          Safety Evaluation of Epimedium koreanum Water Extract in Sprague-Dawley Rats

          음양곽 물추출물의 랫에서의 안전성을 검증하기 위하여 그룹당 5마리씩 총 20마리의 수컷 SD 랫에 각각 음양곽 물추출물을 체중당 10, 100, 1000 ㎎/㎏을 1일 1회 4주간 경구투여 하였다. 대조군으로는 생리식염수를 동량 투여하였다. 임상증상, 체중 및 사료섭취량, 혈액, 혈액생화학 및 조직학적 검사를 실시하였다. 시험기간 중 임상증상, 체중, 사료섭취량의 이상변화를 관찰할 수 없었으며, 투약종료 다음날 부검결과 고환, 부고환의 무게가 용량의존적으로 증가하였으나, 정낭선은 감소경향을 나타내었다. 조직학적 검사결과 일부 변화가 관찰되었으나 건강한 쥐에서도 발견되는 변화였다. 따라서 체중당 1000 ㎎까지의 투여는 임상적으로 안전한 것으로 사료된다. To evaluate of oral toxicity of Epimedium koreanum nakai (EKN) water extract, three doses of EKN water extract (10, 100 and 1000 ㎎/㎏/day) were administered to healthy Sprague-Dawley rats for 4 weeks. The results showed that there are no abnormal signs during the experimental period. The weights of testis and epididymis were increased dose-dependently, but seminal vesicle was decreased compared with control group. In hematology and serum chemistry, changes were observed within the normal ranges. In histological finding, slight to moderated changes were found but those were commonly or rarely encountered in the normal rats. These results suggest that no observed adverse effect level (NOAEL) of the oral application of EKN was considered to be more than 1000 ㎎/㎏ in SD rats under the conditions employed in this study.

          Effect of Beta-Glucan on the Collagen-Induced Rheumatoid Arthritis

          본 실험은 Aureobasidium pullulans SM-2001 유래 베타글루칸의 콜라겐 유발 류마티스 관절염에서의 효과를 알아보고자 하였다. 6 주령 DBA/1J 수컷 마우스를 1주일간 적응기간을 거친 후 콜라겐 200 ㎍를 프로인트 완전면역 보강제와 함께 꼬리부위 피내에 접종하였으며, 21일 후에 콜라겐을 프로인트 불완전면역보강제와 함께 추가접종을 하였다. 베타글루칸 (체중당 21.25, 42.5, 85 ㎎/㎏), 디클로페낙 (체중당 2 ㎎/㎏)을 첫 접종일부터 추가접종 후 1주일간 투여하였다. 관절염 유발군에서는 과증식된 비장(증가된 백색속질)과 연골손상의 증가가 관찰되었다. 이러한 증상은 폴리칸 21.25 ㎎/㎏ 투여군의 일부 항목(경골, 연골)을 제외한 모든 폴리칸 투여군에서 용량의존적으로 유의한 효과를 나타내었다. 아우레오바시디움 플루란스 유래 베타글루칸이 콜라겐으로 유발된 류마티스관절염과 과면역을 효과적으로 억제하는 것을 알 수 있었다. The effect of beta-glucan (Polycan™) derived from Aureobasidium pullulans SM-2001 were observed on collagen-induced rheumatoid arthritis (RA) in DBA mice. Six week-old male DBA/1J mice were immunized by the intradermal injection of 200 ㎍ of bovine type Ⅱ collagen with the equal volume of complete Freund's adjuvant at the tail base on day 1. On day 21, the mice were boosted by the intradermal injection of 200 μg of bovine type Ⅱ collagen with incomplete Freund's adjuvant. From the first immunization, mice had been administered Polycan™ (21.25, 42.5 and 85 ㎎/㎏), diclofenac and vehicle once a day for 4 weeks, respectively. Collagen-induced hyperimmunities and arthritis signs were markedly and dose-dependently inhibited by treatment of Polycan™ compared with RA control except for tibial cartilages of Polycan™ 21.25 group. Polycan™ effectively inhibited the histopathological changes of collagen-induced arthritis and hyper-immunities.

