      • Differences in Epidermal Growth Factor Receptor Gene Mutations and Relationship with Clinicopathological Features in NSCLC Between Uygur and Han Ethnic Groups

        Objective: To investigate differences in mutations of epidermal growth factor receptor (EGFR) gene and relationships with clinicopathological features in patients with non-small cell lung cancer (NSCLC) between Uygur and Han ethnic groups. Methods: The Scorpions amplification refractory mutation system (Scorpions ARMS) was used to measure mutations in exons 18, 19, 20 and 21 of the EGFR gene in paraffin-embedded tumor tissue from NSCLC cases, and statistical analysis was performed to investigate links with clinicopathological features in different histological types of NSCLC. Results: Results from ARMS testing showed EGFR mutations in tumor tissues from six (6) of 50 NSCLC patients of Uygur ethnic group, with a positive rate of 12.0%; four of them (4) had exon 19 deletion in EGFR, and two (2) had L858R point mutation in exon 21 of EGFR. Statistically significant difference was noted in EGFR genetic mutation between adenocarcinoma and non-adenocarcinoma (P < 0.05), but no differences with gender, age group, smoking status, or stage (P > 0.05). EGFR mutations were detected in tumor tissues from 27 of 49 NSCLC patients of Han ethnic group, with a positive rate of 55.1%; 19 of them had exon 19 deletions, seven (7) had L858R point mutations in exon 21 of EGFR and one (1) had mutations in both exon 18 G719X and exon 20 T790M of EGFR. Statistically significant differences were noted in EGFR genetic mutations between genders and between adenocarcinoma and non-adenocarcinoma (P<0.05), but not with age group, smoking status, or stage (P > 0.05). Conclusion: Statistically significant differences were noted in the positive rates of EGFR genetic mutations in NSCLC patients between Uygur and Han ethnic groups, with lower positive rates for the Uygur cases.

        Facile Preparation of Mechanical Reinforced and Biocompatible Silk Gels

        Nontoxic and controllable way to fabricate silk fibroin (SF) gel with high mechanical properties is of critical important to biomaterial in tissue engineering. Electrochemically triggered electrophoretic migration and electric charge of the silk molecules both contributed to SF microspheres and electronic gel (e-gel) formation by sol-gel transition. In this study, a novel silk pH e-gel with higher mechanical property was prepared by combining low-voltage electric fields with isoelectric point (pI) adjustment. This green process was mild and friendly without chemical crosslinker. Compressive modulus of the silk pH e-gel was up to 70 MPa that was significantly higher than that of SF gelation spontaneously. Furthermore, analysis of molecule conformation of the silk pH-e-gel demonstrated that most of random coil structures transformed into α-helix and a little β-sheet structures during this process. The silk pH e-gel was loaded with rhodamine B and showed an obvious sustainable release profile. Accumulation releasing amounts was approximately 60 % at day 9. Cytocompatibility of the silk pH-e-gel was evaluated by epithelial cell. The results showed that the gels could support the cell growth and proliferation in vitro. Finally, gel biodegradation was assessed by protease XIV. After biodegradation for 28 days, remaining weight of the gel was about 20.23±2.59 wt%, indicating its good biodegradability. This novel process was established successfully by combining low voltage field with pH-control, which provided an alternative material for regenerative medicine.

      • Expression of MiR200a, miR93, Metastasis-related Gene RECK and MMP2/MMP9 in Human Cervical Carcinoma - Relationship with Prognosis

        Aim and Background: Cervical cancer remains the third most common cancer in women globally after breast and colorectal cancer. Well-characterized biomarkers are necessary for early diagnosis and to predict metastatic progression and effective therapy. MiRNAs can regulate gene expression, cell growth, differentiation and apoptosis by targeting mRNAs for translational repression or degradation in tumor cells. The present study was conducted to assess expression of miR93, miR200a, RECK, MMP2, MMP9 in invasive cervical carcinoma, and analyze their clinical significance. Method: A total of 116 patients with invasive cervical carcinoma and 100 patients undergoing hysterectomy for benign lesions were retrospectively examined. Quantitative real-time PCR was performed to determine expression of miR93 and miR200a while RECK, MMP2, MMP9 and MVD were assessed by immunohistochemical staining. Results: Cervical carcinoma patients demonstrated up-regulation of miR-93, miR-200a, MMP2 and MMP9, with down-regulation of RECK as compared to benign lesion tissues. RECK was significantly inversely related to invasion and lymphatic metastasis. The 5-year survival rate for patients with strong RECK expression was significantly higher than that with weakly expressing tumors. Conclusion: MiR-93 and miR-200a are associated with metastasis and invasion of cervical carcinoma. Thus together with RECK they are potential prognostic markers for cervical carcinoma. RECK cooperating with MMP2, MMP9 expression is a significant prognostic factor correlated with long-term survival for patients with invasive cervical carcinoma.

