation status of MGMT-B, SFRP2 and APC1A genes by chemical treatment with sodium bisulfite and subsequent MSP. The most frequently methylated locus was MGMT-B (71%; 34 of 48), followed by SFRP2 (66.6 %; 32 of 48), and APC1A (43.7%; 21 of 48). Our study demonstrated for the first time that hypermethylation of the MGMT-B and the SFRP2 gene promoter regions might be involved in IBD development. Methylation of MGMT-B and SFRP2 in IBD patients may provide a method for early detection of IBD-associated neoplasia.

      • Promoter Methylation Status of Two Novel Human Genes, UBE2Q1 and UBE2Q2, in Colorectal Cancer: a New Finding in Iranian Patients

        Background: The ubiquitin-proteasome system (UPS) degrades a variety of proteins which attach to specific signals. The ubiquitination pathway facilitates degradation of damaged proteins and regulates growth and stress responses. This pathway is altered in various cancers, including acute lymphoblastic leukemia, head and neck squamous cell carcinoma and breast cancer. Recently it has been reported that expression of newly characterized human genes, UBE2Q1 and UBE2Q2, putative members of ubiquitin-conjugating enzyme family (E2), has been also changed in colorectal cancer. Epigenetics is one of the fastest-growing areas of science and nowadays has become a central issue in biological studies of diseases. According to the lack of information about the role of epigenetic changes on gene expression profiling of UBE2Q1 and UBE2Q2, and the presence of CpG islands in the promoter of these two human genes, we decided to evaluate the promoter methylation status of these genes as a first step. Materials and Methods: The promoter methylation status of UBE2Q1 and UBE2Q2 was studied by methylation-specific PCR (MSP) in tumor samples of 60 colorectal cancer patients compared to adjacent normal tissues and 20 non-malignant controls. The frequency of the methylation for each gene was analyzed by chi-square method. Results: MSP results revealed that UBE2Q2 gene promoter were more unmethylated, while a higher level of methylated allele was observed for UBE2Q1 in tumor tissues compared to the adjacent normal tissues and the non malignant controls. Conclusions: UBE2Q1 and UBE2Q2 genes show different methylation profiles in CRC cases.

        A review of landform classification methods

        The study of landform can be used to predict specific solid rock and moisture conditions that exist in that landform. Landforms are of significance in engineering because they influence the quality and type of grading, predetermine the drainage requirements, and the soil or rock conditions. They have added significance because there are an infinite number of duplicates of each type. Given that each of the landform units have different characteristics (soil, slope, elevation, vegetation etc.), classification of landforms is importance. Therefore, landforms are widely recognized as natural objects that partition the earth’s surface into essential spatial entities. Landform entities differ from one another in terms of specification such as shape, size, orientation, relief and contextual position. They also differ in terms of the physical processes that were involved in their formation and that continue to operate within them at the present time. This paper aims at providing a review of the landform characteristics and classification. Various definitions and attributes of landforms which focus on aspects of morphometry, geomorphometric context, terrain positions and scale are discussed as well. In addition, the methods developed for the landform classification are assessed.

      • Local joint flexibility equations for Y-T and K-type tubular joints

        It is common that analyses of offshore platforms being carried out with the assumption of rigid tubular joints. However, many researches have concluded that it is necessary that local joint flexibility (LJF) of tubular joints should be taken into account. Meanwhile, advanced analysis of old offshore platforms considering local joint flexibility leads to more accurate results. This paper presents an extensive finite-element (FE) based study on the flexibility of uni-planner multi-brace tubular Y-T and K-joints commonly found in offshore platforms. A wide range of geometric parameters of Y-T and K-joints in offshore practice is covered to generate reliable parametric equations for flexibility matrices. The formulas are obtained by non-linear regression analyses on the database. The proposed equations are verified against existing analytical and experimental formulations. The equations can be used reliably in global analyses of offshore structures to account for the LJF effects on overall behavior of the structure.

