        Transcriptional profiling of mouse cavernous pericytes under high-glucose conditions: Implications for diabetic angiopathy

        Purpose: Penile erection requires integrative interactions between vascular endothelial cells, pericytes, smooth muscle cells, and autonomic nerves. Furthermore, the importance of the role played by pericytes in the pathogenesis of angiopathy has only recently been appreciated. However, global gene expression in pericytes in diabetes mellitus-induced erectile dysfunction (DMED) remains unclear. We aimed to identify potential target genes related to DMED in mouse cavernous pericytes (MCPs). Materials and Methods: Mouse cavernous tissue was allowed to settle under gravity in collagen I-coated dishes, and sprouted cells were subcultivated for experiments. To imitate diabetic conditions, MCPs were treated with normal-glucose (NG, 5 mM) or high-glucose (HG, 30 mM) media for 3 days. Microarray technology was used to evaluate gene expression profiles, and RT-PCR was used to validate sequencing data. Histological examinations and Western blot were used to validate final selected target genes related to DMED. Results: Decreased tube formation and increased apoptosis were detected in MCPs exposed to the HG condition. As shown by microarray analysis, the gene expression profiles of MCPs exposed to the NG or HG condition differed. A total of 2,523 genes with significantly altered expression were classified into 15 major gene categories. After further screening based on gene expression and RT-PCR and histologic results, we found that Hebp1 gene expression was significantly diminished under the HG condition and in DM mice. Conclusions: This gene profiling study provides new potential targets responsible for diabetes in MCPs. Validation studies suggest that Hebp1 may be a suitable biomarker for DMED.

        Reduced Graphene Oxide-Based Assay for Real-Time Monitoring of Cancer Cell Viability

        Viability of cancer cell is an important indicator of physiological state and function of cells, which can be effected by the change of pH in the medium solution, due to the increase of carbon oxide and lactic acid caused by respiration. Although many methods have been developed to detect the viability of cells, mostly based on cytochemical staining and polymerase chain reaction (PCR) technology are time consuming. In this paper, an electronic device was made by thermal reduced graphene oxide (RGO) for detection of cancer cell viability in real-time. This electronic device could be used to monitor the metabolic activity and viability of cancer cells based on the change in pH value. As the pH decreases, colon cancer cells loose viability and the current decreases. This RGO device is simple, sensitive and label-free and could serve as a platform for detection of cells and drug testing.

        Equivalent Magnetic Circuit Model for Consequent Pole Hybrid Excitation Synchronous Machine with AC Field Control

        At present, almost all hybrid excitation machines (HEMs) apply DC field current to control the air-gap flux. However, the DC field control mode results in a problem that the capability of field-weakening is not equal to that of field-strengthening. This paper briefly explains the mechanism of asymmetric bidirectional field control capability in present HEMs, proposes a consequent pole hybrid excitation synchronous (CPHES) machine with AC field control mode to solve the asymmetric problem. The structure and principles of CPHES machine are presented. Additionally, the equivalent magnetic circuit model of CPHES machine is derived to reduce the computational complexity of analyzing CPHES structure by using three-dimensional finite element analysis (3DFEA). The proposed equivalent model and 3D-FEA are compared, the results verify the proposed model is basically consistent with the 3D-FEA, and the proposed model is able to reduce the computational complexity greatly.

        Electrospun SrO-Incorporated Nanofiber Membranes for Biomedical Applications

        Bioactive ceramics have gained increasing interest in biomedical applications. Strontium (Sr)-based ceramics show unique bioactivity in stimulating cell proliferation and migration and promoting angiogenesis. Incorporation of bioactive ceramics into nanofibers represents a facile approach to alter the material properties of electrospun nanofiber membranes. Herein, we prepared SrO-incorporated nanofiber membranes by electrospinning and investigated the effects of SrO particles on the physicochemical properties and cytocompatibility of these membranes. Our results indicate that simple one-stream electrospinning could smoothly introduce SrO particles into poly(L-lactic acid) (PLA)/gelatin nanofibers. SrO-incorporated membranes showed significantly improved wettability but became brittle compared with PLA/gelatin nanofiber membranes. Cells differentially responded to SrO-incorporated membranes: endothelial cells showed greater cell area and coverage on SrO-incorporated membranes; in contrast, fibroblasts were inhibited on these membranes. These findings suggest that SrO-incorporated membranes might find opportunities in specific biomedical applications in which pro-angiogenesis and antiadhesion are simultaneously demanded.

        Efficacy and Adverse Events Associated With Use of OnabotulinumtoxinA for Treatment of Neurogenic Detrusor Overactivity: A Meta-Analysis

        Purpose: OnabotulinumtoxinA is used widely for the treatment of neurogenic detrusor overactivity. We conducted a systematic review and meta-analysis to assess its efficacy and safety for neurogenic detrusor overactivity treatment. Methods: A systematic literature review was performed to identify all published randomized double-blind, placebo-controlled trials of onabotulinumtoxinA for neurogenic detrusor overactivity treatment. MEDLINE, Embase, and the CENTRAL were employed. Reference lists of retrieved studies were reviewed carefully. Results: Six publications involving 871 patients, which compared onabotulinumtoxinA with a placebo were analyzed. Efficacy of onabotulinumtoxinA treatment was shown as a reduction of the mean number of urinary incontinence episodes per day (mean difference, -1.41; 95% confidence interval [CI], -1.70 to -1.12; P<0.00001), maximum cystometric capacity (135.48; 95% CI, 118.22–152.75; P<0.00001), and maximum detrusor pressure (-32.98; 95% CI, -37.33 to -28.62; P<0.00001). Assessment of adverse events revealed that complications due to onabotulinumtoxinA injection were localized primarily to the urinary tract. Conclusions: This meta-analysis suggests that onabotulinumtoxinA is an effective treatment for neurogenic detrusor overactivity with localized advent events

