        Maternal betaine supplementation ameliorates fatty liver disease in offspring mice by inhibiting hepatic NLRP3 inflammasome activation

        BACKGROUND/OBJECTIVES: Previous research has shown maternal betaine supplementation alleviates fetal-derived hepatic steatosis. Therefore, this study examined the anti-inflammatory effect of maternal betaine intake in offspring mice and its mechanism. MATERIALS/METHODS: Female C57BL/6J mice and their offspring were randomly divided into 3 groups according to the treatment received during gestation and lactation: control diet (CD), fatty liver disease (FLD), and fatty liver disease + 1% betaine (FLD-BET). The FLD group was given a high-fat diet and streptozotocin (HFD + STZ), and the FLD-BET group was treated with HFD + STZ + 1% betaine. After weaning, the offspring mice were given a normal diet for 5 weeks and then dissected to measure the relevant indexes. RESULTS: Compared to the CD group, the offspring mice in the FLD group revealed obvious hepatic steatosis and increased serum levels of alanine aminotransferase, interleukin (IL)-6, and tumor necrosis factor (TNF)-α; maternal betaine supplementation reversed these changes. The hepatic mRNA expression levels of IL-6, IL-18, and Caspase-1 were significantly higher in the FLD group than in the CD group. Maternal betaine supplementation reduced the expression of IL-1β, IL-6, IL-18, and apoptosis-associated speck-like protein containing C-terminal caspase recruitment domain (ASC). Maternal betaine supplementation also reversed the increasing protein expressions of nitric oxide dioxygenase-like receptor family pyrin domain containing 3 (NLRP3), ASC, Caspase-1, IL-1β, and IL-18 in offspring mice exposed to HFD + STZ. Maternal betaine supplementation decreased the homocysteine (Hcy) and s-adenosine homocysteine (SAH) levels significantly in the livers. Furthermore, the hepatic Hcy concentrations showed significant inverse relationships with the mRNA expression of TNF-α, NLRP3, ASC, and IL-18. The hepatic SAH concentration was inversely associated with the IL-1β mRNA expression. CONCLUSIONS: The lipotropic and anti-inflammatory effect of maternal betaine supplementation may be associated with the inhibition of NLRP3 inflammasome in the livers of the offspring mice.

        天人合一與生成變易 - 中國傳統文化的精髓和硬核

          천인합일과 생성변역은 중국 고대 역학사유 중에 매우 중요한 두 기본 개념이다. 천인합일이 정체관이라면, 생성변역은 우주생성관이다. 천인합일 혹은 천인감응은 또한 사람과 자연, 주체와 객체, 그리고 마음과 물질의 통일을 말한다. 이것은 유기적 자연관과 정체 사유형식을 가진다. 사람에 의해 출발하여 사람-가족-사회-자연으로 점차 확대되며, 최후에는 천인합일의 유기적 우주상을 완전하게 형성한다. 자연과 인간이 감응하여 만물을 생성하게 되는데, 바로 역학에서 ‘낳고 낳는 것을 역’이라 함을 말한다.<BR>  천인합일과 생성변역은 매우 밀접한 관련성을 가진다. 대대와 유행 두 가지가 서로 결합하여 우주발전과 인류문명 발전의 도식을 형성하였다. 역학의 이러한 독특한 우주 도식은 서양 근대 과학 가운데서 형성되고 발전하였으나 별 다른 효과를 얻을 수 없었다. 그러나 사람과 자연을 결합시키고, 현대와 복잡성을 탐색하는 오늘날 그것은 중국과 서양 각국의 과학기술 발전과 사회 발전을 위한 무한한 원천이 되고 있다. 본 논문에서는 생성성학습을 사례로 하여 천인합일과 생성변역의 현대적 의의를 밝히고자 한다.   Tian Ren He Yi(天人合一, unity of heaven and man) and Sheng Cheng Bian Yi(生成變易, generations and changes) are two fundamental concepts essential to the mentality of the inquiry of I-Ching. The former is a holistic view and the latter an outlook of generative cosmology. Tian Ren He Yi or Tian Ren Gan Ying(天人感應, responses between heaven and man) means the unities of man and nature, subject and object as well as mind and body, which is an organic naturalism and organic manner of thinking. It starts from man and follows the line of gradual amplification of man to family to society and finally to the nature, consequently forming an organic cosmological picture. Everything would be generated according the responses between heaven and man. This is the claim by the inquiry of I-Ching as Sheng Sheng Zhi Wei Yi (“生生之謂易” “Yi”(Changes) takes the way of heaven and earth as its principle and makes things move, progress, change and develop with no end, what is above is the meaning of Yi.").<BR>  The interconnection of Tian Ren He Yi and Sheng Cheng Bian Yi, and the combination of Dui Dai(對待, opposing) and Liu Xing(流行, flowing) result in a basic schema of cosmological development and progress of human civilization claimed in the inquiry of I-Ching. This unique schema of cosmology claimed in the inquiry of l-Ching failed to play the role it has to in the formation and development for the contemporary science of western tradition, however, in the present-day exploring of establishing the new alliance of man and nature and complexity, it has already become the new intellect and inexhaustible resources for the development of science and technology as well as the social progress in China and West countries.<BR>  Finally, generative learning as case study, to expound the contempory significance of Tian Ren He Yi and Sheng Cheng Bian Yi.