        Antinociceptive and gastro-protective effect of the ethanolic extract of the flowering top of Anthocephalus Cadamba Roxb

        The effect of alcoholic extract of Anthocephalus (A.) Cadamba Roxb. was evaluated in experimental models of pain and ulcer. Hot tail flick test, hot plate test and acetic acid induced writhing test were employed for evaluating the peripheral as well as central analgesic mechanism exerted by the extracts. Gastroprotective activity was examined by HCl and ethanol induced gastric damage test. Test group received crude extract 500 mg/kg showed maximum time needed for the response against thermal stimuli (6.26 ± 0.439 s) which is comparable to diclofenac sodium (6.56 ± 0.381 s) in hot tail flick method. These experimental results also followed the experimental results of hot plate test where crude extract 500 mg/kg showed maximum time needed for the response against thermal stimuli (4.74 ± 0.234 s) which is comparable to diclofenac sodium (5.58 ± 0.585 s). The crude extract at 500 and 250 mg/kg showed significant reduction in acetic acid induced writhing in mice with a maximum effect of 68.026% reduction at 500 mg/kg dose which is comparable to standard diclofenac sodium (79.93%). In gastroprotective study the extract of A. Cadamba (250 and 500 mg/kg) significantly inhibited ulceration induced by both HCl and ethanol dose dependently.       • Sensitive and Noninvasive Detection of Aberrant SFRP2 and MGMT-B Methylation in Iranian Patients with Colon Polyps

        Background: The pathogenesis of sporadic colorectal cancer (CRC) is influenced by the patient genetic background and environmental factors. Based on prior understanding, these are classified in two major pathways of genetic instability. Microsatellite instability (MSI) and CPG island methylator phenotype (CIMP) are categorized as features of the hypermethylated prototype, and chromosomal instability (CIN) is known to be indicative of the non-hypermethylated category. Secreted frizzled related protein 2 (SFRP2), APC1A in WNT signaling pathway and the DNA repair gene, O6-methylguanine-DNA methyltransferase (MGMT), are frequently hypermethylated in colorectal cancer. Detection of methylated DNA as a biomarker by easy and inexpensive methods might improve the quality of life of patients with CRC via early detection of cancer or a precancerous condition. Aim: To evaluate the rate of SFRP2 and MGMT hypermethylation in both polyp tissue and serum of patients in south Iran as compared with matched control normal population corresponding samples. Materials and Methods: Methylation-specific PCR was used to detect hypermethylation in DNA extracted from 48 polypoid tissue samples and 25 healthy individuals. Results: Of total polyp samples, 89.5% had at least one promoter gene hypermethylation. The most frequent methylated locus was SFRP2 followed by MGMT-B (81.2 and 66.6 percent respectively). Serologic detection of hypermethylation was 95% sensitive as compared with polyp tissue. No hypermethylation was detected in normal tissue and serum and its detection in patients with polyps, especially of serrated type, was specific. Conclusions: Serologic investigation for detection of MGMT-B, SFRP2 hypermethylation could facilitate prioritization of high risk patients for colonoscopic polyp detection and excision.

        Relationship between groundwater quality and distance to fault using adaptive neuro fuzzy inference system (ANFIS) and geostatistical methods (case study: North of Fars Province)

        The aim of this paper is to use Kriging (spherical, exponential, and Guassian models) and Inverse distance weighted (IDW) methods to prepare the water quality map. In addition, the relationship between water quality and distance to fault is determined in northeast of Fars province, Iran. Adaptive neuro fuzzy inference system method is also used to predict groundwater quality. The measured Sodium adsorption ratio and electrical conductivity parameters that are obtained from 384 wells in 2005 to 2014 are utilized to determine groundwater quality. The results show that the Kriging method (spherical model) has a higher accuracy with lower RMSE value than IDW method. Thereafter, this model is used to prepare the interpolation maps. Moreover, the results indicate the hybrid model in terms of maximum R2 and the minimum error is suitable enough to predict water quality parameters. In addition, the results depict by increasing the number of fault, the groundwater quality is decreased and vice versa.
