Background and Purpose This study investigated the contribution of white-matter hyperintensities (WMH) and lacunar infarcts (LI) to the risk of Alzheimer’s disease (AD) in an elderly cohort in China. Methods Older adults who were initially cognitively normal were examined with MRI at baseline, and followed for 5 years. WMH were classified as mild, moderate, or severe, and LI were classified into a few LI (1 to 3) or many LI (≥4). Cognitive function was assessed using the Mini Mental State Examination and the Activities of Daily Living scale. Results Among the 2,626 subjects, 357 developed AD by the end of the 5-year follow-up period. After adjusting for age and other potential confounders, having only WMH, having only LI, and having both WMH and LI were associated with an increased risk of developing AD compared with having neither WMH nor LI. Moderate and severe WMH were associated with an increased risk of developing AD compared with no WMH. Furthermore, patients with many LI had an increased risk of developing AD compared with no LI. Conclusions Having moderate or severe WMH and many LI were associated with an increased risk of developing AD, with this being particularly striking when both WMH and LI were present.

<P><B>Abstract</B></P> <P>One-dimensional (1-D) carbon nanofibers anchored with partially reduced SnO<SUB>2</SUB> nanoparticles (SnO<SUB>2</SUB>/Sn@C) were successfully synthesized through a simple electrospinning method followed by carbon coating and thermal reduction processes. The partially reduced Sn frameworks, combined with the carbon fibers, provide a more favorable mechanism for sodiation/desodiation than SnO<SUB>2</SUB>. As a result, SnO<SUB>2</SUB>/Sn@C exhibits a high reversible capacity (536mAhg<SUP>−1</SUP> after 50cycles) and an excellent rate capability (396mAhg<SUP>−1</SUP> even at 2C rate) when evaluated as an anode material for sodium-ion batteries (SIBs).</P> <P><B>Highlights</B></P> <P> <UL> <LI> The partially reduced Sn framework provides both favorable Na-ion diffusion dynamics and high specific capacity. </LI> <LI> The conductive Sn framework, bonded with carbon fiber, provides an efficient pathway for electron transport. </LI> <LI> The discrete SnO<SUB>2</SUB>/Sn nanoparticles provide sufficient space to accommodate the large volume expansion. </LI> <LI> The strategy of partial reduction may open up new ways to improve the electrochemical performance of metal oxides. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Separation of bacterial cells from soil is a key step in the construction of metagenomic BAC libraries with large DNA inserts. Our results showed that when combined with sodium pyro-phosphate and homogenization for soil dispersion, sucrose density gradient centrifugation (SDGC) was more effective at separating bacteria from soil than was low speed centrifugation (LSC). More than 70% of the cells, along with some soil colloids,were recovered with one round of centrifugation. A solution of 0.8% NaCl was used to resuspend these cell and soil pellets for purification with nycodenz density gradient centrifugation (NDGC). After purification,more than 30% of the bacterial cells in the primary soil were extracted. This procedure effectively removed soil contamination and yielded sufficient cells for high molecular weight (HMW) DNA isolation. Ribosomal intergenic spacer analysis (RISA) showed that the microbial community structure of the extracted cells was similar to that of the primary soil, suggesting that this extraction procedure did not significantly change the the soil bacteria community structure. HMW DNA was isolated from bacterial cells extracted from red soil for metagenomic BAC library construction. This library contained DNA inserts of more than 200 Mb with an average size of 75 kb.
The discovery of potent immunostimulatory CpG-ODNs widely distributed in bacterial genomes

Oligodeoxynucleotides containing unmethylated CpG dinucleotides (CpG-ODN) can be specifically recognized by Toll-like receptor 9 (TLR9), provoking innate immune responses. Designed according to this structural feature, many synthetic phosphorothioate CpG-ODNs successfully activate macrophages. However, it is difficult to find potent stimulatory CpG-DNA fragments in microbial genomes. Therefore, whether microbial CpG-DNA substantially contributes to infectious and immune diseases remains controversial. In this study, high-throughput scanning was carried out for thousands of bacterial genomes with bioinformatics tools to comprehensively evaluate the distribution of CpG-DNA fragments. A random sampling test was then performed to verify their immunostimulatory properties by experiments in vitro and in vivo. Natural TLR9-dependent and potent stimulatory CpG-DNA fragments were found in microbial genomes. Interestingly, highly conserved stimulatory CpG-DNA fragments were found in 16S and 23S rDNA sequences with multiple copies, while others were species-specific. Additionally, we found that the reported active motifs were mostly nonstimulatory in natural CpG fragments. This evidence indicates that the previous structural descriptions of functional CpG-ODNs are incomplete. Our study has assessed the distribution of microbial CpG-DNA fragments, and identified natural stimulatory CpG-DNA fragments. These findings provide a deeper understanding of CpG-ODN structures and new evidence for microbial DNA inflammatory function and pathogenicity.

