      • In Vivo Inhibition of Bone Morphogenetic Protein-2 on Breast Cancer Cell Growth

        STUDY DESIGN.: In vitro and in vivo study. OBJECTIVE.: To evaluate the role of recombinant human bone morphogenetic protein-2 (rhBMP2) on breast cancer cell (MDA-MB-231 cells) growth. SUMMARY OF BACKGROUND DATA.: Bone morphogenetic proteins (BMPs) are expressed in a variety of human carcinoma cell lines and are known to promote tumor invasion and metastasis. However, their roles in tumor progression have not been fully clarified. In addition, there is no in vivo study regarding the inhibitory effect of BMP2 on breast cancer cell proliferation. METHOD.: Cell proliferation was determined by BrdU incorporation assay and flow cytometry. BMP2 signal transduction pathways were estimated on Western blot. Fifteen animals were divided into 2 groups; 1 (control = 5) was breast cancer cells alone, while the other (experiment = 5) was rhBMP2 + breast cancer cells. Cancer cells were injected into 2 sites (subcutaneous and femur) of nude mice with or without BMP2. Tumor size was determined by direct measurements for subcutaneous tumor formation and by femur radiographs. Histological and immunohistochemical analyses were performed. RESULTS.: RhBMP2 inhibited the proliferation of MDA-MB-231 cells in vitro. Inhibition was associated with changes in both the Smad and Wnt signaling pathways and was ultimately mediated through effects on various cell cycle proteins. Furthermore, rhBMP2 inhibited the growth of MDA-MB-231 cells injected both subcutaneously and intrafemorally. CONCLUSION.: In this model using human breast adenocarcinoma cell line, rhBMP2 has no stimulatory effect of tumor growth. Therefore, we can provide the basic science data to support the utilization in the management of patients with spine tumor in the future.

        A Study on the Problem and Improvement of the Distribution System of Pharmaceuticals in Korea

        The purpose of this study is to examine the development of the pharmaceutical distribution industry. The pharmaceutical industry is are expected to suffer a heavy blow when the Free Trade Agreement (FTA) is introduced, despite its best efforts. Therefore, adequate solutions must be found. Section II introduces the Korean pharmaceutical distribution system and its current situation; Section III explores the distribution system’s strengths and weaknesses. Section IV, identifies the problem and possible solutions for the Korean pharmaceutical distribution system; Section V summarizes and concludes this paper and acknowledges its limitations. Finally, this paper has a clear limitation. The lack of objective information and scientific analysis due to the data being based on interviews with company representatives is its most significant shortcoming. However, it offers implications for new directions for future research.

        White-Matter Hyperintensities and Lacunar Infarcts Are Associated with an Increased Risk of Alzheimer’s Disease in the Elderly in China

        Background and Purpose This study investigated the contribution of white-matter hyperintensities (WMH) and lacunar infarcts (LI) to the risk of Alzheimer’s disease (AD) in an elderly cohort in China. Methods Older adults who were initially cognitively normal were examined with MRI at baseline, and followed for 5 years. WMH were classified as mild, moderate, or severe, and LI were classified into a few LI (1 to 3) or many LI (≥4). Cognitive function was assessed using the Mini Mental State Examination and the Activities of Daily Living scale. Results Among the 2,626 subjects, 357 developed AD by the end of the 5-year follow-up period. After adjusting for age and other potential confounders, having only WMH, having only LI, and having both WMH and LI were associated with an increased risk of developing AD compared with having neither WMH nor LI. Moderate and severe WMH were associated with an increased risk of developing AD compared with no WMH. Furthermore, patients with many LI had an increased risk of developing AD compared with no LI. Conclusions Having moderate or severe WMH and many LI were associated with an increased risk of developing AD, with this being particularly striking when both WMH and LI were present.

        ALKBH5 facilitates CYP1B1 mRNA degradation via m6A demethylation to alleviate MSC senescence and osteoarthritis progression

        Improving health and delaying aging is the focus of medical research. Previous studies have shown that mesenchymal stem cell (MSC) senescence is closely related to organic aging and the development of aging-related diseases such as osteoarthritis (OA). m6A is a common RNA modification that plays an important role in regulating cell biological functions, and ALKBH5 is one of the key m6A demethylases. However, the role of m6A and ALKBH5 in MSC senescence is still unclear. Here, we found that the m6A level was enhanced and ALKBH5 expression was decreased in aging MSCs induced by multiple replications, H2O2 stimulation or UV irradiation. Downregulation of ALKBH5 expression facilitated MSC senescence by enhancing the stability of CYP1B1 mRNA and inducing mitochondrial dysfunction. In addition, IGF2BP1 was identified as the m6A reader restraining the degradation of m6A-modified CYP1B1 mRNA. Furthermore, Alkbh5 knockout in MSCs aggravated spontaneous OA in mice, and overexpression of Alkbh5 improved the efficacy of MSCs in OA. Overall, this study revealed a novel mechanism of m6A in MSC senescence and identified promising targets to protect against aging and OA.

