Objectives: Dendritic cell (DC)-based tumor immunotherapy needs an immunogenic tumor associated antigen (TAA) and an effective approach for its presentation to lymphocytes. In this study we explored whether transduction of DCs with lentiviruses (LVs) expressing the human interleukin-12 gene could stimulate antigen-specific cytotoxic T cells (CTLs) against human lung cancer cells in vitro. Methods: Peripheral blood monocyte-derived DCs were transduced with a lentiviral vector encoding human IL-12 gene (LV-12). The anticipated target of the human IL-12 gene was detected by RT-PCR. The concentration of IL-12 in the culture supernatant of DCs was measured by ELISA.Transduction efficiencies and CD83 phenotypes of DCs were assessed by flow cytometry. DCs were pulsed with tumor antigen of lung cancer cells (DC+Ag) and transduced with LV-12 (DC-LV-12+Ag). Stimulation of T lymphocyte proliferation by DCs and activation of cytotoxic T-lymphocytes (CTL) stimulated by LV-12 transduced DCs pulsed with tumor antigen against A549 lung cancer cells were assessed with methyl thiazolyltetrazolium (MTT). Results: A recombinant lentivirus expressing the IL-12 gene was successfully constructed. DC transduced with LV-12 produced higher levels of IL-12 and expressed higher levels of CD83 than non-transduced. The DC modified by interleukin -12 gene and pulsed with tumor antigen demonstrated good stimulation of lymphocyte proliferation, induction of antigen-specific cytotoxic T lymphocytes and antitumor effects. Conclusions: Dendritic cells transduced with a lentivirus-mediated interleukin-12 gene have an enhanced ability to kill lung cancer cells through promoting T lymphocyte proliferation and cytotoxicity.

<P><B>Abstract</B></P> <P>Selective inhibition of carbonic anhydrase (CA) enzyme is an active area of research for medicinal chemists. In the current account, a hybrid pharmacophore approach was employed to design sulfonamide, amide and amine containing new series of potent carbonic anhydrase II inhibitors. The aromatic fragment associated with pharmacophore was altered suitably in order to find effective inhibitors of CA-II. All the derivatives <B>4a-4m</B> showed better inhibition compared to the standard acetazolamide. In particular, compound <B>4l</B> exhibited significant inhibition with IC<SUB>50</SUB> value of 0.01796 ± 0.00036 µM. The chemo-informatics analysis justified that all the designed compounds possess <10 HBA and <5 HBD. This paper attempts to study the impact of Financial Literacy on Youth Entrepreneurial Intent in Pakistan. A closed-ended self-administered structured questionnaire covering financial literacy, computer knowledge, financial attitude, and financial knowledge with Entrepreneurial Intent was collected from young entrepreneurs. The research tried to investigate the education level with computer skill to inspect the effect of financial literateness on young generation Entrepreneurial Intent in the context of Pakistan. The research model was tested using PLS-SEM and authenticating a measurement model through the advanced methodology and their association with Entrepreneurial Intent. Results revealed that financial literacy and its two parts (financial attitude and financial knowledge) have a positive impact on Entrepreneurial Intent. The size of the joint impact of financial literacy and its components on Entrepreneurial Intent was assessed to be adequate. Entrepreneurial Intent is essential for creating new firms to maintain economic development. Furthermore, it is determined in this research that if youth has better financial knowledge and financial attitude, the probability of Entrepreneurial Intent increases. This suggests that if the youth in Pakistan desire to attain a higher limit of Entrepreneurial Intent, they must implement financial literacy models for enhancing and promoting their current Entrepreneurial Intent.
Gluteal Region Reshaping of Massive Weight Loss Patients—A Decision-Making Strategy

Background Massive weight loss (MWL) is a very common presentation that you may face as a plastic surgeon. Each patient has his own individual criteria, so, you should work according to a well-organized plan, especially when such cases have concerns about their gluteal area contour that were neglected before by many surgeons. A decision-making strategy was used to give a personalized treatment for targeting gluteal region reshaping of MWL patients. Methods This study considered all patients with MWL subjected to buttock reshaping. There was no randomization in treatment; there was a case-by-case assessment. We analyzed the features of the buttocks, the type of surgery performed, the outcomes, and the complications. Results Fifty two patients were included (41 females and 11 males), ages ranged between 21 and 66 years. Demographic data, preoperative body mass index (BMI), duration of surgery, type of surgery, and postoperative complications were collected. Statistically significant improvements were observed in gluteal ptosis and patient satisfaction grades. Conclusion Aesthetic improvement of the buttocks involves either augmentation or contouring that may be obtained by liposculpture, surgical lifting, or combination. Patients with MWL have high expectations and are often treated with multiple procedures. Thus, an easy strategic approach personalized on each patient to treat multiple adjacent areas in one operation is necessary. Adipose tissue distribution, gluteal skin status, and BMI were the main factors that can forcefully affect our plan to guarantee reduction of unpleasant results and complications and improve patient satisfaction.