          Effect of Epimedium Koreanum on the 2-Bromopropane Induced-Reproductive Damage

          음양곽(Epimedicum Koreanum nakai)은 예부터 생식기 장애치료를 위해 사용되어 왔다. 본 연구에서는 2-브로모프로판(2-bromopropane; 2-BP)으로 유발된 생식기 장애에서 음양곽 물추출물의 생식기장애 치료효과를 평가하였다. 2-BP를 체중㎏당 1,355 ㎎씩 28일간 피하로 투여하여 생식기 장애를 유발하였으며, 음양곽 추출물은 용량별(10, 100, 500 ㎎/㎏)로 4주간 경구투여 하였다. 그 결과 일일정자생성(daily sperm production)은 대조군에 비하여 증가하였으나 용량의존적 감소를 나타내었으며, 정세관과 부고환관에서는 조직학적 유의한 변화를 나타내었다. 이러한 결과는 음양곽물추출이 2-BP로 유발된 생식기장애의 치료에 도움이 될 것으로 생각된다. Epimedium koreanum nakai (EKN) has been used for treatment of reproductive disorders. In this study, we evaluated the effects of EKN water extract on the 2-bromopropane (2-BP) induced reproductive dysfunction. The daily sperm production (DSP), sperm counts and histological changes of reproductive organs were investigated after 4 weeks of EKN administration in 2-BP induced reproductive damaged rats. Although the weights of epididymis and seminal vesicle were increased dose-dependently, they were lower than control group. DSP of EKN-dosing groups were increased compared to control group. In EKN-dosing groups, the number of degenerative seminiferous tubules and the number of epididymal tubules showing epithelial cell vacuolation and decreased spermatozoa in the lumen were significantly (p < 0.01) decreased compared to control group dose-dependently. Significant (P < 0.05 or P < 0.01) increases of the number of sertoli cells, spermatogonia, patchytene spermatocytes, round spermatids and elongated spermatids were observed in EKN-dosing groups compared control group. Consequently, based on the results, EKN water extract could treat 2-BP induced reproductive damages dramatically.

          혈액순환과 관련하여 nitric oxide 생성과 angiotensin converting enzyme 활성에 미치는 Acai berry 에탄올 추출물의 영향

          본 연구의 목적은 혈관계 순환을 촉진시키는 약효제를 발굴하는 것이다. 아사이베리(Acai berry) 에탄올 추출물(ABEE)의 항산화 효과 뿐만 아니라 토끼의 폐에서 유래한 angiotensin converting enzyme (ACE)의 활성 및 대식세포와 혈관내피세포에서 nitric oxide 생성에 대한 효과가 본 연구에서 조사되었다. 먼저 환원력과 지질과산화에 대한 ABEE의 항산화효과가 관찰되었다. 뿐만 아니라, ABEE는 hydroxyl radical에 의하여 유발된 DNA 산화에 대한 보호효과를 보여주었다. 더욱이 ABEE는 0.01%의 농도에서 약 50% 정도의 angiotensin converting enzyme 활성에 대한 억제효과를 발휘하는 것으로 나타났다. 혈관내피세포에서 ABEE는 nitric oxide 의 생성을 증가시켰으나 대식세포에서는 lipopolysaccharide에 의하여 생성된 nitric oxide 생성을 감소시켰다. 게다가 superoxide dismutase (SOD)-2와 -3 발현수준은 ABEE 처리에 의하여 증가되었으나 SOD-1의 발현수준은 일정하였다. 더욱이 nitric oxide synthases-1 (NOS-1)의 발현수준은 ABEE 처리에 의하여 증가되었으나, 유발 효소인 NOS-2의 발현수준은 일정하였다. 또한 SOD의 전사인자인 Nrf-2의 발현수준은 ABEE에 의하여 증가되었다. 그러므로 이러한 결과들은 ABEE가 이상의 작용을 경유하여 혈액순환을 촉진시킬 수 있고, 혈관의 건강을 위한 큰 잠재성을 가지고 있다는 것을 암시하고 있다. The aim of this study is to develop a supplementary therapeutic agent capable of promoting vascular circulation. The effects of Acai berry ethanolic extracts (ABEE) on activity of angiotensin converting enzyme from rabbit lung, production of nitro oxide in both murine macrophage cells and vascular endothelial cells as well as antioxidant effects were investigated in this study. First of all, it was observed the direct effects of ABEE on reducing power and antioxidant effect lipid peroxidation. In addition, ABEE showed a protective effect on DNA oxidation induced by hydroxyl radical. Furthermore, ABEE at 0.01% exerted approximately 50% inhibition on activity of angiotensin converting enzyme. ABEE increased the production of nitric oxide in endothelial cells, but decreased the induction of nitric oxide stimulated by lipopolysaccharide in microphage. The expression levels of superoxide dismutase (SOD)-2 and -3 were enhanced by ABEE treatment, however, the expression level of SOD-1 remained constant. Moreover, the expression level of nitric oxide synthases-1 (NOS-1), a constitutive enzyme, was increased by ABEE, but that of NOS-2, a inducible enzyme, was constant. It was also found that the level of Nrf-2, a transcription factor of SOD, was increased by ABEE. Therefore, these results demonstrate that ABEE could promote blood circulation via above actions, suggesting that may be helpful for health of blood vessel.

