MicroRNA-576-3p Inhibits Proliferation in Bladder Cancer Cells by Targeting Cyclin D1

MicroRNAs (miRNAs) are small, endogenous RNAs that play important gene-regulatory roles by binding to the imperfectly complementary sequences at the 3 -UTR of mRNAs and directing their gene expression. Here, we first discovered that miR-576-3p was down-regulated in human bladder cancer cell lines compared with the non-malignant cell line. To better characterize the role of miR-576-3p in bladder cancer cells, we over-expressed or down-regulated miR-576-3p in bladder cancer cells by transfecting with chemically synthesized mimic or inhibitor. The overexpression of miR-576-3p remarkably inhibited cell proliferation via G1-phase arrest, and decreased both mRNA and protein levels of cyclin D1 which played a key role in G1/S phase transition. The knock-down of miR-576-3p significantly promoted the proliferation of bladder cancer cells by accelerating the progression of cell cycle and increased the expression of cyclin D1. Moreover, the dual-luciferase reporter assays indicated that miR-576-3p could directly target cyclin D1 through binding its 3 -UTR. All the results demonstrated that miR-576-3p might be a novel suppressor of bladder cancer cell proliferation through targeting cyclin D1.

The objective was to compare meat quality, muscle fiber characteristics, lipid oxidation and fatty acids of Limousin (LIM), Simmtental (SIM), Luxi (LX), Qinchuan (QC) and Jinnan (JN) offered the same diet in China. After finishing, eight bulls from each breed were randomly selected for slaughter at 18.5 months old. Longissimus dorsi (ld) muscle was taken from the carcass for meat quality evaluations. Breed had little effect on most of meat and fat color parameters except for Hue and $b^*$ in which QC had lower values. LIM showed higher pH (24 h) and better water holding capacity than other breeds. LIM showed the lowest dry matter content but the highest crude protein. LX and LIM had higher percentage and density of red muscle fiber than other breeds. Lipid oxidations were significantly lower in LIM than in QC, with the LX, SIM and JN having the intermediate values. Compared to other four breeds, QC provided the highest values of polyunsaturated fatty acids (PUFA), n-6 fatty acids and n-3 fatty acids. In conclusion, LIM scored better on most of meat quality characteristics; however, local breeds such as LX and QC also had better muscle fiber characteristics and better fatty acids composition.

Purpose: To explore expression of SP and NK1R in basophils of allergic asthma (AA), allergic rhinitis (AR) and AR combined with AA (ARA), and influence of allergens and immunoglobulin E (IgE) mediated mechanisms on SP and NK1R expression. Methods: Expression of SP and NK1R was detected by flow cytometry, NK1R mRNA expression was detected by real time quantitative polymerase chain reaction (qPCR), and mouse AR and AA models were employed for in vivo study. Results: SP+ and NK1R+ cells increased in CCR3+ and CD123+HLA-DR− granulocytes of AA. PPE elevated proportions of SP+ cells in CCR3+ and CD123+HLA-DR− granulocytes, whereas ASWE and HDME augmented SP+ cells in CD123+HLA-DR− granulocytes of AR and ARA patients. ASWE, HDME and PPE increased proportions of NK1R+ cells in CCR3+ PBMC and CD123+HLA-DR− granulocytes of AR patients. OVA, Der p1, IL-33, IL-37, IgE and SP enhanced NK1R expression on KU812 cells. NK1R expressing basophils were increased in blood of OVA sensitized and challenged AR and AA mice. FcεRI-KO AA mice seemed to have less NK1R+ basophils than WT AA mice in their blood. Conclusion: CCR3+ and CD123+HLA-DR− cells are likely involved in AA and AR via SP and NK1R. IgE-related mechanism may participate in upregulation of NK1R expression.
Lyapunov Conditions for Finite-time Stability of Stochastic Functional Systems

In this study, we consider the finite time stability (FTS) and finite time contractive stability (FTCS) of the stochastic functional systems. For the framework of Lyapunov-Ruzumikhin method, the sufficient conditions of FTS and FTCS are given first. Then the theory is applied to FTS and FTCS of linear time-varying stochastic functional systems. Finally, experiments on the numerical examples have demonstrated the superiority of the proposed method.