        Methylation Status of Transcriptional Modulatory Genes Associated with Colorectal Cancer in Northeast China

        Background/Aims: Methylation status plays a causal role in carcinogenesis in targeted tissues. However, the relationship between the DNA methylation status of multiple genes in blood leukocytes and colorectal cancer (CRC) susceptibility as well as interactions between dietary factors and CRC risks are unclear. Methods: We performed a case-control study with 466 CRC patients and 507 cancer-free controls to investigate the association among the methylation status of individual genes, multiple CpG site methylation (MCSM), multiple CpG site heterogeneous methylation and CRC susceptibility. Peripheral blood DNA methylation levels were detected by performing methylation-sensitive high-resolution melting. Results: Total heterogeneous methylation of CA10 and WT1 conferred a significantly higher risk of CRC (adjusted odds ratio [ORadjusted], 5.445; 95% confidence interval [CI], 3.075 to 9.643; ORadjusted, 1.831; 95% CI, 1.100 to 3.047; respectively). Subjects with high-level MCSM (MCSM-H) status demonstrated a higher risk of CRC (ORadjusted, 4.318; 95% CI, 1.529 to 12.197). Additionally, interactions between the high-level intake of fruit and CRH, WT1, and MCSM on CRC were statistically significant. Conclusions: The gene methylation status of blood leukocytes may be associated with CRC risk. MCSM-H of blood leukocytes was associated with CRC, especially in younger people. Some dietary factors may affect hypermethylation status and influence susceptibility to CRC.

      • An Improved Affinity Propagation Clustering Algorithm Based on Entropy Weight Method and Principal Component Analysis

        Traditional affinity propagation algorithm has inefficient results when conducting clustering analysis of high dimensional data because "dimension effect" lead to difficult find the proper class structure .In view of this, the author proposes an improved algorithm on the basis of Entropy Weight Method and Principal Component Analysis (EWPCA-AP). EWPCA-AP algorithm empowers the sample data by Entropy Weight Method, eliminate data irrelevant attributes by Principal Component Analysis, and travel with neighbor clustering algorithm, realization of high-dimensional data clustering in low dimension space. The numerical result of simulation experiment shows that the new EWPCA-AP algorithm can effectively eliminate the redundancy and irrelevant attributes of data and improve the performance of clustering. In addition, the proposed algorithm is applied in the area of the economy in our country and the clustering result is consistent with the real one. This algorithm provides a new intelligent evaluation method for Chinese economy.

        Growth behavior and kinetics of magnetite during magnetization roasting

        The magnetization roasting of hematite with two particle sizes was performed to investigate the growthbehavior and kinetics of magnetite. The magnetite exhibited comparable growth trends throughout themagnetization roasting process at different roasting temperatures, and enhancing the roasting temperaturecan facilitate the growth of magnetite. The nascent magnetite nuclei appeared acicular, predominantlydeveloped along the edges and fissures of the hematite particles before progressively growinginward. Moreover, the temperature stress and magnetite formation led to the loose and porous structureof the nascent magnetite particles. The growth kinetics demonstrated that the growth process of magnetitecould be divided into induction period and growth period based on the different growth rates. For the same hematite sample, the growth rate of magnetite in the induction period was lower than thatin the growth period. The magnetization roasting of hematite with a smaller particle size may be completedin a shorter time, even if increasing the particle size of hematite can accelerate the growth rateof magnetite. The growth kinetic models were established based on the parabolic law, and the reliabilityof describing the growth process of magnetite was verified by comparing the predicted values with theexperimental values.

        De novo transcriptome analysis of an albino mutant Pasphiopedilum pacific shamrock reveals reduced expression of genes related to chloroplast biosynthesis and division

        Paphiopedilum pacific shamrock is an orchid with high ornamental value. Understanding the mechanisms responsible for leaf color in albino mutants is important for ornamental development and breeding. In this study, we compared the leaf photosynthetic pigment content and transcriptome of albino mutants ppa01 and wild-type P. pacific shamrock. Photosynthetic pigment in mutants was less than 2% of the wild type and chl a/b was 60% less than the wild type. Transcriptome sequencing yielded 6.27 Gb and 5.67 Gb clean data from the mutant and wild-type leaves, respectively. De novo assembly yielded 104,763 unigenes with 15,400 greater than 1 kb in length. In unigene expression analysis, 3170 differentially expressed genes (DEGs) were identified with 2231 (70.38% of total DEGs) down-regulated. Results from GO and KEGG enrichment analysis, KEGG pathway analysis and qPCR suggest that the reduction of chloroplast biosynthesis and division in the mutant was due to low expression levels of ppGLK1 and ppFtsz. Mutants were associated with fewer chloroplasts in leaf cells, abnormal chloroplast structures, impaired chlorophyll biosynthesis, and thus reduced total chlorophyll and carotenoid contents. Furthermore, down-regulated expression of ppNYC1 reduced transformation of chlorophyll b into chlorophyll a, leading to a chl a/b decline. The research will guide future studies of leaf pigment mutations and the breeding of P. pacific shamrock.

      • Proteomic Analysis and Extensive Protein Identification from Dry, Germinating Arabidopsis Seeds and Young Seedlings

        Proteins accumulated in dry, stratified Arabidopsis seeds or young seedlings, totaled 1100 to 1300 depending on the time of sampling, were analyzed by using immobilized pH gradient 2-DE gel electrophoresis. The molecular identities of 437 polypeptides, encoded by 355 independent genes, were determined by MALDI-TOF or TOF-TOF mass spectrometry. In the sum, 293 were present at all stages and 95 were accumulated during the time of radicle protrusion while another 18 appeared in later stages. Further analysis showed that 226 of the identified polypeptides could be located in different metabolic pathways. Proteins involved in carbohydrate, energy and amino acid metabolism constituted to about 1/4, and those involved in metabolism of vitamins and cofactors constituted for about 3% of the total signal intensity in gels prepared from 72 h seedlings. Enzymes related to genetic information processing increased very quickly during early imbibition and reached highest level around 30 h of germination.
