        더러브렛 말의 피부병변에서 분리한 Staphylococcus intermedius의 약제 감수성 양상

        Staphylococcus intermedius(S. intermedius) has been rarely isolated from horse. In this study, we investigated the antimicrobial susceptibility patterns of S. intermedius isolated from skin lesions of Thoroughbred horse in Daegu Equestrian Association. The skin lesions were showed with dehairing and slight purulent inflammation. Bacteria were isolated from skin lesions and identified as S. intermedius by biochemical tests and MicroLog (BIOLOG, California, USA). Antimicrobial susceptibility test of S. intemedius isolates was performed with 33 antimicrobial agents (BBL, Maryland, USA) by using the agar disk diffusion method. It showed a high sensibility in the amikacin, amoxycillin/clavuramic acid, ampicillin, cefoxitin, cephalothin, chloramphenicol, ciprofloxacin, enrofloxacin, florfenicol, kanamycin, neomycin, nitrofurantoin, norfloxacin, ofloxacin, oxacillin, penicillin G, tetracycline and vancomycin. The horse was treated with penicillin, and cured completely after two weeks. The present results showed valuable information for treatment and prevention of skin disease in horse.

        국내 말에서 분리된 Staphylococcus aureus의 특성 및 약제 내성 양상

        국내에서 사육중인 말에서 채취한 시료로부터 Stapylococcus aureus를 분리ㆍ동정한 결과 39.2%(58/148)의 분리율을 나타내었다. S. aureus의 혈장응고효소 생산능 시험에서는 98.3%가 양성반응을 나타내었고 신선 혈장의 희석농도는 1:2에서 가장 높은 양성반응을 나타내었다. 본 연구에서 최종 동정된 S. aureus의 기본적인 생화학 성상은 일반적으로 알려져 있는 S. aureus의 특성과 유사한 결과를 얻을 수 있었으나, urease test의 경우 전체 균주의 73%만이 양성 결과를 나타내어 균주별로 다양한 반응 양상을 관찰 할 수 있었다. 또한 34종의 substrate에 대한 분해 양상에 대한 검사 결과 fructose, maltose, α-hydroxy butyric acid, thymidine-5'-mono phosphate, uridine-5'-mono phosphate등의 substrate에 대해서는 모든 균주가 이용능력을 가지고 있었지만 ribose, sorbitol, xylose, xylitol 등의 substrate에 대해서는 모든 균주가 기질 이용능력을 가지고 있지 않는 것으로 확인되었다. 그리고 PCR에 의한 S. aureus를 동정한 결과 108bp 크기의 band를 확인하였다. 국내 말에서 분리한 58주의 S. aureus에 대한 약제 내성 양상을 조사한 결과 spectinomycin, sulfonamides, erythromycin, tetracyclin, ciprofloxacin, neomycin, enofloxacin, penicillin 등의 약제에 높은 저항성을 나타내었다. This study carried out to investigate the antimicrobial resistance and biochemical characteristics of Staphylococcus aureus (S. aureus) isolated from reproductive and respiratory tract in Thoroughbred horse. The specimens were collected from equine vaginal mucosa and upper respiratory tract from March to December 2006 using a culture swab in Korea. S. aureus suspected colonies on blood agar plates were selected and identified as standard biochemical tests and PCR (Applied Biosystems, USA). Antimicrobial resistance test of S. aureus isolates was performed with 30 antimicrobial agents (BBL, USA) by using the agar disk diffusion method. S. aureus isolates were isolated 58 (39.2%) strains of 148 samples: wound 64.7% (11/17), genital discharge 37.0% (37/100) and nasal discharge 32.2% (10/31). Almost isolates showed high resistance to spectinomycin, sulfonamides, erythromycin, tetracyclin, ciprofloxacin and penicillin. These results may provide the basic information to establish strategies for treatment and prevention of reproductive and respiratory disease in Thoroughbred horses in Korea.

        Shoit Communication : Real-time PCR에 의한 equine herpesvirus type 1 (EHV-1) myeloencephalopathy의 진단

        Equine herpesvirus myeloencephalopathy, out of symptoms by equine herpesvirus type 1 (EHV-1) infection,can cause devastating losses on individual farms. Although myeloencephalopathy syndromes of horses in Korea have been recognized for a couple of years in horse populations, there is little study regarding the occurrence of EHV-1 infections. The present study was performed to detect the viral infection of horses with neurological syndrome using real-time PCR. Fifteen horses (27.3%) out of 55 horses with neurological deficiency were positive for EHV-1 viral antigen. Among these 7 horses, 4 horses were detected genotype of A2254/N752 and 3 horses G2254/D752 strain, respectively.

