CC Chemokine CKβ8의 발현 조절 물질 탐색 및 저해활성 물질 분석

CKβ8은 CC 케모카인의 부류에 속하는 사이토카인의 일종으로, 단핵구, 호염구, 호산구, T 림프구, 자연살해세포, 수지상세포 등의 화학주성물질로 작용하는 것으로 알려져 있다. CKβ8은 염증반응 및 혈관신생 등과 같은 여러 가지 생체 반응에 역할을 하는 것으로 알려져 있다. 본 논문에서는 사람 CKβ8 유전자의 전사저해 물질을 선별하기 위하여 CKβ8/Luc 유전자를 발현하는 세포주를 확립하였다. CKβ8 유전자의 전사시작점에서 -299 부터 +10 프로모터 부위를 PCR에 의해 얻은 다음 포유세포 발현 플라스미드에 클로닝하였다. CKβ8/Lvc 유전자는 U937 세포내에서 PMA에 의해 10배 이상 발현이 증가한다. CKβ8/Luc 발현 U937 세포주는 식물 추출물에서 전사 저해제를 탐색하는데 이용되었다. 46 개의 식물 추출물 중에서 6개가 90% 이상 CKβ8/Luc의 전사를 저해하였다. 이들 물질에 대해 세포 독성 실험을 수행한 결과 그 중 한 개만이 세포 독성이 거의 없는 것으로 나타났다. 이 추출물은 붉나무에서 얻은 추출물이며 이 식물 추출물은 염증관련 질환에 대한 가능성이 있는 치료제로 이용될 수도 있을 것으로 추측된다. Ckβ8 belongs to a family of CC chemokines, which is known to act as a chemoattractant for monocytes, basophils, eosinophils, T lymphocytes, NK cells, and dendritic cells. CKβ8 is known to play roles in various biological processes such as inflammation and angiogenesis. In the present study, we developed U937 cell line expressing CKβ8/Luciferase fusion gene for screening of transcriptional inhibitors for human CKβ8 gene. Genomic sequences that contain CKβ8 promoter region from 299 to+10 nucleotide from the transcription start site was amplified by PCR and cloned into mammalian episomal plasmid. The CKβ8/Luc fusion plasmid was introduced into human promonocytic U937 cells and selected for stable U937 cells expressing the fused reporter gene. The expression of the fusion gene was induced by more than 10 folds upon treatment of phorbol myristate acetate. The stable U937 transfectant cells were used for searching transcriptional inhibitors for human CKβ8 gene from plant extracts. Among 46 plant extracts tested, six extracts exerted more than 90% inhibition of PMA-induced CKβ8 transcription. Cytotoxic tests demonstrated that one of extracts, which was extracted from Rhus chinensis Mill, had little cytotoxic effect. The extract may be used for developing a potential therapeutic agent for inflammatory diseases.
Experience with TVT-O for the Surgical Treatment of Female Stress Urinary Incontinence

Objective: We evaluated the clinical efficacy and safety of inside-out transobturator vaginal tape (TVT-O) for the surgical treatment of female stress urinary incontinence. Methods: We analyzed the medical records of 239 women with stress urinary incontinence and who were treated with TVT-O between March 2004 and July 2007. We retrospectively investigated the patient characteristics, the operative time, the number of hospital days, the efficacy, and the complication rate by conducting a 1 year follow-up survey. Results: One hundred eighty-three patients were included in the study. The mean age was 55.7 years (range, 31~74 years) and the mean operative time was 20.1 minutes (range, 12~40 minutes). Of the 76 patients who underwent TVT-O only, 94 patients were discharged the same day of surgery and the mean hospital stay was 0.4 days (range, 0~4.0 days). Of the 183 patients who were followed for 1 year, 162 (88.7%) were assessed as completely dry, 15 (8.2%) showed improvement, and 4 (2.2%) were deemed to have failed. There were no intra-operative bladder, urethral, or bowel injuries; 32 cases (17.5%) showed complications of surgery during the 1 year follow-up period. Thirteen (7.1%) cases were shown to have de novo urgency, 5 (2.7%) cases had urinary retention, 6 (3.3%) cases had vaginal erosions, and 8 (4.4%) cases had wound hematomas and lower extremity discomfort. Conclusion: Our experience showed that TVT-O is an effective and safe procedure for the surgical treatment of female stress urinary incontinence.
쇠뿔현호색 (Corydalis cornupetala Y. H. Kim et J. H. Jeong) 자생지의 생육환경과 식생구조

