        Identification and expression patterns of 20-hydroxyecdysoneresponsive genes from Procambarus clarkii

        Ecdysteroids play an important role in the regulation of molting process in crustaceans. The suppression subtractive hybridization (SSH) technique was employed to search for 20-hydroxyecdysone-responsive genes in the hepatopancreas of the crayfish Procambarus clarkii. Ninety-three putative expressed sequence tags were identified by SSH, including three immune response-related genes, two cell cycle and apoptosis genes, four respiration and energy metabolism genes, four transport-related genes, six metabolism-related genes, two stress response genes, and eight transcription and translation regulation genes. The expression levels of the examined genes were regulated by 20-hydroxyecdysone and varied in the hepatopancreas during the molting stages; data were confirmed by real-time PCR. RNAi of the ecdysteroid receptor had significant effects on the expression levels of 20-hydroxyecdysone- responsive genes. These differentially expressed genes identified by SSH will provide insight into the ecdysteroid signaling pathway in P. clarkii.

      • Gene Microarray Assessment of Multiple Genes and Signal Pathways Involved in Androgen-dependent Prostate Cancer Becoming Androgen Independent

        To study the gene expression change and possible signal pathway during androgen-dependent prostate cancer (ADPC) becoming androgen-independent prostate cancer (AIPC), an LNCaP cell model of AIPC was established using flutamide in combination with androgen-free environment inducement, and differential expression genes were screened by microarray. Then the biological process, molecular function and KEGG pathway of differential expression genes are analyzed by Molecule Annotation System (MAS). By comparison of 12,207 expression genes, 347 expression genes were acquired, of which 156 were up-ragulated and 191 down-regulated. After analyzing the biological process and molecule function of differential expression genes, these genes are found to play crucial roles in cell proliferation, differntiation, cell cycle control, protein metabolism and modification and other biological process, serve as signal molecules, enzymes, peptide hormones, cytokines, cytoskeletal proteins and adhesion molecules. The analysis of KEGG show that the relevant genes of AIPC transformation participate in glutathione metabolism, cell cycle, P53 signal pathway, cytochrome P450 metabolism, Hedgehog signal pathway, MAPK signal pathway, adipocytokines signal pathway, PPAR signal pathway, TGF-${\beta}$ signal pathway and JAK-STAT signal pathway. In conclusion, during the process of ADPC becoming AIPC, it is not only one specific gene or pathway, but multiple genes and pathways that change. The findings above lay the foundation for study of AIPC mechanism and development of AIPC targeting drugs.

        现代汉语“V+C1C2+NP处”结构的使用现象研究

        In Modern Chinese grammar, it is stipulated that the locative object following a verb can only be placed in between the compound directional complement. However, there is a significant phenomenon of the locative object appearing after the compound directional complement in our surroundings. How is this ungrammatical word order phenomenon distributed and what differences exist between it and the normal grammatical expression? Based on previous research, this article presents a comparative analysis of a large corpus of data, enumerating the distribution and syntactic differences between two types of expressions. For instance, the verb category represented by "进" and "回" tends to use the intermediate-object structure, while the verb category represented by "出" and "过" tends to use the post-object structure in the compound directional complement. From an information structure perspective, the intermediate-object structure is often employed to emphasize the locative object at the end of a sentence, while the post-object structure is frequently used in serial verb constructions to highlight the following constituent. Through an analysis of the corpus and a comparison with actual usage through questionnaire surveys, this article discovers that the phenomenon of placing the locative object after the compound directional complement deserves our attention. It is not simply ungrammatical sentences but rather a special word order with usage patterns and tendencies. Moreover, to some extent, it can fulfill the needs of specific syntax and semantics. 现代汉语语法中规定动词后处所宾语只能用在复合趋向补语的中间使用,但在我们周围存在大量处所宾语用在复合趋向补语后的语序现象。这种不合语法的语序现象是如何分布的且与正常的语法表述之间又存在什么样的差异呢?本文在前人研究的基础上,对语料库中大量语料对比分析列举出两种表述格式的分布情况和语序差异。如“进、回”字类动趋式多用中宾式表述;“出、过”字类动趋式多用后宾式表述。从信息结构来看中宾式多用在句末突出处所宾语,而后宾式多用在连动句中突出后面成分。本文在对语料库分析的同时通过问卷调查与人们实际的使用情况对比发现,处所宾语用在复合趋向补语后的语序现象值得我们关注,其并非单纯的病句,而是有使用规律和倾向性的特殊语序,且在一定程度上能满足特殊句法和语义的需要。

