<P>To increase expression of lycopene synthetic genes crtE, crtB, crtI, and ipiHP1, the four exogenous genes were cloned into a high copy pTrc99A vector with a strong trc promoter. Recombinant Escherichia coli harboring pT-LYCm4 produced 17 mg/L of lycopene. The mevalonate lower pathway, composed of mvaK1, mvaK2, mvaD, and idi, was engineered to produce pSSN12Didi for an efficient supply of the lycopene building blocks, isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP). Mevalonate was supplied as a substrate for the mevalonate lower pathway. Lycopene production in E. coli harboring pT-LYCm4 and pSSN12Didi with supplementation of 3.3 mM mevalonate was more than threefold greater than bacteria with pT-LYCm4 only. Lycopene production was dependent on mevalonate concentration supplied in the culture. Clump formation was observed as cells accumulated more lycopene. Further clumping was prevented by adding the surfactant Tween 80 0.5% (w/v), which also increased lycopene production and cell growth. When recombinant E. coli harboring pT-LYCm4 and pSSN12Didi was cultivated in 2YT medium containing 2% (w/v) glycerol as a carbon source, 6.6 mM mevalonate for the mevalonate lower pathway, and 0.5% (w/v) Tween 80 to prevent clump formation, lycopene production was 102 mg/L and 22 mg/g dry cell weight, and cell growth had an OD<SUB>600</SUB> value of 15 for 72 h. © 2006 Wiley Periodicals, Inc.</P>

Background/Aims: Intraductal papillary mucinous neoplasm (IPMN) of the pancreas has a favorable prognosis, but seems to be associated with a high incidence of extrapancreatic tumors. The purpose of this study was to evaluate the incidence and clinicopathological features of extrapancreatic tumors associated with IPMN. Methods: Thirty-seven patients with IPMN of the pancreas, confirmed by surgical resection and typical findings of endoscopic ultrasonography and CT imaging between October 1, 1998 and August 31, 2006 were included. Seventeen patients were diagnosed with surgical resection and biopsy, and others by typical imaging findings of IPMN. These patients were examined for the development of extrapancreatic tumors. Results: Of 37 patients with IPMN, 14 (38%) had 18 extrapancreatic tumors, and 10 (27%) had 13 extrapancreatic malignancies. Five, six, and two extrapancreatic malignancies had diagnosed before during, and after the diagnosis of IPMN. Gastric adenocarcinoma (3 patients, 23%) and colorectal carcinoma (3 patients, 23%) were the most common neoplasms. Other extrapancreatic tumors included lung cancer (n=2), prostatic cancer (n=1), renal cell carcinoma (n=1), cholangiocelluar carcinoma (n=1), urinary bladder cancer (n=1), and gallbladder cancer (n=1), respectively. As benign tumor, there were two gallbladder adenoma, one gastric adenoma, one colonic adenoma and one benign ovarian cystic neoplasm, respectively. Conclusions: IPMN is associated with high incidence of extrapancreatic tumors, particularly gastric and colorectal neoplasms. Upper gastrointestinal endoscopy and colonoscopy should be done, and systemic surveillance for the possible occurrence of other tumors may allow early detection of extrapancreatic tumor in patients with IPMN. In this paper, a loosely coupled integration system is proposed for improving the positioning performance of a mobile robot by fusing the Pseudolite Ultrasonic System (PUS), which is an absolute position measurement system using direct ultrasonic wave, with the odometer of Dead-Reckoning (DR). The PUS and the DR calculate position independently and a Kalman filter estimates the position using position information from the PUS and the DR. To estimate the position of the mobile robot accurately in case the LOS (line-of-sight) between ultrasonic transmitter and receiver is blocked due to obstacles, a positioning failure detection algorithm is proposed. The positioning performances of the proposed loosely coupled integrated system are verified and evaluated by experiments. The experimental results show that the position of mobile robot is estimated accurately by the proposed algorithm.

<P>In this paper, we report a simple and effective method to simultaneously achieve a high charge-carrier mobility and low off current in conjugated polymer-wrapped semiconducting single-walled carbon nanotube (s-SWNT) transistors by applying a SWNT bilayer. To achieve the high mobility and low off current, highly purified and less purified s-SWNTs are successively coated to form the semiconducting layer consisting of poly (3-dodecylthiophene-2,5-diyl) (P3DDT)-wrapped high-pressure carbon mono oxide (HiPCO) SWNT (P3DDT-HiPCO) and poly (9, 9-di-n-dodecylfluorene) (PFDD)-wrapped plasma discharge (PD) SWNT (PFDD-PD). The SWNT transistors with bilayer SWNT networked film showed highly improved hole field-effect mobility (6.18?±?0.85 cm<SUP>2</SUP>V<SUP>−1</SUP>s<SUP>−1</SUP> average), on/off current ratio (10<SUP>7</SUP>), and off current (∼1 pA). Thus, the combination of less purified PFDD-PD (98%–99%) charge-injection layer and highly purified s-P3DDT-HiPCO (>99%) charge-transport layer as the bi-layered semiconducting film achieved high mobility and low off current simultaneously.</P>
Arbekacin의 methicillin 내성 Staphylococcus aureus에 대한 항균 효과

목적 : Arbekacin은 일본에서 개발된 aminoglycoside 제제로 1991년 이후로 methicillin 내성 포도구균(methicillin-resistant Staphylococcus aureus, MRSA) 감염증에 대해 사용되어지고 있다. 저자들은 arbekacin의 MRSA에 대한 항균력을 평가하기 위해 2001년 3월에서 7월까지 아주대학병원 임상검체에서 분리된 MRSA 100균주에 대해 시험관내 항균력을 검사하고자 하였다. 방법 : 임상검체에서 자란 MRSA 100균주를 대상으로 디스크 확산법으로 감수성검사를 시행하고 한천희석법으로 최소발육저지농도(Minimal Inhibitory Concentration, MIC)를 측정하였으며, 항균효과를 평가하기 위하여 bactericidal effect와 post-antibiotic effect를 측정하였다. 결과 : MIC는 8㎍/mL로 측정된 2균주를 제외하고는 전부 4㎍/mL 이하였고, 디스크 확산법 검사상 arbekacin에 내성인 균주는 없었다. arbekacin은 농도에 비례하여 살균효과를 나타내었으며, post-antibiotic effect는 1.6-3.1 시간까지 지속되었다. 결론 : Arbekacin은 우수한 시험관내 항균력을 나타내어 MRSA의 감염증의 치료제로 사용할 수 있을 것으로 사료된다. Background : Arbekacin is an aminoglycoside developed in Japan and has been used since 1991 for the treatment of infections caused by methidllin-resistant Staphylococcus aureus(MRSA). The objective of this study is to determine the in vitro antimicrobial activity of arbekacin against clinical isolates of MRSA obtained from the Ajou University Hospital from March to July, 2001. Method : The minimal inhibitory concentrations(MICs) of the 100 MRSA isolates were determined by the agar dilution method and the antimicrobial susceptibility testing was performed by the disk diffusion method. The bactericidal and the post-antibiotic effects were tested. Results : Two isolates revealed MICs of 8 ㎍/mL, and the others < 4 ㎍/mL. All isolates were considered susceptible to arbekacin. Arbekacin showed concentration-dependent bactericidal activity against MRSA, and the post-antibiotic effect lasted 1.6-5.1 hours. Conclusion : These results suggest that arbekacin could provide an alternative for the treatment of MRSA.