Thermodynamic analysis of the correlation of H_2S and COS has been carried out at the temperature range of 400-650 ℃ at which high temperature desulfurization of coal gas is usually performed. The correlation of the two sulfur species is mainly through the reaction H_2S+CO→COS+H_2. Simulated coal gas with the following composition CO 32.69%, H_2 39.58%, CO_2 18.27%, N2 8.92% and H_2S 0.47% was used in this study, and the equilibrium concentrations of the two species at different temperatures were calculated. The results of Fe-based sorbents during sulfidation were compared with calculations. It is concluded that the above reaction may reach equilibrium concentration in the presence of the Fe-based sorbents, which means the Fe-based sorbents may effectively catalyze the reaction between H_2S and CO. Because of the correlation of the two sulfur species, both can be effectively removed at high temperatures simultaneously, offering high temperature desulfurization some advantages over low temperature desulfurization processes.
MicroRNA-409-3p Inhibits Migration and Invasion of Bladder Cancer Cells via Targeting c-Met

There is increasing evidence suggesting that dysregulation of certain microRNAs (miRNAs) may con-tribute to tumor progression and metastasis. Previous studies have shown that miR-409-3p is dysregulated in some malignancies, but its role in bladder cancer is still unknown. Here, we find that miR-409-3p is down-regulated in human bladder cancer tissues and cell lines. Enforced expression of miR-409-3p in bladder cancer cells significantly reduced their migration and invasion without affecting cell viability. Bioinformatics analysis identified the pro-metastatic gene c-Met as a potential miR-409-3p target. Further studies indicated that miR-409-3p suppressed the expression of c-Met by binding to its 3-untranslated region. Silencing of c-Met by small interfering RNAs phenocopied the effects of miR-409-3p overexpression, whereas restoration of c-Met in bladder cancer cells bladder cancer cells overexpressing miR-409-3p, partially reversed the suppressive effects of miR-409-3p. We further showed that MMP2 and MMP9 may be downstream effector proteins of miR-409-3p. These findings indicate that miR-409-3p could be a potential tumor suppressor in bladder cancer.
A modified error-oriented weight positioning model based on DV-Hop

The distance vector-hop (DV-Hop) is one of the emblematic algorithms that use node connectivity for locating, which often accompanies by a large positioning error. To reduce positioning error, the bio-inspired algorithm and weight optimization model are introduced to address positioning. Most scholars argue that the weight value decreases as the hop counts increases. However, this point of view ignores the intrinsic relationship between the error and weight. To address this issue, this paper constructs the relationship model between error and hop counts based on actual communication characteristics of sensor nodes in wireless sensor network. Additionally, we prove that the error converges to 1/6CR when the hop count increase and tendency to infinity. Finally, this paper presents a modified error-oriented weight positioning model, and implements it with genetic algorithm. The experimental results demonstrate excellent robustness and error removal.
The Detection of Osthole and Application of Cell Imaging Based on Nitrogen-Doped Carbon Dots

A fluorescence probe has been synthesized for the detection of osthole using the nitrogen-doped carbon dots (NCDs) as shown in Fig. 1. The NCDs were fully characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), x-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FTIR). Under the optimal experimental conditions, the NCDs fluorescence probe was highly selective and sensitive to osthole. The linear response range for osthole was 5.0–7 5μM with a detection limit of 38 nM. The mechanism of the interaction of osthole and NCDs was discussed. The fluorescence probe has been applied to the analysis of biological samples. The as-synthesized NCDs with high fluorescence intensity, low toxicity and good biocompatibility were applied to cell imaging.