        Creep properties and damage model for salt rock under low-frequency cyclic loading

        Triaxial compression creep tests were performed on salt rock samples using cyclic confining pressure with a static axial pressure. The test results show that, up to a certain time, changes in the confining pressure have little influence on creep properties of salt rock, and the axial creep curve is smooth. After this point, the axial creep curve clearly fluctuates with the confining pressure, and is approximately a straight line both when the confining pressure decreases and when it increases within one cycle period. The slope of these lines differs: it is greater when the confining pressure decreases than when it increases. In accordance with rheology model theory, axial creep equations were deduced for Maxwell and Kelvin models under cyclic loading. These were combined to establish an axial creep equation for the Burgers model. We supposed that damage evolution follows an exponential law during creep process and replaced the apparent stress in creep equation for the Burgers model with the effective stress, the axial creep damage equation for the Burgers model was obtained. The model suitability was verified using creep test results for salt rock. The fitting curves are in excellent agreement with the test curves, so the proposed model can well reflect the creep behavior of salt rock under low-frequency cyclic loading. In particular, it reflects the fluctuations in creep deformation and creep rate as the confining pressure increasing and decreasing under different cycle periods.

        Surrounding rock pressure of shallow-buried bilateral bias tunnels under earthquake

        By means of finite element numerical simulation and pseudo-static method, the shallow-buried bilateral bias twin-tube tunnel subject to horizontal and vertical seismic forces are researched. The research includes rupture angles, the failure mode of the tunnel and the distribution of surrounding rock relaxation pressure. And the analytical solution for surrounding rock relaxation pressure is derived. For such tunnels, their surrounding rock has sliding rupture planes that generally follow a "W" shape. The failure area is determined by the rupture angles. Research shows that for shallow-buried bilateral bias twin-tube tunnel under the action of seismic force, the load effect on the tunnel structure shall be studied based on the relaxation pressure induced by surrounding rock failure. The rupture angles between the left tube and the right tube are independent of the surface slope. For tunnels with surrounding rock of Grade IV, V and VI, which is of poor quality, the recommended reinforcement range for the rupture angles is provided when the seismic fortification intensity is VI, VII, VIII and IX respectively. This study is expected to provide theoretical support regarding the ground reinforcement range for the shallow-buried bilateral bias twin-tube tunnel under seismic force.

        Attenuation of Peripheral Regulatory T-Cell Suppression of Skin-Homing CD8+T Cells in Atopic Dermatitis

        Purpose: Cutaneous lymphocyte-associated antigen (CLA)-expressing CD8+T cells have been known to play an important role in the pathogenesis of atopic dermatitis(AD). However, the mechanisms underlying the loss of self-tolerance remainunclear. Regulatory T cells (Tregs) play a key role in the development of homeostasisin the immune system. We, therefore, hypothesized that a reduced ability of Tregs to inhibit autologous CD8+CLA+T cells might be underlying mechanism in AD. Materials and Methods: CD8+CLA+T cells and Tregs were obtained from the peripheral blood of AD patients and control volunteers. The frequenciesof CD8+CLA+T cells were evaluated. The proliferative responses of CD8+CLA+T cells were assessed by flow cytometry, and the levels of transforming growth factor-β1 (TGF-β1) and interleukin-10 (IL-10) in culture supernatants were detected by enzyme-linked immunosorbent assay. Results: Our results revealed higher frequency and increased expression of perforin and granzyme-B in peripheralCD8+CLA+T cells in AD, and lower inhibitory ability of Tregs on proliferation of CD8+CLA+T cells in AD. Meanwhile, the levels of TGF-β1 produced by Tregs were significantly lower in AD, and anti-TGF-β1 abolished such suppression. Conclusion: The attenuated inhibitory ability of Tregs on hyper-activated autologousCD8+CLA+T cells, mediated by TGF-β1, plays an important role in the pathogenesis of AD.