        Preparation and antimicrobial assay of ceramic brackets coated with TiO₂ thin films

        Objective: Different methods have been utilized to prevent enamel demineralization and other complications during orthodontic treatment. However, none of these methods can offer long-lasting and effective prevention of orthodontic complications or interventions after complications occur. Considering the photocatalytic effect of TiO₂ on organic compounds, we hoped to synthesize a novel bracket with a TiO₂ thin film to develop a photocatalytic antimicrobial effect. Methods: The sol-gel dip coating method was used to prepare TiO₂ thin films on ceramic bracket surfaces. Twenty groups of samples were composed according to the experimental parameters. Crystalline structure and surface morphology were characterized by X-ray diffraction and scanning electron microscopy, respectively; film thickness was examined with a surface ellipsometer. The photocatalytic properties under ultraviolet (UV) light irradiation were analyzed by evaluating the degradation ratio of methylene blue (MB) at a certain time. Antibacterial activities of selected thin films were also tested against Lactobacillus acidophilus and Candida albicans . Results: Films with 5 coating layers annealed at 700°C showed the greatest photocatalytic activity in terms of MB decomposition under UV light irradiation. TiO₂ thin films with 5 coating layers annealed at 700°C exhibited the greatest antimicrobial activity under UV-A light irradiation. Conclusions: These results provide promising guidance in prevention of demineralization by increasing antimicrobial activities of film coated brackets.

        Certain Polymorphisms in SP110 Gene Confer Susceptibility to Tuberculosis: A Comprehensive Review and Updated Meta-Analysis

        Purpose: Numerous studies have assessed the association of SP110 gene variants with tuberculosis (TB), but the results were inconsistent. Through a comprehensive review and meta-analysis, our study aimed to clarify the nature of genetic risks contributedby 11 polymorphisms for the development of TB. Materials and Methods: Through searching PubMed, web of science, China National Knowledge Infrastructure (CNKI) databases,a total of 11 articles including 13 independent studies were selected. The pooled odd ratios (ORs) along with their corresponding95% confidence interval (CI) were estimated for allelic comparisons, additive model (homozygote comparisons; heterozygotecomparisons), dominant model and recessive model. We also assessed the heterogeneity across the studies and publication bias. Results: The results of combined analysis revealed a significantly increased risk of TB for single nucleotide polymorphism (SNP)rs9061 in all five comparisons (allelic comparisons: OR=1.28, 95% CI=1.14–1.44, p<0.0001; homozygote comparisons: OR=2.84, 95%CI=1.84–4.38, p<0.00001; heterozygote comparisons: OR=1.23, 95% CI=1.05–1.43, p=0.009; dominant model: OR=1.32, 95% CI=1.14–1.53, p=0.0003; recessive model: OR=2.26, 95% CI=1.18–4.34, p=0.01). In subgroup analysis, the risk of TB associated with SNP rs9061appeared to be increased. Moreover, increased risk of TB was also found in Asian subgroup of SNP rs11556887, while decreased riskof TB appeared in large sample size subgroup of SNP rs1135791. No significant association was observed between other SNPs andthe risk of TB. Conclusion: Our meta-analysis suggested that the variant of SNP rs9061 might be a risk factor for TB.

        Trajectory Tracking Control for Nonholonomic Wheeled Mobile Robots with External Disturbances and Parameter Uncertainties

        Aiming at the trajectory tracking problem of nonholonomic wheeled mobile robots (WMR) with bounded external disturbances and parameter uncertainties, a dual-loop attitude tracking robust controller is proposed for WMR. Firstly, a kinematic controller is designed to generate the virtual velocity based on the kinematic error model. Secondly, the sliding mode control with a modified reaching law is adopted to ensure the actual velocity can converge to the virtual velocity in finite time based on the dynamic model. Finally, the stability of the controller is verified through the Lyapunov function. Numerical simulation shows the robustness and effectiveness of the proposed dual-loop tracking controller.

        A Field Theory Based Model for Identifying the Effect of Organizational Structure on the Formation of Organizational Culture in Construction Projects

        A carefully selected organizational structure paves the way for the success of a construction project by fostering a strong and enduring organizational culture. However, very little research has been conducted regarding the effect of organizational structure on the formation of organizational culture. This research employs the field theory to demonstrate organizational culture development and thereafter proposes a novel model for identifying the effect of organizational structure on the formation of organizational culture in a construction project. An empirical analysis based on the Chinese construction industry is performed to verify the proposed model. The primary contribution of this research is twofold. First, it presents a new perspective of examining organizational culture development from organizational structure by using the cultural field theory. Second, it reveals the important effect of organizational structure on the formation of organizational culture and offers project owners a new approach for identifying the effect which could also be a reference for further selecting an appropriate organizational structure.

        Numerical prediction analysis of propeller bearing force for full-scale hullepropellererudder system

        The hybrid grid was adopted and numerical prediction analysis of propeller unsteady bearing force considering free surface was performed for mode and full-scale KCS hullepropellererudder system by employing RANS method and VOF model. In order to obtain the propeller velocity under self-propulsion point, firstly, the numerical simulation for self-propulsion test of full-scale ship is carried out. The results show that the scale effect of velocity at self-propulsion point and wake fraction is obvious. Then, the transient two-phase flow calculations are performed for model and full-scale KCS hullepropellererudder systems. According to the monitoring data, it is found that the propeller unsteady bearing force is fluctuating periodically over time and full-scale propeller's time-average value is smaller than model-scale's. The frequency spectrum curves are also provided after fast Fourier transform. By analyzing the frequency spectrum data, it is easy to summarize that each component of the propeller bearing force have the same fluctuation frequency and the peak in BFP is maximum. What's more, each component of full-scale bearing force's fluctuation value is bigger than model-scale's except the bending moment coefficient about the Y-axis.