          골프 드라이버샷 동작에서 공 속도에 따른 운동학적 변인 비교

          The purpose of this study was to compare three-dimensional imaegs through the analysis of the kinematic variables of the ball speed when the driver shot in competitio nof high school athletes, the analysis for this purpose. For this study, were enrolled in the eight high school athletes who participated in The 13th of Konkuk University President championship. Using a total of two camera sand one Flightscope recorded in Gunsan CC 6 hole Lake Course. Data Base through the recording had Coordinate using KWON3D 3.01. The result of this study can be summarized as follows. Had swing time in accordance with phase has appeared as a lowgroup in a ADD-TOP intervals shorter than the highgroup, in the case of BST-IMP phase showed that the lowgroup shorter than the parent group, the total swing time of this lowgroup than the highgroup It is shown to be short. Compared to a lowgroup of a highgroup head movements are seen as more significant difference between left and right when the back-swing Top statistically about the movement of the hand occurred(p<.05). This appeared to be highgroup caulking angle is lower than the lowgroup, the shoulder angle at impact was found to be greater than the lowgroup is highgroup, the difference in the angle at address and at impact angles showed that the higher greater group. Sacral angle was take the form which bowed all the backswing top, impact, in the case of highgroup showed a form similar to the upright swing impact. X-factor value of this lowgroup was higher than highgroup, the X-factor impact when the value of this lowgroup was higher than the highgroup. Angular velocity body is a lowgroup showed faster than the highgroup, the pelvis showed a higher angular velocity is faster than the lowgroup.

          District Heating System(DH) is one of the most excellent system in terms of energy saving and environmental protection. In order to comply with the global energy crisis and environment problem , it is necessary to research and develop the energy utilization technology with the reduction of the primary energy usage. Although the incresing rate of energy consumption started to attenuate , the entire growth in Korea. The energy demand in Korea increses by annual 3.7% during the period from 2000 to 2006. And it is expected that we should take a charge of the greenhouse gas reduction obligation by the Climatic Charge Convention(Kyoto Protocol) during the 2nd pledge period(2013~2017). This thesis reports something about comparisons between Braze & Gasket heat exchanger.

          In vitro Activities of Polycalcium, a Mixture of Polycan and Calcium Lactate-Gluconate, on Osteoclasts and Osteoblasts

          본 실험에서는 폴리칸(베타-글루칸)과 칼슘 락테이트 글루코네이트 1:9 (g/g) 복합 조성물인 Polycalcium의 시험관 내(in vitro) 골다공증에 대한 효과를 사람 유래 조골세포(human primary osteoblast)와 설치류 유래 파골전구세포(raw264.7 cell)를 이용하여 평가하였다. Polycalcium이 조골세포에 미치는 영향을 확인한 결과, 10 ㎎/㎖ 농도의 polycalcium 처리군에서 무처리 대조군에 비해 유의성 있는 조골세포의 수적 증가가 각각 배양 3, 7 및 10일 후에 확인되었으며, 또한 10 ㎎/㎖ 농도의 polycalcium 처리군에서 무처리 대조군에 비해 유의성 있는 ALP함량의 증가가 확인되었다. Polycalcium이 파골세포에 미치는 영향을 확인한 결과, 각각 10-5, 10-3 및 10?¹ ㎎/㎖ polycalcium 처리군에서 무처리 대조군에 비해 유의성 있는 파골세포의 수적 감소가 배양 4일 후에 확인되었다. 이 같은 결과를 바탕으로, polycalcium이 조골세포의 증식 촉진 효과와 함께 파골세포 형성 억제 효과가 있는 것으로 확인되었다. The present study evaluated the beneficial effects of polycalcium (a mixture of Polycan and calcium lactate-gluconate 1:9 [g/g]) on osteoporosis using in vitro assays. Cell proliferation and alkaline phosphatase activities of osteoblasts (human primary osteoblasts) and osteoclast differentiation of RAW264.7 cells (murine osteoclast progenitor cells) treated with different concentrations of polycalcium for various periods were assessed. Osteoblast proliferation was stimulated and prevented RANKL-induced osteoclast differentiation of RAW264.7 cells. These results support the development of ideal anti-osteoporotic agents, such as polycalcium, that exhibit properties that accelerate bone formation and inhibit bone resorption.