        Prediction model of surface subsidence for salt rock storage based on logistic function

        To predict the surface subsidence of salt rock storage, a new surface subsidence basin model is proposed based on the Logistic function from the phenomenological perspective. Analysis shows that the subsidence curve on the main section of the model is S-shaped, similar to that of the actual surface subsidence basin; the control parameter of the subsidence curve shape can be changed to allow for flexible adjustment of the curve shape. By using this model in combination with the MMF time function that reflects the single point subsidence-time relationship of the surface, a new dynamic prediction model of full section surface subsidence for salt rock storage is established, and the numerical simulation calculation results are used to verify the availability of the new model. The prediction results agree well with the numerical simulation results, and the model reflects the continued development of surface subsidence basin over time, which is expected to provide some insight into the prediction and visualization research on surface subsidence of salt rock storage.

      • Vision-Based Method for Forward Vehicle Brake Lights Recognition

        This paper presents a recognition algorithm combining the vehicle detection and the color difference of RGB color space to recognize the brake-lights state of moving vehicles in order to achieve the intelligent-car’s rear-end collision warning about the vehicle in front of it. Firstly, we train and build AdaBoost cascade classifier by haar features samples and scaling sub-windows are used to detect the target vehicle from the region of interest of the resized image. Then we compare the adjacent frames to recognize brake light status, which including using color, shape, structural features to identify the third brake light; comparing the center of gravity coordinates and the color difference threshold to rear brake lights when vehicles are not red or yellow; according to subtraction of each RGB corresponding channel, binarization, and the color difference threshold of RGB color space to identify the red or yellow vehicles’ brake lights. Finally, experiments show that the algorithm can detect the front vehicle’s braking quickly and accurately.

        An improved Maxwell creep model for salt rock

        The creep property of salt rock significantly influences the long-term stability of the salt rock underground storage. Triaxial creep tests were performed to investigate the creep behavior of salt rock. The test results indicate that the creep of salt rock has a nonlinear characteristic, which is related to stress level and creep time. The higher the stress level, the longer the creep time, the more obvious the nonlinear characteristic will be. The elastic modulus of salt rock decreases with the prolonged creep time, which shows that the creep damage is produced for the gradual expansion of internal cracks, defects, etc., causing degradation of mechanical properties; meanwhile, the creep rate of salt rock also decreases with the prolonged creep time in the primary creep stage, which indicates that the mechanical properties of salt rock are hardened and strengthened. That is to say, damage and hardening exist simultaneously during the creep of salt rock. Both the damage effect and the hardening effect are considered, an improved Maxwell creep model is proposed by connecting an elastic body softened over time with a viscosity body hardened over time in series, and the creep equation of which is deduced. Creep test data of salt rock are used to evaluate the reasonability and applicability of the improved Maxwell model. The fitting curves are in excellent agreement with the creep test data, and compared with the classical Burgers model, the improved Maxwell model is able to precisely predict the long-term creep deformation of salt rock, illustrating our model can perfectly describe the creep property of salt rock.

      • Comparative Serum Proteomic Analysis of Serum Diagnosis Proteins of Colorectal Cancer Based on Magnetic Bead Separation and MALDI-TOF Mass Spectrometry

        Background: At present, the diagnosis of colorectal cancer (CRC) requires a colorectal biopsy which is an invasive procedure. We undertook this pilot study to develop an alternative method and potential new biomarkers for diagnosis, and validated a set of well-integrated tools called ClinProt to investigate the serum peptidome in CRC patients. Methods: Fasting blood samples from 67 patients diagnosed with CRC by histological diagnosis, 55 patients diagnosed with colorectal adenoma by biopsy, and 65 healthy volunteers were collected. Division was into a model construction group and an external validation group randomly. The present work focused on serum proteomic analysis of model construction group by ClinProt Kit combined with mass spectrometry. This approach allowed construction of a peptide pattern able to differentiate the studied populations. An external validation group was used to verify the diagnostic capability of the peptidome pattern blindly. An immunoassay method was used to determine serum CEA of CRC and controls. Results: The results showed 59 differential peptide peaks in CRC, colorectal adenoma and health volunteers. A genetic algorithm was used to set up the classification models. Four of the identified peaks at m/z 797, 810, 4078 and 5343 were used to construct peptidome patterns, achieving an accuracy of 100% (> CEA, P<0.05). Furthermore, the peptidome patterns could differentiate the validation group with high accuracy close to 100%. Conclusions: Our results showed that proteomic analysis of serum with MALDI-TOF MS is a fast and reproducible approach, which may provide a novel approach to screening for CRC.