쇠뿔현호색은 현호색과 현호색속 현호색절의 다년생 초본으로 우리나라 경산시 계정숲에만 제한적으로 분포하는 특산식물종이며, 춘계단명식물로 제한된 생육기간과 자원으로 생활전략을 가지고 있다. 본 연구는 쇠뿔현호색 (Corydalis cornupetala Y.H.Kim et J.H.Jeong) 개체군을 대상으로 자생지의 입지 특성과 식생구조를 파악하여 쇠뿔현 호색 개체군 및 생태연구에 대한 기틀을 마련하고 자생지 보전을 위한 복원 계획 수립의 기초 자료를 제공하고자 한다. 토성은 미사질식양토이며, 토양산도는 pH 5.4~5.9로 약산성, 전기전도도는 0.22~0.50ds/m, 토양유기물함량은 7.61~15.78%로 나타났다. 토양분석결과 양이온치환용량이 우리나라 산림토양의 평균치보다 높게 나타났는데, 이는 방형구에서 유독 칼슘과 마그네슘의 함량이 높게 나타난 결과라 사료된다. 식생분석결과 5개 방형구 내에서 조사된 관속식물은 총 59분류군이었다. 군락분류의 경우 PC-ORD를 이용한 이단계 군락분석 결과 신나무, 갈퀴나물 및 쇠별꽃 군락으로 분류되었다. 쇠뿔현호색의 자생지는 출현종의 수보다는 우점하는 종의 상이성에 따라 집단의 유연관계가 결정되었다. 종다양도는 평균 1.26로 산출되었으며, 우점도와 균등도는 각각 0.08와 0.92이었다. The species of Corydalis cornupetala Y.H.Kim et J.H.Jeong Which is belong to the Corydaliaceae is only distributed at the Gyaejeong Forest in Gyeongsan. As one of the spring ephemeral plants, this species has limited growing period. The present study is on site characteristics and vegetation structure in the wild habitats of Corydalis cornupetala Y.H.Kim et J.H.Jeong and offers basic information on habitat conservation including recovery plan. The soil texture is silty loam and soil acidity is ranged as pH 5.4~5.9. The electric conductivity was 0.22-0.50 ds/m and soil organic content is ranged as 7.61~15.78 %. Fifty nine vascular plants were identified from 5 quadrats. According to the two way cluster analysis applied by the PC-ORD, the community was classified as Acer ginnala, Vicia amoena and Stellaria aquatica. The habitat of Corydalis cornupetala Y.H.Kim et J.H.Jeong was determined by the dissimilarity indices rather than the species number of appearance. Species diversity was 1.26, and dominance and evenness were found to be 0.08 and 0.92, respectively.

$Ag(Ta,Nb)O_3$ materials have a perovskite structure with a low loss tangent. These materials have been widely researched for their applications as high-frequency, passive components. Also, $Ag(Ta,Nb)O_3$ materials have weak frequency dispersion with high dielectric permittivity which gives them enormous potential for use in electronic components, including the filters, and embedded capacitors. Therefore, our research will discuss the structural and electrical relaxation properties of $Ag(Ta_{0.8}Nb_{0.2})O_3$ ceramics for device applications. We will investigate using X-ray diffraction to understand their structural properties and will analyze voltage dependent leakage current and timedependent relaxation behavior to understand their material properties.