        3D buckling analysis of FGM sandwich plates under bi-axial compressive loads

        Based on the Reissner mixed variational theorem (RMVT), finite rectangular layer methods (FRLMs) are developed for the three-dimensional (3D) linear buckling analysis of simply-supported, fiber-reinforced composite material (FRCM) and functionally graded material (FGM) sandwich plates subjected to bi-axial compressive loads. In this work, the material properties of the FGM layers are assumed to obey the power-law distributions of the volume fractions of the constituents through the thickness, and the plate is divided into a number of finite rectangular layers, in which the trigonometric functions and Lagrange polynomials are used to interpolate the in- and out-of-plane variations of the field variables of each individual layer, respectively, and an h-refinement process is adopted to yield the convergent solutions. The accuracy and convergence of the RMVT-based FRLMs with various orders used for expansions of each field variables through the thickness are assessed by comparing their solutions with the exact 3D and accurate two-dimensional ones available in the literature.

        microRNAs (miRNAs) are small non-coding RNAs that regulate gene expression through post-transcriptional interactions with mRNA. miRNAs have recently emerged as key regulators of various cancers. Although miR-27a has been implicated in several other cancers, its role in hepatitis B virus-related hepatocellular carcinoma (HCC) is unknown. In this study, we showed miR-27a to be frequently up-regulated in HCC tissues and HCC cell lines (HepG2 and Huh7). Overexpression of miR-27a enhanced cell proliferation, promoted migration and invasion, and activated cell cycling in HepG2 and Huh7 cells. In summary, our results suggest that up-regulation of miR-27a may play an oncogenic role in the development of HCC and might thus be a new therapeutic target in HCC patients.

        The Characteristics and Distribution of Eukaryotic Phytoplankton Community in Liaodong Bay, China

        Brown tides have occurred in the Bohai Sea of China since 2009 and caused serious losses in aquaculture. In recent years, phytoplankton identification has been greatly promoted by the development of high-throughput sequencing technologies. In this study, V4 region of 18S rDNA was used as the target gene to design the primer V4 (F/R), and high-throughput sequencing technology was adopted to evaluate the diversity of composition and characteristics of the eukaryotic phytoplankton community. A total of 93 alga species were identified, and about 60% (56 out of 93) of them were undetected or unreported in Chinese waters. There were 59% of the total algal species identified as autotrophy in trophic mode while heterotrophy and mixotrophy accounted for 12% and 29% of the total algal species, respectively. 8 species of picophytoplankton that were mainly distributed in the south and a partial area of the east and west formed the scientific basis for the quality and safety early-warning in aquatic products and brown rides study. Diversity index analysis also revealed a trend of fluctuations of marine phytoplankton diversity in different sample sites in Liaodong Bay. In conclusion, our work provides a detailed evaluation of eukaryotic phytoplankton diversity in Liaodong Bay, suggesting the need for long-term monitoring to further study and guard against the potential risk of harmful algal blooms.

        Tissue Quality Comparison Between Heparinized Wet Suction and Dry Suction in Endoscopic Ultrasound-Fine Needle Biopsy of Solid Pancreatic Masses: A Randomized Crossover Study

        Background/Aims: A high-quality sample allows for next-generation sequencing and the administration of more tailored precision medicine treatments. We aimed to evaluate whether heparinized wet suction can obtain higher quality samples than the standard dry-suction method during endoscopic ultrasound (EUS)-guided biopsy of pancreatic masses. Methods: A prospective randomized crossover study was conducted. Patients with a solid pancreatic mass were randomly allocated to receive either heparinized wet suction first or dry suction first. For each method, two needle passes were made, followed by a switch to the other method for a total of four needle punctures. The primary outcome was the aggregated white tissue length. Histological blood contamination, diagnostic performance and adverse events were analyzed as secondary outcomes. In addition, the correlation between white tissue length and the extracted DNA amount was analyzed. Results: A total of 50 patients were enrolled, and 200 specimens were acquired (100 with heparinized wet suction and 100 with dry suction), with one minor bleeding event. The heparinized wet suction approach yielded specimens with longer aggregated white tissue length (11.07 mm vs 7.96 mm, p=0.001) and less blood contamination (p=0.008). A trend towards decreasing tissue quality was observed for the 2nd pass of the dry-suction method, leading to decreased diagnostic sensitivity and accuracy, although the accumulated diagnostic performance was comparable between the two suction methods. The amount of extracted DNA correlated positively to the white tissue length (p=0.001, Spearman̕s ρ=0.568). Conclusions: Heparinized wet suction for EUS tissue acquisition of solid pancreatic masses can yield longer, bloodless, DNA-rich tissue without increasing the incidence of adverse events (ClinicalTrials.gov. identifier NCT04707560).