Objective: This study investigated the impact of two stimulation protocols using highly purified human menopausal gonadotropin (HP-hMG) on the endocrine profile, follicular fluid soluble Fas levels, and outcomes of intracytoplasmic sperm injection (ICSI) cycles. Methods: This prospective clinical trial included 100 normal-responder women undergoing ovarian stimulation for ICSI; 55 patients received concomitant follicle-stimulating hormone (FSH) plus HP-hMG from the start of stimulation, while 45 patients received FSH followed by HP-hMG during mid/late follicular stimulation. The primary outcome was the number of top-quality embryos. The secondary outcomes were the number and percentage of metaphase II (MII) oocytes and the clinical pregnancy rate. Results: The number of MII oocytes was significantly higher in the concomitant protocol (median, 13.0; interquartile range [IQR], 8.5-18.0 vs. 9.0 [8.0-13.0] in the consecutive protocol; p=0.009); however, the percentage of MII oocytes and the fertilization rate were significantly higher in the consecutive protocol (median, 90.91; IQR, 80.0-100.0 vs. 83.33 [75.0-93.8]; p=0.034 and median, 86.67; IQR, 76.9-100.0 vs. 77.78 [66.7-89.9]; p=0.028, respectively). No significant between-group differences were found in top-quality embryos (p=0.693) or the clinical pregnancy rate (65.9% vs. 61.8% in the consecutive vs. concomitant protocol, respectively). The median follicular fluid soluble Fas antigen level was significantly higher in the concomitant protocol (9,731.0 pg/mL; IQR, 6,004.5-10,807.6 vs. 6,350.2 pg/mL; IQR, 4,382.4-9,418.4; p=0.021). Conclusion: Personalized controlled ovarian stimulation using HP-hMG during the late follicular phase led to a significantly lower response, but did not affect the quality of ICSI.
Micro-grid Monitoring and Supervision: Web-based SCADA Approach

Owing to the widespread use of the micro-grid concept to serve many real life applications, the main concern of this paper is to monitor, evaluate and manage the operational performance of an existent, already installed micro-grid that consists of On & Off grid PV systems in addition to the main grid supply. With the aid of customized web based SCADA system fully designed by authors, the performance evaluation of the proposed micro-grid is carried out in terms of energy consumption, power quality indices and energy cost based on Egypt’s energy current tariff . The designed system helps in both micro-grid status monitoring as well as load sharing between the main grid supply and the off grid PV energy system. The data of the on grid PV energy system are accessed and collected online directly from the inverter web server instead of downloading them offl ine from the PV inverter. In this case, data are analyzed with a reasonable refresh rate, which saves time and eff ort. Remote access platform is developed securely through a multi-level security authentication and strong data encryption using the OpenVPN protocol under the Dynamic Domain Name System service. Smart wired and wireless technology—used as integrated sensing devices to the developed SCADA system—enables easy system installation and running cost saving. Finally, to validate the collected data from the newly SCADA designed monitoring system, a performance characterization for the on grid PV system following international standards of yields calculations is carried out for 3 months in 2019.

Natural convection in an F-shaped cavity containing Ag-water non-Newtonian nanofluid (NF) saturatedwith a porous medium was investigated numerically. The left wall of the cavity was maintained at a constant hot temperatureand subjected to a horizontal periodic magnetic field. The right wall was maintained at a constant cold temperature. Both the top and bottom walls of the cavity were kept thermally insulated. In the present work, the Hartmannnumber varied as (0Ha60), the power law index varied as (0.6n1.4), the periodic shape parameter varied as(0.11), the Rayleigh number varied as (103Ra106), the aspect ratio of the cavity varied as (0.1AR0.4), theDarcy number varied as (105Da101) and the solid volume fraction varied as (0 0.06). It was found that theaverage Nusselt number (Nu) increases by increasing the (, Da and Ra), whereas it decreases when (n) and (Ha)increase. Furthermore, it was realized that the relationship between the (Nu) and () exhibits a different behaviordepending on the considered values of (Ra) and (n).

A novel three variable refined plate theory (TVRPT) is developed in this article for laminated composite plates for the first time. The theory takes into account the nonlinear variation of transverse shear deformations, and satisfies the boundary conditions of zero traction on the plate surfaces without considering the “shear correction factor”. The important characteristic of this new kinematic is that the unknowns numbers is only 3 as is employed in “classical plate theory” (CPT). The numerical results of the current theory are compared with 3D-elasticity solutions and the calculations of “first order theories” and other higher order models found in the literature.

The most widely of hydraulic applications power employed source that including low manufacturing costs and good performance are defined as the external gear pumps. In this study, the using of unstandard gears and influence on the performance of gear pumps are studied. These influences include pulsation factor of flow rate, flow rate of pump and trapped volume between two meshing gears. Matlab software has been used to evaluate performance by modifying algorithm and deriving the equations of pump flow rate. The unstandard gear is used in the study undertaken with +0.3 Rack shift coefficient. Results indicated the using of unstandard gears increase flow rate and reduce the trapped volume and pulsation flow rate factor. It was concluded that the factor of pulsation flow rate reduces as increase of teeth number which leads to pulsation flow rate stabilization.
