        The transition and post-transition metal ion chemistry of a wide range of potentially pentadentate dibenzo-substituted macrocyclic ligands incorporating nitrogen, oxygen and/or sulfur donors is reviewed and shown to result in a diverse range of structural types. Aspects of metal ion recognition, bulk membrane transport, systems incorporating appended chromophores, a sulfate binding system, induced Cu(I)/Cu(II) redox switching, coordination polymers, and unsymmetric macrocyclic ligand systems are all discussed.

        Exploring Clinical Subgroups of Participants with Major Depressive Disorder that may Benefit from Adjunctive Minocycline Treatment

        Objective: To explore illness-related factors in patients with major depressive disorder (MDD) recipients of adjunctive minocycline (200 mg/day) treatment. The analysis included participants experiencing MDD from a 12-week, double blind, placebo-controlled, randomized clinical trial (RCT). Methods: This is a sub-analysis of a RCT of all 71 participants who took part in the trial. The impact of illness chronicity(illness duration and number of depressive episodes), systemic illness (endocrine, cardiovascular and obesity), adverse effects and minocycline were evaluated as change from baseline to endpoint (12-week) using ANCOVA. Results: There was a consistent but statistically non-significant trend on all outcomes in favour of the use of adjunctive minocycline for participants without systemic illness, less illness chronicity, and fewer adverse effects. Conclusion: Understanding the relationship between MDD and illness chronicity, comorbid systemic illness, and adverse effects, can potentially better characterise those individuals who are more likely to respond to adjunctive anti-inflammatory medications.

        Experimental Investigation of Mechanical Properties, Formability and Force Measurement for AA7075-O Aluminum Alloy Sheets Formed by Incremental Forming

        Incremental sheet forming (ISF) has demonstrated its high potential to shape complex three-dimensional components without using specific tooling, thus enabling product customization, cost reduction and efficiencies. This paper presents an experimental campaign to investigate AA7075-O aluminum alloy sheet forming using single point incremental forming (SPIF) technology and understand the forming process mechanism. Firstly, tensile tests were carried out to characterize the mechanical properties of AA7075-O sheets with three different thicknesses. Then, the effects of tool path types with different incremental steps on the maximum part draw angle and the successful part height were evaluated to clarify the formability. Additionally, a fracture forming limit diagram was developed to give the design limits for strain. Finally, the trends in forming forces were analyzed considering the influence of different draw angles,sheet thicknesses, step-down sizes and sheet orientations. Experimental results showed that formability is affected by the part draw angle and incremental steps of the tool path. However, the likelihood of successful forming also depends on other geometrical design parameters such as the part height. In addition, the influences of process parameters on tool forces provide further insights into the deformation mechanics of AA7075-O sheets. The slope of the force curve after the peak value can be identified as a forming failure prediction indicator regardless of the tool path types when using a truncated cone as a benchmark. Also the influence of sheet orientations on forming forces was investigated in SPIF. The vertical forming force is found to be smaller in a sheet orientation 45to the rolling direction than that of a sheet orientation 0 for three different sheet thicknesses.

      • The International Mouse Phenotyping Consortium (IMPC): a functional catalogue of the mammalian genome that informs conservation

        <P>The International Mouse Phenotyping Consortium (IMPC) is building a catalogue of mammalian gene function by producing and phenotyping a knockout mouse line for every protein-coding gene. To date, the IMPC has generated and characterised 5186 mutant lines. One-third of the lines have been found to be non-viable and over 300 new mouse models of human disease have been identified thus far. While current bioinformatics efforts are focused on translating results to better understand human disease processes, IMPC data also aids understanding genetic function and processes in other species. Here we show, using gorilla genomic data, how genes essential to development in mice can be used to help assess the potentially deleterious impact of gene variants in other species. This type of analyses could be used to select optimal breeders in endangered species to maintain or increase fitness and avoid variants associated to impaired-health phenotypes or loss-of-function mutations in genes of critical importance. We also show, using selected examples from various mammal species, how IMPC data can aid in the identification of candidate genes for studying a condition of interest, deliver information about the mechanisms involved, or support predictions for the function of genes that may play a role in adaptation. With genotyping costs decreasing and the continued improvements of bioinformatics tools, the analyses we demonstrate can be routinely applied.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (10.1007/s10592-018-1072-9) contains supplementary material, which is available to authorized users.</P>

        Frictional contact analysis in a spherical roller bearing

        Numerical analyses of the roller–raceway contact have been carried out in a spherical roller bearing using frictional contact models of different complexity. The models used in the study include an implementation of Kalker’s exact contact theory named CECT (Conformal Exact Contact Theory) and detailed Finite Element models. The adequacy of the more simplified contact solutions is assessed by contrasting them with the solutions obtained with the more comprehensive models. Additionally, the use of the exact contact theory, well known in the wheel–rail application, is demonstrated in contact mechanics analyses in rolling bearings, describing relevant details of its implementation for this application. Situations with different normal loads and friction levels have been analysed, and two distinct steady equilibrium configurations of the roller have been identified.

        Dietary Supplementation with Pressurized Whey in Patients with Cystic Fibrosis

        Cystic fibrosis (CF) is characterized by malnutrition, chronic pulmonary inflammation, and oxidative stress. Whey protein is rich in sulfhydryl groups and is recognized for its ability to increase glutathione and reduce oxidative stress. Previously, we have shown that supplementation with whey increased intracellular glutathione levels in patients with CF. We have subsequently shown that hyperbaric pressure treatment of whey protein promotes the release of novel peptides for absorption, increases intracellular glutathione in healthy subjects, and reduces in vitro production of interleukin (IL)-8. We hypothesized that pressurized whey supplementation in children and adults with CF could have significant nutritional and anti-inflammatory benefits. A pilot open-label study of 1-month dietary supplementation with pressurized whey in CF patients was undertaken to assess the effects. Twenty-seven patients with CF (nine children, 18 adults) were enrolled. The dose of pressurized whey was 20g/day in patients less than 18 years of age and 40g/day in older patients. Anthropometric measures, pulmonary function, serum C-reactive protein (CRP), whole blood glutathione, and whole blood IL-8 and IL-6 responses to phytohemagglutinin (PHA) stimulation were measured at baseline and at 1 month. Three adults withdrew (one with gastrointestinal side effects, two with acute infection). Both children and adults showed enhancements in nutritional status, as assessed by body mass index. Children showed improvement in lung function (forced expiratory volume in 1 second). The majority of patients with an initially elevated CRP showed a decrease. PHA-stimulated IL-8 responses tended to decrease in the adults. Whole blood glutathione levels did not change. Thus, oral supplementation with pressurized whey improves nutritional status and can have additional beneficial effects on inflammation in patients with CF.