        RAPD Analysis and Antimicrobial Susceptibility of Streptococcus equi subsp. zooepidemicus Isolated from Thoroughbred Horses

        Streptococcus equi subsp. zooepidemicus는 말의 생식기 질환을 유발하는 중요한 병원체 중 하나로서 국내 말의 생산성 향상을 도모할 목적으로 제주도에서 사육중인 더러브렛 암말의 유산태아 및 그 말의 자궁내용물과 장수목장에서 암말의 교배전 생식기 질환이 의심되는 말의 질 내용물로부터 시료를 채취하여 Streptococcus equi subsp. zooepidemicus을 분리하여 그 분리균의 특성 및 항균제 감수성 검사를 실시한 결과, RAPD typing에서는 크게 4개의 cluster로 구분되었다. 특히 제주도에서 분리한 균주와 장수목장에서 분리한 하나의 균주가 동일한 pattern을 나타내었다. 항균제 감수성 검사에서는 대부분의 균주가 ampicillin 등의 약제에 감수성이 있음을 확인하였다. 본 연구의 결과는 국내에서 사육중인 암말의 생식기 질병의 예방 및 치료에 유용할 것으로 생각된다. A total of 68 samples were collected including vaginal mucosa (n=66) from Jangsu stud farm, an equine aborted fetus (n=1), and uterine contents (n=1) from Jeju island. Seventeen Streptococcus equi subsp. zooepidemicus (S. zooepidemicus) strains isolated from horses in Korea were identified as S. zooepidemicus by biochemical tests and sodA?seeI specific multiplex PCR. All isolated strains were divided into 4 clusters: group 1 (No. 2, 3, 5, 6, 7, 11, 12, 13, 14, 15), group 2 (No. 4, 9), group 3 (No. 10, 16, 17), and group 4 (No. 1, 8) by RAPD typing. In group 3, No. 10 isolate that was isolated from vaginal mucosa was indistinguishable from No. 16 and 17 isolates, which were isolated from the equine uterine contents and the equine aborted fetus, respectively. The results of this study suggest that a limited epidemiological relationship exists between the strains from Jangsu (No. 10) and Jeju (No. 16 and No. 17). All isolates showed a high susceptibility to ampicillin, cefoxitin, ceftiofur, cephalothin, florofenicol, gentamicin, nalidixic acid, oxacillin, penicillin, tiamulin, tylosin and vancomycin in antimicrobial susceptibility tests. These results may provide the basic information needed to establish strategies for the treatment and prevention of reproductive diseases in mares in Korea.

        제주지역에서 사육중인 더러브렛 말 호흡기로부터 분리된 병원성 Streptoccus spp.의 생화학성상 및 약제감수성 양상

        This study carried out to investigate the pathogenic Streptococcus spp. isolated from respiratory tract of Thoroughbred in Jeju province. The specimens were collected from nasal mucosa using a culture swab from 113 Thoroughbred horses. Suspected colonies were selected onto blood and MacConkey agar plate, and identified by standard biochemical properties using Vitek 2 system and PCR method. In this study, we isolated S. equi (n=6), S. zooepidemicus (n=31), S. equisimilis (n=5), S. dysgalactiae (n=2), S. agalctiae (n=1), non identified β-hemolytic Streptococcus spp. (n=1) from Thoroughbred horses. In antimicrobial susceptibility test, it showed a high sensibility in the most of antimicrobial except for neomycin, streptomycin, spectinomycin, erythromycin and clindamycin. These results will provide the basic information to establish control measures for the treatment and prevention of respiratory disease by pathogenic Streptococcus spp. in Thoroughbred horses in Korea.

        경북지방 환돈에서 분리한 Streptococcus suis의 생화학적 성상 및 약제감수성

        Streptococcus suis는 사람과 돼지에서 다양한 감염을 유발하는 병원체이다. 2004년부터 2009년까지 경북지방 양돈장에서 병성감정 의뢰한 돼지 가검물을 대상으로 S. suis를 분리하여 생화학적 성상, 약제감수성 시험 및 PCR을 이용한 동정을 실시한 결과는 다음과 같다. 40개 농장에서 분리한 61주의 S. suis에 대하여 생화학적 성상시험 결과 모두 S. suis로 동정되었으며, VP 음성, hippurate, esculin, arginine decarboxylase 양성, lactose 분해능이 있는 것으로 나타났다. Trehalose와 starch 분해능은 비교적 높게 나타났으며 생화학적 성상에 따른 phenotype은 11형으로 나누어졌다. 항생제 감수성 시험 결과 대부분의 균주가 amoxicillin/clavulanic acid, ampicillin, cephalothin, cefoperazone, florfenicol에 높은 감수성을 나타내었지만 amikacin, kanamycin, neomycin, streptomycin, erythromycin, clindamycin, tetracycline에는 높은 내성을 나타내었다. 16S-rRNA 특이유전자 부위를 이용한 PCR 결과 공시균주 모두 304 bp에서 증폭산물이 관찰되었다. 이들 결과는 국내에서 사육되고 있는 돼지에서 S. suis 감염을 예방하는데 도움이 될 것으로 기대된다. Streptococcus suis is a worldwide pathogen of a variety of porcine infection and has also been described as a pathogen for humans. We studied biochemical characteristics, antimicrobial susceptibility, and identification of polymerase chain reaction (PCR) of S. suis isolated from diseased pigs in Gyeongbuk province from 2004 to 2009. Sixty-one isolates were identified as S. suis by biochemical characteristics and PCR from 40 farms. The biochemical characteristics of S. suis isolates were production of VP-negative, hippurate, esculin, and arginine decarboxylase-positive, and fermentation of carbohydrate was variable lactose, trehalose, inulin, and raffinose, which was typeable 11 phenotype. In an antimicrobial susceptibility test, the majority of isolates were highly susceptible to amoxicillin/clavulanic acid, ampicillin, cephalothin, cefoperazone and florfenicol, while being highly resistant to streptomycin, kanamycin, amikacin, neomycin, erythromycin, clindamycin, and tetracycline. The isolates were divided into 11 phenotypes of biochemistry. By using PCR, the 16S-rRNA gene DNA fragment was detected at 304 bp frm all of isolates. These results may provide the basic information needed to establish strategies for the prevention of S. suis infection in pigs.

