Curcumin Analogue A501 induces G2/M Arrest and Apoptosis in Non-small Cell Lung Cancer Cells

Curcumin and its analogues have been reported to exert anti-cancer activity against a variety of tumors. Here, we reported A501, a new curcumin analogue. The effect of A501 on cell viability was detected by MTT assay, the result showed that A501 had a better inhibiting effect on the four non-small cell lung cancer (NSCLC) cells than that of curcumin. Moreover, Colony forming experiment showed A501 significant restrained cell proliferation. Flow cytometry displayed A501 can cause G2/M arrest and induce apoptosis. Western blotting showed that A501 decreased the expression of cyclinB1, cdc-2, bcl-2, while increased the expression of p53, cleaved caspase-3 and bax. In conclusion, curcumin analogues A501 played antitumor activity by inhibiting cell proliferation and inducing apoptosis of NSCLC cells. And it was likely to be a promising starting point for the development of curcumin-based anticancer drugs.

Chemical investigation of the fruits of Alpinia oxyphylla led to an isolation of the two new natural products, 9-hydroxy epinootkatol (1) and (S)-2-pentanol-2-O-β-D-glucopyranoside (2), in addition to the nine known compounds, pinocembrin (3), tectochrysin (4), izalpinin (5), nookatone (6), yakuchinone A (7), protocatechuic acid (8), β-sitosterol (9), daucosterol (10) and β-sitosterol palmitate (11). Their structures were elucidated on the basis of physicochemical constants and NMR spectral data analysis. The effects of the isolated components on nitric oxide production in LPS-induced RAW 264.7 macrophages were examined. The two new natural compounds showed inhibitory activities with IC50 values of 21.8 and 32.8 μg/mL, respectively.

<P>Here we report the synthesis of Pd@Cu core–shell nanocubes <I>via</I> epitaxial growth, where the lattice mismatch is 7.1%. The synthesis involved the use of Pd seeds with different shapes (including cubes, cuboctahedra, and octahedra) for the epitaxial growth of Cu shells. Different from the conventional growth mode, Cu atoms initially nucleated only on a few of the many faces of a Pd seed, onto which more Cu atoms were continuously added to generate Cu blocks. Later, the Cu atoms also started to nucleate and grow on other faces of the Pd seed until the entire surface of the seed was covered by a Cu shell. As a result, the Pd seed was rarely located in the center of each core–shell structure. The final product took a cubic shape enclosed by {100} facets regardless of the type of Pd seeds used because of the selective capping of Cu(100) surface by hexadecylamine. Purpose The purpose of this study was to investigate the prognostic significance of total metabolic tumor volume (TMTV) and total lesion glycolysis (TLG) in patients with follicular lymphoma (FL) at baseline and mid-treatment with 18F-fluorodeoxyglucose positron emission tomography computed tomography (PET-CT) scans. Materials and Methods The study analyzed data from 48 patients with FL who were treated in Jiangsu Province Hospital and reviewed their baseline PET-CT scans. TMTV and TLG were computed by using the absolute value of 2.0, 2.5, and 3.0 thresholding method, respectively. Results Median age was 53 years, 75.0% of patients had stage III to IV disease, 43.8% had a Follicular Lymphoma International Prognostic Index 1 (FLIPI1) score of 3 to 5 and 20.8% had a FLIPI2 score of 3 to 5. Receiver operating characteristic (ROC) curve analysis showed the optimal cut-off values for TMTV3.0 and TLG3.0 were 476.4 (sensitivity, 85.7%; specificity, 78.0%; area under the curve [AUC], 0.760; p=0.003) and 2,676.9 (sensitivity, 71.4%; specificity, 78.0%; AUC, 0.760; p=0.003). On multivariable analysis, TMTV3.0 and TLG3.0 were independent predictors of both progression-free survival (PFS) (hazard ratio [HR], 5.406; 95% confidence interval [CI], 1.326 to 22.040; p=0.019 and HR, 6.502; 95% CI, 1.079 to 39.182; p=0.042) and overall survival (OS) (HR, 4.111; 95% CI, 1.125 to 15.027; p=0.033 and HR, 5.885; 95% CI, 1.014 to 34.148; p=0.049). ROC curve analysis showed the optimal cut-off values for TMTV3.0 and TLG3.0 were 66.3% (sensitivity, 85.7%; specificity, 63.4%; AUC, 0.774; p < 0.001) and 64.5% (sensitivity, 85.7%; specificity, 65.9%; AUC, 0.777; p < 0.001). Conclusion Baseline TMTV and TLG are strong predictors of PFS and OS in FL. Furthermore, interim TMTV (TMTV > 66.3%) and TLG (TLG > 64.5%) reduction are valuable tools for early treatment response assessment in FL patients.
Deformation Feature of Sheet Metals During Inclined Hole-Flanging by Two-Point Incremental Forming