Purpose: Asthma is a common chronic inflammatory respiratory tract disease with high morbidity and mortality. The global trends in asthma burden remain poorly understood, and asthma incidence has increased during the worldwide coronavirus disease 2019 (COVID-19) pandemic. This study aimed to provide a comprehensive view of the global distribution of asthma burden and its attributable risk factors from 1990 to 2019. Methods: Based on the Global Burden of Disease Study 2019 Database, asthma incidence, deaths, disability-adjusted life years (DALYs), the corresponding age-standardized incidence rate (ASIR), age-standardized death rate (ASDR), age-standardized DALY rate, and estimated annual percentage change were analyzed according to age, sex, sociodemographic index (SDI) quintiles, and locations. Risk factors contributing to asthma deaths and DALYs were also investigated. Results: Globally, the asthma incidence increased by 15%, but deaths and DALYs decreased. The corresponding ASIR, ASDR, and age-standardized DALY rate also decreased. The high SDI region had the highest ASIR, and the low SDI region had the highest ASDR. The ASDR and age-standardized DALY rate were negatively correlated with the SDI. The low-middle SDI region, particularly South Asia, showed the highest asthma-related deaths and DALYs. The incidence peak was under 9 years old, and more than 70% of all deaths occurred in the population over 60 years old. Smoking, occupational asthmagens, and a high body mass index were the main risk factors for asthma-related mortality and DALYs, and their distributions varied between sexes. Conclusions: Globally, the asthma incidence has increased since 1990. The greatest asthma burden is borne by the low-middle SDI region. The 2 groups that need special attention are those under 9 years old and those over 60 years old. Targeted strategies are needed to reduce the asthma burden based on geographic and sex-age characteristics. Our findings also provide a platform for further investigation into the asthma burden in the era of COVID-19.
Antibacterial Polyketides from the Jellyfish-Derived Fungus <i>Paecilomyces variotii</i>

<P>Four new polyketides (<B>1</B>–<B>4</B>) were isolated from the fungus <I>Paecilomyces variotii</I>, which was derived from the jellyfish <I>Nemopilema nomurai</I>. The planar structures and relative configurations of these polyketides were elucidated on the basis of spectroscopic analyses, including 2D NMR experiments. Influence of Swirl Injector Diameter on the Process of Conical Sheet Breakup

High-speed shadowgraph was used to capture the process of conical liquid sheet ejected from the central hole in coaxial swirl injector of gas generator; and the impact of injector diameter was especially analyzed. With the increasing of injector diameter, breakup length shortens and the cone angle becomes larger. For this kind of injector, the breakup lengths are around 50mm and the cone angles are less than 60 degree though the pressure drop exceeds 3.0 MPa. The experimental cone angles were far less than the therotical formula when compared with Abromvich and Lefebvre`s result, while they have the same trend that cone angle increases with injector`s characteristic parameter A. The understanding of conical sheet breakup is beneficial to master the atomization character of coaxial swirl injector.
An Anacardic acid Analog from the Jellyfish-derived Fungus Paecilomyces variotii

An anacardic acid analog (1) was isolated from the fungus Paecilomyces variotii which was derived from the giant jellyfish Nemopilema nomurai. Compound 1 was isolated from a natural source for the first time and was evaluated for antibacterial activity against human and marine pathogens, including MDR (multidrug-resistant) strains. Compound 1 exhibited mild antibacterial activity against Escherichia coli DC 2, Streptococcus iniae, and methicillin-resistant Staphylococcus aureus 3089 (MRSA).
