      • Transient Knock Down of Grp78 Reveals Roles in Serum Ferritin Mediated Pro-inflammatory Cytokine Secretion in Rat Primary Activated Hepatic Stellate Cells

        Chronic liver diseases, including cancer, are characterized by inflammation and elevated serum ferritin (SF). However, the causal-relationship remains unclear. This study used primary rat hepatic stellate cells (HSC) as a model to investigate effects of physiological SF concentrations (10, 100 and 1000 pM) because HSCs play a central role in the development and progression of liver fibrosis. Physiological concentrations of SF, either horse SF or human serum, induced pro-inflammatory cytokine $IL1{\beta}$, IL6 and $TNF{\alpha}$ secretion in rat activated HSCs (all p<0.05). By contrast, treatment did not alter activation marker ${\alpha}SMA$ expression. The presence of SF markedly enhanced expression of Grp78 mRNA (p<0.01). Furthermore, transient knock down of Grp78 by endotoxin EGF-SubA abolished SF-induced $IL1{\beta}$ and $TNF{\alpha}$ secretion in activated HSCs (all p<0.05). In conclusion, our results showed that at physiological concentrations SF functions as a pro-inflammatory mediator in primary rat HSCs. We also provide a molecular basis for the action of SF and identified Grp78-associated ER stress pathways as a novel potential therapeutic target for resolution of fibrosis and possible prevention of liver cancer.

        Anti-metastatic Effect of Garlic Hexane Extract on Lung Metastasis Induced by Melanoma B16F10 Cells in Mice

        암전이는 현재까지 적당한 치료제가 거의 없었기 때문에 암에 의한 사망의 주요한 원인 중의 하나로 인식되고 있다. 최근 본 연구팀은 마늘 추출물과 순수분리한 성분에 대한 암전이 억제 시험 결과 마늘의 추출물 또는 성분이 암전이를 억제시켰으며, 역학조사에서도 마늘을 많이 섭취한 사람은 암의 발생을 억제시키는 것으로 보고되어 있다, 본 연구의 암전이 실험에서는 C57BL/6 mouse의 꼬리 정맥에 melanoma B16F10세포를 주사하여 폐에 전이를 유도하였다. 암세포 주사 1일 후에 마늘의 헥산 추출물 50, 100 및 200 mg/kg body weight를 2일 간격으로 21일 동안 구강투여 한 다음 암전이 억제효과를 조사하였다. GHE를 처리하지 않은 대조구에서는 폐에서 암 colony가 97.4±30.2으로 대량 생성되었다. GHE를 50, 100 및 200 mg/kg의 농도로 경구투여시에 암전이 빈도는 각각 6.93, 46.80 및 50.53% 억제하였다. 또한 100 mg/kg body weight 경구투여 시에는 폐로 암전이 억제율이 약 53% 이상으로 매우 높았다. 폐에서 melanoma cell colony의 발생율과 면적은 마늘 헥산 추출물의 농도가 높을수록 감소하였다. 결론적으로 C57BL/6 mice의 암전이 모델에서 마늘 헥산추출물의 구강투여는 폐에 암전이를 억제시켰으나, 향후 그 기작에 대한 연구가 수행되어야 할 것으로 생각된다. Metastatic cancer is one of the main causes of cancer-related death since they rarely respond to available treatments. There is epidemiologic evidence that high garlic consumption decreases the incidence of cancer. Recent studies of our laboratory have revealed that a garlic-extracts is effective in suppressing metastasis. For experimental metastasis, C57BL/6 mice were injected intravenously with melanoma B16F10 cells in the tail vein, and were orally administered various concentrations (0, 50, 100 or 200 mg/kg body weight) of garlic hexane extract (GHE) for 21 days. The incidence and the area of the melanoma cell colony occupied by the poorly differentiated carcinoma were significantly lower in dose-dependent in 50, 100 and 200 mg/kg BW GHE–treated mice compared with control mice. In conclusion, the results of the present study show that GHE administration prevents lung metastasis in C57BL/6 mice.

        Inhibition of Insulin-Like Growth Factor-1-Induced Sebum Production by Bilobetin in Cultured Human Sebocytes