Incremental hole-flanging is an applicable extension of incremental sheet forming (ISF), but the knowledge on the deformation of metallic sheets during the course is lack, especially when the axes of the flanged necks are not perpendicular to the sheet plane. The current study used Al 6061 and SUS 304 stainless steel sheets with the thickness of 1 mm and 0.3 mm to examine the inclined hole-flanging processes on the flat plates by two-point ISF processes. It was found that, although as a whole the thickness variation is thinning, the decreasing degrees within upper and lower domain are not equal, and the difference increases with the inclination angle. Crack, buckling and local deformity are typical defects found in the experiments. The maximum springback increases with tool diameter, inclination angle and feeding pitch. Due to the material properties and a thinner thickness, local deformity is easier to take place on SUS 304 sheets during forming. Utilizing the tool feeding path along the oblique plane can obtain even force and consequently, more uniform deformation. Moreover, the equations for preliminarily determining the dimension of precut holes were derived according to the radial developed length of the flanged neck.

Pebax® MH 1657 (Pebax)-based blend membranes with different polyether-amine (PEA) loadings were designed and fabricated for efficient CO2 separation. The CO2 separation performance of Pebax/PEA blend membranes was greatly improved in comparison with that of pure membranes. This was mainly because the introduced PEA tailored the physical and chemical microenvironments in blend membranes. Specifically, PEA was a liquid-like additive, which was beneficial to reduce the mass transfer resistance of gases and increase CO2 permeability. Meanwhile, PEA contained amino groups that acted as mobile carriers to tailor the chemical microenvironment in blend membranes. The mobile carriers preferentially reacted reversibly with CO2 molecules, facilitating CO2 transport in membranes. Compared with CO2/CH4 separation performance of pure Pebax membrane, CO2 permeability and CO2/CH4 separation factor of Pebax/PEA-3 increased by 144.8% and 29.4%, respectively. This study suggests that PEA is a promising membrane material for tailoring the physical and chemical microenvironments in blend membranes for efficient CO2 separation.

This paper presented solutions of displacement and stress for a circular opening which is reinforced with grouted rock bolt. It satisfies the Mohr-Coulomb (M-C) or generalized Hoek-Brown (H-B) failure criterion, and exhibits elastic-brittle-plastic or strain-softening behavior. The numerical stepwise produce for strain-softening rock mass reinforced with grouted rock bolt was developed with non-associative flow rules and two segments piecewise linear functions related to a principle strain-dependent plastic parameter, to model the transition from peak to residual strength. Three models of the interaction mechanism between grouted rock bolt and surrounding rock proposed by Fahimifar and Soroush (2005) were adopted. Based on the axial symmetrical plane strain assumption, the theoretical solution of the displacement and stress were proposed for a circular tunnel excavated in elastic-brittle-plastic and strain-softening rock mass compatible with M-C or generalized H-B failure criterion, which is reinforced with grouted rock bolt. It showed that Fahimifar and Soroush's (2005) solution is a special case of the proposed solution for n = 0.5. Further, the proposed method is validated through example comparison calculated by MATLAB programming. Meanwhile, some particular examples for M-C or generalized H-B failure criterion have been conducted, and parametric studies were carried out to highlight the influence of different parameters (e.g., the very good, average and very poor rock mass). The results showed that, stress field in plastic region of surrounding rock with considering the supporting effectiveness of the grouted rock bolt is more than that without considering the effectiveness of the grouted rock bolt, and the convergence and plastic radius are reduced.