        MLVA를 이용한 경북지역 소에서 분리된 Brucella abortus의 유전형별

        Brucella (B.) abortus is a facultative intracellular pathogen that infects a wide variety of animal species and human. Brucellosis is the zoonosis and an extremely important disease around the world. Although the eight species can be differentiated by conventional phenotypic tests, these species display a high degree of DNA homology in DNA-DNA hybridization assay (>90%). Various methods have been established for genotyping of Brucella species, but most of analytical methods are lack reproducibility and limited capability to differentiate them. The attempt of this study was to evaluate multiple- locus VNTR analysis (MLVA) for use of epidemiological trace-back analysis in bovine brucellosis. Ninety-four B. abortus isolates from Gyeongbuk province during 2006~2010 were analyzed using 16 VNTR locus. High resolution automatic capillary electrophoresis system was used for more throughput, simpleer, faster, and better discriminable than conventional gel electrophoresis. As a result, 13 different genotypes were identified from 94 B. abortus isolates. MLVA could contribute to epidemiological trace-back analysis of bovine brucellosis.

        말에서 분리한 Escherichia coli의 특성 및 항생제 감수성

        This study was conducted to investigate characteristics and antimicrobial susceptibility of Escherichia (E.) coli isolates isolated from vaginal mucosa and clitorial fossa of 105 Thoroughbred mares suspicious of the genital disease in Korea during the period from March 2006 to July 2007. Ninety six E. coli isolates were identified as standard biochemical properties and using BIOLOG system. Fifty three isolates (55.2%) could be classified into a total of 21 O serotypes and forty three isolates (44.8%) were non-typeable with 51 O antisera used in this study. The verotoxin 1 (VT 1) and verotoxin 2 genes were analyzed by multiplex PCR. Among them, one isolate was detected VT 1 gene (130 bp). Most of isolates showed a high susceptibility in ciprofloxacin (100%), enrofloxacin (100%), norfloxacin (100%), cefoxitin (96.9%), gentamicin (96.9%), sulphamethoxazole (96.9%), nitrofurantoin (94.8%), amikacin (93.8%), nalidixic acid (92.7%) and tetracycline (90.6%). These results may provide the basic information to establish strategies for the treatment and prevention of reproductive disease in Thoroughbred mares in Korea.

        돼지 폐렴병소에서 분리한 Pasteurella multocida의 특성 및 항생제 감수성 양상

        영남지방 5개 농장에서 사육중인 돼지 251두의 폐렴병소로부터 P. multocida를 분리한 결과 91주의 P. multocida가 분리되어 분리율은 36.3%로 나타났다. 생화학적 특성을 조사한 후 PCR로 최종 확인한 결과 P. multocida species-specific 유전자 460 bp의 특이 증폭산물을 관찰하였다. 분리균 91주의 P. multocida에 대한 약제 감수성 검사 결과 amikacin (91.2%), cephalothin (87.9%), cefoxitin (84.6%), florofenicol (93.4%), norfloxacin (65.9%), ofloxacin (80.2%) 등의 약제에 비교적 높은 감수성을 나타내었다. 또한 대부분의 분리주는 5종 이상의 약제에 대해 다재 내성 양상을 나타내었다. The present study was conducted to investigate the species-specific gene detection and antimicrobial susceptibility of Pasteurella (P.) multocida isolated from pneumonic lung lesions of Youngnam swine herds during the period from July 2006 to September 2007. A total of 91 (36.3%) strains of P. multocida were isolated from 251 pneumonic lung lesions. The species-specific P. multocida gene was detected at 460 bp amplicons by PCR. The P. multocida tested was susceptible to florofenicol (93.4%), amikacin (91.2%), cephalothin (87.9%), cefoxitin (84.6%), ofloxacin (80.2%) and norfloxacin (65.9%) in 27 antimicrobial susceptibility tests. Most of strains were resistant to more than 5 drugs.