        Background: Sebocytes are the major cells of sebaceous gland. The essential role of sebocytes is the production of sebum, a specific lipid mixture, that covers the body surface and provides the barrier function. At puberty, sebum production increases under the effects of various stimuli including androgens and insulin-like growth factor-1 (IGF-1). Excessive sebum production changes the microenvironment surrounding hair follicle, often leading to the onset of acne. Objective: We previously performed screening test using cultured human sebocytes, and found that bilobetin had a potential for inhibiting lipid production. The aim of this study is to demonstrate the effects of bilobetin on IGF-1-induced lipogenesis in sebocytes. Methods: We pretreated simian virus 40 T (SV40T)-transformed sebocytes with bilobetin then stimulated with IGF-1. Effects of bilobetin on lipogenesis of sebocytes were examined by thin layer chromatography and Western blot. Results: Bilobetin markedly inhibited IGF-1- induced lipid production in sebocytes, especially in terms of production of squalene and wax ester. Supporting these results, bilobetin showed significant inhibitory effect on squalene synthase promoter activity. In addition, bilobetin significantly down-regulated lipogenic transcription factors such as sterol response element binding protein (SREBP)-1 and SREBP-2. To delineate the possible action mechanism, we investigated the effect of bilobetin on intracellular signaling. As a result, bilobetin inhibited IGF-1-induced phosphorylation of AKT. Conclusion: Together, these results suggest that bilobetin has an inhibitory potential on sebum production in sebocytes, being applicable for acne treatment. (Ann Dermatol 31(3) 294∼299, 2019)

        동충하초균주로 발효한 감초의 주요성분 함량 변화 및 NO 생성 억제 효과

        약용식물의 발효는 새로운 식품의 소재 개발이 가능하나, 발효 균주는 대부분 이스트, 유산균, 박테리아 등이 이용되고 있다. 본 연구에서는 감초와 번데기 단독 또는 감초에 발효원인 번데기를 20%와 50%로 첨가한 혼합물의 배지에 눈꽃 동충하초(Paecilomyces tenuipes)와 밀리타리스 동충하초(Cordyceps militaris)를 이용하여 고체배양방법을 확립하였다. 동충하초 발효물을 식품소재로 개발하기 위하여 식용 가능한 용매인 에탄올 95%, 70%, 50%, 25% 및 물로서 추출한 다음 동충하초로부터 생성된 cordycepin과 감초의 지표성분인 liquiritin, liquiritigenin과 glycirrhizin의 함량 및 NO생성 억제효과를 조사하였다. Cordycepin함량은 감초에 번데기를 50%로 혼합한 배지에 밀리타리스 동충하초 균주을 접종하여 발효한 발효물을 70% EtOH추출하였을 경우에 가장 많았으며, 번데기를 첨가하지 않은 밀리타리스 동충하초 발효물 추출물보다 함량이 33배 정도 증가하였다. 또한 추출용매의 극성이 70% EtOH보다 높거나 낮아지면 감소하는 경향이었으며, 특히 발효원으로서 번데기의 첨가는 cordycepin의 함량을 현저하게 증가시켰다. Liquiritin의 함량은 발효하지 않은 감초보다 눈꽃 동충하초와 밀리타리스 동충하초로 발효한 모든 추출물에서 감소하였다. Liquiritigenin의 함량은 눈꽃 동충하초로 발효한 추출물이 밀리타리스 동충하초 발효 추출물보다 현저히 증가하였으나, 밀리타리스 동충하초 균쥬의 발효 추출물은 발효하지 않은 감초추출물과 거의 차이가 없었으며, 두 균주 모두 번데기의 첨가량이 증가할수록 liquiritigenin의 함량이 감소하는 경향이었다. 감초에 번데기의 첨가량 또는 추출 용매의 극성이 증가하면 liquiritin과 glycyrrhizin의 함량은 현저히 감소하였다. 이상의 결과로부터, cordycepin 함량은 C. militaris 균주로 liquiritigenin은 P. tenuipes로 발효시에 현저하게 증가하였으나, liquiritin과 glycyrrhizin은 감소하였다. 감초를 동충하초로 발효시에 번데기의 첨가는 주요 성분의 변화를 현저하게 유도하였다. 동충하초 발효 추출물은 NO생성 억제효과가 증가하였으며, 고극성 용매추출물에서 그 효과가 현저하였다. 감초의 발효시에 생성된 cordycepin과 liquiritin, liquiritigenin 및 glycyrrhizin의 함량은 발효원으로서 첨가되는 번데기, 추출용매의 극성, 발효 균주의 종류 등에 따라서 현저한 차이가 있었다. 이러한 결과는 동충하초 균주를 이용한 기능성 식품 소재를 개발하기 위한 기초 자료로서 활용이 가능할 것으로 기대된다. Traditional Korean fermented herbal plants are potential sources of new food that promote health, but they are still produced by yeast, fungi or bacteria fermentation. In the present work, mushroom (Paecilomyces tenuipes and Cordyceps militaris) fungal dongchunghacho were used to fermented Glycyrrhiza uralensis Fischer (licorice) or mixed with pupa. The pupa were tested as solid substrates for the production of corcycepin, liquiritin, and liquiritigenin. The fermented substrates were analyzed the content of cordycepin, liquiritin, liquiritigenin, and glycirrhizin productivity and inhibitory activity of NO. The cordycepin content of 70% EtOH extract from the fermented mixture of licorice and 50% pupa with C. militaris increased maximum at 33 times. Pupa was very excellent for the production of cordycepin. The liquiritin content was decreased in all the assays inoculated with P. tenuipes and C. militaris dongchunghachos. The liquiritigenin content was higher when fermented with P. tenuipes than C. militaris. The addition of pupa significantly reduced the liquiritin content and glycyrrhizin production. As a result, the liquiritigenin content increased in fermented P. tenuipes and C. militaris, and liquiritin and glycyrrhizin decreased. The inhibition of NO production in the different ethanolic extracts fermented with licorice and pupa was also significantly increased and higher than that of a nonfermented extract in higher polar solvent extracts. The contents of cordycepin and biological active compounds were altered in accordance with the concentration of pupa and fungi. This study provides basic data for use in developing dongchunghacho fungi as a functional food resource.

