Vac8p is a vacuolar protein that plays pivotal roles in both vacuole inheritance and the formation of nucleus vacuole junction (NVJ) in yeast. The Vac8p directly interacts with Atg13p, a component of the autophagy machinery, and mediates cytoplasm-to-vacuole targeting (Cvt) pathway, resulting in the maturation of aminopeptidase I (Ape1p). Here, we coexpressed and purified Saccharomyces cerevisiae Vac8p complexed with Atg13p in Escherichia coli bacteria cells, and crystallized the complex proteins under the condition of 25% (v/v) PEG 400, 100 mM Tris pH 8.5, 2% (v/v) Ethylene glycol, 2% (w/v) PEG 3350, 1.5% (w/v) PEG 20000, 5 mM DTT at 293K. X-ray diffraction data of the crystals were collected to 2.9 Å resolution at the synchrotron radiation. The crystals belong to the orthorhombic space group P2 1 2 1 2 1 with unit cell parameters a = 62.7 Å, b = 92.4 Å, and c = 139.9 Å. The asymmetric unit contains one Vac8p-Atg13p heterodimer with a corresponding V M of 2.92 Å 3 Da -1 and solvent content of 57.8%.

Ethical and safety issues have rendered mesenchymal stem cells (MSCs) popular candidates in regenerative medicine, but their therapeutic capacity is lower than that of induced pluripotent stem cells (iPSCs). This study compared original, dental tissue-derived MSCs with re-differentiated MSCs from iPSCs (iPS-MSCs). CD marker expression in iPS-MSCs was similar to original MSCs. iPS-MSCs expressed higher in pluripotent genes, but lower levels in mesodermal genes than MSCs. In addition, iPS-MSCs did not form teratomas. All iPSCs carried mtDNA mutations; some shared with original MSCs and others not previously detected therein. Shared mutations were synonymous, while novel mutations were non-synonymous or located on RNA-encoding genes. iPS-MSCs also harbored mtDNA mutations transmitted from iPSCs. Selected iPS-MSCs displayed lower mitochondrial respiration than original MSCs. In conclusion, screening for mtDNA mutations in iPSC lines for iPS-MSCs can identify mutation-free cell lines for therapeutic applications. [BMB Reports 2019; 52(12): 689-694]

Lonicera Caerulea(Honey berry) has been used in medical treatment in Russia, Japan, China and Korea. It has high level of vitamin C and polyphenolics. Polyphenolics can improve anti-inflammatory effect and prevent cancer, diabetes mellitus type 2. Also, Vitamin C is a representative anti- oxidant. however, It is still unknown what effect it will have on the oxidation stress of the reproductive system. In previous studies, ROS can be produced when it is exposed to heat stress and has negative effect on sperm's maturation, capacitation, hyperactivation, acrosome reaction and fusion of egg and sperm. Therefore, The purpose of this study is to investigate the antioxidant effects of L.caerulea on the sperm and egg cells of mice. At first, it conducted using ICR mouse(n=20) during 4 weeks. There are four groups of mouse(n=5 per group). Also, L.caerulea was taken by oral gavage. Group Ⅰ(control) kept at 23℃~27℃ and administer D.W(0.5ml/day), Likewise, Group Ⅱ(HB) kept at room temperature but gave HB(0.5ml/day), Group Ⅲ (HB+HS) received heat stress (40℃) using hyperthermia induction chamber and gave HB at same dose. and Group Ⅳ(HS) exposed heat stress only. Mainly, we showed degree of gene expression using Western blot in SOD, HSP 70, 17β-HSD and Real time PCR. It can find correlation between intracellular activity like steroid hormone, apoptosis under ROS and antioxidant activity of L.caerulea.

The effect of deprivation on depression across different age groups in Korea

This study aimed to verify whether deprivation is experienced differently across different age groups and whether these differences change the effect of deprivation on depression. In this study, the CES-D 11 scale was used to measure depression. Additionally, to verify the relationship between deprivation and depression by age group, logistics analysis was conducted. This study analyzed the survey data of 4,000 individuals from Korea; respondents were classified into four age groups: ≤34, 35–49, 50–64, and ≥65 years old. The results showed that there was a significant relationship between deprivation and depression and that specific areas of deprivation affected depression differently for each age group. The deprivation ratios of older individuals were higher in all areas. Further, the deprivation ratio of the ≤34 group in housing and living conditions was higher than that of the other groups. Additionally, respondents in the 50–64 and the ≥65 groups were alienated from the social security network, which poses an increased risk for chronic deprivation or poverty. The results of this study suggest that policies and practical interventions are needed, taking into account that the effects of absolute and relative deprivation on depression vary by age group.

The objective of this study is to analyze the elements of presentation fluency perceived by learners, targeting foreign university students. For this, targeting foreign learners taking the course of academic speech at two universities in Seoul, the evaluation of fellow students presentation during class hours were analyzed. The evaluation form was itemized into contents/constructive element, linguistic element, paralinguistic element, and non-verbal element, including five questions for contents/constructive element, three questions for linguistic element, four questions for paralinguistic element, and five questions for non-verbal element. This study examined differences in the mean value and total value of impression evaluation of each element of presentation fluency of learners in each group, and also examined the elements of fluent presentation in the position of learners through the analysis of each item. As a result of the study, the learners judged the fluency element of presentation in order of linguistic element, content / constructive element, paralinguistic element and non-verbal element. Especially non-verbal elements showed different patterns compared to other elements and it was necessary to strengthen education with its importance. 본고는 대학에서 수학하는 외국인 학부생들을 대상으로 하여 학습자들이 생각하는 발표하기 유창성에 관여하는 요소가 무엇인지 분석해 보는 데 목적이 있다. 이를 위해 서울 소재 2개 대학에서 학업 말하기를 수강하는 외국인 학습자들을 대상으로 수업시간에 학우들의 발표를 평가한 평가지를 분석하였다. 평가지는 내용·구성적 요소, 언어적 요소, 준언어적 요소, 비언어적 요소로 항목화하여 작성하였고, 내용·구성적 요소 5문항, 언어적 요소 3문항, 준언어적 요소 4문항, 비언어적 요소 5문항으로 평가를 측정할 수 있도록 하였다. 이를 통해 각 집단에서 학습자들의 발표하기 유창성에 대한 각각의 요소들의 평균값과 전체 인상평가의 값이 차이가 있는지 살폈고, 각각의 항목 분석을 통해 학습자들의 입장에서 유창한 발표의 요소가 무엇인지를 살펴보았다. 연구 결과 학습자들은 언어적 요소를 가장 중요한 요소로 그 다음으로 내용/구성적 요소, 준언어적 요소, 비언어적 요소의 순서로 판단하였다. 특히 비언어적요소는 다른 요소들에 비해 다른 패턴을 보여 그 중요성과 더불어 교육을 강화할 필요가 있었다.
