Simple Sequence Repeat (SSR) and GC Distribution in the Arabidopsis thaliana Genome

We have mined each of the five A. thaliana chromosomes for the presence of simple sequence repeats (SSRs) and developed custom perl scripts to examine their distribution and abundance in relation to genomic position, local G/C content and location within and around transcribed sequences. The distribution of repeats and G/C content with respect to genomic regions (exons, UTRs, introns, intergenic regions and proximity to expressed genes) are shown. SSRs show a non-random distribution across the genome and a strong association within and around transcribed sequences, while G/C density is associated specifically with the coding portions of transcribed sequences. SSR motif repeat number shows a high degree of variation for each SSR type and a high degree of motif sequence bias reflecting local genome sequence composition. PCR primers suitable for the amplification of identified SSRs have been designed where possible, and are available for further studies.
Identification of Mg2Cu Particles in Cu-Alloyed Austempered Ductile Iron

In the present work, the Mg2Cu precipitates in copper-alloyed austempered ductile iron (ADI) were identified byanalyzing techniques such as TEM and SEM with EDS. It was revealed that, in castings made of ADI-containingcopper, highly dispersed particles of Mg2Cu are formed, whose size does not exceed <1 μm. The research workwas carried out on ductile iron that was austenitized at 900 °C, followed by austempering at 380 °C. The microstructurewas investigated using various techniques, including optical microscopy, XRD, SEM, and TEM. In additionto this, the exhibited impact properties of castings with Cu, Ni, and Cu+Ni were also determined. This studycasts a new light on the formation of the structure of Cu-alloyed ADI. The highly-dispersive and brittle Mg2Cu particlesthat are located in the vicinity of the graphite nodules have a negative effect on the impact properties of ADI. It has also been shown that impact strength decreases from levels of 160-180 J (for copper-free ADI) to 90-120 J(for copper- and copper-nickel-alloyed ADI).

<P>Abstract</P><P>The R2R3-type <I>OsMyb4</I> transcription factor of rice has been shown to play a role in the regulation of osmotic adjustment in heterologous overexpression studies. However, the exact composition and organization of its underlying transcriptional network has not been established to be a robust tool for stress tolerance enhancement by regulon engineering. <I>OsMyb4</I> network was dissected based on commonalities between the global chilling stress transcriptome and the transcriptome configured by <I>OsMyb4</I> overexpression. <I>OsMyb4</I> controls a hierarchical network comprised of several regulatory sub-clusters associated with cellular defense and rescue, metabolism and development. It regulates target genes either directly or indirectly through intermediary MYB, ERF, bZIP, NAC, ARF and CCAAT-HAP transcription factors. Regulatory sub-clusters have different combinations of MYB-like, GCC-box-like, ERD1-box-like, ABRE-like, G-box-like, as1/ocs/TGA-like, AuxRE-like, gibberellic acid response element (GARE)-like and JAre-like <I>cis</I>-elements. Cold-dependent network activity enhanced cellular antioxidant capacity through radical scavenging mechanisms and increased activities of phenylpropanoid and isoprenoid metabolic processes involving various abscisic acid (ABA), jasmonic acid (JA), salicylic acid (SA), ethylene and reactive oxygen species (ROS) responsive genes. <I>OsMyb4</I> network is independent of drought response element binding protein/C-repeat binding factor (<I>DREB/CBF</I>) and its sub-regulons operate with possible co-regulators including nuclear factor-Y. Because of its upstream position in the network hierarchy, <I>OsMyb4</I> functions quantitatively and pleiotrophically. Supra-optimal expression causes misexpression of alternative targets with costly trade-offs to panicle development.</P>

The Long Baseline Array is an array of radio telescopes using the technique of Very Long Baseline Interferometry to achieve milli-arcsecond-scale angular resolution. The core telescopes are located in Australia, with telescopes in New Zealand and South Africa also participating regularly. In this paper the capabilities of the Long Baseline Array are described, and examples of the science undertaken with the array are given.

The rhizome of Curcuma longa (turmeric) is often used in Asia as a spice and as a medicine. Its most wellstudied component, curcumin, has been shown to exhibit poor bioavailability in animal studies and clinical trials. We hypothesized that the presence of lipophilic components (e.g., turmerones) in turmeric extract would affect the absorption of curcumin. The effects of turmerones on curcumin transport were evaluated in human intestinal epithelial Caco-2 cells. The roles of turmerones on P-glycoprotein (P-gp) activities and mRNA expression were also evaluated. Results showed that in the presence of a- and aromatic turmerones, the amount of curcumin transported into the Caco-2 cells in 2 hours was significantly increased. a-Turmerone and verapamil (a P-gp inhibitor) significantly inhibited the efflux of rhodamine-123 and digoxin (i.e.,inhibited the activity of P-gp). It is interesting that aromatic turmerone significantly increased the rhodamine-123 efflux and Pgp (MDR1 gene) mRNA expression levels. The effects of a- and aromatic turmerones on curcumin transport as well as P-gp activities were shown here for the first time. The presence of turmerones did affect the absorption of curcumin in vitro. These findings suggest the potential use of turmeric extract (including curcumin and turmerones), rather than curcumin alone, for treating diseases.

Background: Virtual reality (VR) distraction is a nonpharmacological method to prevent acute pain that has not yet been thoroughly explored for anesthesiology. We present our experience using VR distraction to decrease routine intravenous sedation for patients undergoing preoperative perineural catheter insertion. Methods: This 1-month quality improvement project involved all elective unilateral primary total knee arthroplasty patients who received a preoperative adductor canal catheter. Clinical data were analyzed retrospectively. For the first half of the month, all patients received usual care; intravenous sedation was administered at the discretion of the regional anesthesiologist. For the second half of the month, patients were offered VR distraction with intravenous sedation upon request. The primary outcome was fentanyl dosage; other outcomes included midazolam dosage, procedure-related pain, procedural time, and blood pressure changes. Results: Seven patients received usual care and seven used VR. In the VR group, 1/7 received intravenous sedation versus 6/7 who received usual care (P = 0.029). The fentanyl dose was lower (median [10th–90th percentiles]) in the VR group (0 [0–20] μg) versus the non-VR group (50 [30–100] μg; P = 0.008). Midazolam use was lower in the VR group (0 [0–0] mg) than in the non-VR group (1 [0–1] mg; P = 0.024). Procedure-related pain was lower in the VR group (1 [1–4] NRS) versus the non-VR group (3 [2–6] NRS; P = 0.032). There was no difference in other outcomes. Conclusions: VR distraction may provide an effective nonpharmacological alternative to intravenous sedation for the ultrasound-guided placement of certain perineural catheters.
