Callus from tissue- cultured petioles of celery (Apium graveolens var. dulce) plants on modified MS medium were employed for cell suspension culture. When sufficient number of cells were obtained, they were pipetted to regeneration medium. The presence of auxin prevented regeneration, but addition of zeatin made no difference in the number of plants regenerated. Highest number of plants regenerated was obtained when cell clusters were used as inoculum to the regeneration medium. One of the objectives of present study is to observe DNA base changes induced through tissue culture by recently developed RAPD method. However, all 12 primers used in this experiment foiled to detect genetic variation in regenerated celery plants. Further study with more primers under better environmental control is proposed.
二面交雜에 의한 무(Raphanus sativus l.)의 收量 形質들에 대한 量的遺傳 分析

Eight parents and a set of 28 diallel crosses were employed to investigate heterosis, combining ability and inheritance of yield characters in radish(Raphanus sativus L.). Using model 1 of Griffing's method 2, heterosis and combining abilities of above characters were estimated. With Jinks and Hayman's method, genetic variance component analysis were performed. The results obtained are summarized as follows. 1.Heterosis was highest for root weight(156.0%). Characters such as leaf weight (55.5%), root length(45.9%), root diameter of middle portion(39.2%), root diameter of lower portion(37.3%) and root diameter of upper portion(32.3%) showed positive heterosis, while T/R ratio(-41.9%) showed negative heterosis. 2.In the analysis of combining ability, mean squares of GCA and SCA were highly significant for the all characters investigated, except leaf length character. 3.The significant positive GCA effects for root length, root weight, number of leaves and root shape index were shown in Tokinashi and Minowase. Yonghyeon and Jungkukcheongpi showed the significant positive GCA effects for root diameter, root weight and leaf length. However, Jinjudaepyeong and Wulsanjaerae showed a negative GCA effect for root length and root weight and, especially Taimayhua showed a negative GCA effect for all characters investigated. 4.The combinations of Tokinashi x Taimayhua, Tokinashi x Wulsanjaerae and Jinjudaepyeong x Minowase indicated the significant positive SCA effects for root length, root diameter and leaf weight. 5.In the degree of dominance, root length, root diameter, T/R ratio, number of leaves and root shape index demonstrated partial dominance, while leaf length showed overdominance. 6.Dominance effects of genes were greater than additive ones for root length, root diameter and leaf weight. Number of leaves, T/R ratio and root shape index expressed larger additive effects. But root weight and leaf length showed overdominance. 7.Heritabilities in broad sense(h2B) were very high for all characters investigated. Heritabilities in narrow sense (h2N) were highs for root length, number of leaves and root shape index, but low for leaf length.

우리나라 고추재배에 큰 문제가 되고 있는 TMV, 역병, 더뎅이병에 대한 저항성품종 육성의 처음단계로써 이 3가지 병에 대한 고추의 계통 및 품종에 대한 저항성 정도를 검정하였다. TMV에 있어서 37계통중 5개가 저항성으로 나타났고 역병의 34계통 중 4개, 더뎅이병에서는 외국 도입계통들이 저항성으로 나타났다. 이 성적은 2차년도 교배 조합을 작성할 때 도움이 될 것으로 보인다.

Six orient varieties of N.tabacum and 15 $F_1$ hybrids, not including reciprocal crosses, were evaluated for the degrees of heterosis and the combining abilities of 6 characteristics in Suweon, Korea, 1981. The values 9f general combining abilities tiers greater than those of specific combining abilities in all traits but significant SCA was also observed for Plant height, days to flowering and yield. In general, CCA effect was large in Samsun, Kavala and Izmir varieties. Large SCA values were observed for number of leaves in Samsun x Izmir, days to flowering in Samsun x Serves, leaf shape in Basma x Xantlli, and yield in Basma x Kavala and Xanthi x Izmir. Heterosis of favorable directions in $F_1$ was generally observed for all traits. Especially the $F_1$ of Samsun x Kavala flowered earlier than its midparents. Large heterosis effects were manifested for the number of leaves in Samsun x Izmir and yield in Samsun x Izmir as well as Xanthix Izmir.
배추(Brassica campestris ssp. pekinensis ) 약 배양시 약의 발달 단계, 온도 처리 및 배지의 종류가 배 발생에 미치는 영향

결구배추(Brassica campestris ssp. pekinensis)의 약배양을 통한 반수체의 유기율을 높히고 반수체의 발달과정을 알아보기 위해 본 연구를 수행하였다. 결구배추의 일대잡종 및 순계 의 약을 수정된 B/sb 5/ 배지에 전처리 및 후처리를 하여 액체 및 고체배지에 배양하였다. 접종후 9일부터 13일까지 반수체가 생겼는데 저온처리 효과는 없었으나 고온인 4$0^{\circ}C$ 1시간, 45$^{\circ}C$ 1시간 처리는 반수체 유기율을 높였다. 육성계통간에 따라 유기율이 큰 차이가 있었으며 배지는 큰 차이가 없었다. 후처리 효과는, 35$^{\circ}C$ 1일 처리후 3$0^{\circ}C$ 1일 처리효과가 있었으며 4$0^{\circ}C$ 이상 고온처리는 효과가 없었다. 배양한 약을 단계별로 조직학적으로 관찰한 결과 체세포배 유기는 약내 화분으로부터 유기되는 것을 관찰할 수 있다. Anthers of hybrids and inbred lines of Chinese cabbage (Brassica campestris ssp. pekinensis) were inoculated on the modified solid or liquid B$_{5}$ medium after several pre-treatments. Anthers were then maintained at 25 $^{\circ}C$ after being subjected to various post-treatment. Somatic embryos began to appear 9 days after inoculation and ended at 13th days. Low temperature pre-treatment did not increase embryoid production whereas high post-treatment temperature of 4$0^{\circ}C$ or 45$^{\circ}C$ enhance the production. There were considerable levels differences in embryogenesis between lines used, but not between the culture methods. Somatic embryo yield was also increased by subjecting anthers to one day at 35$^{\circ}C$ and then another one day at 3$0^{\circ}C$ after plating. Histological observations showed several stages in haploid development ranging from a few celled to large multiple-celled embryoids.s.

This experiment was carried out to investigate the degrees and directions of dominance, and gene distributions by analysing diallel crosses of oriental varieties. The analysis of Wr-Vr indicated non-allelic gene interaction for days to (lowering and number of leaves. Five Plant characteristics showed different degrees of dominance : incomplete dominance for Plant height and leaf shape, over dominance for , wield and sugar, and complete dominance for nicotine. It was shown that additive genetic variance was predominant for plant height, leaf shape, and total sugar. More number of dominant genes were present in Kavala and canthi for Plant height; Basma, Samsun and Izmir for leaf shape ; and Basma and Samsun (or nicotine. Their directions of dominance were tall height, narrow leaves, and low nicotine, respectively.