        <P><B>Abstract</B></P> <P>We synthesized highly porous hypercross-linked polymers of (4, 4′-bis ((chloromethyl)-1, 1′-biphenyl-benzyl chloride)) [designated as HCP-BCMB] and (1, 3, 5-triphenyl benzene-1, 4-bis (chloromethyl) benzene) [designated as HCP-TPB] as sorbents for the removal of volatile organic compounds such as chloroform, toluene and ethyl methyl ketone under humidified conditions. The micro-porous nature of the hypercross-linked polymers (pore diameter 0.8nm) was confirmed by Brunner–Emmet–Teller (BET) and scanning electron microscopy (SEM) techniques. HCP-TPB showed a maximum adsorption of 32.8 wt% for toluene. In addition, both the HCPs exhibited CO<SUB>2</SUB> adsorption close to 6 wt% at 25°C and 1atm.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Synthesized highly porous hypercross-linked polymers as a sorbent for the removal of VOCs. </LI> <LI> Porous nature of the HCPs was confirmed by BET and SEM techniques. </LI> <LI> Maximum toluene adsorption of 32.8 wt% by HCP-TPB. </LI> <LI> DRS UV–visible spectroscopy confirms conjugated quinonoid matrix in both the polymers. </LI> </UL> </P>

        SDR front-end를 위한 Complex Bandpass Sampling

        Bandpass sampling(BPS) 기술은 나이퀴스트(Nyquist) 샘플링 주파수보다 낮은 주파수를 사용하여 RF 대역의 신호를 직접 하향변환 할 수 있다는 장점을 가지고 있지만, 나이퀴스트 영역에서 self-image의 중복을 피하기 위해서는 샘플링 주파수의 선택에 제약이 따른다. 2개의 ADC를 사용하는 2차(second-order) BPS는 self-image를 제거하기 위한 신호처리가 추가 된다는 조건으로 샘플링 주파수의 선택이 자유롭다. 하지만 RF 대역이 바뀌면 신호처리를 위한 파라미타를 재구성해야 한다. 본 논문에서는 2차 BPS의 한 형태인 quadrature BPS의 구조를 가지면서, 재구성이 필요 없는 간단한 보상 필터만을 사용하여 임의 RF 대역으로부터 나이퀴스트 영역으로 하향 변환하는 complex BPS 기반의 SDR front-end에 대하여 기술한다. Bandpass sampling technique has an advantage that it uses lower sampling frequency than Nyquist criterion. But special care is required in choosing sampling frequency to avoid self-image overlapping in the first Nyquist region. Recently, the second-order BPS techniques which can suppress possible self-image by using an additional ADC and by employing digital signal processing have been proposed. This paper addresses a complex BPS based SDR front-end. Unlike general second-order BPS, it needs simple FIR filter to compensate delay in the second ADC. We show a method to find proper sampling frequencies to down convert RF signals selected by tunable RF filter operating in arbitrary frequency range.

        Potential for Augmentation of Fruit Quality by Foliar Application of Bacilli Spores on Apple Tree

        Previous studies have addressed the management of phyllosphere pathogens by leaf and root-associated microbes. The present study evaluated the effect of the foliar application of three strains of Bacillus spp. on plant growth and fruit quality. The application of a bacilli spore preparation significantly improved leaf growth parameters such as leaf thickness and photosynthesis capacity, indicating that bacilli treatment directly promoted leaf growth. In addition, foliar treatment resulted in an improvement in the key indicators of fruit quality including water, glucose, and sucrose contents. The present results suggest that foliar spraying of beneficial bacilli is a potential treatment of wide application for the improvement of apple quality. Foliar application of bacilli preparation as effective plant growth-promoting rhizobacteria broadens the spectrum of their availability for orchard application.
