Enhancement of amino acid contents by germination in a Korean weedy rice germplasm

Germination can improve the nutritional value of cereals. A weedy rice, WD3, showed the highest antioxidant activity among Korea-native weedy rice germplasm (Cho et al. Korean K Crop Sci 64:219–228, 2018). Thus, it was expected that WD3 might contain the higher functional and nutritional compounds. The purpose of this study is to examine the amino acid contents of WD3 in germinated and non-germinated seeds compared with a cultivated rice, Sindongjin (SDJ). Amino acid contents were measured using ultra-performance liquid chromatography (UPLC) coupled with tandem mass spectrometry (MS/MS). The amino acids were grouped, and the percentage and ratio of the respective amino acid group were evaluated. As a result of the experiment, the contents of all kind of free amino acids increased after germination in both WD3 and SDJ, but their increasing rates were much higher in WD3 than in SDJ except only two (GABA, Glu) among 21 kinds of amino acids. The total amino acid (TAA) content increased only 1.3 times in SDJ, while it increased 4.2 times in WD3 after germination. Especially, the branched-chain amino acid (TBCAAss) content after germination increased 18.2 times in WD3, compared to 4.1 times increase in SDJ. Additionally, as the ratio of the non-essential amino acid (NEAA) to the essential amino acid (EAA) of WD3 changed from 4.0 to 1.1 after germination, germinated WD3 grain could get well balance between EAAs and NEAAs. In conclusion, the nutritional value of the weedy rice, WD3, was much better than that of the cultivated rice, SDJ, when it is used as a germinated grain. Thus, WD3 could be used as a potential functional and nutritional food source for health benefits.
Core Promoter Mutation of ntC1731T and G1806A of Hepatitis B Virus Increases HBV Gene Expression

Chronic infection by hepatitis B virus (HBV) greatly increases the risk for liver cirrhosis and hepatocellular carcinoma (HCC). The outcome of HBV infection is shaped by the complex interplay of the mode of transmission, host genetic factors, viral genotype, adaptive mutations, and environmental factors. The pregenomic RNA transcription of HBV for their replication is regulated by the core promoter activation. Core promoter mutations have been the reason for acute liver failure and are associated with HCC development. We obtained HBV genes from a patient in Myanmar who was infected with HBV and identified gene variations in the core promoter region. For measuring the relative transactivation activity of the core promoter, we prepared the core-promoter reporter construct. Among the gene variations of the core promoter, the mutations of C1731T and G1806A were associated with increase in the transactivation of the HBV core promoter. Through computer analysis for searching for a tentative transcription factor binding site, we showed that the mutations of C1713T and G1806A newly created C/EBPβ and XBP1-responsive elements of the core promoter, respectively. The ectopic expression of C/EBPβ largely increased the HBV core promoter containing the C1713T mutation and that of XBP1 activated the M95 promoter containing the G1806A mutation. Our efforts to treat and prevent HBV infections are hampered by the emergence of drug-resistant mutations and vaccine-escape mutations. Our results provide the biological properties and clinical significance of specific HBV core promoter mutations.
Antioxidant Activities in Germinated and Non-germinated Seeds of Korean Weedy Rice

Weedy rice (Oryza sativa) is a semi-wild rice grown in paddy fields and is more adapted to the environment than cultivated rice. Therefore, it is expected that the seeds of weedy rice might contain some chemicals related to its environmental adaptation, such as antioxidants, which may be useful for nutritional and medicinal purposes. This study was carried out to investigate the antioxidant activities of weedy rice compared to a Korean-bred rice cultivar, Sindongjin (SDJ), and to screen lines that show higher antioxidant activity in 199 accessions of weedy rice germplasm collected in Korea. DPPH (1,1-diphenyl-2- picrylhydrazyl) radical scavenging activity ranged from 31.0 to 91.7%, with an average of 82.5%, and 19 accessions that showed more than 91% antioxidant activity were selected. The 19 accessions were re-screened against non-germinated brown rice (BR) and germinated brown rice (GBR) using four assays, total phenol determination, DPPH radical scavenging, ABTS radical scavenging, and reducing power activities. The results showed that accession ‘WD3’ had the highest antioxidant capacity in both BR and GBR, suggesting that WD3 is a promising potential source of antioxidants and could be developed as a potentially functional substance material.

This study was carried out to develop an evaluation method to predict rice seedling establishment (SE) under low-temperature conditions. Two Korean-bred japonica cultivars, Shindongjin and Hopum were used in the experiment. Fresh seeds were treated with an accelerated aging (AA) at 40°C and 100% RH for 1-15 days. The SEs of the fresh and AA seeds were evaluated in nursery beds at 17°C, and their correlation coefficients with seed vigor values measured by 9 test methods including standard germination test (SGT), cool germination test (CLT), cold germination test (CDT), seedling growth rate test (SGRT), ascorbate peroxidase (APX), peroxidase (POX), superoxide dismutase (SOD), catalase (CAT), and α ‐amylase (AMY) activities. The percentage of SE decreased slowly from 75 to 0% with an increasing of AA period from 0 to 15 days. The result of nine vigor tests showed different correlations with the SE. SGT, CLT, SGRT, and POX were significantly correlated with the SE. In the correlation analysis with only short-term aging seeds (1-7 days), the SE was very highly correlated with SGT, CLT, CDT, SGRT, POX, and CAT. These results suggest that seed vigor values measured by several methods including SGT and POX could be used as a reference value to secure SE at low temperatures in nursery bed rice seedling culture.