        One-Step Hydrothermal Synthesis of Bi2S3-TiO2-RGO Composites with Enhanced Visible Light Photocatalytic Activities

        The Bi2S3-TiO2-RGO composites were synthesized by a facile one-step hydrothermal method and applied for the photocatalytic degradation of Rhodamine B (Rh B) under the visible light. The Bi2S3-TiO2-RGO composites were characterized by transmission electron microscopy, X-ray diffraction, Raman and Fourier transform infrared spectrometer. The results indicated that the Bi2S3-TiO2-RGO composites were successfully prepared, and Ti-O-C and S-C bonds were existing among Bi2S3, TiO2 as well as RGO. Furthermore, the photocatalytic ability of Bi2S3-TiO2-RGO composites was excellent under visible light due to its responding to the whole visible light region, low recombination rate of photogenerated electron–hole pairs and relatively negative conduction band. Rh B photocatalytic degradation rate was 99.5% after 50 min and still could reach 98.4% after five cycles. Finally, a formation mechanism as well as a photocatalytic mechanism of Bi2S3-TiO2-RGO composites were proposed based on the experimental results.

        Role of Transcutaneous Electrical Nerve Stimulation in Treating Children With Overactive Bladder From Pooled Analysis of 8 Randomized Controlled Trials

        Purpose: Transcutaneous electrical neural stimulation (TENS), as a non-invasive modality, has been clinically used as an alternative treatment for children with overactive bladder (OAB). We conducted a pooled analysis to explore the effect of TENS on OAB. Methods: The Preferred Reporting Items for Systematic Reviews and Meta-analysis guideline was followed in this study. The MEDLINE, Embase, and Cochrane Central Register of Controlled Trials databases, as well as the reference lists of the retrieved studies, were used to find trials relevant for assessing the use of TENS to treat OAB. Results: Of the 246 records identified, 8 publications were analyzed in our study. Our analysis found that TENS resulted in a greater decrease of wet days/wk, daily voiding frequency, daily incontinence episodes, and daily number of voids than was observed in the control group. Furthermore, TENS-treated patients showed similar visual analogue scale (VAS) scores to patients in the control group, demonstrating that the application of TENS did not increase patients’ discomfort and pain. TENS had a relative advantage in the number of partial responses, but no clear differences were found in frequency of no response or a full response compared to the control group. In urodynamic testing, TENS led to obvious improvements in average voided volume and maximum voided volume in children with OAB. Conclusions: TENS had a remarkable effect on the improvement of urodynamic indexes and objective OAB symptoms without a significant increase in VAS scores for children with OAB.

        Bioinformatic and integrated analysis identifies an lncRNA–miRNA–mRNA interaction mechanism in gastric adenocarcinoma

        Background lncRNAs–miRNAs–mRNAs networks play an important role in Gastric adenocarcinoma (GA). Identifcation of these networks provide new insight into the role of these RNAs in gastric cancer. Objectives Biological information databases were screened to characterize and examine the regulatory networks and to further investigate the potential prognostic relationship this regulation has in GA. Methods By mining The Cancer Genome Atlas (TCGA) database, we gathered information on GA-related lncRNAs, miRNAs, and mRNAs. We identifed diferentially expressed (DE) lncRNAs, miRNAs, and mRNAs using R software. The lncRNA–miRNA–mRNA interaction network was constructed and subsequent survival examination was performed. Representative genes were selected out using The Biological Networks Gene Ontology plug-in tool on Cytoscape. Additional analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) terms were used to screen representative genes for functional enrichment. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) were used to identify the expression of fve candidate diferential expressed RNAs. Results Information of samples from 375 cases of gastric cancer and 32 healthy cases (normal tissues) were downloaded from the TCGA database. A total of 1632 DE-mRNAs, 1008 DE-lncRNAs and 104 DE-miRNAs were identifed and screened. Among them, 65 DE-lncRNAs, 10 DE-miRNAs, and 10 DE-mRNAs form lncRNAs–miRNAs–mRNAs regulatory network. Additionally, 10 lncRNAs and 2 mRNAs were associated with the prognosis of GA. Multivariable COX analysis revealed that AC018781.1 and VCAN-AS1 were independent risk factors for GA. GO functional enrichment analysis found DE-mRNA was signifcantly enriched TERM (P<0.05). The KEGG signal regulatory network analysis found 11 signifcantly enrichment networks, the most prevailing was for the AGE-RAGE signaling pathway associated with Diabetic complications. Results of RT-qPCR was consistent with the in silico results. Conclusions The results of the present study represent a view of GA from a analysis of lncRNA, miRNA and mRNA. The network of lncRNA–miRNA–mRNA interactions revealed here may potentially further experimental studies and may help biomarker development for GA.
